Water-extract of Helianthus annuus Seed Exhibits Potent Anti-asthma Activity In Vitro and In Vivo (해바라기씨 추출물의 천식 완화 효과)

  • In order to investigate whether antioxidant biomaterials inhibits IL-4 and/or IL-13 expression in vitro and in vivo, we carried out antioxidant assays by enzyme or cell-based assays with Helianthus annuus extract. Antioxidant assays include DPPH, FRAP, hydroxyl radical assays. Helianthus annuus extract exhibited SOD scavenging activity, and had different patterns by each solvent extracted reaction. DW extract inhibited oxidative stress by $H_2O_2$ that induced apoptosis. We measured $CD4^+$ cell and IL-/13 cytokine expression in a classical mouse animal model. The result show that Helianthus annuus extract showed strung inhibition of immune response in the lung. These result suggest that Helianthus annuus extract can reduce inflammation induced by n mouse asthma model.

Effect of Amino Acids Addition on Stability and Antioxidative Property of Anthocyanins (아미노산의 첨가가 anthocyanins 색소의 안정성과 항산화능력에 미치는 영향)

  • Effects of amino acids (Arg, Lys, Gly, Ile, Glu, Asp, and Met) on the color intensity, stability and antioxidative properties of anthocyanins extracted from grape skins were investigated. Intensity of anthocyanins was significantly increased by the addition of Asp. Except for basic amino acids such as Arg and Lys, stabilities of anthocyanins were significantly improved by the addition of other amino acids including neutral, acidic and sulfur containing amino acids during the storage at $30^{\circ}C$ at pH 3.5. In case of control anthocyanins was remained unchanged the intensity of red color decreased significantly during the storage whereas their antioxidative activity were unchanged. Although effects of amino acids addition on electron donating abilities of anthocyanins were not differentiated by DPPH (2,2-diphenyl-1-picrylhydrazyl) method, the addition of Asp or Met resulted in increased ferric reducing ability which measured by FRAP (ferric reducing ability of plasma) assay.

Effect of Harvest Time on Antioxidant Activity and Functional Constituents of Oriental Melons (수확시기가 참외의 항산화 활성 및 기능성 성분에 미치는 영향)

  • This study compared the antioxidant activity and functional constituents of the peel, flesh,and placenta of oriental melons according to the harvest time. The sample oriental melons were harvested during the major harvest time (from June to August) and divided into peel, flesh, and placenta. To examine the antioxidant activity and functional constituents, the sample melons were extracted using EtOH. As a result, the functional constituents and antioxidant activity of the oriental melons were found to change according to the harvest time. The total phenol and total flavonoid contents were highest in the samples harvested in June, and lowest in the samples harvested in July. Meanwhile, the antioxidant activities were estimated using an ABTS and FRAP assay, where the samples harvested in June also showed the strongest the antioxidant potential, while the samples harvested in July showed significantly lower antioxidant activities. This means that the climate influences the synthesis of secondary metabolites. The above data also suggests that oriental melons harvested in June contain more functional phytochemicals, making them more beneficial for human health.

Comparison between the Antioxidant Activity and the Index Content of ACTS002 according to the Extraction Solvent (추출용매에 따른 ACTS002의 항산화 활성 및 지표성분의 함량 비교)

  • Objectives: Samul-tang is commonly used to alleviate the side effects of chemotherapy. This study aimed to establish an efficient method of extracting ACTS002 based on Samul-tang using the yield, high-performance liquid chromatography (HPLC), and antioxidant assay. Methods: ACTS002 was extracted from each extraction solvent, and the contents of 5-hydroxymethyl-2-furaldehyde (5-HMF), paeoniflorin, and ferulic acid were quantitatively analyzed and compared using HPLC. Moreover, the antioxidant activities of ACTS002 were measured using total flavonoids, total phenolic compounds, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and Ferric reducing/antioxidant power (FRAP). Results: All of the components were set as the index contents because they were easy to process. The antioxidant activity of total flavonoids was the highest in 70% ethyl alcohol extracts, and total phenolic compounds were the highest in 50% ethyl alcohol extracts. In DPPH, 50% ethyl alcohol extracts showed the highest activity, and in ABTS 70% ethyl alcohol extracts were the highest. In FRAP, 70% ethyl alcohol extracts showed the highest activity. Conclusions: ACTS002 can control quality by setting 5-HMF, paeoniflorin, and ferulic acid as the index contents. The antioxidant activity measurement was relatively high in the 50% and 70% ethyl alcohol extracts. Our results can predict the possibility of a pharmacological activity and the standardization of ACTS002.

Effect of Storage Temperature and Water Activity on Antioxidant Activities of Powdered Green Tea Extracts (분말녹차 추출물의 항산화 활성에 미치는 수분활성도 및 저장온도의 효과)

  • The antioxidant properties of green tea leaves and powder extracts were determined using several tests including estimation of reducing power, DPPH(1,1-diphenyl-2- picrylhydrazyl) radical-scavenging activity, and FRAP(Ferric reducing/antioxidant power) assay. All tests indicated that extracts of green tea powder had higher antioxidant activities than extracts of green tea leaves, and the activities were concentration-dependent. However, each test yielded somewhat different results with respect to storage conditions. The reducing power of green tea leaves was highest at $1,000{\mu}g/mL$, storage at $4^{\circ}C$, and an Aw(water activity) value of 0.23. However, the reducing power of green tea powder, assayed at $1,000{\mu}g/mL$, was high under all storage conditions(with variations in temperature and Aw), and was about 1.5.2-fold greater than that of green tea leaves. Radical-scavenging activity, as assessed by the DPPH assay, increased in a dose-dependent manner over the range $15{\sim}125{\mu}g/mL$. At higher concentrations, activities were $80{\sim}90%$ of maximal were attained. The FRAP activity of green tea extract also increased with rising concentration. Particularly in the case of green tea leaves, antioxidant activity was most greatest with storage at $-20^{\circ}C$ and Aw values of 0.69 and 0.23 when assayed at a concentration of $1,000{\mu}g/mL$. These results indicate that the most important factor during storage of green tea is not the Aw value but rather temperature, and that use of refrigeration($4^{\circ}C$) is preferable to increase or maintain the antioxidant activities of biological components in green tea.

Antioxidant Activities and Changes in trans-Resveratrol and Indigestible Oligosaccharides according to Fermentation Periods in Cheonggukjang (청국장의 발효기간에 따른 trans-Resveratrol과 비소화성 올리고당의 변화 및 항산화활성)

  • Cheonggukjang was manufactured using three different kinds of soybeans, after which changes in the content of phytoalexins such as trans-resveratrol (3,5,4'-trihydroxy-trans-stilbene) were measured. Along with phytoalexins, changes in the content of functional oligosaccharides such as stachyose and raffinose were also measured, and the corresponding antioxidant activities were studied. The content of trans-resveratrol was found to be higher in fermented beans than in raw beans. Generally, the content was higher as the fermentation period increased. After 48 hours of fermentation, the contents of trans-resveratrol were $50.06{\pm}0.82$, $39.04{\pm}0.49$, and $34.00{\pm}0.54{\mu}g/g$ (Nulchan, Daewon, and Taekwang), respectively, which is 4 times higher than the contents of raw beans. The contents of stachyose were $10.84{\pm}0.42{\sim}13.05{\pm}0.13mg/g$ in raw beans, $11.37{\pm}0.03{\sim}12.05{\pm}0.52mg/g$ immediately after boiling, and $0.16{\pm}0.01{\sim}0.33{\pm}0.02mg/g$ after 12 hours of fermentation, which is a 97% decrease from those of raw beans. After 24 hours of fermentation, no amount of stachyose was detected. The contents of raffinose were the lowest in raw beans at $2.66{\pm}0.09{\sim}3.54{\pm}0.05mg/g$, but they increased 3~4 times between boiling and 24 hours of fermentation to $10.61{\pm}0.16{\sim}12.66{\pm}0.17mg/g$. However, raffinose content tended to decrease to $8.28{\pm}0.17{\sim}11.83{\pm}0.44mg/g$ after 48 hours of fermentation. From FRAP, DPPH, and ABTS assays, antioxidant activities according to fermentation period of Cheonggukjang were rather low in boiled soybeans compare to raw soybeans. However, the activities were higher as the fermentation period increased. The antioxidant activity of trans-resveratrol showed an $RC_{50}$ value of $4.71{\pm}0.36{\sim}8.46{\pm}0.05{\mu}g/mL$ from the DPPH, ABTS, and FRAP assays. This could be partly due to the significant increase in trans-resveratrol according to fermentation periods. However, changes in functional oligosaccharides (stachyose and raffinose) during fermentation appear to not be related to the antioxidant effects of Cheonggukjang.

Quality characteristics of ground coffee extracts according to physical and chemical defects (커피빈의 물리화학적 결점에 따른 추출액의 품질특성)

  • To examine the utilization possibility of defective coffee beans, non-defective and defective coffee bean were compared by means of its physiochemical properties and antioxidant capacities measured by DPPH radical scavenging activity, FRAP assay, total phenol contents, functional component (trigonelline, caffeine, chlorogenic acid) contents. After roasting process, pH and soluble solid contents of coffee extracts decreased; $L^*$ value decreased while $a^*$ and $b^*$ values increased. DPPH radical scavenging activities of defective green bean extracts were higher than that of non-defective green bean extracts. Immature green bean extract showed the highest radical scavenging activity. In FRAP assay, green bean extracts ranged from 15.28~21.80 mM TE which was higher than roasted bean extracts which showed 14.81~16.38 mM TE. Total phenol contents of green bean extracts ranged 191.06~256.25 mg% GAE which was higher than that of roasted bean extracts showed 161.91~173.44 mg% GAE. The contents of trigonelline, caffeine, chlorogenic acid in immature green bean extract were the highest, which showed 895.20 mg/L, 825.85 mg/L and 3,836.94 mg/L respectively. Each contents were decreased after roasting process. Results of this study suggest that defective coffee bean can be used as a functional food material.

Changes in phytoestrogen contents and antioxidant activities during fermentation of soybean-powder milks prepared from different soybean cultivars by Lactobacillus plantarum P1201 (Lactobacillus plantarum P1201에 의한 콩 품종별 콩-분말 두유 발효 과정에서의 식물성 에스트로젠 함량과 항산화 활성의 변화)

  • This study evaluated the changes of phytoestrogen contents and antioxidant activities of soybean-powder milk (SPM) prepared from yellow soybean during fermentation with Lactobacillus plantarum P1201. In consequence, the levels of total phenolic and isoflavone-aglycone contents, ABTS and DPPH radical-scavenging activities, and FRAP assay values increased, while isoflavone-glycoside contents decreased during fermentation. The highest levels of daidzein, glycitein, and genistein were present in the Daepung SPM at concentrations of 177.92, 20.64, and $106.14{\mu}g/g$, respectively after 60 h of fermentation. Moreover, Daepung SPM showed the highest DPPH radical-scavenging activity of 48.54%, an ABTS radical-scavenging activity of 99.25%, and a FRAP assay value of 0.84 at the end of fermentation. The fermented Daepung SPM possessed highest isoflavone aglycone contents and antioxidant activities, which can be utilized for the development of functional foods.

Stability of Ethanolic Extract from Fermented Cirsium setidens Nakai by Bioconversion during Different Storing Conditions (저장조건에 따른 생물전환 발효고려엉겅퀴 주정추출물의 안정성 조사)

  • This study investigated the stability of ethanolic extract from fermented Cirsium setidens Nakai (FCSN) in order to develop functional materials during different storage conditions. We evaluated pectolinarin and pectolinarigenin contents, total phenol content and antioxidant activity (DPPH radical scavenging activity and FRAP assay) of ethanolic extract from FCSN obtained by bioconverison at various temperatures (4, 25 and $50^{\circ}C$) and pHs (4.0, 7.0 and 10.0). Our results show that the pectolinarin, pectolinarigenin, and total phenol contents in ethanolic extract from FCSN were decreased during the storage periods. Moreover, the DPPH radical scavenging activity did not significantly change at $4^{\circ}C$ and $25^{\circ}C$. Pectolinarin and pectolinarigenin contents, total phenol content and DPPH radical scavenging activity of ethanolic extract from FCSN at acidic pH (pH 4.0) and neutral pH (pH 7.0) were higher than those at the alkaline pH range. These results indicate that the optimum storage condition of the ethanolic extract from FCSN was $4^{\circ}C$ and pH 4.0~7.0 range.

Antioxidant Activity of the Ethanol Extract from Cooking Drips of Thunnus thynnus by Gamma Irradiation (참치 자숙액의 항산화 활성에 감마선 조사가 미치는 영향)

  • This study was conducted to examine the effect of a gamma irradiation on the antioxidant activity of ethanol extract from cooking drips of Thunnus thynnus (ECT). The electron donating ability against 1,1-diphenyl- 2-picryl-hydrazyl (DPPH), authentic hydroxyl radical scavenging activity by Ultra Weak Luminescence Analyzer, oxidative protection factors by Racimat method, ferric reducing antioxidant power (FRAP), and ${\beta}$-carotene bleaching activity were measured as indices of antioxidant activity. All of the results shown that irradiation increased the antioxidant activity of ECT. These results suggested that ECT could be used as an antioxidative component in food and gamma irradiation could enhance these antioxidative properties.
