Anti-thrombosis Activity of the Aerial Part of Aruncus dioicus var kamtschaticus (눈개승마 지상부의 항혈전 활성)

  • The oriental traditional medicine, Aruncus dioicus var kamtschaticus (ADK) is used for hemostasis (blood stopping) and the promotion of blood circulation. Recently, the demands of the aerial part of ADK as edible mountain herbs are rapidly increased due to its unique fragrance and bioactivity. In this study, to evaluate the anti-thrombosis activity of ADK, ethanol extract and organic solvent fractions were prepared from aerial parts of ADK, and their anticoagulation and anti-platelet aggregation activities were determined. In an anticoagulation activity assay, the ethanol extract of ADK increased the thrombin time, prothrombin time, and activated partial thromboplastin time (aPTT) 1.4-2.3 times at a concentration of 5 mg/ml. Among the fractions, the ethylacetate fraction showed strong inhibitory effects against blood clotting factors, as shown in an extension of the aPTT. In contrast, the butanol fraction strongly promoted blood clotting. In an anti-platelet aggregation assay, the activity of the ethanol extract was comparable to that of aspirin, a commercial anti-platelet aggregation agent, and the butanol fraction showed 2-fold higher aggregation inhibitory activity than aspirin. The aforementioned ethanol extract and active fractions have ignorable hemolytic activity against human red blood cells up to a concentration of 0.5 mg/ml. Considering the high content of total polyphenol, total flavonoid, and total sugar of the ethylacetate and butanol fractions, the purified active substances have potential as safe and novel anti-thrombosis agents. This report provides the first evidence of anti-thrombosis activity of ADK.

Evaluation on efficacy of β-hemolytic Streptococcus iniae vaccine on olive flounder, Paralichthys olivaceus (β-용혈성 Streptococcus iniae 불활화백신의 넙치에 대한 효능 평가)

  • Olive flounder, Paralichthys olivaceus is one of the most important cultured fish in Korea, its farming has been negatively impacted by viral, bacterial and parasitic diseases. Streptococcal infection was considered as a serious problem because of significant economic losses in olive flounder farm industry. The development and evaluation of vaccine for protection against infection by this agent were required. We evaluated the safety and efficacy of ${\beta}$-hemolytic Streptococcus (S.) iniae vaccine on olive flounder Three hundreds of flounders (weight $119.8{\pm}20.7g$, body length $22.6{\pm}1.4cm$) were reared in 0.5 tons aquaria in land-marine tank system. Seawater was provided from the sea of Inchon in Korea, and water temperature was set to $22^{\circ}C$ and $25^{\circ}C$ in the vaccination and challenge test, respectively. We used the formalin-inactivated ${\beta}$-hemolytic S. iniae (F2K) vaccine (M VAC INIAE; Kyoritsu seiyaku, Japan) originated in Japan. The vaccine was intraperitoneally administered to fish. Both of vaccinated group and control group were challenged with intraperitoneally injection by virulent S. iniae SI-36 isolates with $1.0{\times}10^7CFU/fish$ at 3 weeks after vaccination. Difference on mortality of control and vaccinated group (90.0 and 15.0%, 76.5 and 8.0% respectively) at two trials were found significant (p<0.05), and relative percent survival were 83.4% and 89.5%, respectively. The dead fishes were showed dark pigmentation of skin, abdominal extension, hemorrhagic ascites, and liver necrosis, and isolated the S. iniae strain from ascites, liver and kidney. We confirmed the safety and efficacy of ${\beta}$-hemolytic S. iniae vaccine by determinations of the optimal management condition and artificial challenge test in olive flounder.

Detection of Pseudomonas tolaasii Causing Brown Blotch Disease of Mushroom with Species-specific DNA Probe (종 특이 DNA probe를 이용한 버섯 세균성 갈반병 병원균(Pseudomonas tolaasii)의 검출)

  • This study was carried out to develop the molecular marker for the detection of Pseudomonas tolaasii, a causative agent of bacterial brown blotch disease of oyster mushroom (Pleurotus ostreatus). When several primers designed from repetitive sequences and pectin lyase genes of bacteria were used to produce DNA polymorphism from different Pseudomonas spp. isolated from edible mushrooms, PEU1 primer derived from pectin lyase gene produced polymorphic bands differentiating P. tolaasii strains from other Pseudomonas species. Two bands, 1.0kb and 0.4kb, found commonly in 6 isolates of P. tolaasii were cloned into pGEM-T vector which were designated as pPTOP1 and pPTOP2, respectively, to use as probe. The 0.4 kb insert of pPTOP2 hybridized to only 6 isolates of P. tolaasii, but did not to the other Pseudomonas species. As few as $1.5{\times}10^3$ colony forming unit (cfu) of P. tolaasii could be detected by dot blot hybridization with the cloned 0.4kb DNA in pPTOP2.

  • extensible Markup Language (XML) is a new markup language for data exchange on the Internet. In this paper, we propose a language extensible Rule Markup Language (XRML) which is an extension of XML. The implicit rules embedded in the Web pages should be identifiable, interchangeable with structured rule format, and finally accessible by various applications. It is possible to realize by using XRML. In this light, Web based Knowledge Management Systems (KMS) can be integrated with rule-based expert systems. To meet this end, we propose the six design criteria: Expressional Completeness, Relevance Linkability, Polymorphous Consistency, Applicative Universality, Knowledge Integrability and Interoperability. Furthermore, we propose three components such as RIML (Rule Identification Markup Language), RSML (Rule Structure Markup Language) and RTML (Rule Triggering Markup Language), and the Document Type Definition DTD). We have designed the XRML version 0.5 as illustrated above, and developed its prototype named Form/XRML which is an automated form processing for disbursement of the research fund in the Korea Advanced Institute of Science and Technology (KAISI). Since XRML allows both human and software agent to use the rules, there is huge application potential. We expect that XRML can contribute to the progress of Semantic Web platforms making knowledge management and e-commerce more intelligent. Since there are many emerging research groups and vendors who investigate this issue, it will not take long to see XRML commercial products. Matured XRML applications may change the way of designing information and knowledge systems in the near future.

Isolation and Characterization of Pepper mottle virus Infecting Tomato in Korea

  • A peculiar virus-like disease of tomato showing yellow mosaic and necrotic spots on leaves and necrosis on veins, petioles and stems was observed at the Tomato Experimental Station (TES), Buyeo, Chungcheongnamdo, Korea. The disease incidence at TES fields ranged from 21 to 35% infecting different tomato cultivars. For this reason, to identify the virus infecting tomato and to characterize the virus based on biology, serology, cytology and at molecular level. Here, leaf samples were randomly collected from different infected tomato cultivars at TES fields and greenhouses and tested by ELISA using Pepper mottle virus (PePMoV) and Tomato mosaic virus (ToMV) antisera. Infected saps were mechanically inoculated in different host plants to test for pathogenicity, symptomatology and host ranges. Infected tissues and ultrathin sections were examined by electron microscopy. Finally, putative coat protein and 3'-untranslated region (CP/3'-UTR) fragment was amplified and cloned for sequence determination and analyzed its genetic relationship to existing PepMoV and PVY sequences at the Genbank. Results showed 69% of the samples were positive with PepMoV, 13% with ToMV and 19 % were doubly infected with PepMoV and ToMV. Symptoms greatly varied from different host plants inoculated with tomato leaf sap infected with PepMoV alone and discussed in detailed in this paper. Electron microscopy from infected tissues showed filamentous particles of 720-750nm in length, a typical morphology and size of PepMoV. In addition, cylindrical inclusion bodies, pinwheels, scrolls and laminates with masses of fibrillar inclusions were also found in ultrathin sections. Alignment of the sequences of the CP/3'-UTR revealed >96% sequence identity with PepMoV and only <61% with PVY. Taken together, all these evidences presented clearly indicated that the causal agent infecting tomato at TES was PepMoV and we designated this PepMoV infecting tomato as Tom-sd2 strain in this study.

Isolation and Characterization of Actinomycete Strain BK185 Possessing Antifungal Activity against Ginseng Root Rot Pathogens (인삼 뿌리썩음병균에 항균활성이 있는 방선균 BK185의 분리 및 특성)

  • Ginseng (Panax ginseng C. A. Meyer) is an economically valuable pharmaceutical crop in Korea. In order to find promising biocontrol agents for soil-borne fungal pathogens which infect ginseng roots, we have isolated actinomycete, BK185 from soil. The isolate was investigated for the antifungal activity against to ginseng rot pathogens prior to testing genetic and chemical properties. The strain was identified as Streptomyces sp. using phylogenetic analysis based on 16S rRNA gene sequence. The most closely related species was S. sporoclivatus and S. geldanamycininus with high similarities (>99%). The isolate, BK185 showed positive reaction for PCR detection targeting biosynthetic gene clusters of PKS (Type-I polyketide synthase) and NRPS (Non-ribosomal polypeptide synthetase) genes. Major metabolite from the BK185 was analyzed by The LC/MS and identified to geldamycin, which was known to contained broad antibacterial, antifungal or anticancer activities. The results provide evidences that the strain, BK185 can be promising biocontrol agent for ginseng organic farming.

Inhibitory Effect of Extracts from 33 Medicinal Herbs Against TMV and CMV Infection (33종 생약 추훌물의 담배모자이크바이러스(TMV)와 오이모자이크바이러스(CMV)에 대한 감염 억제효과)

  • Extracts of 33 medicinal herbs belonging to 28 different families used as precious resources in the Northeast Asia were tested for their antiviral activities against two major plant viruses. Twenty one methanol extracts from 19 different families were found to have a antiviral activity against tobacco mosaic virus (TMV) and cucumber mosaic virus (CMV) when tested on indicator plants under greenhouse conditions. Most of these extracts have weak activities at the concentration used. But the methanol extracts of Phellinus linteus exhibited potent ($98.7{\pm}1.3%$) antiviral activity against TMV infection and also showed $97.2{\pm}2.2%$ activity against CMV infection. The methanol extracts of the gall of Rhus javanica also showed strong inhibitory efficacy over $98.7{\pm}1.1%$ against TMV or CMV infection. Further research is needed to elucidate the active constituents of these medicinal herbs which may be useful in the development of new and effective antiviral agent against plant viruses.

Compressive Strength Characteristics of Light-weight Air Foamed Soil Using Dredged Silty Soils (준설 실트질 점토를 이용한 경량기포혼합토의 압축강도 특성)

  • In this research, laboratory tests were carried out to investigate the engineering properties of Light-Weight Air Foamed Soil (LWAS) based on silty clays with the animal foaming agent and cement. LWAS has been used as an embankment material over soft ground for road and side extension of the existing road. In field, unit weight and flow value is measured right after producing in mixing plant in order to control the quality of LWAS, and laboratory tests are carried out to confirm the quality through compressive strength of LWAS as well. In this research, direct estimation of the specification requirement of strength using flow values in field is the main purpose of the study together with other characteristics. From the test results, it can be seen that flow values increase with the initial water content and unit weight increases with the depth due to material segregation. Compared to the upper specimen, lower end of 60 cm specimen shows about 2 times higher compressive strength. Relationship between flow values and normalized factor presented by Yoon & Kim (2004) was presented. With that relationship, compressive strength can be predicted from flow values in field. From the relationship, the normalized factor was calculated. Thereafter calculated compressive strengths according to the flow values were compared to measured strengths in the laboratory. The higher the initial water content of the dredged soil has, the better relationship between predicted and measured shows. Therefore it is necessary to predict the compressive strength in advance through the relationship between the flow value and the normalized factor to reflect it in the design stage.

  • Today, Korean medical community faces rapid changes in medical environment due to opening of medical market, more emphasis on making profit and introduction of private medical insurance. Therefore, it is apparent that around, major university hospitals, efforts are being made to adapt to such changes by establishing mid to long range strategic plans. We want to keep pace with changing times and diverse demand of patients by introducing state of the art system, utilizing Ubiquitous technologies for improvements. In doing so, we want to distinguish our hospital services from others. However, Hospital Information System that integrates ubiquitous technologies are introduced in limited basis due to problems like standardization and limits on medical use etc. Particularly, problems like absence of tag design suitable for medical environment, compatibility and extension issue with RFID system need to be addressed on application of RFID technologies. In order to solve such problems, this paper implemented RFID tag system, RFID-SIP UA program, location tracking program. This paper provides a comprehensive basic review of RFID and SIP model suggests the evolution direction of further advanced RFID application service. The design of RFID-SIP application offers advance RFID system with drawback and reduction search time of medical object. so there is a improvement of hospital information system in ubiquitous environment.

Control of Powdery Mildew on Solanaceous Crops by Using COY (Cooking Oil and Yolk Mixture) in the Greenhouse (난황유를 이용한 가지과 작물의 흰가루병 방제)

  • Cooking oil and yolk mixture (COY), a environmentally acceptable plant protection agent, and COY+$CaCO_3$+neem oil mixture were studied to control the powdery mildew occurring on eggplant, paprika, cherry tomato and maturity tomato in glass houses and vinyl houses during 2005 to 2007. The morphological changes of the pathogenic fungi on the leaf surface before and after treatment of COY were observed. COY made of rape seed oil and COY+$CaCO_3$+neem oil mixture were sprayed three times with 5 days interval to foliar parts of eggplant, paprika and tomato and the disease development were examined 5 days after final spray. In eggplant, the control efficacy of COY to powdery mildew was 94.6%. In paprika, the control efficacy of COY to powdery mildew was 91.6% and that of COY+$CaCO_3$+neem oil mixture was 96.2% that revealed little higher than COY itself. In tomatoes(cherry or maturity tomato), the control efficacy of COY were about 91 %, however, when COY mixture were sprayed to tomato leaves and stems the powdery mildew was controlled completely. Typical and healthy mycelia, conidiophores and condia were observed through scanning electron microscope in COY unsprayed leaf surface, on the other hand destroyed and winkled mycelia and conidiophores were observed in COY treated leaves regardless host plants nor taxonomic differences of fungi.
