고지방 식이로 유도된 비만쥐에서 식이 레반이 체지방 형성 및 혈청 렙틴과 UCP 발현에 미치는 영향 (Effects of Dietary Levan on Adiposity, Serum Leptin and UCP Expression in Obese Rats Fed High Fat Diet)

  • The effects of dietary levan, high-molecular-weight $\beta$-2,6-linked fructose polymer, on adiposity, serum leptin and UCP expression in rats fed high fat diet were studied. The adipose tissue hormone, leptin has been proposed to be involved in the regulation of food intake and energy expenditure. Uncoupling protein (UCP), a mitochondrial protein that uncouples the respiratory chain from oxidative phosphorylation, generates heat instead of ATP, thereby increase energy expenditure. To determine whether the dietary levan may have the anti-obesity effect, 4 wk old Sprague Dawley male rats fed high fat diet for 6 wks to induce obesity, and subsequently fed one of three diets for further 6 wks: 1) high fat (40% of calories) diet without levan 2) with 3% (w/w) levan 3) with 5% levan. For the comparison, control group fed AIN-76A diet. Visceral and peritoneal fat masses were lower in high fat diet with levan groups compared to high fat diet group. The effect of levan was dose-dependent. Adipocyte size was significantly reduced in the levan diet groups compared to the no levan diet group. Serum cholesterol level was not affected by levan containing diet, while the serum HDL cholesterol level was higher in leven diet groups. In addition, serum triglyceride level was markedly reduced by levan containing diet, thus lower than that of control group. Serum leptin was reduced by levan containing diet and lower in 5% levan group compared to 3% levan group (p < 0.001), as a result, serum leptin and insulin levels of 5% levan group were reduced to level of control group. Futhermore, the serum leptin level reflected the adiposity. The expression of UCP 1, and UCP 2 in brown adipose tissue was up-regulated by levan containing diet. In conclusions, levan containing diet reduced adiposity and serum triglyceride but increased UCP expression in the obese rats fed high fat diet. (Korean J Nutrition 35(9) : 903~911, 2002)

Effect of High Dietary Carbohydrate on the Growth Performance and Physiological Responses of Juvenile Wuchang Bream, Megalobrama amblycephala

  • An optimum dietary carbohydrate content is important for maximum fish growth. In this study, we fed Wuchang bream (Megalobrama amblycephala) with either control diet (30.42%) or high carbohydrate diet (52.92%) for 90 d. Fish were fed to apparent satiation three times daily in an aquarium with automatic temperature control and circulated water. Growth performance, plasma biochemical parameters, hepatic morphology and enzyme activities were determined. It was shown that compared to fish fed control diet, fish fed high carbohydrate diet had higher plasma triglyceride and cortisol levels for d 90, and lower alkaline phosphatase level for d 45, lower hepatic superoxide dismutase and total antioxidative capacity for d 90, higher malondialdehyde for d 45 and glycogen content for d 45 and 90 (p<0.05). Histological and transmission electron microscopy studies showed that hepatocytes of fish fed high carbohydrate diet contained large lipid droplets, causing displacement of cellular organelles to periphery of hepatocytes. The relative level of hepatic heat shock protein 70 (HSP70) mRNA of Wuchang bream fed high carbohydrate diet was significantly higher than that of fish fed the control diet for 90 d (p<0.05). These changes led to decreased specific growth rate and increased feed conversion ratio (p<0.05). Upon hypoxia challenge, fish fed high carbohydrate diet had higher cumulative mortality than those fed the control diet (p<0.05). These results suggested that high dietary carbohydrate (52.92%) was detrimental to the growth performance and health of Wuchang bream.

SBY-III이 비만 및 비만 후 식이조절 흰쥐에 미치는 영향 (Anti-obesity Effects of SBY-III in High Fat Diet-Fed Obese Rats Continued by High Fat Diet and Regulated by Normal Diet)

  • Objectives : This experimental study was designed to investigate the effect of SBY-III extract on the weight, cell size of epididymal fat-pad, fat accumulation area in liver, serum lipid level and UCP1 mRNA in brown adipose tissue of high fat diet-fed obese rats continued by high fat diet and regulated by normal Diet. Methods : The body weight gain, weight of the internal organs(epididymis, liver, brown adipose tissue), insulin, triglyceride, total cholesterol, total lopod, free fatty acid, expression of UCP1 mRNA were measured in high fat diet-fed obese rats continued by high fat diet and regulated by normal diet. The experimental study are divided into exp-I and exp-II. Each study was administered normal diet, high fat diet and SBY-III according to each situation. Normal group is normal diet for 8 weeks. Exp-I are divided into control group(high fat diet for 8 weeks) and sample group(high fat diet for 8 weeks and SBY-III for last 2 weeks). Exp-II are divided into control group(high fat diet for 6 weeks and normal diet for 2 weeks) and sample group(high fat diet for 6 weeks and normal diet with SBY-III for 2 weeks). These were then compared mutually. Results : 1. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease of weight gain than control group and diet control-fed sample group with SBY-III showed the more effective decrease of weight loss including weight gain than control group. 2. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease cell size of epididymal fat-pad, fat accumulation area in liver than control group. 3. Non diet control-fed sample group taken SBY-III showed the more effective decrease of serum triglyceride, total lipid, free fatty acid than control group and diet control-fed sample group taken SBY-III showed the decrease of serum triglyceride, free fatty acid than control group. 4. Only diet control-fed sample group taken SBY-III showed the decrease of UCP1 volume. Conclusions : These results shows that SBY-III has effects on anti-obesity, especially keeping pace with diet control.

식이섬유의 종류와 섭취수준이 고지방식을 섭취한 흰쥐의 체내 지질대사에 미치는 영향 (Effects of Dietary Fiber Sources and Levels on Lipid Metabolism in Rats fed High Lard Diet)

  • These works were designed to examine the effects of dietary fiber sources and levels on lipid metabolism in rats fed high lard diet. Rats were divided into three($\alpha$-cellulose, polydextrose, carrageenan) groups and were fed with containing 6% fiber diet. Serum, liver and fecal lipid contents were examined after four weeks of dieting. Also, in order to investigate whether hypocholesterolemic effect of dietary fiber at 4 weeks is continued at 8 weeks, rats were fed with diets containing same sources with different levels of each dietary fiber and were sacrificed after eight weeks of dieting. Serum cholesterol in rats fed carrageenan diet and polydextrose diet were reduced compared with $\alpha$-cellulose-fed group at both four and eight weeks dieting. Liver cholesterol in both polydextrose-fed and carrageenan-fed group were lower than $\alpha$-cellulose-fed group. Fecal total group were increased compared with $\alpha$-cellulose-fed group. On the other hand, there was no significant difference between polydextrose-fed and carrageenan-fed group. On the basis of the results, it was suggested that the hypocholesterolemic effect of soluble fiber used in this study (polydextrose, carageenan) on rats was due to the inhibition of cholesterol absorption on the intestinal lumen and acceleration of cholesterol catabolism in the liver and enhancement of bile acids excretion. On the other hand, insoluble fiber($\alpha$-cellulose) showed no hypo-cholesterolemic effect.

칼슘과 철 보충제의 과다섭취가 빈혈모델 흰쥐의 체내 철 이용성 및 간과 신장기능에 미치는 영향 (Effect of Excess Calcium and Iron Supplement on Iron Bioavailability, Liver and Kidney Functions in Anemic Model Rats)

  • This study examined the effects of excess intake of calcium(Ca) and iron(Fe) supplements on iron bioavailability, liver and kidney functions in anemic model rats. Seven-week-old female rats were first fed and Fe-deficient diet for ten weeks, and then fed one of nine experimental diets for an additional eight weeks, containing three levels of Ca, normal (0.5%) or high(1.5%) or excess (2.5%) and three levels of Fe, normal(35ppm) or high(210 ppm) or excess(350ppm). In anemic model rats, serum Fe, total iron binding capacity(TIBC), hemogolin(Hb), hematocrit(Hct) and liver Fe contents were significantly decreased. Apparent Fe absorption significantly increased with increasing dietary Fe levels, and decreased with increasing dietary Ca levels. serum Fe concentration significantly increased in rats fed a high- and excess-Fe diet, and decreased in rats fed a excess-Ca diet. TIBC was decreawed in rats fed a excess-Ca diet, and transferrin saturation(%) increased in rats fed ahigh- and excess-Fe diet. Hb and Hct were decreased in rats fed an excess-Ca diet regardless of dietary Fe levels. Fe and thiobarbituric acid reactin gsubstance(TBARS) Contents of liver significantly increased in rats fed a high- and excess0-Fe diet, and decreased in rats fed a high- and excess-Ca diet. Fe content of the spleen showed similar results. Urinary creatinine and GFR increased in rats fed an excess-Ca diet regardless of dietary Fe levels. GOT, GPT and LDH were not significantly affected by dietary Ca and Fe levels. These results suggest that excess intake of Fe may increase liver Fe deposits and TBARS, and excess intake of Ca may decrease Fe bioavailability and kidney function leading to potential health problems in anemic model rats.

구기자 섭취에 의한 고지방식이를 하는 흰쥐의 혈중 지질상태 변화 (Changes of Serum Lipid Profiles after Eating Lycii Fructus in Rats Fed High Fat Diet)

  • The purpose of this study was to investigate the effect of Lycii fructus on the serum lipid in rats fed high fat diet. We compared the effects of L. fructus and L.fructus water extract both adminstered with high fat diets on rats that had previously been on high fat or standard diets. Two separate experiments were conducted for 6 weeks. respectively. In experiment I, 4 groups of rats were fed experimental diets consisting of either \circled1 6 weeks of a standard diet(control), \circled2 6 weeks of a high-fat diet(HHC), \circled3 3 weeks of a high-fat diet followed by 3 weeks of a high-fat diet containing L. fructus(HHL) or \circled4 6 weeks of a high-fat diet with L. fructus extract in place of water for the last 3 weeks (HHT). In the second set of experiments, a high-fat diet (SHC), high-fat diet containing L.fructus(SHL) or high-fat diet with L. fructus extract in place of water (SHT) were fed for 3 weeks after 3 weeks of standard diet feeding. Rats fed L. fructus diet consumed more diets than high-fat diets. THe results of experiment I showed significant decreases(p<0.05) in serum triglyceride(TB) and low density lipoprotein-cholesterol (LDL-C) levels with L. fructus feedings, but did not show andy changes in total cholesterol (TC) level. High density lipoprotein-cholesterol (HDL-C) level was increased significantly(p<0.05) with L. fructus feedings. Therefore, the ration of LDL-C to HDL-C(LDL-C/HDL-C) which is used as an atherosclerosis index was significantly (p<0.05) low, while the HDL-c/TC ration was significantly(p<0.05) high with L.fructus intake. However, no significant were found in serum cholesterols and TG levels in experimentII. The results of these experiments indicate that , regardless of the feeding from, L. fructus can be beneficial in lowering serum TG and LDL-C levels for habitual high-fat diet intakers. L.fructus also seems to be effective in elevating serum HDL-C level, theregy having beneficial effects on atherosclerosis by influencing the serum lipoprotein profile.

Effects of Dietary Protein Sources and Levels on Heat Production and Thermoregulatory Responses of Sheep Exposed to a High Ambient Temperature

  • Four Suffolk ewes were used in Latin Square switch over design to study the effects of varying levels and sources of protein on heat production and thermoregulatory responses at daytime high ($33^{\circ}C$ temperature. They were fed Italian ryegrass hay supplemented with fishmeal and/or urea, providing three different levels of crude protein (CP) (low/unsupplemented: 7.9, medium: 11.6, and high: 15.8%) at $1.5{\times}maintenance$. Feeds were distributed at 0900 (30%) and 1700 (70%). Urea diet caused higher heat production and increased vaginal temperature compared to fishmeal and fishmeal-urea mix diets. Time spent standing, skin temperature and respiration rate of sheep fed urea were similar with those of sheep fed fishmeal. Sheep fed diet with low CP level had higher heat production, increased vaginal and skin temperature than sheep fed diet with medium CP content. Sheep on high CP diet produced significantly more heat than sheep fed medium CP diets. Their vaginal temperatures were similar with those of sheep fed medium CP diet but lower than those of sheep fed low CP diet. Respiration rates of sheep and time spent by them for standing on all diets did not differ significantly. These results suggest that urea is inferior protein supplement for thermoregulation of animal at hot environment, as it induced higher heat production than fishmeal and fishmeal-urea mix. Thermoregulatory response on fishmeal-urea mix diet was similar to fishmeal diet. Increasing CP of the diet from low to medium gives advantage for thermoregulation of animal. Increasing CP further to high level was not beneficial as it resulted in the responses of sheep similar to those on low protein diet.

  • The effects of indigestible dextrin on lipids in serum, tissue, and feces were investigated in two experiments carried out with 64 male Sprague-Dawley rats for eight weeks. Experiment I diet(normal fat diet) containing 5% corn oil and experiment II diet(high fat diet) containing 18% fat(13% beef tallow and 5% corn oil) and 1% cholesterol were divided into four groups with 0.5% cellulose(0.5CL : control), 10% cellulose(10CL), 10% Indigestible dextrin I(ID-I) and 10% indigestible dextrin II(ID-II), respectively. ID-I and ID-II diets produced a decrease in serum total lipid and cholesterol concentrations in both normal and high fat diets but more significantly in the groups fed high fat diet. ID-I, ID-II, and 10CL of high fat diet decreased serum glucose concentration. ID-I and ID-II diets also decreased liver cholesterol and triglyceride concentrations in rats fed normal and high fit diets. The fecal excretion of cholesterol in ID-II/high fat diet was significantly higher than that of 0.5CL diet. Bile acid excretion was increased in the rats fed 10CL and ID-II diets of normal and high fat diets. These observations indicat the hypolipidemic and hypocholesterolemic effects of indigestible dextrin on the rats. These effects are mediated by the increase of facal cholesterol and bile acid excretion. (Korean J Nutrition31(6) : 981-990, 1998)

  • Objectives : To identify the effect of Taeyeumjowee-tang on the treatment of obesity Methods : After having been fed a high-fat diet for 6 weeks, obese white rats were classified into three groups. Control group 1 was fed a high-fat diet for 2 more weeks. Control group 2 was fed a normal diet for 2 weeks. The Sample group was fed Taeyeumjowee-tang extract with a normal diet for 2 weeks. The result of the comparison among the three groups was as follows. Results : The Sample group decreased significantly as compared with the Control 1 and 2 groups in changes of the body weight, fat cell size, fat accumulation, insulin, triglyceride, total cholesterol, total lipid, free fatty acid and UCP1. Conclusions : Taeyeumjowee-tang has significant effect on the treatment of obesity resulting from a high-fat diet. Especially, it was a sure remedy for Control group 2 that was fed normal1y after having a high-fat diet.

Grape skin improves antioxidant capacity in rats fed a high fat diet

  • This study was conducted to investigate the effect of dietary grape skin on lipid peroxidation and antioxidant defense system in rats fed high fat diet. The Sprague-Dawley rats were fed either control (5% fat) diet or high fat (25% fat) diet which was based on AIN-93 diet for 2 weeks, and then they were grouped as control group (C), control + 5% grape skin group (CS), high-fat group (HF), high fat + 5% grape skin group (HFS) with 10 rats each and fed corresponding diets for 4 weeks. The hepatic thiobarbituric acid reacting substances (TBARS) were increased in high fat group as compared with control group, but reduced by grape skin. The serum total antioxidant status, and activities of hepatic catalase and superoxide dismutase, xanthine oxidase and glucose-6-phosphatase were increased by supplementation of grape skin. Glutathione peroxidase activity was significantly higher in CS group than in C group. Grape skin feeding tended to increase the concentration of total glutathione, especially in control group. The ratio of reduced glutathione to oxidized glutathione was lower in high fat groups than in control groups. The ratio was increased by dietary supplementation of grape skin in control group. These results suggest that dietary supplementation of grape skin would be effective on protection of oxidative damage by lipid peroxidation through improvement of antioxidant defense system in rats fed high fat diet as well as rats with low fat diet.
