Effect of Different Volume of Microdrop Culture on B6D2F1 Mice Oogenesis (배양액 용량이 B6D2F1 마우스 배아발생능력에 미치는 영향)

  • This study was conducted to evaluate the effects of different volume ($100{\mu}l$ vs. 2 ml) of microdrop culture on B6D2F1 mice oogenesis. In the present study, B6D2F1/CrljOri $F_1$ mice were utilized in order to maximize oogenesis. Also we used TCM-199, Dulbecco's medified Eagle's medium (DMEM), embryo culture medium (Fertilization medium, Cleavage medium, Blastocyst medium), G series medium and One step medium. Blastulation rate was not different between groups ($58.4{\pm}2.9%$ vs. $61.2{\pm}4.8%$). Zona hatched rate ($38{\pm}15.4%$ vs. $27{\pm}3.4%$) and attached rate ($55{\pm}13.9%$ vs. $46{\pm}3.9%$) did not differ by the volume of culture media. Total cell numbers ($59.8{\pm}9.7$ vs. $70.3{\pm}8.7$), ICM cell numbers ($15.8{\pm}0.6$ vs. $16.8{\pm}1.5$), TE cell numbers ($44.0{\pm}9.7$ vs. $53.6{\pm}7.3$), % ICM ($26.4{\pm}2.9%$ vs. $23.8{\pm}3.3%$) and ICM:TE ratio ($1:2.8{\pm}0.4$ vs. $1:3.2{\pm}0.6$) were not different between groups (i.e., $100{\mu}l$ vs. 2 ml). These results show that the capacity of the culture medium did not effect the cell numbers of B6D2F1 mice blastocysts. In summary, these results can provide fundamental data to maximize culture condition for in vitro fertilization on B6D2F1 mice.

  • The ascomycete fungus Fusarium fujikuroi causes bakanae disease in rice and this disease has been reemerging in Korea. Other fungal species including F. graminearum and Magnaporthe oryzae are often associated with F. fujikuroi, hampering pure isolation of F. fujikuroi from rice. In this study, we modified a selective medium for F. fujikuroi as supplementing both pentachloronitrobenzene and hydrogen peroxide into minimal medium. This medium efficiently suppressed the vegetative growth of F. graminearum and M. oryzae, but did not significantly reduce F. fujikuroi growth, providing an efficient tool for isolating F. fujikuroi.

Effect of Culture Medium on Results of Maerobroth Dilution Antifungal Susceptibility Testing of Candida albicans (Candida albicans의 시험관 희석법에 의한 항균력 검사시 배지가 항균력에 미치는 영향)

  • A total of 42 strains of Candida albicans were examined for susceptibility to three antifungal agents, amphotericin B(AMB), 5-fluorocytosine(5-FC), and ketoconazole(KTZ), using defined medium, synthetic amino acid medium-fungal(SAAM-F), supplemented yeast nitrogen base(SYNB) and undefined medium Sabouraud's dextrose broth(SDB) and Kimmig broth media. A tube dilution method was used with minimum inhibitory concentrations(MICs) determined after incubation for 24 hour and 48 hours. All testes were performed in duplicate. In general, MICs were more reproducible after 48 hour of incubation. Forthermore, MICs determined after incubation for 48 hours were significantly higher than those determined after 24 hours. The actural MICs obtained with the different antifungal agents were clearly influenced by the test medium used. The rank order of AMB MICs according to the test medium was as follows: SAAM-F>SYNB>SDB>Kimmig broth. With 5-FC, the following pattern was observed: SYNB>SAAM-F>SDB>Kimmig borth. For ketoconazole, the MICs according to the test medium was SAAM-F>SDB>SYNB> Kimmig broth. In amphotericin B, the MICs mean value with the test medium was as follows: SDB, 0.24 mcg/ml; Kimmig broth, 0.29 mcg/ml; SYNB, 0.21 mcg/ml and SAAM-F, 0.15mcg/ml. The actural value of 5-FC was; SDB, 37.20 mcg/ml; Kimmig broth, 67.41mcg/ml; SYNB, 21.29 mcg/ml and SAAM-F, 24.61 mcg/ml and in ketoconazole, the MICs value was; SDB, 1.83 mcg/ml; Kimmig broth, 4.08 mcg/ml; SYNB, 1.95 mcg/ml and SAAM-F, 1.41 mcg/ml. The results of this investigation suggested that broth dilution susceptibility testing of yeast and yeast-like fungi are best performed with an incubation period of 48 hours. Furthermore, medium composition can significantly influence the results of such testing.

The Effect of EDTA and Fetal Cord Serum Supplementation to Ham's F-10 Culture Medium on Developmental Potential of Mouse Embryos In Vitro (Ham's F-10 배양액에 첨가된 태아제대혈청과 EDTA가 백서 수정란의 분할에 미치는 영향)

  • It is the most important to select optimal culture conditions to promote safe embryo growth in the technique of human in vitro fertilization and embryo transfer. It has been shown that the addition of biologic fluids, such as blood serum, of various origins, improved fertilization and early cleavage rates in numerous species. The purpose of this study is to attempt to measure developmental potential of mouse eggs fertilized and cleaved in Ham's F10 culture medium containing a chelating agent, EDTA and fetal cord serum. In this study, we selected 40 female mice and 20 male mice, and investigated optimal serum concentrations for mouse embryo growth. Two cell stage mouse embryos were cultured in Ham's F-10 medium, Ham's F-10 medium with various concentrations of EDTA, or Ham's F-10 medium with EDTA and 10% human cord serum. Developmental ratios to morula in Ham's F-10 medium containing various concentrations of EDTA and/or 10% fetal cord serum were significantly higher than in unsupplemented Ham's F-10 medium (p<0.05). Developmental ratios to blastocyst in Ham's F-10 containing 10% fetal cord serum and $50{\mu}M$ or $100{\mu}M$ EDTA were significanltly higher than in unsupplemented Ham's F-10 medium (p<0.05). Developmental ratios to morula in Ham's F-10 containing 10% fetal cord serum and $100{\mu}M$ EDTA were significanltly higher than in Ham's F-10 with 10% fetal cord serum used commonly in many human IVF centers(p<0.05). Developmental ratio to blastocyst in Ham's F-10 containing 10% fetal cord serum and $100{\mu}M$ EDTA was significanlty higher than in Ham's F-10 with $200{\mu}M$ EDTA(P<0.05). In summary, embryo development to morula and blastocyst was significanlty higher in the presence of human cord serum or EDTA than in the unsupplemented medium. The most significanly development to morula and blastocyst was obtained at Ham's F-10 medium with $100{\mu}M$ concentration of EDTA and 10% fetal cord serum. These results suggest that Ham's F-10 medium containing 10% fetal cord serum and optimal concentrations of EDTA significantly promoted early cleavage of mouse zygotes, and these will be useful as basic data for the selection of culture medium in human in vitro fertilization.

Development of a Selective Medium for the Fungal Pathogen Fusarium graminearum Using Toxoflavin Produced by the Bacterial Pathogen Burkholderia glumae

  • The ascomycete fungus Fusarium graminearum is a major causal agent for Fusarium head blight in cereals and produces mycotoxins such as trichothecenes and zearalenone. Isolation of the fungal strains from air or cereals can be hampered by various other airborne fungal pathogens and saprophytic fungi. In this study, we developed a selective medium specific to F. graminearum using toxoflavin produced by the bacterial pathogen Burkholderia glumae. F. graminearum was resistant to toxoflavin, while other fungi were sensitive to this toxin. Supplementing toxoflavin into medium enhanced the isolation of F. graminearum from rice grains by suppressing the growth of saprophytic fungal species. In addition, a medium with or without toxoflavin exposed to wheat fields for 1 h had 84% or 25%, respectively, of colonies identified as F. graminearum. This selection medium provides an efficient tool for isolating F. graminearum, and can be adopted by research groups working on genetics and disease forecasting.

Rapid Quenching Dynamics of F Center Excitation by $OH^-$ Defects in KCI

  • The rapid quenching dynamics of F center excitation by OH- defects in KCl crystals are investigated by monitoring ground state absorption bleach recovery, using a picosecond streak camera absorption spectrometer. F center absorption bleach in OH--doped crystals shows three distinguishable recovery components with the current temporal resolution, designated as slow, medium and fast components. The slow one is due to the normal relaxation process of F* centers as found in OH--free crystals. The others are consequent on energy transfer from electronically excited F centers to OH--vibrational levels. The fast component is a minor energy transfer process and resulting from the relaxation of somewhat distant, not the closest, associated pairs of F* and OH- defects. The energy transfer between widely separated F* and OH- defects opens up a recovery process via the medium component which is assisted by OH- librations, lattice vibrations and OH- dipole reorientations. The quenching behaviors of F* luminescence and photoionization by OH- are explained well by the relaxation process of the medium component.

Holding Strength of Screw in Domestic Particleboard and Medium Density Fiberboard(II) -Predicting Formula of Withdrawal Strength of Screw- (국산(國産) 파티클보드와 중밀도섬유판의 나사못유지력(維持力)(II) -나사못 유지력(維持力) 예측식(預測式)-)

  • This study was carried out to determine the withdrawal strength of various screws according to root diameter of screw and embeded length on the face and edge of domestic particleboard and medium density fiberboard. The obtained results were as follows: 1. The withdrawal strength of screw in domestic particleboard and medium density fiberboard was closely related to embeded length of the screw but less dependent on root diameter of the screw. 2. The withdrawal strength on the face and edge of domestic particleboard could be predicted by means of the following expression: $F_{Pf}=4.60{\times}D^{0.24}{\times}L^{1.14}(R^2=0.87)$ $F_{Pe}=0.54{\times}D^{0.43}{\times}L^{1.73}(R^2=0.84)$ Where: $F_{Pf}$ : withdrawal strength on the face of particleboard(kgf) $F_{Pe}$=withdrawal strength on the edge of particleboard(kgf) D=diameter of the screw(mm) L=embeded length(mm) 3. The withdrawal strength on the face and edge of domestic medium density fiberboard could he predicted by means of the following expression: $FM_f=1.53{\times}D^{0.53}{\times}L^{1.39}(R^2=0.93)$ $F_{Me}=1.14{\times}D^{0.66}{\times}L^{1.36}(R^2=0.87)$ where: $F_{Mf}$ = withdrawal strength on the face of medium density fiberboard(kgf) $F_{Mf}$=withdrawal strength on the edge of medium density fiberboard(kgf).

Effects of Nurse's Parenting Stress and Parenting Efficacy on Turnover Intension of Nurses at Medium-sized Hospitals (중소병원 간호사의 양육스트레스와 양육효능감이 이직의도에 미치는 영향)

  • Objectives : The aim of this research was to identify the relationships among parenting stress, parenting efficacy and turnover intension among nurses working in medium-sized hospitals. Methods : Two-hundred three nurses were recruited in 10 medium-sized hospitals in B city. They were asked to complete a questionnaire, and data were analyzed using SPSS program. Results : Subjects perceived a moderate level of turnover intension. Turnover intension showed significant differences by age (F=3.29, p=.039), working department (F=5.11, p=.007), working form (t=0.36, p=.037), reason for going to work (F=3.13, p=.027), and satisfaction with the job (F=17.94, p<.001), support from colleagues (F=12.82, p<.001). Factors affecting turnover intension were satisfaction of working, parenting support from colleagues, reason for working. The model was statistically significant explaining 24.1% of varience (F=10.141, p<.001). Conclusions : Nursing administrators should consider the experience of married nurses at medium-sized hospitals as a significant human resource, and be aware that supplementing this can reduce nurses' turnover intension.

The Effect of E.G.F. and Human Follicular fluid on the Maturation of Mouse Oocytes in in vitro Culture (생쥐난의 체외 배양에서 인간난포액과 표피 성장 인자가 난성숙에 미치는 영향)

  • The human follicular f1uids(F.F.) may be considered to contribute the maturation of the oocytes on the in vitro culture. To investigate the effects of epidermal growth factor (E.G.F.), which is present in mature and immature follicular fluids, we had experiments of mouse oocytes maturation in vitro. The endpoints assayed were rated as percentage of oocytes undergoing germinal vesicle breakdown(G.V.B.D.) and polar body(P.B.) formation at 12 hours after in vitro culture. The rates of G.B.B.D. were 87% in mature F.F. 68% in immature F.F. and 78% in Ham's F-10 medium respectively. And overall the mature F.F. seem to stimulate on in vitro oocyte maturation compared with either immature F.F. or Ham's F-10 medium. As the concentration of addition of E.G.F. in immature F.F., the rates of G.V.B.D. and P.B. formation were 82 %, 23% in addition with 2 ng/ml while 84%, 32% in addition with 4 ng/ml respectivly. And at the concentration of addition of E.G.F. in Ham's F-10 media as well, the rates of G.V.B.D. and P.B. formation were 84%, 40% and 82%, 44% in addition with each 2ng, 4ng. AccordinglY there was no influence on the oocytes maturation at the addition of E.G.F. to each immature F.F. and Ham's F-10 medium. In conclusion, the E.G.F. is not able to induce oocyte maturation independent of it's effects in immature F.F. and Ham's F-10 media.

Cultural Conditions for Pretense Production by a feather-Degrading Bacterium, Bacillus megaterium F7-1 (우모분해세균 Bacillus megaterium F7-1에 의한 단백질 분해효소 생산에 영향을 미치는 배양조건)

  • The effects of inorganic salts and feather concentrations on pretense production by Bacillus megaterium F7-1 were investigated. Pretense production was dependent on the presence of phosphates in the medium. Supplementation of medium with calcium ion slightly increased protease production. The highest protease production was obtained at $1.4\%$ feather. The optimal medium contained $2.0\%$ glucose, $0.8\%$ skim milk, $0.06\%\;K_{2}HPO_{4}\%,\;0.04\%\;KH_{2}PO{4},\;0.06\%\;NaCl,\;0.03\%\;MgCl_{2}\cdot6H_{2}O,\;0.002\%\;CaCl_{2}\cdot2H_{2}O,\;and\;1.4\%$ whole feather. By using this optimized medium, increased production of the protease was achieved compared with the cases of using basal medium.
