Cryoprotective Properties of Exopolysaccharide (P-21653) Produced by the Antarctic Bacterium, Pseudoalteromonas arctica KOPRI 21653

  • Twenty-five bacterial strains that secrete mucous materials were isolated from sediment obtained from King George Island, Antarctica. Seven of these strains proved capable of producing cryoprotective exopolysaccharides. The strain KOPRI 21653 was selected for the further study of an anti-ice-nucleating polysaccharide (ANP), which originated from a polar region. KOPRI 21653 was identified as Pseudoalteromonas arctica as the result of 16S rRNA analysis. The exopolysaccharide, P-21653, was purified completely from the KOPRI 21653 cell culture via column chromatography and protease treatment. The principal sugar components of P-21653 were determined to be galactose and glucose, at a ratio of 1:1.5, via GC-MS analysis. The cryoprotective activity of P-21653 was characterized via an E. coli viability test. In the presence of 0.1% (w/v) P-21653, the survival ratio of E. coli cells was as high as 82.6% over three repeated freeze-thaw cycles. The survival ratio decreased drastically to 71.5 and 48.1 %, respectively, in five and seven repeated cycle conditions; however, the survival ratios were greater over three (96.6-92.1%) to seven (100.5-91.6%) freeze-thaw cycles in the presence of 0.5 and 1.0% (w/v) P-21653. In addition, at much lower concentrations (0.1-1.0%), P-21653 resulted in survival ratios (83.1-98.4%) similar to those of two commercially available cryoprotectants ($V_{EG}$ plus X-1000, 92.9% and $V_{M3}$, 95.3%), which were utilized at the recommended concentrations (90%). The biochemical characteristics of exopolysaccharide P-21653 reflect that this compound may be developed as a useful cryoprotectant for use in medical applications and in the food industry.

막걸리 유래 미생물의 활용을 위한 연구 동향 (Application of Functional Microbial Strains Isolated from Traditional Rice Wine in Korea)

  • 우리나라는 예전부터 전통적인 방법으로 누룩을 제조하고, 이를 발효제로 막걸리를 만들어 왔다. 막걸리는 여과 또는 살균 과정없이 다양한 미생물을 살아 있는 상태로 섭취하기 때문에 영양학적으로나 기능적인 측면에서 가치가 높다. 최근 많은 연구에서 막걸리로부터 미생물을 분리동정하고 다양한 기능성에 대해 스크리닝한 결과, 높은 프로바이오틱스 활성과 다양한 스펙트럼의 항균활성을 가진 균주들이 선별되었다. 특히 일부 유산균들은 GABA와 EPS 등의 기능성 물질을 생성하기도 했다. 또한, 일부 유산균과 효모의 경우 각각 bacteriocin 및 killer toxin을 통해 항균활성을 나타내는 것으로 여겨진다. 이러한 막걸리 유래 기능성 미생물과 그 대사산물은 기능성 식품 소재나 안전한 식품첨가물 및 다양한 산업분야에서 활용될 수 있을 것이다.

Isolation and Characterization of Mucous Exopolysaccharide (EPS) Produced by Vibrio furnissii Strain VB0S3

  • Marine bacterial strains were isolated trom coastal regions of Goa and screened for the strains that produce the highest amount of mucous expolysaccharide (EPS). Our screening resulted in the identification of the strain Vibrio furnissii VB0S3 (hereafter called VB0S3), as it produced the highest EPS in batch cultures during the late logarithmic growth phase. The isolate was identified as VB0S3 based on morphological and biochemical properties. Growth and EPS production were studied in mineral salts medium supplemented with NaCl (1.5%) and glucose (0.2%). The exopolymer was recovered from the culture supernatant by using three volumes of cold ethanol precipitation and dialysis procedure. Chemical analyses of EPS revealed that it is primarily composed of neutral sugars, uronic acids, and proteins. Fourier-transform infrared (FT-IR) spectroscopy revealed the presence of carboxyl, hydroxyl, and amide groups, which correspond to a typical heteropolymeric polysaccharide, and the EPS also possessed good emulsification activity. The gas chromatographic analysis of an alditol-acetate derivatized sample of EPS revealed that it was mainly composed of galactose and glucose. Minor components found were mannose, rhamnose, fucose, ribose, arabinose, and xylose. EPS was readily isolated from culture supernatants, which suggests that the EPS was a slime-like exopolysaccharide. This is the first report of exopolysaccharide characterization that describes the isolation and characterization of an EPS expressed by Vibrio surnissii strain VB0S3. The results of the study contribute significantly and go a long way towards an understanding of the correlation between growth and EPS production, chemical composition, and industrial applications of the exopolysaccharide in environmental biotechnology and bioremediation.

Influence of Agitation Intensity and Aeration Rate on Production of Antioxidative Exopolysaccharides from Submerged Mycelial Culture of Ganoderma resinaceum

  • The present study investigated the influence of the aeration rate and agitation intensity on the production of the mycelial biomass and antioxidative exopolysaccharide (EPS) in Ganoderma resinaceum. In submerged cultures with varying agitation speeds and aeration rates in a stirred-tank reactor, the maximum mycelial biomass and maximum EPS concentration were achieved at 50 rpm and 300 rpm, respectively. Under varying aeration rates, the highest amount of mycelial biomass (18.1 g/l) was accumulated at the lowest aeration rate (0.5 vvm) and the maximum EPS production (3.0 g/l) obtained at 1.0 vvm. A compositional analysis revealed that the five different EPSs were protein-bound heteropolysaccharides, consisting of 87.17-89.22% carbohydrates and 10.78-12.83% proteins. The culture conditions had a striking affect on the carbohydrate composition of the EPS, resulting in different antioxidative activities. All the EPSs showed strong scavenging activities against superoxide and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, whereas no clear trend in antioxidative activity was observed against hydroxyl radicals and lipid peroxides. Although the precise reason for this difference is still unclear, the high glucose moiety of EPS is probably linked to its broad spectrum of antioxidative activity.

적조 살상 해양 미생물 Hahella chejuensis의 유전체 구조 (Lessons from the Sea : Genome Sequence of an Algicidal Marine Bacterium Hahella chehuensis)

  • Harmful algal blooms (HABs or red tides), caused by uncontrolled proliferation of marine phytoplankton, impose a severe environmental problem and occasionally threaten even public health. We sequenced the genome of an EPS-producing marine bacterium Hahella chejuensis that produces a red pigment with the lytic activity against red-tide dinoflagellates at parts per billion level. H. chejuensis is the first sequenced species among algicidal bacteria as well as in the order Oceanospirillales. Sequence analysis indicated a distant relationship to the Pseudomonas group. Its 7.2-megabase genome encodes basic metabolic functions and a large number of proteins involved in regulation or transport. One of the prominent features of the H. chejuensis genome is a multitude of genes of functional equivalence or of possible foreign origin. A significant proportion (${\sim}23%$) of the genome appears to be of foreign origin, i.e. genomic islands, which encode genes for biosynthesis of exopolysaccharides, toxins, polyketides or non-ribosomal peptides, iron utilization, motility, type III protein secretion and pigment production. Molecular structure of the algicidal pigment was determined to be prodigiosin by LC-ESI-MS/MS and NMR analyses. The genomics-based research on H. chejuensis opens a new possibility for controlling algal blooms by exploiting biotic interactions in the natural environment and provides a model in marine bioprospecting through genome research.

Functional Characterization of an Exopolysaccharide Produced by Bacillus sonorensis MJM60135 Isolated from Ganjang

  • The present study focused on the production, characterization, and in vitro prebiotic evaluation of an exopolysaccharides (EPS) from Bacillus sonorensis MJM60135 isolated from ganjang (fermented soy sauce). Strain MJM60135 showed the highest production ($8.4{\pm}0.8g/l$) of EPSs compared with other isolates that were screened for EPS production based on ropy culture morphology. Furthermore, MJM60135 was cultured in 5 L of medium and the EPS was extracted by ethanol precipitation. The emulsification activity of the EPS was higher in toluene than in o-xylene. Fourier transform infrared spectroscopy analysis showed the presence of hydroxyl and carboxyl groups and glycosidic linkages. The isolated EPS contained mannose and glucose, as observed by thin-layer chromatography analysis of the EPS hydrolysate. Lactic acid bacteria (LAB) and pathogenic E. coli K99 and Salmonella enterica serovar Typhimurium were tested for their growth utilizing the EPS from B. sonorensis MJM60135 as the sole carbon source for its possible use as a prebiotic. All the tested LAB exhibited growth in the EPS-supplied medium compared with glucose as carbon source, whereas the pathogenic strains did not grow in the EPS-supplied medium. These findings indicate that the EPS from B. sonorensis MJM60135 has potential application in the bioremediation of hydrocarbons and could also be used as a prebiotic.

Zoogloea ramigera l15SLR의 다당류 생산에 영향을 미치는 인자 (Factors Affecting on the Production of Exopolysaccharides from Zoogloea ramigera l15SLR.)

  • Zoogloea ramigera l15SLR로부터 zooglan다당류의 생산을 위한 최적조건에 관한 연구를 수행하였다 탄소원으로 2.5%의 glucose와 lactose를 이용하였을 때 각각 10.7, 10.5 g/L의 다당류를 생산하였으며 , galactose, white sugar, brown sugar의 경우는 각각 6.4, 6.7, 8.7 g/L의 다당류 생산 을 보였다. 2.5% glucose가 첨가된 제한배지에서 종균 배양액(TSB 배지)을 7ml 이상 접종함으로서 종균 1 ml를 접종 하석 생산된 다당류(2.37 gt) 보다 약 4배의 높은 다당류 생산(10.28 g/L)을 보였다. 종균 배양액을 5 ml이상 접종시에 배양 36시간 이후부터 다당류를 생산하였으며 48시간 배양시에 10g/L이상의 다당류 생산이 가능하였다. 탄소원으로서 또한, 종균 배양액의 흡광도가 1.0~l.7일 경우에 10g/L 이상의 다당류를 생산하였으며, 이 범위를 벗어나는 경우에는 다당류 생산이 저해되었다. 배양액에 rifampicin첨가는 12 g/L이상의 다당류 생산을 보였으며, 다당류 분자량은 1.367$\times$106(g/mol)로서 rifampicin을 가하지 경우 1.711$\times$106(g/mol)와 약간의 차이를 보였다.

  • Demand for a rice cake, a popular traditional food in Korea, is rising, but its industrial-scale production is extremely difficult due to its short shelf-life caused by starch retrogradation and microbial spoilage. By means of the sourdough fermentation technique, we attempt to develop rice cakes with a longer shelf-life. Heterofermentative lactic acid bacteria (Weissella koreensis HO20, Weissella kimchii HO22) isolated from kimchi were used to ferment wet-milled rice flour for their abilities to produce exopolysaccharides and to inhibit the microbial spoilage of rice cakes. After 24 hr of fermentation at $25^{\circ}C$, viable cell counts in rice dough increased from $10^6$ CFU/g to $10^8$ CFU/g and total titratable acidity increased from 0.05% to 0.20%, whereas pH decreased from 6.5 to 5.1. Fermented rice flour showed significantly lower peak, trough, and final viscosities as well as breakdown and setback viscosities measured by rapid viscoanalyzer. Both lactic acid bacteria showed in vitro antifungal activity against Penicillium crustosum isolated from rice cakes. The antifungal activity remained constant after the treatments with heat, proteinase K and trypsin, but fell significantly by increase of pH. Rice cakes made of fermented rice flour were found to retard mycelial growth of P. crustosum. The degree of retrogradation as measured by the hardness of the rice cake was significantly reduced by the use of fermented rice flour. The results suggest that use of fermented rice flour has a beneficial role in retarding starch retrogradation and in preventing fungal growth, hence extending the shelf-life of rice cakes.

Zoogloea Ramigera 115SLR로부터 다당류 생합성에 관여하는 유전자의 분리 및 염기서열 결정 (Cloning and Sequencing of a Gene Involved in the Biosynthesis of Exopolysaccharide in Zoogloea Ramigera 115SLR)

  • Zoogloea ramigera 115SLR로부터 다당류 생합성에 관여하는 유전자를 분리하기 위해서 균주의 genomic DNA로부터 제조된 gene bank로부터 plasmid pLEX3이 얻어졌다. 이로부터 재조합된 5.0 kb DNA fragment를 포함하는 plasmid pLEX10은 다당류의 형태를 변환시키는 유전자를 포함하고 있으며, 이중에서 upstream 영역에 해당하는 1.7 kb DNA fragment가 분리되었다. 1.7 kb DNA 염기서열의 결과로부터 단백질을 인지 할 수 있는 2개의 ORF가 존재하였으며, 50 kDa 단백질을 인지 할 수 있는 ORF3은 X. campestris의 다당류 생합성 유전자들인 gumC와 R. meliloti의 exoP와 아미노산의 동질성을 나타내었다. ORF4는 N-terminal 영역이 결여된 단백질을 인지하며, Thermotoga maritime의 다당류 export에 관여하는 단백질과 동질성을 보였다. Z. ramigera 115SLR and Z. ramigera 115SLR/pLEX10은 각각 slime또는 capsule 형태의 다당류를 생합성하며 이들로부터 생합성된 다당류양은 각각 0.26% (w/v) and 0.16% (w/v)였다.

Activation and immobilization of phenol-degrading bacteria on oil palm residues for enhancing phenols degradation in treated palm oil mill effluent

  • The presence of phenols in treated palm oil mill effluent (POME) is an environmental concern due to their phytotoxicity and antimicrobial activity. In this study, phenol-degrading bacteria, Methylobacterium sp. NP3 and Acinetobacter sp. PK1 were immobilized on oil palm empty fruit bunches (EFBs) for removal of phenols in the treated POME. The bacterial exopolysaccharides (EPS) were responsible for cell adhesion to the EFBs during the immobilization process. These immobilized bacteria could effectively remove up to 5,000 mg/L phenol in a carbon free mineral medium (CFMM) with a greater degradation efficiency and rate than that with suspended bacteria. To increase the efficiency of the immobilized bacteria, three approaches, namely activation, acclimation, and combined activation and acclimation were applied. The most convenient and efficient strategy was found when the immobilized bacteria were activated in a CFMM containing phenol for 24 h before biotreatment of the treated POME. These activated immobilized bacteria were able to remove about 63.4% of 33 mg/L phenols in the treated POME, while non-activated and/or acclimated immobilized bacteria could degrade only 35.0%. The activated immobilized bacteria could be effectively reused for at least ten application cycles and stored for 4 weeks at $4^{\circ}C$ with the similar activities. In addition, the utilization of the abundant EFBs gives value-added to the palm oil mill wastes and is environmentally friendly thus making it is attractive for practical application.
