The composition of Hwoangbaec-tang has been traditionally used in Korea to treat cancer. Hwoangbaec-tang I is the water extracts prepared from Angelica dahurica, Fritillariae verticillata, Ailanthus altissima, Viscum coloratun, Scutellaria Radix, Ginseng Radix, Astragalus membranaceus, and Glycyrrhizae Radix. Hwoangbaec-tang II also is the water extracts prepared from Ginseng Radix, Astragalus membranaceus, and Glycyrrhizae Radix. The anti-leukemic effects of human promyelocytic leukaemia (HL-60 cells) by Hwoangbaec-tang I or II was accessed by propidium iodide staining flow cytometric analysis, and apoptosis-inducing activity was further confirmed by a nuclear morphological change, a ladder pattern of DNA fragmentation, and an activation of caspase-3 and 9. Hwoangbaec-tang I was found to induce the apoptosis of HL-60 cells via caspase-3 and 9 pathway. In the other side, Hwoangbaec-tang II was found to inhibit the growth of HL-60 cells by inducing these cells to differentiate toward granulocytes. These results indicate that the different anti-leukemic effects of Hwoangbaec-tang in HL-60 cells can be induce the apoptosis or differnetiation of HL-60 cells in Hwoangbaec-tang composition dependent manner.
This study was designed to investigate the protective effect of Bojungbangam-tang Ethanol Extracts (EBJT) on cisplatin and hydrogen peroxide-induced cytotoxicity of human endothelial cell line ECV304 cells. After cells were treated with cisplatin and hydrogen peroxide, MTT assay was performed for cell viability test. To explore the mechanism of cytotoxicity, we used the several measures of apoptosis to determine whether this processes was involved in cisplatin and hydrogen peroxide-induced cell damage in ECV304 cells. Also, cells were treated with EBJT and then, followed by the addition of cisplatin or hydrogen peroxide. Cisplatin or hydrogen peroxide decreased the viability of ECV304 cells in a dose-dependent manner. ECV304 cells treated cisplatin or hydrogen peroxide were revealed as apoptosis characterized by nuclear staining. EBJT protected ECV304 cells from cisplatin or hydrogen peroxide-induced nuclear fragmentation and chromatin condensation. Also, EBJT inhibited the cleavage of poly(ADP-ribose) polymerase (PARP) in cisplatin or hydrogen peroxide-treated ECV304 cells. According to above results, EBJT may protect ECV304 cells from the apotosis induced by cisplatin or hydrogen peroxide.
이 연구에서는 살아있는 질편모충의 단백질녈해효소가 인체 면역글로불린을 분해하는지 알아보고 질편모충에 의한 조직세포 독성에 있어서 단백질분해효소의 역할을 시험관내에서 관찰하였다 실험에 사용한 질편모충은 질염환자로년터 분리한 KT9 분리주이었으며 세포독성을 알아보기 위한 표적세포로는 HeLa 세포를 사용하였다 질편모충 단백질분해효소가 인체 면역글로불린을 분해하는지 관찰하고자 인체의 분비 IP. 혈청 IgA 및 IgG를 살아있는 원충. 원충의 용출액 및 분비-배설 액과 DTT를 넣어 반응시켰다. 여러 계열의 단백질분해효소 저해제(aminopeptidase, serine, metallo, cystelrle계열)를 살아있는 질편모충과 미리 반응시킨 후 세척하고 면역글로불린의 분해 단백질분해효소 활성 및 조직세포독성에 미치는 영향을 관찰하였다 살아있는 질편모충은 인체의 분비 IgA. 혈청 IgA 및 IgG를 분해하였는데 질편모충 수가 증가할수록 반응시간이 길수록 분해가 더 잘 이루어졌다 질푄모충의 용출액과 분비-배설액도 분비 IgA. 혈청 IgA 및 IgG를 분해하였다. Cysteine, serine계 열의 단백질분해효소 저해제 (I-64 antipain, iodoacetic acid, iodoacetamide, TLCK)를 처리한 질편닐충은 분비 IgA의 분해를 저해하였으며. 단백질분해효소저해제로 처리한 경우 질편모충의 단백질븐해효소 활성은 감소하였고 HeLa세포에 대한 독성이 감소하였다. 이상의 성적을 종합하면 질편모충에서 분비되는 단백질분해효소는 시험관내에서 조직세포에 세포독성을 나타내며 또한 인체면역글로불린을 분해하여 숙주의 방어기전에 대한 도피물질로 작용하는 것으로 생각된다.

습지의 생태-사회적 가치는 인간사회에 혜택을 준다. 많은 나라들이 습지를 보전하고 보호하기 위해 노력하고 있다. 하지만 많은 습지들은 자연적 교란과 함께 인위적 교란으로 인해 소실되거나 구획화되었다. 구획화된 습지는 경관생태학적 연구의 주 대상이 되는데 이는 습지를 효율적으로 관리하고 보전하는 데 매우 중요하다. 본 연구는 행정구역상 경상남도 습지의 공간-사회적 분포양상을 파악하기 위해서 습지의 분포와 크기, 습지 간의 거리, 습지의 경관학적 형태, 토지이용현황, 공시지가 등을 연구하였다. 경상남도 내에는 146개(3,598.85 ha)의 습지가 분포해 있고 하천습지가 76개(1,955.60 ha), 범람습지가 49개(1,282.28 ha), 산지습지가 21개(1,282.28 ha)이었다. 대부분의 습지들이 크기가 작고(<2 ha) 농경지에 둘러 쌓여 있었다. 많은 습지들이 구획화되어 있고 훼손상태가 심각하지만 하천습지와 농업용 수로를 이용하여 연결할 수 있는 것으로 보인다. 습지의 연결성 확대를 통한 습지서식처의 확장은 습지에 의존하여 서식하는 생물들의 보전에 매우 유의할 것으로 생각된다. 많은 습지들이 현재 농업용수확보를 위한 유지로 이용되고 있고 국유지의 비율이 높아 추후 습지의 훼손과 소실은 국가단위의 대규모 계획공사로 인할 가능성이 높으므로 생태적으로 민감한 행정과 법률의 개발이 습지의 보전과 관리를 위해 중요하다.
재배 와송의 항암효과를 입증하기 위해 SW480 대장암세포에서 재배 와송 추출물의 apoptosis 유도 효과 및 그 기전에 미치는 영향에 대해 알아보았다. 재배 와송과 천연 와송 메탄올 추출물의 암세포 성장 억제능을 측정한 결과, 재배 와송은 천연 와송과 유사하게 농도 의존적으로 높은 암세포 증식억제효과를 보였으며 특히 농도 300 ${\mu}g$/mL의 재배 와송 메탄올 추출물에서는 천연 와송 메탄올 추출물 보다 더 높은 억제 효과를 나타내었다. 또한 재배 와송 메탄올 추출물을 분획하여 얻은 각각의 물, 헥산, 클로로포름, 에틸아세테이트 및 부탄올 분획물 중 와송 클로로포름 분획물에서 암세포의 증식 억제효과가 가장 높게 나타났다. 이러한 와송 클로로포름 분획물의 apoptosis 유도 효과는 Sub-G1기의 함량 증가와 핵의 응축 및 apoptotic bodies 생성 그리고 DNA 분절을 통해 나타났다. 한편, 와송 클로로포름 분획물의 caspase 활성은 caspase inhibitor 처리군에서 암세포의 증식 억제효과가 확연히 저해됨을 나타내었으며 각각의 caspase 활성 중 caspase-3 및 -9의 활성을 증가시켰다. 또한 와송 클로로포름 분획물은 apoptosis 관련 단백질들인 Bid, Bcl-2 및 PARP의 발현을 농도 의존적으로 감소시켰으며 tBid 및 Bax 단백질의 발현을 현저하게 증가시켰다. 본 연구 결과, 재배 와송과 천연 와송은 유사한 암세포 성장 억제 효과를 보였으며 재배 와송의 클로로포름 추출물은 caspase 의존적인 경로를 통해 apoptosis를 일으켜 세포 사멸 효과를 나타내는 것을 확인할 수 있었다.
Purpose. Free radicals are implicated in the pathophysiology of aging, ischemic injury and neurodegenerative disorders. To deform]no whether Hominis Placenta extract prevents $H_2O_2$-induced apoptosis, we have performed morphological and biochemical analyses for the detection of apoptotic phenomena in the pineal tumor cell line $PGT-{\beta}$ We have also peformed cytochemical and immunocytochemical analyses for the detection of changes in nitric oxide synthase (NOS) activity and estimated the expression . of apoptotic genes using reverse transcription-polymerase chain reaction (RT-PCR) Methods. $PGT-{\beta}\;cells$ were pretreated with Hominis Placenta extracts $(0,\;10^{-2}\;{\mu}g/ml)$ for 2 hours and then exposed to $H_2O_2\;(0,\;50\;{\mu}M)$ for 3 hours. Appearance of apoptotic characteristics were monitored using 4, 6-diamidino-2-phenylindole dihydrochloride (DAPI) staining assay, terminal deoxynucleotidyl transferase-mediated dUTP-digoxigenin nick end labeling (TUNEL) assay and flow cytometric analysis. NOS activity was measured by NADPH-diaphorase cytochemistry. Expression of inducible NOS (iNOS) and nuclear factor kappa B (NF k B) was assessed via immunocytochemistry. The expression of apoptotic genes was examined by RT-PCR. Results. After 3 flours of exposure to $H_2O_2$, it was shown that $PGT-{\beta}\;cells$ treated with $H_2O_2(50\;{\mu}M)$ exhibit classical apoptotic features and increases in NOS activity and caspase-3 expression. Treatment with Hominis Placenta extract resulted in a reduced occurrence of apoptotic features. DAPI staining, TUNEL and flow cytometric assays revealed decreases in the occurrence of nuclear fragmentation and in the sub-Gl fraction in the $PGT-{\beta}\;cells$ treated with Hominis Placenta extract. Cells treated with Hominis Placenta extract also showed lower activity of NADPH-diaphorase and immunoreactivities of both iNOS and NF k B than those of $H_2O_2$-treated cells which were not treated with Hominis Placenta extract. By RT-PCR, it was shown that the level of caspase-3 mRNA was derreased In the cells treated with Hominis Placenta . extract. Conclusions. This study shows that Hominis Placenta extract prevents $H_2O_2$-induced apoptosis in $PGT-{\beta}\;cells$; inhibitions of iNOS and caspnse-3 are possible mechanisms of the protection against apoptosis.
Background/Objectives: Although anaplastic thyroid carcinoma (ATC) is rare, it is one of the deadliest forms of thyroid cancer. The fatality rate for ATC is high, and the survival rate at one year after diagnosis is <20%. The present study aimed to investigate the anti-tumor activities of paclitaxel, radiation, and tyrosine kinase inhibitor (TKI) combined therapy in anaplastic thyroid cancer cells both in vitro and in vivo and explore its effects on apoptotic cell death pathways. Materials & Methods: ATC cell line was exposed to TKI, lenvatinib in the presence or absence of paclitaxel with radiation, and cell viability was determined by MTT assay. Effects of the combined treatment on cell cycle and intracellular signaling pathways were assessed by flow cytometry and western blot analysis. The ATC cell line xenograft model was used to examine the anti-tumor activity in vivo. Results: Our data revealed that the combined administration of paclitaxel, TKI, and radiation decreased cell viability in ATC cells, and also significantly increased apoptotic cell death in these cells, as demonstrated by the cleavage of caspase-3 and DNA fragmentation. This combination therapy reduced anti-apoptotic factor levels in ATC cells, while significantly decreasing tumor volume and increasing survival in ATC xenografts. Conclusion: These results indicate that administering the combination of paclitaxel, TKI, and radiation therapy may exert significant anticancer effects in preclinical models, potentially suggesting a new clinical approach for treating patients with ATC.
과일의 방사선 조사 여부를 DNA comet assay로 확인하였다. 포도, 자두, 딸기, 복숭아, 사과, 천도복숭아를 구입하여 1.0kGy이하의 저선량으로 조사하고 비조사 시료와 조사시료간의 DNA 손상청도를 육안 검사 및 측정된 comet tail length로 비교하였다. 모든 시료에서 비조사 시료보다 조사시료의 tail length가 더 길었으며 포도와 자두는 $0{\sim}0.5kGy$에서 뚜렷한 증가가 관찰되었다. 특히 자두의 비조사 시료는 포도, 딸기, 복숭아, 사과, 그리고 천도복숭아의 비조사 시료에 비해서 손상된 세포의 comet 모양의 핵이 많이 관찰되기는 하였으나 항상 비손상된 세포의 원형모양의 핵이 동반되었으며 조사된 시료에서는 모두 전반적으로 comet모양의 핵이 관찰되어 비조사 시료와 조사시료 간에 comet 양상을 비교할 수 있었다. Comet 분석을 이용한 과일의 방사선 조사 유무의 확인은 비조사 시료와 조사시료의 tail length의 측정 및 육안 검사 그리고 통계분석으로 확인이 가능하였다. 한편, 이미지 분석기와 같은 정밀한 시스템이 동반되면 저선량으로 조사된 시료들간의 comet 양상을 정확하게 확인하는 것이 가능하다고 생각된다. 따라서 식품의 'DNA comet assay'는 시료의 저장상태 및 화학적 물리적 충격에 대한 몇가지 제한점을 가지고 있지만 간단하고 신속하게 검지할 수 있으므로 다양한 식품의 방사선 조사 유무를 확인하는데 유용하게 활용될 수 있을 것으로 기대한다.
Purpose: Ionizing radiations have been reported as an apoptosis initiating stimulus in various cells and it has established that sustained elevations in [Ca/sup 2+/] can lead to DNA fragmentation by Ca/sup 2+/-dependent endonucleases, ultimately resulting in apoptotic cell death. The previous experiments have been reported by using primarily thymocytes and lymphocytes and the change of [Ca/sup 2+/] was measured only by minutes or hours respectively. We need to evaluate [Ca/sup 2+/] in both several minutes and hours after irradiation of radiation of radiation therapy and verify the apoptotic cells. Materials and Methods: We have measured [Ca/sup 2+/] in human gingival epitheloid cancer cell with 10Gy irradiation, at minutely intervals and hourly intervals using digitized video-intensified fluorescence microscopy and the fluorescent Ca/sup 2+/ indicator dye, fura-2. In order to find out that the transient rise in [Ca/sup 2+/] could induced apoptosis, cells were incubated for 1 hour at 37℃ with TdT enzyme, rinsed and resuspended containing fluorescence and observed under a confocal fluorescence microscope. MTT assay was done to determine cell activity and LDH assay was done to determine the amount of necrotic cells. Results: After irradiation, the transient and temporal increasing of [Ca/sup 2+/] in the KB cells was founded. Though, there was no change in the intracellular [Ca/sup 2+/] at 30 minutes and 2 hours after irradiation. We could detect of DNA fragmented cells at 4 hours after 10Gy irradiated cells. There were no significant differences between 4 hour, 1 day, 3 day cells. There were no significant differences in MTT and LDH assay between the irradiated group and the control group after 4 hours and 1 day. Though after 3 days there were differences in MTT and LDH assay between the irradiated group was significantly decreased than the control group, in LDH assay the number of necrotic cell death of the irradiated was higher than the control group. Conclusion: In KB cells there were incipient and temporal increasing of the [Ca/sup 2+/] with 10Gy irradiation and the apoptosis was founded from 4 hours later which was earlier than seeing of the change of the amount of the cellular ability and necrosis.

제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
