Studies on the Antimutagenicity of extract from Barley (Hordeum vulgare) (보리(Hordeum vulgare) 추출물의 항변이원성)

  • This study was performed to determine the effects of antimutagenicity from Barley (Hordeum vulgare). In Salmonella typhimurium reversion assay (In vitro test), the extract of Barley (Hordeum vulgare) inhibited mutagenic activity of 4-NQO and Trp-p-1 with 59 mix. in Salmonella typhimurium TA98 and TA100. In Micronucleus test (In vivo test), the methanol extract of Barley (Hordeum vulgare) inhibited micronucleus formation in bone marrow by cyclophosphamide. The $\beta$-glucan of Barley (Hordeum vulgare) showed inhibitory effects of 59-77% in mutagenic activity of 4-NQO by Salmonella typhimurium TA100. The mutagenicity of Trp-p-1 with S9 mix. by Salmonella typhimurium TA98 showed inhibitory effects of 24-56%. The methanl extract (M) was fractionated with ether (MI), ethylacetate (M2), buthanol (M3) and water (M4). The Antimutagenicity of Trp-p-1 with 59 mix. by Salmonella typhimurium TA98 in Barley fraction showed the following: methanol extract (99.58%)>ether fraction (98.05%)>buthanol fraction (56.90%)>water fraction (56.72%)>ethyl acetate fraction (28.72%). Among them, ether fraction in TA 98 showed strong antimutagenicity effects (85.56%, 98.05%) against mutation induced by 4-NQO and Trp-p-1. As concentration of the methanol extract increased (1.25~5 g/kg/10 cc), micronucleus formation in bone marrow by chemical mutagen (CP) showed inhibitory effects of 50% (p< 0.05).

An Antifungal Compound Against Phytophthora capsici Produced by Streptomyces sp. 3D3 (Streptomyces sp. 3D3 균주가 생산하는 항고추역병성 항생물질)

  • During the screening for the antifungal compounds against Phytophthora capsici causing phytophthora blight of red pepper, we isolated a strong active compound, bafilomycin $C_1$, produced by strain 3D3. The producing organism was identified as Streptomyces sp. based on taxonomic studies. The antifungal compound was purified from culture broth by HP-20 column chromatography, ethylacetate extraction, silica gel column chromatography and HPLC, and was identified as bafilomycin $C_1$ by color reaction, UV and $^{1}H$-NMR spectral data analysis. Bafilomycin $C_1$ showed strong antifungal activity against various phytopathogenic fungi.

  • This study was conducted to investigate the effect of acorn extracts on the antioxidative enzyme system. Male Sprague-Dawley rats(110$\pm$10g) were fed on containing normal and high fat diets. They were orally administrated (0.02g/100g B.W) of acorn ethylacetate-extract and water-extract at the same time once a day, respectively. Net weight gain and feed efficiency ratio were increased in high fat diet groups and decreased by acorn extracts administration. The effect of acorn extracts on hepatic glutathione peroxidase(GSH-Px), glutathione S-transferase(GST) and catalase activities did not show significancy in normal fat diet groups. GST and catalase activities and lipid peroxidation content(LPO) were significantly increased in high fat diet groups and this increment were decreased by acorn extracts administration. However GSH-Px activity and GSH content were decreased in high fat diet groups but increased by acorn extracts administration. The activities of lactate dehydrogenase(LDH), alkaline phosphatase(ALP) and aminotransferase in serum were significantly increased in high fat diet group but these increment reduced in acorn extracts administration groups. These results indicate that acorn extracts could improve the liver function and prevent the metabolic diseases by hyperlipidemia.

  • Alnus japonica Steud (Betulaceae) has long been used as a Korean traditional medicine for gastric disorders, hepatitis, and fatty liver. As a part of our study on the identification of secondary metabolites of naturally occurring bioactive compounds, we isolated 1,7-bis(4-hydroxyphenyl)-3,5-heptanediol(1), 5-hydroxy-1,7bis(4-hydroxyphenyl)-3-heptanone(2), 5,3'-dihydroxy-7,4'-dimethoxyflavone(3) and 3,5,7,3',4'-pentahydroxyflavone(4) from the dichloromethane and ethylacetate-soluble fractions of Alnus japonica Steud. These compounds showed significant antioxidant activity in a concentration-dependent manner. The $IC_{50}$ values of compounds 1, 2, 3 and 4 were 30.1, 37.4, 20.2 and 13.7 ${\mu}g/mL$, respectively, through the scavenging capacity of 1,1-diphenyl-2-picrylhydrazyl radical assay.

Antimutagenic and Cytotoxic Effects of Ethanol Extract from the Inonotus obliquus (차가버섯 분획물의 항돌연변이 활성 및 암세포 성장억제효과)

  • This study was performed to determine the antimutagenic and cytotoxic effects of ethanol extract, ethylacetate fraction, water fraction and water soluble and insoluble polysaccharides I and II isolated from Inonotus obliquus using Ames test and SRB assay. Each sample itself did not show mutagenic effect. Among samples, the water unsoluble polysaccharides II in the presence of 50 $\mu\textrm{g}$/plate showed the strongest antimutagenic effect with over 90% against MNNG, 4NQO, B(a)P and Trp-P-l. However, ethyl acetate fraction (1 mg/mL) which had 90.8%, 94.3% and 83.5% inhibitory effect against on MCF-7, A549, AGS showed the strong cytotoxic effect compared to other samples.

Screening of Antimicrobial Substances against Brachyspira hyodysenteriae (Brachyspira hyodysenteriae에 대한 항균물질 탐색)

  • The extracts from 70 different plant natural products were used to investigate the antimicrobial activities against Brachyspira hyodrsenteriae, the causative agent of swine dysentery. Among them, the extracts from Terminalia chehula exhibited the highest antimicrobial activities against B. hyodysenteriae. It showed the antimicrobial activity against B. hyodysensteriae mostly in parts of its shell. Minimum inhibitory concentration (MIC) of the water extracts of T chehula was 12.5 mg/ml. After organic solvent extractions, the ethylacetate extracts exhibited higher antimicrobial activities against B. hyodysenteriae, and their antimicroactivities were similar to that of the tannin. In conclusion, these findings suggest that the extracts from T chebula may be used as animal feed additives for their antimicrobial activities against B. hyodysenteriae.

Studies on the Efficacies of Water Extract of Propolis (프로폴리스 물추출물의 약효연구)

  • Propolis is a lipophilic, natural product prepared by mixing the exudates collected from various plants by honeybees with beeswax for the purpose of using to seal hive walls and to strengthen the borders of combs. Because of its versatile bioactivities, propolis has been attracting many investigators'interest. But the pharmacological studies on propolis has, to date, been exclusively performed for an alcohol extract, there is few information of water extract. Therefore, in this study, we investigated the various effects of water extract of Chinese propolis. The water extract of propolis and its fractions of organic solvents showed strong antioxidative activities, especially ether and ethylacetate fractions, and reduced the lipid peroxidation of rat liver in viro. Additionally the ether fraction of propolis (10 mg/ml) inhibited the activity of hyaluronidase by 50%. In vivo, the water extract of propolis considerably decreased s-GOT, s-GPT and s-LDH activities which represent for the hepatotoxicity induced by $CCl_4$ in rats, and prolonged the MST (Medium revival tinge) and ILS (Increasing in MST over control) by 18% in mice which inoculated with sarcoma 180 ascites cells. These results suggest that the water extract of propolis has various bioactivities as well as the alcohol extract.

  • In the course of phytochemical studies of Rumex acetosella L. (Polygonaceae), the MeOH extract of the whole plants was fractionated with $CH_2Cl_2$ and $H_2O$, and the $CH_2Cl_2$ layer was fractionated again with 90% MeOH and hexane. Whereas $H_2O$ layer was fractionated with EtOAc and then with n-BuOH again. By repeated column chromatography, compound 1 was isolated from the 90% MeOH fraction, compounds 1, 2, 3, 4 and 5 were from the ethylacetate fraction, and compound 5 was from the n--BuOH fraction. On the basis of physico-chemical, spectroscopic evidences and in comparison with authentic samples, the structures of them were confirmed to emodin, citreorosein, $chrysophanol-8-O-{\beta}-D-glucopyranoside$, luteolin and $luteolin-7-O {\beta}-D-glucopyranoside$.

  • To elucidate antidiabetic activity of Lycii Fructus. The extract of the Lycii Fructus was subjected to bioactivity-guided fractionation. The extract of the Lycii Fructus was partitioned suceesively with hexane, dichloromethane, ethylacetate and butanol. The butanol fraction, which is most abundant in the Lycii Fructus was subjected to silica gel column chromatography and resulted in seven fractions namely Fr. I - Fr. VII. Among tested several subfractions, Fr IV. Showed the highest antidiabetic activity in the streptozotocin-induced model rat.

  • Aurantii Fructus has been used in traditional medicine as a digestant, expectorant, and in the treatment of diarrhea, anal prolapse. For the quality control of this crude drug, hesperidin was isolated from the ethylacetate fraction of Citrus aurantium (Rutaceae) and identified by the spectroscopic evidences. A quantitative analysis of hesperidin using HPLC method exhibited that the average contents of hesperidin were $2.13{\pm}1.16%$ in 57 samples collected throughout the various regions of Korea.
