Improved choleretic effect of Benachio-F®-based formula enriched with fennel extracts

  • Functional dyspepsia (FD) is a gastrointestinal disorder with diverse symptoms but no structural or organic manifestations. Benachio-F® (herein named 'BF-1') is an over-the-counter liquid digestive formulated with multiple herbal extracts, which has been reported to improve symptoms of FD. A total two experiments were conducted. First, we examined whether BF-1 can modulate the progression of FD through two experimental rat models. A total of three doses (0.3x, 1x, 3x of the human equivalent dose) were used. In the gastric emptying model, both 1x (standard) or 3x (3-fold-concentrated) BF-1 enhanced gastric emptying was compared with that of vehicle-treated animals. In a feeding inhibition model induced by acute restraint stress, treatment with 1x or 3x BF-1 led to a similar degree of restoration in food intake that was comparable to that of acotiamide-treated animals. Among the constituents of BF, fennel is known for its choleretic effect. Thus, we next investigated whether a novel BF-based formula (named 'BF-2') that contains an increased amount of fennel extract (3.5-fold over BF-1), has greater potency in increasing bile flow. BF-2 showed a superior choleretic effect compared to BF-1. Furthermore, the postprandial concentration of serum secretin was higher in animals pretreated with BF-2 than in those pretreated with BF-1, suggesting that the increased choleretic effect of BF-2 is related to secretin production. Our results demonstrate that BF-1 can modulate the pathophysiological mechanisms of FD by exerting prokinetic and stress-relieving effects, and that BF-2 has a better choleretic effect than BF-1.

Erythrocyte Micronucleus Test of Pomace Schisandra chinensis Extracts Using ICR Mouse (ICR 마우스를 이용한 오미자박 추출물의 소핵 시험)

  • Objectives : In this study, erythrocyte micronucleus test of pomace Schisandra chinensis extracts was conducted in order to up-cycling to a high value-added industry using by-products discarded in the production process of Schisandra chinensis products and active ingredients such as dibenzocyclooctadiene lignans in Schisandra chinensis. Methods : The micronucleus test was performed according to the 'OECD Guidelines'. Including the negative control group(0 mg/kg) and the positive control group(CPA 70 mg/kg), pomace Schisandra chinensis extracts were orally administered to ICR mouse at doses of 500, 1,000, and 2,000 mg/kg. After sacrificing the experimental animals bone marrow cells were collected and micronucleated polychromatic erythrocyte were counted. And genetic toxicity was confirmed according to the frequency of micronucleus. Results : As a result of the micronucleus test, there were no changes in body weight, clinical signs, or death in any group. But, a significant increase was observed in the frequency of micronucleated polychromatic erythrocyte among polychromatic erythrocytes in the positive control group administered with CPA compared to the negative control group(p<0.05). Whereas, no significant increase was observed in the group administered with pomace Schisandra chinensis extracts compared to the negative control group. Conclusions : Pomace Schisandra chinensis extracts did not induce micronucleus in bone marrow cells of ICR mouse up to a concentration of 2,000 mg/kg, and it was judged that no genetic toxicity was observed.

Embryotoxic and Teratogenic Effects of Scolopendra Water Extract in Mice (오공(蜈蚣) 추출물의 태아 기형 및 모체 독성 마우스 시험)

  • Objective : Scolopendra, a dried body of Scolopendra subspinipes mutilans, is one of Korean medicine. Several reports revealed that Scolopendra has therapeutic effects for arthritis, neuroinflammatory diseases and neuropathic pain. However, the fetal adaptive response or teratogenicity associated with administration of Scolopendra is unclear. Therefore, this study aimed to investigate the fetal toxicity effects that were induced following oral administration of Scolopendra water extract (SWE) in pregnant mice. Methods : The pregnant mice were administrated SWE at dosed of 0, 100, 500 and 1000 mg/kg/day during gestation day 0-18. The mortality, body weight and clinical signs of pregnant mice were observed throughout experimental period. Also, the mortality and malformations in foetus were examined. Results : No meaningful changes were observed in the mortality and clinical signs of pregnant mice between the normal control group and SWE administrated groups. Additionally, there are no significant changes in fetal mortalities, and malformations by SWE administration. conclusion : These results suggest that oral exposure to SWE during pregnancy at oral dosages up to 1000 mg/kg/day did not induce teratogenic toxicity in regard to fetal mortality and morphology.

  • In this research, the microwave oven drying method was investigated as a potential accurate testing approach for determining the water content of fresh concrete. To do so, water content estimation formulas employed in three prominent oven drying test methods were selected, and the calculation principles for each equation, along with potential error factors arising during the actual testing process, were considered. Moreover, a concrete test was conducted to validate the possible error factors. Consequently, it was confirmed that estimation errors in the water content of fresh concrete can occur due to sample deviations arising during the wet screening process for creating mortar specimens or deviations in the coarse aggregate sampling quantity during the sample collection process.

  • The allowable mean overtopping discharge is used as a design parameter for coastal structures. The crest elevation of coastal structures should ensure the wave overtopping discharge within acceptable limits for structural safety and the safety of pedestrians, vehicles, operations, and so on. In this study, two-dimensional physical model tests on typical rubble-mound structure geometries were performed and the the mean wave overtopping discharges under various water depth and wave conditions were measrued. The various test conditions were applied to the tests with the change of the wave steepness, relative freeboard and relative wave height. An empirical formula from the experimental data was proposed to predict the mean wave overtopping volumes.

Safety Verification through Repeated Dose 90-Day Oral Toxicity Test of Schisandra Fruit Extract Powder(SFEP) (오미자추출물(SFEP)의 90일 반복경구투여 독성 시험을 통한 안전성 검증)

  • Objectives : This study conducted a repeated dose 90-day oral toxicity test in order to up-cycling Schisandra fruit extract powder(SFEP) using discarded Schisandra chinensis by-products and evaluated the NOAEL of SFEP. Methods : SD-rats were orally administered SFEP at concentrations of 0, 62.5, 125, and 250 mg/kg once daily for 90 days. Body weights and clinical signs were observed during the administration period. After completion of the experiment, the experimental animals were autopsied to observe necropsy findings and organ weights changes, and hematological parameters and blood chemistry values were measured. Results : During the SFEP administration period, clinical signs such as salivation, wounds, and erosion were sporadically observed in 1 to 2 animals. In the SFEP 250 mg/kg administered group, weights of the liver and thyroid gland significantly increased compared to the control group, but no significant changes were observed in organ weights according to body weights. As a result of measuring hematological parameters and blood chemistry values, a decrease in RDW, T-BIL, and TBA, and an increase in TP, ALB, and Ca were observed due to SFEP administration. However, these changes following SFEP administration were accidental and not dose-dependent. Additionally, no correlation was found between gender and other parameters. Conclusions : Therefore, the NOAEL of SFEP was confirmed to be 250 mg/kg.

Effect of Hwanggeumjackyak-tang (HJT) on the DSS-induced ulcerative colitis mouse model : including changes in intestinal metabolites (황금작약탕이 DSS로 유발된 궤양성 대장염 생쥐 모델에 미치는 영향 : 장내 대사물질 변화를 포함하여)

  • Background : To investigate the effect of Hwanggeumjackyak-tang (HJT) on Dextran sulfate sodium (DSS) induced ulcerative colitis. Methods : The experimental animals were divided into three groups; group 1, normal group(Normal); group 2, DSS-induced colitis and untreated group(UT+DSS); group 3, DSS-induced colitis and HJT 200 mg-treated group(HJT200+DSS). We evaluated cytotoxicity after HJT administration and confirmed the anti-inflammatory effect by histological changes in the intestine and genetic analysis of mucosal cells after HJT administration for each group. In addition, microbiological weapons and metabolites in faeces were examined, and the correlation between gut microbiome and metabolites was also investigated. Result : HJT was not observed to be cytotoxic, even at relatively high concentrations, and was effective in protecting the barrier and preventing intestinal inflammation by suppressing the increase in mucus secretion and the expression of inflammatory factors in mucosal cells. HJT treatment affected the increase in the amount and diversity of the gut microbiome in faeces and the increase in metabolites thought to be involved in alleviating inflammation in the gut. Conclusion : This study demonstrates the therapeutic potential of HJT in ulcerative colitis. Further studies should be carried out to confirm our findings.

Derivation of Coherent Reflection Coefficient at Mid and Low Frequency for a Rough Surface (불규칙 경계면에 대한 중저주파수 간섭 반사 계수 유도)

  • When we apply a propagation model to the ocean with boundaries, we can calculate reflected wave using reflection coefficient suggested by Rayleigh assuming the boundaries are flat. But boundaries in ocean such as sea surface and sea bottom have an irregular rough surface. To calculate the reflection loss for an irregular boundary, it is needed to compute the coherent reflection coefficient based on an experimental formula or scattering theory. In this article, we derive the coherent reflection coefficients for a fluid-fluid interface using perturbation theory, Kirchhoff approximation and small-slope approximation respectively. Based on each formula, we can calculate coherent reflection coefficients for a rough sea surface or sea bottom, and then compare them to the Rayleigh reflection coefficient to analyze the reflection loss for a random rough surface. In general, the coherent reflection coefficient based on small-slope approximation has a wide valid region. Comparing it with the coherent reflection coefficients derived from the Kirchhoff approximation and perturbation theory, we discuss a valid region of them.

Fatigue Life Evaluation of Welded Joints by a Strain-life Approach Using Hardness and Tensile Strength

  • To evaluate the fatigue lifetime of structures, it is necessary to identify the values of parameters through tests. From the viewpoint of time and cost it is difficult for engineers to get the necessary data through tests. In this study, we surveyed literature and proposed a procedure to identify the fatigue parameters expressed with the Brinell hardness and elastic modulus. After obtaining stress concentration factors by finite element analysis, we calculated fatigue notch factors using Peterson's formula. Taking into account the welding residual stress, which was also obtained by finite element analysis, we evaluated the fatigue lifetime of four kinds of welded joints using the proposed approach. The estimated results are in a good agreement with the experimental results.

  • This paper presents three methods for ship hull form improvement on the basis of wave pattern analysis. These methods are primarily based on the assumption that a linear relation exists between the small difference of hull sections and the difference of the wave amplitude functions. The improved hull form of ship is made by superposing a thin hull form on that of a parent model. The first method use the simplification of framelines and Michell's Theory to the formula of wave resistance for superposed thin ship. The second method use tent functions and Michell's Theory to that. And the third is experimental method.
