Antimicrobial Activities of Chestnut Flower Extracts(Castanea crenata) (밤꽃 추출물의 항균성)

  • Ethyl acetate, methanol, water extract and their fractions from chestnut flower(Castancea crenata) were tested for antimicrobial activities. Yields of prebloomed chestnut flower extracts were 13.84, 12.90 and 1.82% in methanol, water and ethyl acetate, and those of the postbloomed were 13.12, 11.75 and 1.18%, respectively. Methanol extract from the chestnut flower was fractionated by solvents using hexane, chloroform, ethyl acetate, n-butanol and water, yields of those fractions were 0.16, 0.08, 1.94, 4.75 and 6.91% in the prebloomed, and were 0.90, 0.13, 1.40, 3.42 and 7.18% in the postbloomed. In the solvent extracts of water, ethyl acetate and methanol, methanol extract showed the most effective antimicrobial activity, antimicrobial activity of ethyl acetate fraction of methanol extract was stronger than others. Minimum inhibitory concentration of ethyl acetate fractions from the prebloomed showed 100, 140, 100 and 90ppm against Staphylococcus aureus, Streptococcus mutans, Echerichia coli and Salmonella typhimurium, in fractions from the postbloomed were 140, 140, 100 and 150ppm, respectively. Growth of all the strains was completely inhibited to 30 hours in a 150ppm concentration. E. coli sells treated with ethyl acetate fraction was collapsed severely.

The Effects of Fractions of Houttuynia cordata THUNB on the Accumulation of Cadmium and Induction of Metallothionein in Rats(IV) (어성초 분획이 흰쥐 장기내 카드뮴 축적과 Metallothionein 생성에 미치는 영향(VI))

  • This study was conducted to investigate the antitoxic effects of Houttuynia cordata THUNB with chloroform and ethyl acetate fractions. The results were as follows: 1. Detoxication effects of chloroform and ethyl acetate fractions of Houttuynia cordata THUNB were increased in proportion to the dosages. Detoxication effects of ethyl acetate fraction of Houttuynia cordata THUNB were higher than chloroform fraction of Houttuynia cordata THUNB's results and detoxication effects in the kidneys were higher than results for the liver. 2. Metallothionein concentrations in the liver were higher than concentrations in the kidneys, and ethyl acetate fraction of Houttuynia cordata THUNB was higher than chloroform fraction of Houttuynia cordata THUNB in induction of metallothionein. 3. After the administration of chloroform and ethyl acetate fractions of Houttuynia cordata THUNB, body weights increased in proportion to chloroform and ethyl acetate fraction s dosage of Houttuynia cordata THUNB but changes of body weight dminished after 3 weeks. From the above results, this study suggests that chloroform and ethyl acetate fraction of Houttuynia cordata THUNB increased metallothionein induction to cadmium intoxication in rat s kidney and liver and decreased the toxicity of cadmium in rats.

Antioxidant Activity of Ethyl acetate Fraction of Mallotus japonicus Twigs in Caenorhabditis elegans (예덕나무 가지의 Ethyl Acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과)

  • Caenorhabditis elegans model system was used to investigate the antioxidant activity of methanol extract of Mallotus japonicus twigs. The ethyl acetate soluble fraction of the M. japonicus methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using a transgenic strain. As a result, the ethyl acetate fraction increased SOD and catalase activity, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

  • A series of bimetallic Cu-Zn/$SiO_2$ catalysts were prepared via thermal decomposition of the as-synthesized $CuZn(OH)_4(H_2SiO_3)_2{\cdot}nH_2O$ hydroxides precursors. This highly dispersed Cu-solid base catalyst is extremely effective for hydrogenation of ethyl acetate to ethanol. The reduction and oxidation features of the precursors prepared by coprecipitation method and catalysts were extensively investigated by TGA, XRD, TPR and $N_2$-adsorption techniques. Catalytic activity by ethyl acetate hydrogenation of reaction temperatures between 120 and $300^{\circ}C$, different catalyst calcination and reduction temperatures, different Cu/Zn loadings have been examined extensively. The relation between the performance for hydrogenation of ethyl acetate and the structure of the Cu-solid base catalysts with Zn loading were discussed. The detected conversion of ethyl acetate reached 81.6% with a 93.8% selectivity of ethanol. This investigation of the Cu-Zn/$SiO_2$ catalyst provides a recently proposed pathway for ethyl acetate hydrogenation reaction to produce ethanol over Cu-solid base catalysts.

The Antioxidative Effects and Isolation and Characterization of the Extracts from Morus alba L. (뽕잎 추출물의 항산화 효과와 항산화 성분 분리 및 동정에 관한 연구)

  • This study identified the formula of the antioxidant substance separated from the ethyl acetate and the butanol extract and tested the antioxidant properties with the electron donating ability(EDA). Each phase with the fractionated methanol extract from mulberry leaf was screened in advance for the antioxidant substance with EDA. As the result, activity appeared in the ethyl acetate and butanol phase and the antioxidant component was separated. As the consequence, 2 components from the ethyl acetate phase and 1 from the butanol phase were separated, among which the structures of the components from ethyl acetate were determined by wogonin and linarin, whereas the structure of the component from the butanol phase was determined by pectolinarin. In the screening of antioxidant activity by the scavenging effect of the DPPH radical, the wogonin and linarin components from ethyl acetate phase showed more powerful antioxidant property than the component from butanol. The results from this study indicate that the chemical compound separated from the ethyl acetate extract has more powerful antioxidant property than the one separated from the butanol extract. The components separated from the ethyl acetate extract were wogonin and linarin, which are flavonoids, whereas the component from butanol was pectolinarin. Therefore, this study suggested that the feasibility of mulberry leaf as a functional food additive and its value as a natural antioxidant is very high.

Antioxidant Activity of Ethyl acetate Fraction of Oat in Caenorhabditis elegans (귀리 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과)

  • Oat, the seeds of Avena sativa L. (Gramineae), is an important dietary staple for people in many countries. Previous studies reported that A. sativa had various pharmacological effects such as anti-inflammatory, antitumor, neurotonic, and antispasmodic activities. In this study, Caenorhabditis elegans model system was used to investigate the antioxidant activity of methanol extract of oat. The ethyl acetate soluble fraction of the oat methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using GFP-expressing transgenic worm. As a result, the ethyl acetate fraction increased SOD and catalase activities, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity compared to the control.

Selection of Organic Carbon (Different Form of Acetate Compounds and Concentration) for Cultivation of Anabena under Mixotrophic Cultivation Mode (혼합영양 배양조건에서의 Anabena 배양을 위한 유기탄소(acetate 종류 및 농도) 선정 연구)

  • The main objective of this study was to evaluate the effects of acetate on the cultivation of anabena under mixotrophic condition. Four different types of acetates were used for the anebena cultivation. Among them, ethyl acetate was found to be the most effective and the growth rates linearly increased as the amount of ethyl acetate increased. When 40 mM of ethyl acetate was used, the highest values of specific growth rate of $0.979day^{-1}$ and maximum biomass productivity of $0.293g\;L^{-1}\;d^{-1}$ were obtained. On the contrary, input of acetic acid and butyl acetate inhibited the growth of anabena. For aeration tests, 0.54 vvm was optimum for anabena cultivation. For a semi-continuous cultivation test, ethyl acetate was used after 0.54 vvm test was finished. Then, test continued under 0.54 vvm and 40 mM of ethyl acetate. Lower specific growth rate and maximum biomass productivity were obtained compared to those from batch cultivation tests. However, the greatest maximum concentration of 5.91 g/L was obtained during the semi-continuous cultivation test.

Esterification of Alcohols with Organic Acids during Distilled Spirit Distillation (증류식 소주 증류중 유기산에 의한 에스테르화)

  • The esterification of alcohols with acetic acid, lactic acid and citric acid was carried out in batch during the second distillation. Effect of various parameters, e.g. pH of reactant base spirit, temperature of distillation, time of distillation were studied. The parameter of distillation temperature was modified by atmospheric distillation and pressure distillation. The pressure(1.9 atm.) distillation was used in order to react and distill at high temperature. Compared with the base spirit concentration, ethyl acetate, ethyl lactate and iso-amyl acetate in esterification distillate increased by 2,890%, 6,410% and 52%, respectively. Major factors of the esterification with organic acids in distilled spirit making were molecular weight of the organic acid and pH of reactant base spirit.
