on-line system for preconcentration and separation of lead(II) is presented. The method is based on the complex formation of Pb(II) with adsorbed Methyl thymol blue on activated carbon. The conditions of preparing the solid phase reagent and of quantitative recovery of Pb(II) from diluted solutions, such as acidity of aqueous phase, solid phase capacity, and flow variables were studied as well as effect of potential interfering ions. After preconcentration step, the metal ions are eluted automatically by 5 ml of 0.5 M HNO3 solution and the lead ions content was determined by flame atomic absorption spectrometry. Under the optimum conditions, the lead ions in aqueous samples were separated and preconcentrated about 1000-fold by the column. The detection limit was 0.001 g mL-1. Lead has been determined in river and tap water samples, with recovery of 98 to 102%.

Taste-active compounds (e.g., amino acids and nucleotides) play an important role in contribution to the gustatoty sensation of food. The current study aimed to examine the differences in taste-active compounds between different beef muscles, breeds and aging periods. We have chosen the longissimus dorsi and semitendinosus muscles of Hanwoo breed and longissimus dorsi muscle of Angus breed for the investigation of the aforementioned compounds. Hanwoo muscles were aged for 7 or 28 d, and Angus samples were aged for 28 d at $4^{\circ}C$. Results revealed that 8 out of the 18 detected free amino acids (FAA) showed significant (p<0.05) differences between the two Hanwoo muscles. Twelve FAAs showed aging effect (p<0.05) in which the amounts of 8 FAAs significantly increased as aging time increased. Inosine 5-monophosphate (IMP), hypoxanthine (Hx) and inosine showed significant (p<0.05) differences between the Hanwoo muscles, aging resulted in an increase in amounts of these nucleotides. Hanwoo beef had significantly (p<0.05) higher total amount of sweet amino acids than the Angus ones in that 15 amino acids showed differences (p<0.05) between the two breeds. Amounts of guanosine 5-monophosphate (GMP) and Hx were significantly higher (p<0.05) for Angus beef. Current study indicated that muscle type, breed and aging period had large variations in free amino acid and nucleotide contents, which may subsequently affect the taste attributes of cooked beef.
An enrichment/separation system for atomic absorption spectrometric determinations of Cu(Ⅱ), Fe(Ⅲ), Ni(Ⅱ) and Co(Ⅱ) has been established. The procedure is based on the adsorption of the analytes as calmagite chelates on Chromosorb-102. The effects of some parameters including pH, amount of ligand, salt matrix, flow rates of sample and eluent solutions were investigated. Under optimized conditions, the relative standard deviation of the combined method of sample treatment, preconcentration and determination with FAAS (N=5) is generally lower than 5%. The limit of detection (3σ) was between 6.0-112.9 ㎍/L. The results were used for preconcentration of analytes from some sodium and ammonium salt.

Chemical compounds, hydrogen peroxide and nitrite-scavenging activities of Phellinus baumii (PB) were investigated to expand the utilization of PB as functional food material. Total mineral contents of PB was 534.3 mg% and potassium was the highest content being 224 mg%. Total and reducing sugars were 56.2% and 9.8%, respectively The contents of free amino acids (FAAs) were in a range of $16.9{\sim}765.5mg%$ with the major FAAs of phenylalanine, aspartic acid, glutamic acid, leucine, serine and valine. The contents of total phenolic compounds in methanol and hot water extracts of PB were 33.3 and 20.7 mg/100 mL, respectively and were higher than those of other solvent extracts. Hydrogen peroxide-scavenging activity (80%) of methanol extract at $10{\mu}g/mL$ for 30 min was similar to tocopherol (83.1%) as control. Nitrite-scavenging activity of extracts of methanol and hot water at 500 mg/mL and pH 1.2 were 57.3% and 51.8%, respectively and then their effects were increased by lowering pH. The present results showed that the methanol and water extracts of Phellinus baumii exhibited strong hydrogen peroxide and nitrite-scavenging activities.
Rice flour reference materials were prepared from the unpolished rice grown in Korea and certified for elemental composition. The reference materials consist of two samples containing normal and high level. The reference material at elevated level was prepared by spiking to the normal rice flour six toxic elements of As, Cd, Cu, Cr, Hg, Pb with $1.0{\mu}g/g$ on a dry weight basis. Homogeneity of the prepared materials was evaluated through the determination of Ca, Cu, Fe, Mn, Zn by instrumental neutron activation analysis (INAA) and atomic absorption spectrometry (AAS). Small variance of elemental composition among interbottled samples assured homogeneity of the prepared materials. The materials were decomposed by high pressure digestion and microwave digestion method. INAA, AAS, inductively coupled plasma-atomic absorption spectrometry (ICP-AES), ICP-mass spectrometry (MS) and vapour generation techniques were employed to analyze the reference materials. From this independent analytical results, the certified or reference values are determined for As, Ca, Cd, Cr, Cu, Fe, Hg, K, Mg, Mn, Mo, Na, P, Pb, Se, Zn.

This study aims to assess the water quality of the Eoran and the Imha aquafarm with different aquafarm environments in Haenamn-gun, and analyzed the composition of total amino acids (TAAs) and free amino acids (FAAs) in three Neopyropia dentata cultivars (Yuldo, Supum1 and 2) in two aquafarms. Mean water temperature ranged from 22.9 to 10.9℃ during September to November 2018. In Eoran aquafarm, the water quality analysis showed that NO2-N was high in September, NH4-N and COD in October, and NO3-N, DIN, and DIP in November. In Imha aquafarm, the water quality analysis showed that NH4-N and COD was high in September, NO3-N and DIN in October, and NO2-N and DIP in November. We confirmed the proximate composition, amino acid composition (TAA/FAA) in two auqufarms. In the Eoran aquafarm, the 'Yuldo' cultivar had significantly higher crude lipid content than two other cultivars (Supum 1 and 2). The 'Supum1' cultivar had significantly higher moisture content, whereas the highest content of crude protein, crude lipid, crude ash, and carbohydrates was found in the 'Supum2' cultivar. In the Imha aquafarm, the content of crude lipid and crude ash was highest in the 'Yuldo' cultivar. The highest content of crude protein and carbohydrates was found in the 'Supum1' cultivar, while the 'Supum2' cultivar had the highest content of moisture. The highest concentration of glutamic acid belong to TAAs is observed in all cultivars from Eoran and Imha aquafarm, while all cultivars in two aquafarm also contained higher content of alanine among the detected FAAs.
Magnetite nanoparticles ($Fe_3O_4$ NPs) were synthesized by co-precipitating method under optimized condition. The $Fe_3O_4$ NPs coated with sodium dodecyl sulfate-thenoyltrifluoroacetone ($Fe_3O_4$ NPs-SDS-TTFA) were then exerted as the magnetic solid phase extraction (MSPE) adsorbent for the extraction process prior to introducing to a flame atomic adsorption spectrometry (FAAS). The synthesized $Fe_3O_4$ NPs-SDS-TTFA were applied for the extraction of Pb(II) ions from different water samples. The characterization studies of nanoparticles were performed via scanning electron microscopy-energy dispersive micro-analysis (SEM-EDAX), X-ray diffraction (XRD) and vibrating sample magnetometer (VSM) techniques. The substantial parameters affecting the extraction efficiency were surveyed and optimized. A dynamic linear range (DLR) of $10-400{\mu}g\;L^{-1}$ was obtained and the limit of detection (LOD, n=7) and relative standard deviation (RSD%, n= 6, $C=20{\mu}g\;L^{-1}$) were found to be $2.3{\mu}g\;L^{-1}$ and 1.9%, respectively. According to the results, the proposed method successfully applied for the extraction of Pb(II) ions from different environmental water samples and satisfactory results achieved.
In this research, a novel and efficient quinoline thioacetamide functionalized magnetic graphene oxide composite ($GO@Fe_3O_4@QTA$) was synthesized and utilized for dispersive magnetic solid phase preconcentration of Cd(II) and Ni(II) ions in urine and various food samples. A number of diverse methods were employed for characterization of the new nanosorbent. The design of experiments approach and response surface methodology were applied to monitor and find the parameters that affect the extraction performance. After sorption and elution steps, the concentrations of target analytes were measured by employing FAAS. The highest extraction performance was achieved under the following experimental conditions: pH, 5.8; sorption time, 6.0 min; $GO@Fe_3O_4@QTA$ amount, 17 mg; 2.4 mL $1.1mol\;L^{-l}$$HNO_3$ solution as the eluent and elution time, 13.0 min. The detection limit is 0.02 and $0.2ng\;mL^{-1}$ for Cd(II), and Ni(II) ions, respectively. The accuracy of the new method was investigated by analyzing two certified reference materials (sea food mix, Seronorm LOT NO 2525 urine powder). The interfering study revealed that there are no interferences from commonly occurring ions on the extractability of target ions. Finally, the new method was satisfactorily employed for rapid extraction and determination of target ions in urine and various food samples.
We displayed four types of Solanum nigrum metallothionein (SMT) for the first time on the surface of Saccharomyces cerevisiae using an α-agglutinin-based display system. The SMT genes were amplified by RT-PCR. The plasmid pYES2 was used to construct the expression vector. Transformed yeast strains were confirmed by PCR amplification and custom sequencing. Surface-expressed metallothioneins were indirectly indicated by the enhanced cadmium sorption capacity. Flame atomic absorption spectrophotometry was used to examine the concentration of Cd2+ in this study. The transformed yeast strains showed much higher resistance ability to Cd2+ compared with the control. Strikingly, their Cd2+ accumulation was almost twice as much as that of the wild-type yeast cells. Furthermore, surface-engineered yeast strains could effectively adsorb ultra-trace cadmium and accumulate Cd2+ under a wide range of pH levels, from 3 to 7, without disturbing the Cu2+ and Hg2+. Four types of surfaceengineered Saccharomyces cerevisiae strains were constructed and they could be used to purify Cd2+-contaminated water and adsorb ultra-trace cadmium effectively. The surface-engineered Saccharomyces cerevisiae strains would be useful tools for the bioremediation and biosorption of environmental cadmium contaminants.

A simple and reliable method has been developed to selectively separate and concentrate trace amounts of thallium ion from real samples for the subsequent measurement by flame atomic absorption spectrometry (FAAS). Thallium ions are absorbed quantitatively during passage of aqueous real samples through an octadecyl bonded silica membrane disk modified by 4-(4-Chloro-phenylazo)-2-[(4-hydroxy-phenylamino)-methyl]-phenol. The retained $Tl^+$ ions are then stripped from the disk quantitatively with a minimal amount of thiosulfate solution as eluent. The proposed method permitted large enrichment factors of about 130 and higher. The relative standard deviation for ten replicate extraction of thallium from 1 L samples containing 5 ${\mu}g$ thallium is 1.2%. The break through volume for 5 ${\mu}g$ thallium is 1000 mL. The limit of detection of the proposed method is 11.2 ng of $Tl^+$ per 1000 mL. The effects of various cationic interferences on the recovery of thallium in binary mixtures were studied. The method was applied to the recovery of $Tl^+$ ions from natural water and human hair samples.
