Effect of microorganisms collected from uterus of Hanwoo cattle with low conception rate on the development of IVF-derived embryos (저수태 한우 암소 자궁에서 회수된 미생물이 체외수정란 발달율에 미치는 영향)

  • The cause of infertility is either fertilization failure or early embryonic death. The aetiology may involve a combination of many factors, e.g. genetic factors, abnormalities in the gametes nutritional disorders, inadequate luteal function, and delayed ovulation. This study was conducted to investigate the effect of microorganisms collected from uterus of Hanwoo cattle on early embryonic development. Microorganisms isolated from the uterus of Hanwoo cattle included Bacillus cereus (Bc), Bacillus thuringiensis (Bt), Staphylococcus warneri (Sw) and Enterococcus faecalis (Ef). When cultured with Bc, Bt, Sw, and Ef, oocytes were not developed into a blastocyst in vitro. The proportion of blastocyst was dramatically increased after reducing the number of microorganisms ($10^4CFU/ml$). Interestingly, the proportion of blastocyst was decreased by adding the Sw and Ef. These results indicate that among intrauterine microorganisms, Sw and Ef strains negatively affect theearly embryonic development, thereby aggravate the rates of implantation and pregnancy. These findings will provide useful information for association studies in other pig populations.

Fertility Prolongation of the Solid Typed Organic Fertilizer from Cattle Manure (축분 유기질비료의 고형화에 의한 비효 연장)

  • Solid typed organic fertilizer from cattle manure may Increase plant root growth with the residual effect on soil fertility if it was applied directly into root system. It may also increase labour efficiency by simplifying the work process of manure application to soil. This research was carried out from May to October 1999 to study the fertility prolongation effects of bar typed organic fertilizer from poultry manure by analysing plant growth model compare with those of the chemical fertilizer or powder typed manure. The results showed that the bar typed organic fertilizer increased growth rates of shoot and root system with extending the effect of its fertility by slow releasing on pepper plant. Especially, solidifying organic manure into the bar type made possible the application of cattle manure under plastic mulching. And it also has benefits on simplifying the application process of the fertilization and effect on an extending soil fertility.

Production of Hybrid and Allotriploid between Rainbow Trout (Oncorhynchus mykiss) and Coho Salmon (O. kisutch) (무지개송어(Oncorhynchus mykiss)와 은연어(O. kisutch)간의 잡종 및 잡종 3배체 생산)

  • Hybird and allotriploid between female rainbow trout (Oncorhynchus mykiss) and male coho salmon (O. kisutch) were produced by artificial fertilization and heat shocks. Hatching and survival rate of allotriploid at 2 month after hatching was $77.6\%$ and $54.5\%$ respectively, and these rates clearly exceeded those of their hybrid. Cell and nuclear sizes of the erythrocyte of hybrid were intermidiate of their parents and those of allotriploid were larger than thier hybird. The somatic chromosome number of viable hybrid was 2n = 60 and that of allotriploid was $90\~93$ with chromosomal polymorphism. Allotriploid karyotpe was constituted by two sets of rainbow trout chromosome and one set of coho salmon chromosome.

Effect of Pentoxifylline on the ICSI Program Undergone in Severe Asthenozoospermia (심한 무력정자증 환자의 ICSI 시행시 Pentoxifylline을 사용한 정자처리법이 임상결과에 미치는 영향)

  • Objective: The aim of this study was to evaluate the effect of pentoxifylline (PF) on the conventional ICSI program undergone in severe asthenozoospermia. Method: Total 348 cycles of ICSI programs undertaken at CHA General Hospital from January, 1996 to September, 2000, were divided into two groups - injected with pentoxifylline-treated sperm (PFT, 204 cycles) or non-treated sperm (NPFT, 144 cycles) and the clinical results of PFT group were compared with those of NPFT. Results: PF-treatment on sperm increased their motility of normozoospermia and severe asthenozoospermia. Fertilization rate of PFT group was higher than those of ICSI programs undertaken using sperm of NPFT (70.6% vs. 62.9%, p<0.01). And, ET and clinical pregnancy rates of PFT were slightly higher than those of NPFT (93.1%, 44.2% vs. 90.3%, 36.2%). Conclusion: These results showed that treatment of pentoxifylline has a beneficial role on selection of viable sperm in severe asthenozoospermia.

Effect of Nitrogen Fertilization on Growth, Dry Matter Yield and Nitrogen Use of Orchardgrass (질소기비에 대한 Orchardgrass의 생육 및 수량반응과 질소이용성)

  • Field experiment with 0, 100, 200, 400, and 800 kg-N/ha-year application levels was carried out to study the nitrogen response in the early stage of orchardgrass pasture establishment at College Fann, SNU, in 1979 and 1980. Both the highest dry matter yield and maximum percent of N recovery were obtained at the same level of 200 kg-N/ha in the year of seeding. but those of the next year were obtained at 400 kg-N/ha level. Leaf area indices (LAI) and net assimilation rates (NAR) during each regrowth periods as well as total nitrogen contents of forage at each cutting time increased with applied N in both years. The maximum crop growth rate (CGR) over two years was estimated to be obtained when LAI reached to about 5. The accumulation of NO_3 -N in forage started from 400kgㆍN/ha application in 1980. and exceeded the safe level for ruminants at the level of 800 kg-N/ha.

Combined application of oil cake and rice bran reduced the number of weeds and increased the yield of paddy rice in a paddy field incorporated with white clover

  • The combined application of oil cake and rice bran into the soil surface was found useful for weed control in our previous pot study. The present study was undertaken to evaluate the performance of white clover (Trifolium repens L.) while incorporated in the paddy field and effects of combined fertilizer on weed control and rice yield. A plot was divided into two parts i.e. white clover incorporated and not incorporated. The nitrogen content of the incorporated white clover was $12.5gm^{-2}$. Chemical fertilizer and combined fertilizer plots were compared with non-fertilizer conditions. The mixed ratio of combined fertilizer was oil cake 1.35 and rice bran 1.0. Combined fertilizer was applied to the soil surface, and chemical fertilizer was mixed in the soil. Nitrogen application rate was $8gm^{-2}$ for any fertilizer. The weed numbers were significantly reduced in the white clover plot irrespective of application condition both at heading and harvest time. Also, weed control ability was improved by the use of combined fertilizer. In the not incorporated plot, the number of weeds was suppressed about 90% by applying combined fertilizer. The rice yield was markedly increased by the incorporation with white clover under all fertilization conditions. Contribution rates of increased rice yield by white clover and combined fertilizer were about 55% and about 25%, respectively. The rice yield was increased by the incorporation with white clover, and the number of weeds remarkably decreased as well. Also, these effects were improved due to combined application of oil cake and rice bran.

  • Embryo transfer (ET) technology is of high importance in modern cattle breeding programs. ET is one step in the process of removing one or more embryos from the reproductive tract of an outstanding donor female and transferring them to one or more recipient females. Embryos also can be produced in the laboratory via techniques such as in vitro fertilization (IVF). But the actual transfer of an embryo is only one step in a series of processes that may include some or all of the following: superovulation and insemination of donors, collection of embryos, isolation, evaluation and short-term storage of embryos, micromanipulation and genetic testing of embryos, freezing of embryos and embryo transfer. Cryopreservation and direct transfer of frozen-thawed embryos is common-place with pregnancy rates near that of fresh embryos. Polymerase chain reaction (PCR) technology is currently being used for sexing embryos, and this technology will be used for "embryo diagnostics" and "embryo genomics" in the future. Although, many limitations and problems remain to overcome, these and other new technologies promise to change livestock breeding drastically in the next decade.

Effect of Monothioglycerol on ROS Inhibition, Mitochondrial Activity, and DNA Integrity in Frozen-thawed Miniature Pig Sperm (Monothiolglycerol이 동결 융해 후 미니돼지 정자의 활성산소 억제, 미토콘드리아 활성 그리고 DNA Integrity에 미치는 영향)

  • Cryopreservation and in vitro fertilization (IVF) protocols are important in genetic studies and applications to transgenic animals. Various studies about boar sperm cryopreservation have been studied for a long time. Those were about the use of extenders, the choice of sugars, the cooling and warming rates. The factors that influence the boar sperm are the dramatic changes in temperatures, osmotic and toxic stresses, and reactive oxygen species (ROS) generation. Among these factors, ROS generation is the main damage to DNA which is a principal genetic material and the most important for the practical applications. So we wondered whether ROS generation could be reduced. In previous study, monothioglycerol (MTG) was essential for the culture of embryo stem cells. Therefore we added MTG in the freezing extender based on lactose-egg yolk (LEY) with trehalose. For the assessment of the frozen-thawed sperm, we focused onmotility, membrane integrity and DNA damage. First, we used a computer-aided sperm analysis system for overall conditions of sperm such as motility and viability. Then we performed the sperm chromatin structure assay for DNA integrity and hypo-osmotic swelling test for membrane integrity. And our result showed the existence of MTG in the freezing extender caused less damage to DNA and higher motility in frozen-thawed boar sperm. Also we checked a relative antioxidant activity of MTG in modified Modena B extender. We concluded that this reagent can activate sperm mitochondria at MTG $0.2{\mu}M$, contribute to sperm motility and DNA integrity but there was no significant difference on membrane integrity. Also antioxidant activity of MTG in modified Modena B extender was proved.

Correlation of Oct4 and FGF4 Gene Expression on Peri-implantation Bovine Embryos Reconstructed with Somatic Cell

  • This study was carried out to investigate the developmental rates of embryo reconstructed with different cell type and to estimate correlation of transcriptional level of octamer-binding transcription factor 4 (Oct4) and fibroblast growth factor 4 (FCF4) gene on peri-implantation stage embryos. Donor cells were transferred into perivitelline space of enucleated oocytes. The karyoplast-cytoplast couplets were accom- plished by cell to cell fusion and activated with ionomycin and 6-dimethylaminopurine. Reconstructed embryos were co-cultured with bovine oviduct epithelial cells in CR 1 aa medium. There is no difference in blastocyst formation rate following nuclear transfer UT) with fetal fibroblast cell (16/50; 32.0%), cumulus cell (16/49; 32.6%) and ear cell (17/52; 32.6%). The expression level of Oct4 and FCF4 in peri-implantation bovine embryo derived from in vitro fertilization (IVF) and NT were determined by reverse-transcription polymerase chain reaction (RT-PCR) technique. In peri-implantation of IVF result in a transient increased of FCF4 paralleled by an increased expression of Oct4. However, Oct4 gene was highly expressed in hatching blastocysts derived from NT compared to IVF. Also, FGF4 expression level in hatching blastocysts and outgrowth stage derived from NT was lower than that of IVF. In conclusion, it is suggested that the different transcription patterns observed in nuclear transfer embryos may lead to a lower rate of embryo development, implantation and pregnancy.

Effect of High Stocking Rates on Growth and Survival of the Endangered Rio Grande Silvery Minnow Hybognathus amarus

  • The endangered Rio Grande silvery minnow Hybognathus amarus was cultured in 2.44-m-diameter outdoor tanks in a 131-day yield trial to assess growth, survival, and percentage of taggable-sized fish (${\geq}35mm$ total length) when stocked at 500/tank (1.07 million/ha), 1,000/tank (2.14 million/ha), and 1,500/tank (3.21 million/ha). At harvest, fish averaged 45.6 mm and 0.94 g in the 500/tank treatment, 42.6 mm and 0.74 g in the 1,000/tank treatment, and 38.4 mm and 0.55 g in the 1,500/tank treatment; the differences were significant (P = 0.05). Survival in the three treatments was 70%, 64%, and 52%, respectively, but the differences were not significant. Percent taggable-sized fish was 86%, 89%, and 65%, respectively, but the differences were not significant. Yield was 672.5 kg/ha, 1,026.6 kg/ha, and 887.8 kg/ha, respectively; yield in the 1,000/tank treatment was significantly greater than that in the 500/tank treatment, but was not significantly greater than that of the 1,500/tank treatment. This facility is a conservation facility and a major goal is to raise fish without formulated feed. Fertilization produced good growth for the first month, but little growth occurred during the second month so supplemental feed had to be used for the final 60 d of the yield trial.
