Evaluation of Sanitation Management Practices and Microbiological Quality of Foods in Kindergarten Foodservice Settings (유치원 급식의 위생관리 실태조사 및 미생물적 품질평가)

  • This research aims to audit foodservice sanitation management practices and to assess microbiological quality of foods and their food contact environments in kindergartens. Sanitation auditing was conducted in 10 kindergartens in Seoul, Gyeonggi, and Incheon areas to assess the levels of safety practices. Results revealed that the surveyed kindergartens scored 41.4 out of 100 points, on average. The average scores of each category were 6.4/11 (58.1%) for facilities sanitation, 4.2/12 (35.0%) for equipment sanitation, 2.4/10 (24.0%) for personal hygiene, 5.1/10 (51.0%) for food ingredients management, 6.0/17 (35.3%) for production process, 5.4/10 (54.0%) for environmental sanitation, 2.0/6 (33.3%) for kitchen utensils sanitation, and 2.2/6 (96.7%) for safety management. Microbiological quality of raw, prepared foods, personal sanitation (hands), environmental sanitation, and drinking water were assessed. Total plate counts (TPC) of the following menus exceeded the critical limit: seasoned leek (5 log CFU/g), cucumber (5.0 log CFU/g), panbroiled fish paste (TNTC at $10^4$), tangpyeongchae (5.3 log CFU/g), egg rolls (6.1 log CFU/g), panbroiled sausage (TNTC at $10^4$), and soft tofu pot stew (TNTC at $10^4$). Coliform which exceeded the standard limit were detected from seasoned leek (2 log CFU/g), cucumber (2.5 log CFU/g), panbroiled fish paste (2.0 log CFU/g), egg roll (3.8 log CFU/g), tangpyeongchae (4.0 log CFU/g), panbroiled sausage (2.3 log CFU/g), and soft tofu pot stew (3.7 log CFU/g). For seasoned foods (muchim), S. aureus ranged 2.2~2.9 log CFU/g. In food workers' hands, microbial profiles ranged 3.8~7.9 log CFU/hand for TPC, ND~4.5 log CFU/hand for coliforms, ND~4.7 log CFU/hand for S. aureus, and ND~5.3 log CFU/hand for Enterobacteriaceae. Microbiological profiles of food contact surface of knives, cutting boards, dish-clothes, and trays showed possibilities of cross-contamination. General bacteria were 2.1~4.5 logCFU/ml in 4 purified water samples and E. coli were found in the kitchen of one kindergarten. These results suggested that environmental sanitation management practices need more strict improvement: effective sanitation education methods and practices were strongly required, and more strict sanitation management for cooking utensils and equipment were required.

Induction and resuscitation of viable but nonculturable Edwardsiella tarda (Edwardsiella tarda의 비배양성 생존상태(VBNC) 유도 및 소생 특성)

  • Bacteria in the viable but nonculturable (VBNC) state fail to produce colonies on routine bacteriological media, but are still alive in the state of very low metabolic activity. The aim of the present study was to induce the VBNC state of the Edwardsiella tarda using sea water microcosm under starvation conditions at $10^{\circ}C$ and to investigate resuscitation of the VBNC cells in temperatures changed from 10 to $25^{\circ}C$, with and without additives. E. tarda entered into the VBNC state within about 42-84 days of incubation in the microcosm. Throughout this period, the total cell counts as determined using acridine orange direct counting remained near the original inoculum level of ${\sim}10^8cells/ml$. The live cell counts measured with direct viable counting, on the other hands, declined to ${\sim}10^4cells/ml$. When the VBNC cells were incubated with addition of yeast extract, fish muscle extract or serum at $25^{\circ}C$, the ratios of resuscitated samples were 37%, 23%, and 37%, respectively. The characteristics of resuscitated E. tarda were consistent with those of the original E. tarda. When the resuscitated E. tarda were intraperitoneally injected into olive flounders, all fishes died within 5 days, indicating that the VBNC E. tarda might retain its pathogenic potential. Therefore, E. tarda under starvation conditions in the winter enter into the VBNC state and the VBNC E. tarda cells resuscitated at summer and autumn seawater temperature are considered to be pathogen continuously to olive flounder on the southern coast of Korea.

A Survey of the Safety in Seasoned Dried Fishes (조미건어포류의 안전성에 대한 조사연구)

  • A survey of food preservatives, sulfur dioxide and microbial contamination levels was conducted on 73 seasoned dried fishes(41 squids, 17 filleted fish, 6 Alaska pollack, 9 others) collected from domestic markets and internet. Among preservatives, only sorbic acid was detected in 25 samples including 23 seasoned dried squids, 1 seasoned filleted fish and 1 seasoned dried Alaska pollack with a range $0.01{\sim}0.92g/kg$. Sulfur dioxide was detected from one sample(seasoned dried squid) and its content was 10.5 mg/kg. All samples were contaminated by total aerobic bacteria, and the degree was $3.00{\sim}6.45\;log_{10}$ CFU/g(average level was $5.09\;log_{10}$ CFU/g). Escherichia coli, Salmonella spp. and Staphylococcus aureus were not detected in all samples. On the basis of the results above, most of the seasoned dried fishes were appropriate for Korea food standard, yet biological hygiene control will be needed constantly.

  • This paper deals with the distribution of coliform group bacteria and the environmental factors in Suyeong Bay from November 1990 to February 1991. The ranges of environmental factors were as follows : Salinity 16.84~34.93%, Dissolved Oxygen $0.58{\sim}6.79m{\ell}/{\ell}$, COD 0.48~5.06 ppm, Nitrite $0.42{\sim}18.82{\mu}g-at/{\ell}$, Nitrate $0.50{\sim}53.23{\mu}g-at/{\ell}$, Ammonia $3.73{\sim}56.16{\mu}g-at/{\ell}$, Silicate $5.18{\sim}147.29{\mu}g-at/{\ell}$. The coliform group and fecal coliform group ranged $1.1{\times}10^2{\sim}2.4{\times}10^5/100m{\ell}$, $1.7{\times}10{\sim}3.5{\times}10^4/100m{\ell}$ each other. The ratio of TC/FC was 1.73~58.49 during study periods. Among the composition of coliform group, E. coli was dominent(64.4%), which was appeared all studying stations. E. coli I type was accounted for 97.4% in the E. coli.

Changes in the Microflora of Marine Fishes during Storage by Partial Freezing (해산어(海産魚)의 부분동결(部分凍結)에 의한 Microflora의 변화(變化))

  • Marine fishes, sardine(Sardinops melanosticta), scad kingfish(Caranx equula), horse mackerel(Trachurus japonicus) and file fish(Navodon modestus), were stored for fifty days with partial freezing at$-3.5^{\circ}C$. During the storage, the changes in microflora and volatile nitrogen content was investigated. The fishes exhibited $10^4\;to\;10^6$ of bacterial cells per square centimeter of their skin just before they were submitted to the storage. The bacterial cell number was increased as $10^6\;to\;10^8$ cells as the storage time passed over twenty-two days. Offensive odor which is typical in the spoilage of fishes became strong as increase the bacterial cell numbe. The major isolates among the three hundred strains of bacteria isolated from the fish skins were identified as Pseudomonas I/II, III/IV-H, Vibrio and Moraxella. The same was found in the spoiled fishes, however, Pseudomonas I/II, was predominant on contrast to that of fresh fishes. Pseudomonas III/IV-NH, Flavobacterium, Cytophaga and Micrococcus were also found in early period of storage, but they disappeared as the progress of storage. Nine per cent of isolates were unidentified.

Effect of the Polychaete Antimicrobial Peptide as feed Additives on Olive Flounder and Black Rockfish Immune Activity (넙치 및 조피볼락 면역 활성에 대한 사료첨가제로서 갯지렁이 항균펩타이드의 효과)

  • In this study, the effect of the polychaete antimicrobial peptide as feed additives on fish, olive flounder (Paralichythys olivaceus) and black rockfish (Sebastes schlegelii), immune activity was described. The antimicrobial peptide of the polychaete was induced by peptidoglycan from Micrococcus luteus. The fish were fed an experimental diet supplemented with 0%, 0.05%, 0.1%, 0.5% or 1% of immune induced the polychaete to a commercial diet. Haematological parameters, nonspecific immunes and stress were evaluated 2, 4, 6 and 8 weeks during fed. The resistance against bacteria, Edwardsiella tarda and Streptococcus iniae, were analysed on after 8 weeks. The haematological parameters were not significantly changed among tested groups. But the lysozyme activities were significantly high in the 0.1% and 0.5% supplement group of olive flounder and black rockfish, respectively. Additionally, cortisol in plasma was low in the 0.1% and 0.5% supplement group of olive flounder and black rockfish, respectively. And resistance of these supplement groups were significantly induced against bacterial injection.

Histopathological observation of the gill of the crucian carp, Carassius auratus by the leech, Limnotrachelobdella sinensis (거머리, Limnotrachelobdella sinensis의 기생으로 인한 붕어, Carassius auratus 아가미의 조직병리학적 관찰)

  • On the inner side of each operculum of the crucian carp, Carassius auratus (n=10), the leech, Limnotrachelobdella sinensis of 1-4 individuals were parasitic. The leeches had approximately 41.0 mm in total length and 11 mm in width. These body was composed with anterior sucker, neck, trunk and posterior sucker and average length was 2.3 mm, 7.2 mm, 23.3 mm and 8.7 mm respectively. To both sides of the trunk lateral vesicle of 11 pair existed. When observed by SEM, anterior sucker was hemisphere shape and the mouth where proboscis comes out existed with the its center. Proboscis was connected the esophagus directly. Under light microscopy, bloodsucking gill of C. auratus showed lamella fusion, hypertrophy the epithelial cell of the filament and lamella, increased mucocytes and congested capillaries. On the other hand, necrotic and hydropic degeneration epithelial cell of the lamella, and infiltration of the macrophages from some individuals were suggested the secondary infection with the bacteria or virus after bloodsucking activity of the leech.

Effects of a Commercial Activating Treatment Agent on Cultured Porphyra yezoensis thalli (양식 방사무늬김, Porphyra yezoensis의 활성처리제 처리 효과)

  • The use of activating treatment agent (formerly acid treatment agent) has been an effective strategy to remove deleterious epibiont organisms such as diatoms and green seaweeds, and it has greatly contributed to increase in Porphyra production. Although many manufacturers supply many kinds of activating treatment agent with different components in these days, no report about their effects on Porphyra culture was found. In this paper, effects of a commercial activating treatment agent were evaluated for practical use in Porphyra culture. No difference was found in dead cell ratios(%) of Porphyra yezoensis thalli between treated and control groups. However, dead cell ratios of Monostroma nitidum thalli were increased from 0~4.6% to 99.0~100% after the treatment. Bathing Porphyra thalli in activating treatment agent resulted in a great decrease in epiphytic bacterial number attached to the thalli from $10^2{\sim}10^{11}$ cells/g to $0{\sim}10^5$ cells/g but did not change the colour of the thalli. These results suggest that bathing Porphyra thalli in activating treatment agent could be a promising strategy to remove green algae, diatoms and bacteria.

Studies on a Toxin/Antitoxin System in Streptococcus iniae (어류병원균 Streptococcus iniae의 toxin/antitoxin system에 대한 연구)

  • Streptococcus iniae is a typical fish pathogen causing streptococcosis and it can also cause zoonotic infectious diseases. We studied S. iniae FP5228 isolated from infected olive flounder in Wando, Korea. In a study to find virulence factors in FP5228, we found that the number of live bacteria decreased dramatically in culture medium containing S. iniae FP5228 for more than 24 hr. This phenomenon was hypothesized to be related to Toxin ${\zeta}$ and Antitoxin ${\varepsilon}$ genes, components of the Toxin/ Antitoxin (TA) system on the 14 kb plasmid of FP5228. We used a protein overexpression system to identify it. The pBP1140 vector system was constructed to regulate the expression of Toxin ${\zeta}$ and Antitoxin ${\varepsilon}$ by IPTG and Arabinose. E. coli/pBP1140 strain grew slowly in early growth under toxin expression condition, and it was confirmed by microscopic observation that the strain became longer. S. iniae CK287, lacking a 14 kb plasmid of S. iniae FP5228 strain, was constructed. CK287 bacterial cells did not show rapid killing during culture, and the ability to produce biofilm was also decreased, and toxicity was weakened in cytotoxicity test and fish test. These results suggest that the TA system is involved in physiological regulation and expression of virulence factors in S. iniae FP5228.

Withdrawal time study of lincomycin administered intramuscularly to olive flounder Paralichthys olivaceus, and in vitro efficacy evaluation against Gram-negative bacterial pathogens (넙치에서 lincomycin 근육투여에 따른 휴약기간 도출 및 병원성 그람음성 세균에 대한 약효 평가)

  • This study was performed to delineate pharmacokinetic residue characteristics of lincomycin (LM) in olive flounder at low water temperature (13℃), and to examine whether LM is effective against main Gram-negative pathogens. It was observed that the times for residue declined below the maximal residue limit (0.1 mg/kg) in the muscle was, respectively, 32, 33 and 55 days following 10, 20 and 40 mg/kg intramuscular LM injections. The in vitro MIC value of LM against Edwardsiella piscida was higher than 256 ㎍/mL and against Vibrio harveyi 32 ㎍/mL when examined with several clinical isolates. These results suggest that the withdrawal time of LM should be more than 30 days in consideration of the 10 mg/kg employed as the regular clinical dose. It was also found that LM administered to treat streptococosis in olive flounder is not likely to be effective against some pathogenic Gram-negative bacteria.
