Inhibitory Action of Tsunokaori Tangor Peel on the Lipopolysaccharide-Induced Inflammatory Response in RAW 264.7 Macrophage Cells

  • We evaluated the effects of extracts of Tsunokaori tangor peel on lipopolysaccharide (LPS)-induced nitric oxide (NO) and prostaglandin $E_2\;(PGE_2)$ in RAW 264.7 cells. The ethyl acetate fraction of Tsunokaori tangor peel (EA-TTP) markedly inhibited the production of NO and $PGE_2$ in LPS-stimulated RAW 264.7 cells. Consistent with these findings, the expression levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) proteins were down-regulated in a dose-dependent manner. Additionally, EA-TTP decreased the expression iNOS mRNA but not COX-2 mRNA. To determine the upstream signaling mechanism for the down-regulation of LPS-induced iNOS expression, we investigated the effect of EA-TTP on the degradation and re-synthesis of $I{\kappa}B{\alpha}$. EA-TTP dose-dependently delayed $I{\kappa}B{\alpha}$ degradation and increased $I{\kappa}B{\alpha}$ re-appearance following degradation, suggesting this as the mechanism by which EA-TTP suppressed iNOS gene expression. The EA-TTP also dose-dependently reduced the expression of the cellular stress-response protein heme oxygenase-1, and inhibited the LPS-induced sustained activation of extracellar signal-regulated kinase (ERK).

Evaluation of Lipid Peroxidation Inhibition and Nitrogen Oxide Scavenging Activity from Peel of Gardenia jasminoides Ellis Fructus Extracted by Various Solvents (치자(Gardenia jasminoides Ellis fructus) 껍질 용매 별 추출물의 지질과산화 저해 및 질소산화물 소거능)

  • The aim of this study was to investigate the bioactivity and antioxidant activity of peel from Gardenia jasminoides Ellis fructus (GJE). We were separated into GJE peel. After that, we determined anthocyanin. GJE peel were extracted by 70% methanol, distilled water (DW) and ethyl acetate (EA) three solvents. To investigate by the solvent extract of total phenol content and value as a functional food ingredient of GJE peel through nitrogen oxide scavenging activity, antioxidant activity, reducing power and lipid peroxidation inhibition were performed. Solvent extract bioactivity of increasing concentrations (0.2, 0.4, 0.6 mg/mL) were significantly increased (p<0.05). GJE peel extracts showed lower activity than positive control (ascorbic acid, BHA, trolox). The total phenol contents of GJE peel extracts were highest in EA extract. However, the order of total phenol content of the solvent in the GJE peel and the results of analysis of various physiological activities were inconsistent. Considering the extraction yield and various physiological activities, it is expected to be effective when extracted from 70% methanol and DW extract. The results suggest that GJE peel is highly expected to be useful as a functional foods and natural antioxidant.

Inhibitory Effect of Methanolic Extract from Adenophorae Radix on Melanogenesis (사삼 메탄올 추출물의 멜라닌생성 억제효과)

  • Effects of methanolic extract from Adenophorae Radix (AR) on melanogenesis were investigated in mouse melanoma B16F10 cells. The methanol extract of AR was partitioned into Hexane, Ethyl acetate (EA), Butanol, H₂O, and exogenously added to the culture medium for 72 hours at the concentration of 10, 50, 100 and 200 ㎍/㎖. Of the four partitions, Hexane and EA partion of AR reduced tyrosinase activity, which is the key enzyme for a melanogenesis, as well as melanin contents. But the EA partition was less toxic for B16F10 cells and has more efficient melanin-reducing effect than the former. In addition, the EA partition dramatically lightened the color of cell pellet and significantly decreased the level of tyrosinase protein expression. In these results, EA partition of AR reduced melanin synthesis of B16F10 mouse melanoma cells by down regulating the tyrosinase activity and tyrosinase protein expression. Therefore, it is anticipated that AR is a candidate for an efficient whitening agent which supresses melanogenesis.

Antifungal Activity of Bacillus sp. BCNU 2003 against the Human Pathogenic Fungi (인체 병원성 진균에 대한 Bacillus sp. BCNU 2003의 항진균 효과)

  • An antifungal antibiotic-producing strain, BCNU 2003, was isolated from forest soil in Korea. The morphological and physiological characters, and 16S rRNA sequences analysis of strain BCNU 2003 identified this strain as Bacillus genus. The Bacillus sp. BCNU 2003 showed strong antifungal activities against Aspergillus niger, Trichophyton mentagrophytes and Trichophyton rubrum with inhibition ranging from 62.05 to 63.49% by using dual culture technique. Bacillus sp. BCNU 2003 produced a maximum level of antifungal substances under aerobic incubation at 28oC and pH 6.5-7.2 for 6 days in LB broth. Ethyl acetate extract of the cultured broth showed strong antifungal activity and a broad antifungal spectrum against various pathogenic fungi. The minimum inhibitory concentration (MIC) values for its active extracts ranged between 0.0625 mg/ml and 1 mg/ml. In addition, Bacillus sp. BCNU 2003 was determined to have the ability to produce enzymes such as amylase, protease, gelatinase and catalase.

Effects of Blend Composition and Compatibilizer on the Mechanical Properties of Polypropylene/Acrylonitrile-Butadiene-Styrene Blends (블렌드 조성과 상용화제가 폴리프로필렌/ABS 블렌드의 기계적 물성에 미치는 영향)

  • Polymer blends containing polypropylene (PP) with compatibilizers were prepared using twin screw extruder. Physical properties were investigated using universal test machine (UTM) and Izod impact tester. In the PP/acrylonitrilebutadiene-styrene (ABS) blends, mechanical strength was increased with the addition of PP-g-styrene acryloritrile (PP-g-SAN) compatibilizer, and the ductility was increased with the addition of ethylene-ethyl acrylate-maleic anhydride (E-EAMAH-g-SAN) compatibilizer. For the PP/ABS/ polycarbonate (PC)/Nylon-6,6 blends, impact strength was increased with the addition of ethylene glycidylmethacrylate (E-GMA compatibilizer) up to 0.5 phr. In the case of the PP/ABS/PC/Nylon-6,6/poly(methyl methacrylate) (PMMA)/poly(oxymethylene) (POM)/poly(vinyl acetate) (PVC)/poly(butylene terephthalate) (PBT) blends, mechanical properties were increased by the complex compatibilizing effects of PP-g-SAN, E-EA-MAH-g-SAN and E-GMA, respectively.

Antioxidant Activities and Lipid Peroxidation Inhibition Ability of Gardenia jasminoides Ellis Fructus Seed Extracts (치자(Gardenia jasminoides Ellis fructus) 종자 추출물의 항산화 활성 및 지질과산화 저해능)

  • The purpose of this study was to investigate the bioactivity of extracts from the seeds of Gardenia jasminoides Ellis fructus (GJE) found in Namhae, Korea. Extraction was performed using three solvents, 70% methanol, Distilled Water (DW), and Ethyl Acetate (EA). We determined the total phenol and phytic acid contents of the extracts to evaluate their nitrogen oxide scavenging activity, antioxidant activity, reducing power, and lipid peroxidation inhibition ability. The phytic acid content of GJE was found to be 1.157 mg PAE (Phytic Acid Equivalent) /g DW. The yields of the three extraction processes were as follows: DW, 36.61%; 70% methanol, 30.10%; and EA, 20.40%. The physiological activities of the extract solvents increased significantly with increasing concentrations (0.2, 0.4, and 0.6 mg/mL) (p<0.05), but were lower than those of ascorbic acid, BHA, and trolox. Total phenol content was the highest in the 70% methanol extract, followed by DW and EA extracts. Further, nitrogen oxide scavenging activity and antioxidant activity were the highest for the 70% methanol extract followed by DW and EA extracts. Based on these results, the bioactivities of the 70% methanol and DW extracts of GJ seeds were excellent. These extracts can be used as natural antioxidants.

In vitro anti-oxidant, anti-wrinkle and whitening effect of solvent fraction from takli-san (탁리산 용매 분획물의 in vitro 항산화, 주름 억제 및 미백 효과)

  • In this study, the activity of Takli-san for anti-oxidation, anti-wrinkle and whitening effect was verified and its applicability as a cosmetic material was confirmed. 1-1-diphenyl-2-picryl-hydrazyl radical scavenge, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenge, elastase and tyrosinase inhibitory activity were investigated by solvent fraction, and ethyl acetate fraction of water extract from Takli-san (TW-EA) showed the highest inhibitory activity. Western blot was performed to confirm anti-wrinkle and whitening effect in the cells, and it was investigeted that the TW-EA inhibition effect was superior to that of the positive contol used for the comparison test. Through the results of the experiments, the applicability as a cosmetic material of Takli-san was verified.

UV-induced skin damage improvement effects of solvent fraction from Maekmoondong-tang (맥문동탕 용매 분획물의 UV에 의한 피부손상 개선 효능평가)

  • In this study, we verified the 1-1-diphenyl-2-picryl-hydrazyl radical scavenge, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenge, elastase, tyrosinase inhibitory effect by using the solvent fractions of Maekmoondong-tang hot water extract. As a result, the ethyl acetate fraction (MW-EA) showed the highest inhibitory activity. In cell-based assays, MW-EA treatment confirmed a 34% ($100{\mu}g/mL$) efficacy in reactive oxygen species inhibitory activity, and at the same concentration, MMPs showed more than 50% inhibition and tyrosinase inhibited 25% ($50{\mu}g/mL$). Therefore Maekmoondong-tang is considered high development potential as a material to improve the skin.

Whitening Effects of Solvent Fractions Isolated from Vitex rotundifolia (만형자 용매 분획물의 미백 개선 효과)

  • Recently many effort focused to understand the mechanical insights of melanogenesis to develop the agent for hyper-pigmentation. So this study was performed to investigate the depigmentation of Vitex rotundifolia. With B16F10 mouse melanoma cell, we have seen inhibition of the tyrosinase, MITF, TRP-1, TRP-2, and melanin synthesis, which eventually were dose dependently decreased by Vitex rotundifolia. Specially, Vitex rotundifolia decreased the protein levels of tyrosinase and TRP-1. In conclusion, Vitex rotundifolia showed the whitening activity in all the experiments mentioned above and we expect that it can be used for preventing melanin synthesis.
