Comparative Efficacy of Antifungal Agents for Aquaculture Fish and their Eggs (양식 어류와 이들 난에 대한 항곰팡이성 약물들의 효과 비교)

  • In fresh water fish hatcheries and farms, Saprolegniales often cause serious mortality to the fish and their eggs. Malachite green is an effective antifungal agent, but is carcinogenic to fish and humans. Alternative antifungal agents are needed. Presently, we tested various concentrations of MBT-01108 (Opuntia ficus-indica extracts) alone and in combination with bronopol, formalin and sodium chloride (MBT-01108 mixture) on in vitro mycelial growth and in vivo remediation of adult eel, Anguilla japonica, infected with Saprolegnia sp. and fertilized eggs of chum salmon, Oncorhynchus keta, to evaluate the compounds' antifungal efficacy on eyed-egg and hatching rates. MBT-Oll08 mixtures incorporating bronopol and formalin at respective concentrations of 50 and 30 parts per million (ppm), and 100 and 20 ppm were most effective in controlling Saprolegnia in vitro and in vivo (P<0.05). Repeated daily exposures to 50 ppm and 100 ppm MBT-01108 were more effective than exposure every 2-3 days post-fertilization for the inhibition of Saprolegnia infection of rainbow trout, Oncorhynchus mykiss eggs as compared with control (0 ppm).

Quantitative fluctuation and species composition of ichthyoplankton in Gwangyang Bay, Korea (광양만 연안에 분포하는 부유성 난 및 자치어의 종조성과 양적변동)

  • Species composition and yearly variation of fish eggs and larval fishes were investigated from 2014 to 2016. During the study period, the fish eggs were identified as belonging to 14 taxa. The dominant species of fish eggs were Leiognathus nuchalis, Engraulis japonicus, and Konosirus punctatus. These three species accounted for 97.4% of the total number of individuals collected. The collected larval fishes were identified into 37 taxa, 22 families, and seven orders. The dominant species of larval fish were Leiognathus nuchalis, and Gobiidae fishes. These three species accounted for 85.4% of the total number of individuals collected. The diversity index of the larval fishes was the highest in 2014 (H' = 1.60) and the lowest in 2015 (H' = 1.15). The evenness index was the highest in 2016 (J = 0.51) and the lowest in 2015 (J = 0.39). The dominance index was the highest in 2015 (D = 83.5%) and the lowest in 2016 (D = 70.9%). In addition, the economically important species in this area were Engraulis japonicus, Konosirus punctatus, Hippocampus coronatus, Sebastes schlegelii, Acanthopagrus schlegeli, and Cynoglossus joyneri.

Seasonal Variation of Species Composition and Distribution of Fish Eggs and Larvae in the Western Part of Jeju Island, Korea (제주도 서부 해역의 난·자치어 분포와 종조성의 계절변동)

  • To find out the seasonal variation of species composition and distribution of fish eggs and larvae in the western part of Jeju Island, investigations were carried out with plankton net during five different months from September 2004 to August 2005. The mean abundance of fish eggs and larvae showed a maximum values in summer (August). During the survey period, total 59 species (51 genus, 26 families and 10 orders) of fish larvae were identified. Among the 10 orders, Perciformes (41 species, 15 families), Scorpaeniformes (7 species, 2 families) and Gasterosteiformes (2 species, 2 families) were the dominant species in this study area. The number of species was higher in high water temperature period than in low one, showing a peak in August as 25 species, and the lowest in May as 6 species. Engraulis japonicus, Scomber japonicus and Scombridae spp. which were migratory fish species occurred abundantly in summer. On the other hand, Sebastiscus marmoratus and Sebastes inermis which were sedentary fish species occurred abundantly in winter.

Temporal and Spatial Variations and Species Composition of Ichthyoplanktons in a Sea Area, with the Construction of Artificial Upwelling Structure (인공용승류 해역에서 채집한 난자치어의 종조성 및 시공간 변동)

  • Temporal and spatial variations and species composition of ichthyoplanktons were investigated by Bongo net in a sea area with the construction of artificial upwelling structure at 13, June, 21 September, and 23 September in 2012. A total of fish eggs was 4,849 ind./$1,000m^3$, and fish larvaes was 641 ind./$1,000m^3$ in the study area. The common fish eggs was Engraulis japonicus which was composed of 57.5%, and unidentified I, II, III and IV were occurred with decreased number of individuals order. For fish larvaes, Pictiblennius yatabei, Repomucenus sp.A, sp.B and Scomber japonicus were dominated, however, other species were Engraulis japonicus, Konosirus puntatus, and unidentified I, II. Temporal variations of fish eggs and larvae were remarkable differed. Higher numbers of fish eggs and larvaes were July 2012, whereas those were lower at August 2012. Spatial variations of those were also significantly differed with higher numbers were at station 2 and 3 during study periods. The total number of fish eggs and larvae was about 4 fold against numbers of those in the compared area far from study area. These results indicated that higher numbers of fish eggs and larvaes were due to influx much more nutrients and food organisms with artificial upwelling.

  • This study was designed to find out the mineral content in Korean foods. The data will be used as a fundamental guide in planning sodium and potassium restricted diet in Korea. The study is part 6 sodium and potassium content in meat, eggs and fish. The previous study is s follows: part1 and part 5 on vegetables and fruit, part 2 on cereals, part 3, on milk and soft drinks and part 4 on orange juice and nectars. part 6 includes 10 beef cuts, 5 retail items of pork, 4 parts of chicken, 2 kinds of eggs and 15 individual kinds of fish were collected from the market and analyzed for the content of sodium and potassium by atomic absorption spectrophotometry. Egg Quality and Amino Acid Composition of Fertilized Eggs of Sevenband Grouper, Epinephelus septemfasciatus per Farming Condition (사육조건에 따른 능성어, Epinephelus septemfasciatus, 수정란의 난질 및 아미노산 조성)

  • This study aims to investigate egg quality and amino acid composition of buoyant and non-buoyant eggs and evaluate egg quality of sevenband grouper, Epinephelus septemfasciatus. Amino acid analysis of eggs was conducted to investigate what elements were necessary for the survival and good quality of egg depending on farming condition and different diet. We analyzed amino acid from buoyant eggs and non-buoyant eggs, farming conditions (tank and sea cage), and different dietary conditions (formulated feed, formulated feed+raw fish-based moist pellets, and raw fish-based moist pellets). Egg quality was the best in a sea cage and when raw fish-based moist pellets (MP) were fed. In addition, egg quality with formulated and MP was better than that with formulated feed. As a result of amino acid analysis of eggs, buoyant eggs were containing more free amino acid than non-buoyant eggs. Also, eggs with MP were containing more free amino acid than those with formulated feed and MP. Eggs with mixed formulated feed and MP were containing more free amino acid than formulated feed. In conclusion, amino acid was helpful to improve egg quality, and egg quality can be controlled by farming conditions and feed.

Effects of Size and Degree of Abdomen Inflation of the Mother Fish on Ovulation Induction of Red Spotted Grouper, Epinephelus akaara

  • We determined the morphologic characteristics (body weight and degree of abdomen inflation) of the red spotted grouper, Epinephelus akaara, mother fish producing healthy eggs. Experimental fish were chosen from fish reared in a sea cage. The fish were divided into four size groups by body weight: 400~600, 600~800, 800~1,000, and 1,000~1,200 g and four stages (I~IV) of the degree of abdomen inflation. After hormone treatment, we observed the amount of ovulation-induced eggs, and rates of buoyancy, fertilization, embryonic survival, and hatching. As a result, mother fish with a body weight of 600 g or more spawned, and the fertilization rate, embryonic survival rate, and hatching rate were high in the 800~1,000 g range, thus showing effective ovulation induction. As a result of dividing the degree of abdomen inflation based on the anal fin of the mother fish into I-IV stages and determining hormone treatment time, the GSI was $0.9{\pm}0.2%$ at stage I, $2.3{\pm}0.2%$ at stage II, $5.6{\pm0.2%$ at stage III, and $7.9{\pm}0.9%$ at stage IV. The flotation rate and hatching rate were highest at stage III, and the fertilization rate and embryonic survival rate were highest at stage IV. Therefore, in terms of egg quality, the amount of eggs collected per mother fish, maturation, and histology were different depending on the degree of abdomen inflation. At stage III, where the abdomen inflation degree of the mother fish was based on the basal part of the dorsal fin relative to the height of the anal fin was 1, the egg quality was highest.

Fish and Efficiency on Attached Fish Eggs of Artificial Floating Island in Lake Soyang, Korea (소양호 인공식물섬의 어류 서식 및 어란 부착 효과)

  • The fish and attached fish eggs by artificial floating island in lake Soyang was investigation from May to June 2013. The observed fish at the floating island during the survey period were Zacco platypus (5.2 %), Siniperca scherzeri (1.0 %), Lepomis macrochirus (90.8 %) and Micropterus salmoides (3.0 %) at the Cheongpyeong-ri, Cyprinus carpio (0.4 %), Z. platypus (54.6 %), L. macrochirus (20.3 %) and M. salmoides (24.7 %) at the Ungjin-ri. The collected species at the neighboring facility were 22 species belonging the seven families. Among them, exotic species such as Carassius cuvieri, L. macrochirus and M. salmoides. Also, Z. platypus (30.4 %), L. macrochirus (18.9 %), Opsariichthys uncirostris amurensis (15.4 %) and Hemibarbus labeo (9.3 %) were numberous. Attached fish eggs number were 708,800 (800 per $m^2$) at the Cheongpyeong-ri and 974,500 (1,100 per $m^2$) at the Ungjin-ri. The fish eggs number were 504,920 of the Carassius auratus (212,640 at the Cheongpyeong-ri, 292,280 at the Ungjin-ri) and 1,178,380 of the C. carpio (496,160 at the Cheongpyeong-ri, 682,220 at the Ungjin-ri). The hatched eggs number were 350,505 of the C. auratus(102,067 at the Cheongpyeong-ri, 248,438 at the Ungjin-ri) and 811,222 of the C. carpio (238,157 at the Cheongpyeong-ri, 573,065 at the Ungjin-ri).

Spawning, Hatching and Larval Growth of Red and Black Clownfish Amphiprion melanopus (Red and Black Clownfish Amphiprion melanopus의 산란, 부화 및 자어성장)

  • We studied spawning, hatching and larval growth of Amphiprion melanopus under laboratory conditions. Each time a fish spawned about 440 to 650 eggs. The eggs were hatched 9 days later. Nest colour of the eggs changed from clear yellow orange to greyish violet with processing of embryogenesis. With regard to spawning according to the substrates, the fish prefered red clay pot and ceramic pot to volcanic scoria and live rock. Hatching rate (%) of the eggs on the former substrates was significantly higher than that on the latter. The number of eggs varied with the food stuff supplied to the fish. The fish fed Tetra bits (commercial diet), meat of short necked clams and mussels, and astaxanthin showed significantly higher fecundity with 638 eggs than that fed only Tetra bits (477 eggs) or Tetra bits with meat of shellfish without astaxanthin ($549{\sim}553\;eggs$). Higher temperature ranging from 24 to $30^{\circ}C$ induced shorter incubation period and smaller total length, but higher hatching rate. The optimum salinity and L:D cycle with $60\;{\mu}mol$ photon $m^{-2}s^{-1}$ for hatching were 33 psu and 16:8, respectively. The newly hatched larvae about 4.6 mm in total length grew to 7.5 mm in one week, 18.8 mm in a month and 37.9 mm in 70 days after hatching.

Molecular Identification and Distribution Pattern of Fish Eggs Collected around Jejudo Island (제주 연안해역 어란의 분자동정과 분포양상)

  • Monthly variation of species composition and abundance of fish eggs were examined to know the spawning time and location of the fishes inhabiting the coastal region of Jejudo Island. Samplings had been performed at the four locations (Jeju port, Seongsanpo, Seogwipo port and Chagwido) with a bongo net which was towed monthly at the sea surface from August 2006 to July 2007. The fish eggs were identified based on phylogenetic analyses with the DNA sequences generated through PCR-amplification and sequencing of the mitochondrial cytochrome b gene. Among a total of 43 taxa classified during the study period, 34 were identified to species, 4 to families, and the remaining 5 were unidentified. Of them, 23 taxa were occurred at Jeju port, 21 at Seongsanpo, 19 at Seogwipo port and 18 at Chagwido, whereas 15 taxa were found in September 2006, 12 in June 2007, 6 to 8 in August 2006 and January~May 2007, 5 in each October and November 2006, 3 in each December 2006 and July 2007. Among 34 species of fish eggs, Engraulis japonicus and Callanthias japonicus most frequently appeared at 16 times out of 48 observations in total. When those eggs were collected, the surface seawater temperature ranged $14.0{\sim}28.6^{\circ}C$ for E. japonicus and $14.9{\sim}20.5^{\circ}C$ for C. japonicus. The success rates of PCR-amplification and species identification in this study were 68.3% and 79.1%, respectively.
