Optimization of Olive Flounder Paralichthys olivaceus Size as a Raw Material for Sikhae and Quality Characteristics of Sikhae with Suitable Olive Flounder Paralichthys olivaceus Weight (식해 소재로서 넙치(Paralichthys olivaceus) 크기의 최적화 및 이를 활용한 식해의 품질 특성)

  • This study was conducted to optimize the size of olive flounder Paralichthys olivaceus (OF) as a material of sikhae and to investigate the quality characteristics. The results on the protease activity of OF meat, protein and ash contents of the bone, and yields and hardness of fish bone during fermentation time suggest that the suitable fish weight for sikhae was less than 250 g. The proximate compositions of the OF sikhae fermented under optimum condition (fermentation for 9 days at 15℃), were 73.0% moisture, 12.0% crude protein, 1.3% crude fat and 2.4% ash. The salinity, titration acidity and amino acid nitrogen contents per 100 g sikhae were 1.7 g, 2.46 g, and 311.3 mg, respectively. The lactic acid bacteria concentration in the sikhae were 8.84 log CFU/g, which were higher than those (5.78-6.62 log CFU/g) of 5 kind of commercial flounder sikhae. The functional properties, such as ACE inhibitory activity (69.0%), antioxidative activity (69.3%), α-glucosidase inhibitory activity (22.7%), xanthine oxidase inhibitory activity (88.2%), and nitrite scavenging activity (96.4%) of the sikhae were superior to those of 5 kind of commercial flounder sikhae.

Effects of Rhodobacter sp. SA16 on Lettuce(Lactuca sativa L.) in Plastic Film House (시설 상추에 대한 Rhodobacter sp. SA16 처리 효과)

  • This experiment was conducted to determine the development of mixed organic fertilizer using photosynthetic bacteria and mass production of mixed microbial compound for the environment-friendly agriculture. Photosynthetic bacteria, Rhodobacter sp. SA16 was isolated from soil collected by plastic film house. The SA16 strain was identified based on 16S rDNA sequence analysis and it is closely related to Rhodobacter sp.(100% similarity). The mixed organic fertilizer using SA16 was made of $N-P_2O_5-K_2O=60-10-20\;g\;kg^{-1}$ with combined soybean cake, sesame cake, powdered blood, fish meal, powdered bones and red-yellow soil. The mixed organic fertilizer 0.45, 0.90 and 1.35 Mg $ha^{-1}$ application in Ihyeon series was treated based on soil testing for lettuce cultivation in plastic film house. These results showed that the yield was increased the 18 and 19%over control by the mixed organic fertilizer application 0.45 and 0.90 Mg $ha^{-1}$, respectively. In the physical properties of the soil, the porosity of mixed organic fertilizer 1.35 Mg $ha^{-1}$ treatment was highest at 58.8%. Our results clearly revealed that the organic fertilizer using Rhodobacter sp. SA16 and mass production of mixed strains could be a useful technology in pursuing environment-friendly agriculture.

The Quality Characteristics of Kimchi Prepared with Salt-fermented Toha Jeot Juice (염장토하 숙성시 나오는 액즙을 이용한 김치의 품질연구)

  • The purpose of this study was to investigate the growth of lactic acid bacteria and the self-life of Kimchi prepared with Toha sauce(TK) and different kinds of jeot-kal, a traditional fermented fish sauce. The Kimchi samples were stored for 42 days at 12$\pm$2$\^{C}$. The results were as follows: The pH value was decreased in the order of TK, AK(prepared with fermented anchovy juice), SK(prepared with fermented shrimp) and CK(control), and the acidity was increased in the opposed order. Redox potentials decreased until 4 days of fermentation but increased thereafter. TK showed the lowest value in redox potential as well as the lowest change in the number of yeast; however, it had the highest contents of vitamin C and reducing sugar. The total number of lactic acid bacteria was higher in jeot-kal-added Kimchi than control throughout the fermentation period. In sensory evaluation. AK got the highest score in overall preference in the early stage of fermentation, but TK was preferred in the late stage.

Isolation and Identification of a Histamine-degrading Barteria from Salted Mackerel (자반고등어에서 histamine 분해능을 가진 세균의 분리 동정)

  • Histamine can be produced at early spoilage stage through decarboxylation of histidine in red-flesh fish by Proteus morganii, Hafnia alvei or Klebsiella pneumoniae. Allergic food poisoning is resulted from the histamine produced when the freshness of Mackerel degrades. Conversely it has been reported that there are bacteria which decompose histamine at the later stage. We isolated histamine decomposers from salted mackerel and studied the characteristics to help establish hygienic measure to prevent outbreak of salted mackerel food poisoning. All the samples were purchased through local supermarket. Histamine decomposers were isolated using restriction medium using histamine 10 species were selected. Identification of these isolates were carried out by the comparison of 16S rDNA partial sequence; as a result, we identified Pseudomonas putida strain RA2 and Halomonas marina, Uncultured Arctic sea ice bacterium clone ARKXV1/2-136, Halomonas venusta, Psychrobacter sp. HS5323, Pseudomonas putida KT2440, Rhodococcus erythropolis, Klebsiella terrigena (Raoultella terrigena), Alteromonadaceae bacterium T1, Shewanella massilia with homology of $100\%,{\;}100\%,{\;}99\%,{\;}99\%,{\;}99\%,{\;}99\%,{\;}100\%,{\;}95\%,{\;}99\%,{\;}and{\;}100\%$respectively. Turbidometry determination method and enzymic method were employed to determine the ability of histamine decomposition. Among those species Shewanella massilia showed the highest in ability of histamine decomposition. From these results we confirmed various histamine decomposer were present in salted mackerel product in the market.

Characteristics of Citrus By-Product Ferment Using Probiotics as Starter (Probiotics를 이용하여 발효시킨 감귤 가공부산물 발효물의 특성)

  • In this study, we investigated the biological activity of antioxidant and antibacterial activity of citrus by-product ferment. Among the six probiotic bacteria, Bacillus subtilis and Saccharomyces cerevisiae had the highest antioxidant activity. Hot water extracts from citrus by-product of ferment were screened for antibacterial activity fish pathogenic bacteria by paper disc method. Among the various hot water extracts, the Bacillus subtilis, Saccharomyces cerevisiae, Pediococcus pentosaceus, Lactobacillus rhamnosus, Lactobacillus plantarum, Enterococcus faecium showed relatively strong antibacterial activities in the order. The reducing activity on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and $O^{2-}$ and OH radical scavenging potential were sequentially screened, in search for antioxidant activities of citrus by-product ferment.

Bacterial Community Analysis and Antibacterial Activity Isolated from Umbraulva japonica (초록갈파래(Umbraulva japonica)에서 분리한 세균의 군집 구조 분석 및 항균 활성)

  • In this study, 79 bacterial isolates were collected from the surface of marine algae Umbraulva japonica. As a result of analysis of 16s rRNA gene sequence, the 79 isolated bacteria were divided into 4 major groups: [Proteobacteria (74.69%), Actinobacteria (2.53%), Fimicutes (2.53%), and Bacteroidetes (20.25%)] - 7 classes (Actinobacteria, Flavobacteria, Sphingobacteria, Baciili, Alphaproteobacteria, Betaproteobacteria, and Gammaproteobacteria), 12 orders, 17 families and 31 genera. The newly isolated 3 strains could be novel species because of less than 97% similarity in 16s rRNA sequence. Therefore, it is considered that additional experiments should be conducted together with the standard strain. Analysis of 79 bacterial antibacterial activity against human and fish pathogens, such as Edwardsiella tarda, Vibrio harveyi, Streptococcus iniae, Steptococcus parauberis, Escherichia coli, Steptococcus mutans, Listeria monocytogenes and Vibrio vulnificus, was performed by using the supernatant liquid and pellet. As a result, pellet of UJT9, UJT20 and UJR17 showed antibacterial activity against V. vulnificus, UJR17 also showed antibacterial activity against S. parauberis. UJT7 and UJT20, UJR17 have been identified as Bacillus sp. and Pseudomonas sp. and it may be safely assented that it's beneficial to carry out additional experiments for various applications.

Isolation and Characterization of Lactic Acid Bacteria from Fermented Goat Milk in Tajikistan

  • The lactobacilli associated with a fermented goat milk product from Tajikistan were isolated to characterize their technological properties and antibiotic resistances in order to assess their suitability for development as starter cultures. In this study, twenty three strains were identified by 16S rRNA sequencing as typical dairy-associated lactic acid bacterial strains, i.e. L. plantarum, L. pentosus, L. delbrueckii, L. helveticus and L. paracasei. These strains were generally susceptible to most antibiotics tested in this study and this allowed a selection of strains as safe starters. The draft genomes of four representative strains were sequenced and the number of contigs of the four assembled genomes ranged from 51 to 245 and the genome sizes ranged from 1.75 to 3.24 Mbp. These representative strains showed differences in their growth behavior and pH-reducing abilities in in vitro studies. The co-inoculation of these Lactobacillus spp. strains together with a yeast Kluyveromyces marxianus MBT-5698, or together with the yeast and an additional Streptococcus thermophilus MBT-2, led to a pH reduction to 3.4 after 48 h. Only in the case of fermentation inoculated with the co-culture, the viscosity of the milk increased noticeably. In contrast, fermentations with single strains did not lead to gelation of the milk or to a decrease in the pH after 24h. The results of this study provide a comprehensive understanding of the predominant lactobacilli related to Tajikistani fermented milk products.

Environmental Factors Related to Mass Moralities of Young Red Seabream (Pagrus major) in the Artificial Seed Production (일본산 참돔, Pagrus major 종묘생산과정중 사육수 변화로 인한 대량폐사)

  • As seed production program is growing prosperously in various fishes in southern Korea, disease problems in larval and juvenile stages have emerged as a new research object. The following results were obtained from investigation about environmental factors related to mass mortalities of young Kinki red seabream, Pagrus major in process of artificial seedling production. Total length of red seabream larvae hatched was 2.93mm, and became 18.83~20.12mm at day 40. The first noticeable mortality of red seabream larvae (7.98~9.37mm) occurred in 25~30 day-old fish with the survival rates of 59.8~60.3%. Thereafter the mortality of larvae decreased, survival rate was 20.5~25.45% in day 40. After 20~30 days, the quality of pond water was bellow II class. During the experimental period COD, $PO_4$-P, $NO_2$-N, $NO_3$-N and $NH_$-N increased up to 3, 7, 34, 6 and 8 times, respectively, compared to initial ones. The number of viable bacteria in pond water and seabream larvae were $6.3{\times}10^6$~$2.3{\times}10^7$ cfu/ml, 4.3~$7.4{\times}10^6$ cfu/g in day 25, respectively. Among the isolated bacteria from the diseased red seabream in day 25, Vibrio spp. was considered to be the causative organism. External symptoms of this disease were floated, spined near the surface and inflated abdomen. When the isolated strain of the Vibrio was bathed to seabream larvae, $LD_50$ of seabream larvae was over $10^6$ cfu/ml of Vibrio spp.

Short -term changes of microbial communities after control of Cochlodinium polykrikoides by yellow clay and chemical compound dosing in microcosm experiments (황토와 화학물질 살포에 의한 적조생물Cochlodinium polykrikoides 제어에 따른 미소생물그룹의 단주기변화)

  • This study aimed to understand the changes in microbial community after algicide dosing to control the fish-killing dinoflagellate Cochlodinium polykrikoides in 10L microcosm. Based on our microcosm experiments, the algicidal activity for C. polykrikoides of yellow clay at the concentrations of 4g and 10g per 10 L was < 20%. At $0.8{\mu}M$ concentration of thiazolidinedione(TD49), the population of C. polykrikoides was controlled to be > 85%. In microbial community, a significant increase in heterotrophic bacterial (HB) abundance was observed at day 1 in the TD49 and yellow clay treatments including control. The HB remained high for 2 days and then gradually decreased. In contrast, the abundance of heterotrophic nanoflagellates (HNFs) increased significantly on days 3 and 5 in the TD49 treatments, indicating that the decline in HB was probably a result of predation by the high density of HNFs. In addition, fluctuations in the aloricate ciliate Uronema sp., which feed on bacteria, was clearly correlated with fluctuations in HB abundance, with a lag period of 1-3 days. Therefore, the short-term responses of the HNF and Uronema sp. may have been a result of the rapidly increasing of HB abundance, which is related to degradation of the dense C. polykrikoides bloom, particularly in the TD49 treatment. In addition, large aloricate ciliate Euplotes sp. was significantly increased after reproduction of HNFs and Uronema sp. Consequently, the algicide TD49 had positive effect on the microbial communities, which indicates that the microbial loop was temporarily enhanced in the microcosm by energy flow from HB through HNFs to ciliate.

Marine Metatranscriptome Profiling in the Sea Adjacent to Jeju Island, Korea, by RNA-sequencing (RNA-sequencing을 이용한 제주도 인접 바다의 메타전사체 프로파일링)

  • The Ocean is a rich source of diverse living organisms include viruses. In this study, we examined the microbial communities in the sea adjacent to Jeju Island in two seasons by metatranscriptomics. We collected and extracted total RNA, and, using the next-generation sequencing HiSeq 2000 and de novo transcriptome assembly, we identified 652,984 and 163,759 transcripts from the March and December samples, respectively. The most abundant organisms in March were bacteria, while eukaryotes were dominant in the December sample. The bacterial communities differed between the two samples, suggesting seasonal change. To identify the viruses, we searched the transcripts against a viral reference database using MegaBLAST with the most identified being bacteriophages infecting the marine bacteria. However, we also revealed an abundance of transcripts associated with diverse herpesviruses in the two transcriptomes, indicating the presence or possible threat of infection of fish in the sea around Jeju Island. This data is valuable for the study of marine microbial communities and for identifying possible viral pathogens.
