Purpose: Glabridin (GD) is a bio-available isoflavane isolated from the root extract of licorice (Glycyrrhiza glabra L.). It exhibits a variety of pharmacological activities such as anti-inflammatory and anti-oxidant activities. However, extracellular vesicles (EVs) secretion and the anti-cancer mechanism of action remains largely unknown. The present study investigates the anticancer effects of GD by determining the inhibition of EVs secretion in the human breast cancer cell line, MDA-MB-231. Methods: Cell viability, reactive oxygen species (ROS) production, migration, invasion rate, and vascular endothelial growth factor (VEGF) concentration were assessed in MDA-MB-231 cells treated with increasing concentrations of GD (0.1, 1, 5, 10, 20 µM). Subsequently, EV secretion and exosomal DEL-1 protein expression were evaluated to determine the anticancer effects of GD. Results: The results showed that GD significantly inhibited the cell proliferation of MDA-MB-231 cells in a dose- or time-dependent manner. Also, ROS production and apoptosis marker protein cleaved caspase-3 were significantly increased in GD-treated MDA-MB-231, compared to control. Furthermore, GD exposure resulted in significantly decreased not only migration and invasion rates but also the VEGF concentration, thereby contributing to a reduction in angiogenesis. Interestingly, the concentration and number of EVs as well as EV marker proteins, such as CD63 and TSG101, were decreased in GD-treated MDA-MB-231 cells. Markedly, extracellular matrix protein DEL-1 as angiogenesis factor was decreased in EVs from GD-treated MDA-MB-231 cells. Conclusion: This study identifies that the anti-cancer molecular mechanism of GD is exerted via inhibition of angiogenesis and EVs secretion, indicating the potential of GD as a chemotherapeutic agent for breast cancer.
Mucosal immunity is a well-designed defense system that builds precise and dynamic relationships against pathogens, and the gastrointestinal tract is the most important organ with this system, acting as a guardian at the forefront of its activity. Salmonella spp. cause food poisoning, entering the body orally and mainly invading the Peyer's patches of the small intestine. Although Salmonella strains share similar mechanisms for inducing innate immunity, different serotypes may have different effects on the intestinal mucosa due to host specificities and pathogenicity. In this study, we evaluated the effects of Salmonella enteritidis infections in mouse intestine and observed significantly reduced dose-dependent survival rates in a challenge test. Flow cytometry data showed no significant differences in intestinal immune cell populations, although histology indicated increased mucin production and decreased goblet cell counts in the Salmonella-treated groups. Furthermore, Claudin expression was significantly decreased in the samples with Salmonella. To investigate the relationship between S. enteritidis infection and inflammatory response, dextran sodium sulfate (DSS) was administered after infection and the results indicate lower survival rate after DSS treatment. In conclusion, we were able to identify the optimal concentration of S. enteritidis to modulate the intestinal mucosal immunity of mice and inflammatory response.
Solenopsis japonica, which is belonging to Formicidae in Hymenoptera, is a native ant species in Korea. However, it had not been studied for cold hardiness of S. japonica to understand on its overwintering mechanisms in field so far. Cold tolerance on developmental stages was measured at different cold temperature with various exposure times. Workers showed more survival at 5℃ and 10℃ compared with other stages and elevated cold tolerance when workers were exposed at 15℃ for more than 12h incubation as a rapid cold hardening (RCH) condition. RCH treatment not only increased survival of workers at cold temperatures, but also decreased supercooling point (SCP) and freezing point (FP). RCH group increased the survival rate by 44% at 10℃ compared with Non-RCH group. SCP and FP were depressed from -10.0 to -14.2℃ and from -11.3 to -15.3℃, respectively, after RCH treatment. Cold temperature increased expression level of cold- and stress-related genes such as glycerol kinase and heat shock protein. These results indicate unacclimated cold tolerance of S. japonica and its acclimation to low temperature by RCH.
The purpose of this study was to consider composition of the movement program for the elderly with dementia to develop visit-customized movement program. For the study, 20 experts were selected including 8 dance therapists, 2 dementia-specialized doctors, 5 physical education leaders of the elderly, and 5 geriatric care helpers. First Delphi data, which listed items, was used to analyze content. Depression has a negative impact on social functioning due to its high prevalence and increased suicide rate, and is a disease with a high economic burden. Depression is related to diverse brain-related phenomena, such as neuroinflammation, synaptic dysfunction, and cognitive deficit. As antidepressant drugs used in clinical trials have shown poor therapeutic effects, antidepressant drugs that show rapid efficacy urgently need to be developed. Although studies on various genes, proteins, and signaling pathways related to depression have been conducted, the pathogenesis of depression has not been clearly elucidated. Sirtuin 1 is a nicotinamide-adenine dinucleotide- (NAD+-) dependent histone deacetylase and is involved in cell differentiation, apoptosis, autophagy, and cancer metabolism. Recent genetic studies found that sirtuin 1 is a potential target gene for depression. In addition, preclinical studies reported that sirtuin 1 signaling affects depression-like behavior. In this review, we attempt to present up-to-date knowledge of depression and sirtuin 1. We describe the various roles of sirtuin 1 in the regulation of glial activation, circadian rhythm, neurogenesis, and cognitive function and the effects of its expression on depression. Further, we discuss the effect of sirtuin 1 on the impairment of neural plasticity, one of the key mechanisms of depression, and the associated mechanisms of sirtuin 1.

A buccal pit is a prominent point-like depression that appears at the cervical end of the mandibular molar developmental grooves. A defective buccal pit can be defined as a buccal pit in which the continuity of the dentinoenamel junction is broken and the pit extends to the dentinal level. This study aimed to determine the frequency of buccal pits and defective buccal pits in un-erupted mandibular first and second molars using cone-beam computed tomography (CBCT). The analysis was performed on CBCT images taken from 417 Korean children and adolescents who visited the Department of Pediatric Dentistry, Yonsei University Dental Hospital between 2004 and 2020. Based on cross-sectional views of CBCT images, buccal pits were categorized into 4 classes according to the depth of the pits. The expression rate of the buccal pits was 29.1%. The prevalence of defective buccal pits was 7.9%. The buccal pits tended to develop bilaterally. To date, this is the most comprehensive study on the frequency of buccal pits with the largest sample size. This was the first attempt worldwide to analyze the depth of the buccal pit using CBCT images and to define a defective buccal pit worldwide.
Recently, the demand for the cultivation of upland soil has been increasing, and the rate of conversion of paddy soil into upland soil is also increasing. Theincrease in uneven precipitation due to climate change has resulted in dramatic effects of waterlogging stress on upland crops. Therefore, the present study was conducted to investigate the changes in growth characteristics and the expression patterns of proteins at the two-leaf stage of adzuki beans. The domestic cultivar, Arari (Miryang No. 8), was used to test waterlogging stress. At the two-leaf stage of adzuki beans, plant height slightly decreased androot fresh weight showed significant changes after 3 days of waterlogging treatment. Chlorophyll content was also significantly different after 3 days of waterlogging treatment compared to its content in control plants. Using two-dimensional gel electrophoresis, more than 400 protein spots were identified. Twenty-one differentially expressed proteins from the two-leaf stage were analyzed using linear trap quadrupole-Fourier transform-ion cyclotron resonance mass spectrometry. Of these 21 proteins, 9 were up-regulated and 12 were down-regulated under waterlogging treatment. Protein information resource (https://pir.georgetown.edu/) categories were assigned to all 49 proteins according to their molecular function, cellular component localization, and biological processes. Most of the proteins were found to be involved in the biological process, carbohydrate metabolism and were localized in chloroplasts.
Hand-foot-and-mouth disease (HFMD) is a viral infectious disease that occurs in children under 5 years of age. Its main causes are coxsackievirus (CV) and enterovirus (EV). Since there are no efficient therapeutics for HFMD, vaccines are effective in preventing the disease. To develop broad coverage against CV and EV, the development of a bivalent vaccine form is needed. The Mongolian gerbil is an efficient and suitable animal model of EV71 C4a and CVA16 infection used to investigate vaccine efficacy following direct immunization. In this study, Mongolian gerbils were immunized with a bivalent inactivated EV71 C4a and inactivated CVA16 vaccine to test their effectiveness against viral infection. Bivalent vaccine immunization resulted in increased Ag-specific IgG antibody production; specifically, EV71 C4a-specific IgG was increased with medium and high doses and CVA16-specific IgG was increased with all doses of immunization. When gene expression of T cell-biased cytokines was analysed, Th1, Th2, and Th17 responses were found to be highly activated in the high-dose immunization group. Moreover, bivalent vaccine immunization mitigated paralytic signs and increased the survival rate following lethal viral challenges. When the viral RNA content was determined from various organs, all three doses of bivalent vaccine immunization were found to significantly decrease viral amplification. Upon histologic examination, EV71 C4a and CVA16 induced tissue damage to the heart and muscle. However, bivalent vaccine immunization alleviated this in a dose-dependent manner. These results suggest that the bivalent inactivated EV71 C4a/CVA16 vaccine could be a safe and effective candidate HFMD vaccine.

In this study, stem cells were extracted from the callus derived from the aloe cambium, and the antioxidant activity and effect of skin elasticity improvement were assessed. The aloe cambium-derived stem cell extract, AloStem and callus showed the survival rate of each 98.27% and 71.31%. In the results of the DPPH antioxidant activity of AloStem and aloe extract, it was confirmed that the antioxidant effect of AloStem was more than twice that of Aloe extract. AloStem did not affect the cytotoxicity of normal human dermal fibroblasts (NHDF) cells up to 0.25% concentration. Also, AloStem increased elastin, COL1A1 and HAS2 mRNA expression levels dose-dependently. Furthermore, to examine lifting effect of skin elasticity using a sheet mask containing AloStem, 21 adult men and women applied the sheet mask in the face, once a day for 2 weeks. As a result, after 2 weeks the skin length was 116.75 ± 5.58 mm before the use of the sheet mask, but after 2 weeks of use, it was confirmed that 0.59% increased to 117.44 ± 5.17 mm. Thus, we concluded that the sheet mask containing AloStem can help the lifting effect of skin elasticity.

ASL estimation of public building is based on how appropriate the maximum age of the asset is derived based on the age record of the asset in the statistical data owned by public institutions. This is because we get a 'constrained' ASL by that number. And it is especially true because other studies have assumed that the building is an Iowa curve R3. Also, in this study, the survival rate is 1% as the threshold value at which the survival curve and the predictable life curve almost coincide. Rather than a theoretical basis, in the national statistical survey, the value of residual assets was recognized from the net value of 10% of the acquisition value when the average service life has elapsed, and 1% when doubling the average service life has elapsed. It is based on the setting mentioned above. The biggest constraint in fitting statistical data to the Iowa curve is that the maximum ASL is selected at R3 150%, and the 'constrained' ASL is calculated by the proportional expression on the assumption that the Iowa curve is followed. In like manner constraints were considered. First, the R3 disposal curve for the RCC(reinforced cement concrete) building was prepared according to the discarding method in the 2000 work, and it was jointly worked on with the National Statistical Office to secure the maximum amount of vintage data, but the lacking of sample size must be acknowledged. Even after that, the National Statistical Office and the Bank of Korea have been working on estimating the Iowa curve for each asset class in the I-O table. Another limitation is that the asset classification uses the broad classification of buildings as a subcategory. Second, if there were such assets with a lifespan of 115 years that were acquired in 1905 and disposed of in 2020, these discarded data would be omitted from this ASL calculation. Third, it is difficult to estimate the correct Iowa curve based on the stub-curve even if there is disposal data because Korea has a relatively shorter construction history, accumulated economic wealth since the 1980's. In other words, "constrained" ASL is an under-estimation of its ASL. Considering the fact that Korea was an economically developing country in the past and during rapid economic development, environmental factors such as asset accumulation and economic ability should be considered. Korea has a short period of accumulation of economic wealth, and the history of 'proper' architectures faithful to building regulations and principles is short and as a result, buildings 'not built properly' and 'proper' architectures are mixed. In this study, ASL of RCC public building was estimated at 70 years.
