Hypoglycemic and Antioxidative Effects of Functional Rice Goami and Nokwon in High Fat-Fed Mice

  • Effects of newly developed functional rice grains Goami (high-amylose rice) and Nokwon (green-kerneled rice) on the glucose metabolism and antioxidative defense system in C57BL/6N mice under high fat diet condition were investigated. Animals were randomly divided into five groups (n = 8) and given experimental diets for eight weeks: normal control diet (NC), high fat diet (HF), and high fat diet supplemented with white rice (HF-WR), Goami rice (HF-GR), and Nokwon rice (HF-NR). At the end of the experimental period, the HF group exhibited markedly higher blood glucose level, insulin concentration, plasma lipid peroxidation and lower hepatic glycogen concentration than that exhibited by NC group. However, diet supplementation of Goami and Nokwon suppressed the high fat diet-induced hyperglycemia and oxidative stress through inhibition of the glucose-regulating enzymes and enhancement of the antioxidant enzymes activities. The results illustrate that the new functional rice Goami and Nokwon may be useful in the development of functional foods with preventive effect against high fat diet-induced hyperglycemia and oxidative stress.

  • This study was attempted to observe the possibility of body fat distribution and fasting serum insulin as potential predictive factors for obesity and energy intake. Subjects participated in this study were seventeen pairs of upper body type women and lower body type women whose age weight BMI daily energy expenditure per body weight were equally matched, Waist to hip girth ratio(WHR) was measured as a criteria of body fat distribution. Comparison of eating behavior between upper body type and lower body type women did not show any significant differences in meal size meal duration and energy intake per minute While serum free fatty acid level was lower in upper body type women percentage of body fat and fasting serum insulin triglycerde level of upper body type women were statistically higher than those of lower body type women(p<0.05) Our data may indicate the possibility that sympathetic nervous system activity was suppressed in upper body type women which needs ti be examined in future studies. In addition since the upper body type women were dieting over the period of survey with the intention of weigh loss we can conclude that upper body type women had larger variabilities than lower body type women in terms of daily energy intake level.

Secondary Fermented Extract of Chaga-Cheonggukjang Attenuates the Effects of Obesity and Suppresses Inflammatory Response in the Liver and Spleen of High-Fat Diet-Induced Obese Mice

  • Cheonggukjang and chaga mushrooms have numerous health benefits, and have been used in alternative medicine. Therefore, a powder mixture of 98: Cheonggukjang and 2: Chaga extracts was fermented with Lactobacillus acidophilus KCTC3925 (FCC) and its anti-obesity effects in high-fat diet (HFD)-induced obese mice were determined. Five-week-old male ICR mice were fed a normal diet or HFD in the presence or absence of 3% and 5% FCC by weight (n = 10 per group). After 12 weeks, the mice were sacrificed, and the serum and tissue samples were collected for analysis. Body weight and epididymal fat pad weight were significantly lowered in the 3% and 5% FCC groups compared with those in the HFD control group (p < 0.01). FCC supplementation suppressed serum triglyceride and increased serum HDL-C levels (p < 0.01). Serum GOT, GPT, and leptin levels, hepatic COX-2 mRNA expression, and splenic COX-2 and IL-4 mRNA expression were significantly higher in the HFD groups than in the control group (p > 0.05); however, except for splenic IL-4 levels, the increases were significantly attenuated by FCC supplementation. Expression of ICAM-1, an aortic inflammatory marker, was significantly increased in the HFD group; this effect was suppressed in the 3% FCC group (p < 0.01) but not in the 5% FCC group. FCC suppressed the body weight and epididymal fat pad weight gain, as well as inflammatory responses in the liver and spleen of HFD-fed mice. Thus, FCC supplementation will be beneficial for the treatment of obesity-related effects.

  • Objectives This study aimed to investigate the efficacy of Ephedra sinica (E. sinica), Panax ginseng (P. ginseng), and Alisma orientale (A. orientale) Extract (MIT) on insulin resistance induced by Non-alcoholic fatty liver disease (NAFLD). Methods C57BL /6 male mice (8-week-old, 20 g) were divided into four groups: control group (Ctrl), high-fat diet group (HFDF), high fat diet with metformin administration group (METT), and high fat diet with MIT administration group (MITT). Each 10 mice were allocated to each group (a total of 40 mice). All mice were allowed to eat fat-rich diet freely throughout the experiment. To examine the effect of MIT, we observed Cannabinoid receptor type 1 (CB1), Cannabinoid receptor type 2 (CB2), G protein-coupled receptor 55 (GPR55), and Transforming growth factor-β (TGF-β). Results In the MITT group, positive reactions of the CB1, CB2, and GPR55 were significantly was significantly suppressed compared to the HFDF group. The positive reactions of the CD36 and TGF-β in the liver tissue were significantly suppressed in MITT. Conclusions MIT has the effect of improving NAFLD induced insulin resistance through the regulation of the lipid metabolism.

IL-10 Expression-Inducing Gut Bacteria Alleviate High-Fat Diet-Induced Obesity and Hyperlipidemia in Mice

  • In the present study, we examined the effects of interleukin (IL)-10 expression-inducing bacteria Bifidobacterium adolescentis HP1, Lactobacillus mucosae HP2, and Weissella cibaria HP3 on high-fat diet (HFD)-induced obesity and liver steatosis in mice. Oral gavage of HP1, HP2, and HP3 reduced HFD-induced bodyweight gain, triglycerides, and total cholesterol levels in the blood and liver. They also suppressed HFD-induced colitis and the fecal δ,γ-Proteobacteria population. Of the tested bacteria, HP2, which most potently inhibited IL-10 expression, also suppressed HFD-induced bodyweight gain, liver steatosis, and colitis most effectively. These findings suggest that IL-10 expression-inducing gut bacteria can suppress obesity and liver steatosis.

Postcontrast T1-weighted Brain MR Imaging in Children: Comparison of Fat-suppressed Imaging with Conventional or Magnetization Transfer Imaging

  • To assess the merits and demerits of postcontrast fat-suppressed (FS) brain MR imaging in children in the evaluation of various enhancing lesions, compared with postcontrast conventional or Magnetization Transfer (MT) imaging. 대상 및 방법: We reviewed patients with enhancing lesion on brain MR imaging who underwent both FS imaging and one of conventional or MT imaging as a postcontrast T1-weighted brain MR imaging. Inclusion criteria of our study were as follows: MR studies should be peformed within one-year interval and showed no significant interval change of imaging findings. Thirty-four patients (21 male, 13 female; mean age, 8 years) with 43 enhancing lesions (19 intra-axial, 19 extra-axial, and 5 orbital location) were included in this study, Twenty-one pairs of FS and conventional imaging, and 15 pairs of FS and MT imaging were available. Two radiologists visually assessed the lesion conspicuity and the presence of flow or susceptibility artifacts in a total of 36 pairs of MR imaging by consensus. For 21 measurable lesions (19 pairs of FS and conventional imaging, 5 pairs of FS and MR imaging), contrast ratio between the lesion and the normal brain( [SIlesion-SIwater]/[SInormal brain-SIwater]) were calculated and compared.

인삼 및 틀의 초 saponin분획의 항당뇨 및 항고지혈 효과 (Hypoglycemic and Hypolipidemic Effects of Crude Saponin Fractions from Panax ginseng and Gynostemma pentaphyllum)

  • Crude saponin fractions were isolated using non-ionic resin chromatography from Ginseng Radix Alba (PG) and Gynostemmae Herba+ (GP). These saponin fractions were orally administered to strep- tozotocin (STZ)-induced diabetic rats for 2 weeks and to high-fat diet-induced obese rats for 4 weeks. Treatment with either PG saponin or GP saponin at a dose of 1 mg/kg significantly decreased the plasma glucose level to that of glibenclamide treated or normal groups. The increased plasma triglyceride (TG) level in diabetic rats was decreased by 50% with PG or GP saponin treatment. Combined administration of PG and GP saponins with different ratios (total dose of 1 mg/kg) also had the similar effects on the blood glucose and TG levels with that of PG or GP alone. Treatments with GP (1 mg/kg) or GP (0.5 mg/kg) and PG (0.5 mg/kg) together significantly suppressed the rise in TG levels induced by high-fat diet whereas slightly suppressed the rise in the total cholesterol level. The body weight gain was also decreased both in the two saponin treated groups. These results demonstrate that either alone or mixture of PG and GP have similar degree of effects on hyperglycemia and hyperlipidemia.

  • The nutritional status of host has a profound effect on immune response and its ability to defend aganinst invading pathogen. Almost all nutrient dificiencies impaired host defence, and more than recommended levels of certain nutrient enhance immune response beyond that observed with 'adequate'. But high-fat diets have been associated with various types of immune dysfunction in experimental animal models and humans. Also, high linoleic acid suppressed immune function and growth and metastasis of tumor than saturated fatty acids. The present review focused on the effect of dietary lipid on immune fuction, cytokine production and clinical conditions like infection, autoimmune disease and cancer.

Effects of ᴅ-allulose on body fat accumulation in rats fed severely carbohydrate-restricted diets containing beef tallow or soybean oil

  • Purpose: The carbohydrate-restricted diet has been recognized to be effective into preventing and alleviating lifestyle-related diseases, such as obesity and type 2 diabetes. The rare sugar ᴅ-allulose is a functional monosaccharide with anti-obesity effects. In the present study, we examined the effects of dietary ᴅ-allulose on body fat accumulation in rats fed severely carbohydrate-restricted diets containing high concentrations of different fats, beef tallow, or soybean oil. Methods: Male Wistar rats (n = 35, 3-week-old) were divided into 5 groups: One chow-fed control (C) group, and four carbohydrate-restricted groups, namely, beef tallow (B), beef tallow + ᴅ-allulose (BA), soybean oil (S), and soybean oil + ᴅ-allulose (SA), with free access to the diet and water for 8 weeks. The B and BA diets contained 23% beef tallow and 2% soybean oil, whereas the S and SA diets contained 25% soybean oil. Furthermore, the BA and SA diets contained 5% ᴅ-allulose. Results: The final body weight, weight gain, and food intake were significantly higher, and food efficiency was significantly lower in the control group compared to the other carbohydrate-restricted groups. Intra-abdominal adipose tissue, carcass fat, and total body fat weights were not influenced by dietary fat type or ᴅ-allulose supplementation, except for the epididymal adipose tissue weight. In contrast, carbohydrate restriction suppressed body weight gain in rats, but remarkably increased body fat accumulation. Conclusion: Under carbohydrate-restricted conditions, no anti-obesity effects of dietary ᴅ-allulose were observed, regardless of the dietary fat type. The causes of these effects are unknown. However, they may be influenced by a very low carbohydrate and high protein diet. Further research is required to elucidate the effects of ᴅ-allulose under various nutrient compositions with different fat, carbohydrate, and protein energy ratios.

High Molecular Weight Poly-Gamma-Glutamic Acid Regulates Lipid Metabolism in Rats Fed a High-Fat Diet and Humans

  • We investigated the effect of high molecular weight polygamma- glutamic acid (hm ${\gamma}$-PGA) on adiposity and lipid metabolism of rats in the presence of an obesity-inducing diet. Thirty-two Sprague-Dawley rats were fed either a normal-fat (11.4% kcal fat, NFC) or high-fat (51% kcal fat, HFC) diet. After 5 weeks, half of each diet-fed group was treated with hm ${\gamma}$-PGA (NFP or HFP) for 4 weeks. The HFC group had significantly higher body weight, visceral fat mass, fasting serum levels of total cholesterol, LDL cholesterol, and leptin, and lower serum HDL cholesterol level compared with those of the NFC group (p < 0.05). Treatment with hm ${\gamma}$-PGA decreased body weight gain and perirenal fat mass (p<0.05), fasting serum total cholesterol, and mRNA expression of glucose-6- phosphate dehydrogenase (G6PD), regardless of dietary fat contents (p < 0.01). However, hm ${\gamma}$-PGA increased serum HDL cholesterol in the HFC group (p < 0.05). In vitro, 3-hydroxy-3-methylglutaryl coenzyme-A (HMGCoA) reductase activity was suppressed by the addition of hm ${\gamma}$-PGA. In agreement with observations in animal study, the supplementation of hm ${\gamma}$-PGA (150 mg/day) to 20 female subjects in an 8-week double-blind, placebocontrolled study resulted in a tendency to decrease total cholesterol and LDL cholesterol concentrations. We thus conclude that dietary supplementation of hm ${\gamma}$-PGA may act as a hypocholestrolemic agent, secondary to its inhibitor effect on HMG-CoA reductase, and decrease abdominal adiposity by decreasing hepatic lipogenesis. The present study is an important first step in establishing the effect of hm ${\gamma}$-PGA on cholesterol levels in rats and humans.
