Anti-oxidant effect of ethanol extract from mixture including Caryophylli Flos, Aucklandiae Radix and Angelicae Dahuricae Radix (정향 및 목향과 백지를 혼합한 한약재 에탄올 추출물의 항산화 효능)

  • Objectives : Herbal medicinal mixture (JMB) are consisted of Caryophylli Flos, Aucklandiae Radix and Angelicae Dahuricae Radix. Each of herbal medicines has studied on anti-oxidant effect. So this study was conducted to investigate efficacy and potency of JMB on anti-oxidation. Methods : The JMB was extracted at room temperature by 80% ethanol. And total polyphenol contents, total flavonoid contents in JMB ethanol extract were determined by colorimetric method. Also, DPPH, ABTS free radical scavenging capacity and reducing power of JMB ethanol extract were measured at 100, 500, 1000, $5000{\mu}g/m{\ell}$ concentrations by spectrometric assay. Positive control was used BHA (butylated hydroxyanisole). Results : The total polyphenol contents and total flavonoid contents of the extract were 55.38 mg/TAEg, 513.72 mg/RUEg, respectively. Also, DPPH free radical scavenging capacity and reducing power of JMB ethanol extract in treated concentrations (100, 500, 1000, $5000{\mu}g/m{\ell}$) increased dose dependently. In particular, DPPH free radical scavenging capacity of JMB ethanol extract at 500, 1000, $5000{\mu}g/m{\ell}$ was similar to positive control (BHA) at high concentration (50, $100{\mu}g/m{\ell}$). ABTS free radical scavenging capacity of JMB ethanol extract at 500, 1000, 5000 ug/ml was similar to BHA at high concentration (50, $100{\mu}g/m{\ell}$). Also, reducing power was showed that JMB ethanol extract at $5000{\mu}g/m{\ell}$ was similar to BHA at high concentration (50, $100{\mu}g/m{\ell}$). Conclusions : These results suggest that JMB ethanol extract has effects to scavenge free radicals. Therefore, JMB has potential and applicable benefits for development of materials and products to have anti-oxidation functions.

Physiological activity of the extract from Dolwoe (Gynostemma pentaphyllum Makino) leaves tea by different ethanol concentrations (에탄올 농도에 따른 돌외 잎 차 추출물의 생리활성)

  • The objective of this study was to compare the effects of different concentration of ethanol extraction on the total phenol and flavonoid contents and physiological activities. The total phenol content of the extracts ranged from 35.54 to 71.52 mg GAE/g. An increase in the ethanol concentration of the solvent led to an increase in the phenol content, with the highest content being found in the 80 and 99.5% ethanol extract. The same trend was observed for flavonoid content. DPPH, nitric oxide, superoxide, hydroxyl radical scavenging activity and TEAC, FRAP and ORAC were measured by antioxidant assay. Radical scavenging activity of aqueous ethanol extracts was better than that of water and 99.5% ethanol extracts. TEAC and FRAP were highly dependent on ethanol concentration and ORAC showed high activity in 40 ~ 80% ethanol extract. Antioxidant activity of Dolwoe leaves tea showed different results among the assay systems. In most experiments, the activities of water and 99.5% ethanol extracts was relatively low. ${\alpha}$-Glucosidase inhibitory activity and microorganism inhibitory activity were highest in the 80% and 99.5% ethanol extracts. Therefore, it was considered that extraction with 80% ethanol was appropriate when considering the antioxidative and physiological activities of Dolwoe leaves tea. Based on these results, it can be used as a basic data for the development of food of Dolwoe leaves tea.

Potential of Red Ginseng Marc for Ethanol Production as a Fermentation Medium (에탄올 발효 배지로서 홍삼박의 활용)

  • The potential of the red ginseng marc (RGM) for the production of bio-ethanol using enzymatic hydrolysis and fermentation without any additional nutrients was investigated. Reducing sugar content in RGM treated with Viscozyme and Flavourzyme was 101.1 g/L and was much higher than that (7.2 g/L) in intact RGM. When enzymatically hydrolyzed red ginseng marc (ERGM) was fermented with commercially available dry yeast at $25^{\circ}C$ for 7 days, the final ethanol concentration reached 29.3 g/L with ethanol yield at 0.274 g of ethanol per 1 g of solubilized total sugar. Ethanol concentration and ethanol yield of ERGM were drastically increased over 1000% and 50%, respectively than those of RGM.

Development of a Fluorescent Sensor Based on Resazurin and Hydrotalcite for the Determination of Ethanol in Alcoholic Beverages

  • In this study, a fluorescent ethanol sensor is developed to determine the ethanol concentration in the liquid phase. The sensor is developed using a complex of resazurin (RA)/resorufin (RO) and a hydrotalcite (HT) catalyst in a sol-gel matrix of methyltrimethoxysilane (MTMS) to produce a fluorescent ethanol-sensing membrane (RA/RO*HT membrane). The operation mechanism of the RA/RO*HT membrane is based on (i) the oxidation of ethanol to acetaldehyde and (ii) the reduction of RA to RO, through electron flows followed by EtOH ↔ HT ↔ RA/RO ↔ EtOH interactions. These possible redox reactions can lead to an increased fluorescence intensity of the RA/RO*HT membrane as the ethanol concentration increases. The RA/RO*HT membrane shows a linear detection range of 1-20 vol.% EtOH with limit of detection (LOD) of 0.178%. Additionally, the RA/RO*HT membrane has high sensitivity and accuracy for determining the alcohol content in several Korean alcoholic beverages.

Effect of Fermentation Temperature on the Production of high content Alcohol (고농도 알코올 생성을 위한 온도의 영향)

  • The effect of fermentation temperature on the production of high content alcohol has been investigated with high substrate concentration. The maximum specific growth rate, ${\mu}max\;was\;0.461hr^{-1}\;at\;35^{\circ}C$ which was the highest, whereas the maximum biomass concentration waas 8.7g/l at $25^{\circ}C$, at the growth rate lower than at $35^{\circ}C$. Approximately 140g/l of ethanol was produced in the temperature range of 20 to $25^{\circ}C$ with nearly complete comsumption of the substrate. Extended fermentation time has been required at lower temperatures, however, for the maximum values of biomass concentration and alcohol content, hence higher ethanol productivity, as the temperature was elevated to $40^{\circ}C$. The viability of yeasts was greatly improved by lowering the fermentation temperature down to $25^{\circ}C$ and also extended survival of the cells has been observed at lower fermentation temperatures, although the ethanol concentration of both waas higher.

Beauty Food Activities of Isolated Phenolic Compounds from Ulmus pumila (유근피(Ulmus pumila)로부터 분리한 페놀성물질의 미용식품활성)

  • Phenolic compounds of $17.9{\pm}1.0mg/g$ were extracted from Ulmus pumila with 70% ethanol. The elastase inhibitory activity related with forming wrinkle was shown an excellent wrinkle improvement effect in 70% ethanol extracts as 55.5-69.5% at phenolic concentration of $50-200{\mu}g/mL$. The tyrosinase inhibitory activity related with skin-whitening was 24% in 70% ethanol extracts at phenolic concentration of $200{\mu}g/mL$. The astringent activity of 70% ethanol extracts was shown activity of 71% at phenolic concentration of $200{\mu}g/mL$ therefore it is judged that there is a high effect on pores reduction of the skin. The hyaluronidase inhibitory activity of U. pumila extracts was confirmed anti-inflammation effect of 80% at phenolic concentration of $50{\mu}g/mL$. Antimicrobial activity of U. pumila water extracts was shown each 8.7, 10.0, 11.1 and 11.8 mm clear zones on Propionebacterium acnes at phenolic concentration of $50-200{\mu}g/mL$. The stability of the multi-functional cosmetic (lotion) added U. pumila extracts was very stable for 28 days without changing of pH and viscosity also it's stable on temperature and sun lights. As the concentration of extracts was increased, the color of lotion was getting dark, but the sensory evaluation was high at score of 8.5.

  • This study provided the evidence activity for biological of phenolic compounds from Trachelospermum asiaticum var. intermedium nakaier as a beauty food. The contents of phenolic compounds in water and 70% ethanol extracts were 16.8 mg/g and 38.1 mg/g, respectively. DPPH free radical scavenging activities of water and ethanol extracts at $100{\mu}g/mL$ of phenolic concentration were 80.9% and 83.1%, respectively. ABTS radical decolorization activity was 95.0% in water extracts and 95.8% in ethanol extracts at $200{\mu}g/mL$ phenolic concentration. Antioxidant protection factor (PF) was determined to 2.43 and 2.45 PF in water extracts and ethanol extracts at $100{\mu}g/mL$ phenolic concentration. TBARs of water and ethanol extracts were 89.9% and 89.3% each at $200{\mu}g/mL$ phenolic concentration. The inhibitory effect of ethanol extracts at $200{\mu}g/mL$ phenolic concentration on xanthine oxidase was 50.5%. The inhibitory activity of a-glucosidase was 92.6% in ethanol extracts at $200{\mu}g/mL$ phenolic concentration. As a result, this study will provide valuable information as a functional material with antioxidant activity, inhibitory activities of xanthin oxidase and ${\alpha}-glucosidase$.

  • The surface of porous alumina membrane was modified with silane coupling agent in order to enhance hydrophobicity. The contact angle of water to the surface-modified alumina membrane was greater than $90^{\circ}$. The surface-modified membrane was tested in vapor permeation for the concentration of aqueous ethanol. With the increase of ethanol concentration in the feed, permeation flux increased due to the greater affinity of ethanol with surface-modified alumina membrane than that of water. The experimental results showed that the permeation rate of surface-modified alumina membrane was 15~1000 times greater than that of polymer membranes.

Extract Yields and Saponin Contents of Red Ginseng Extracts prepared with various Concentrations of Ethanol (홍삼의 알콜 농도별 추출 엑스의 수율과 사포닌 함량)

  • Red ginseng extracts were prepared with various concentrations of ethanol. Extract yields were examined and saponins in the extracts were identified and determined by TLC and HPLC, respectively. Yields of the extracts, $19.7{\sim]50.3%$, were the highest in water extract and showed significant decrease with the increase of ethanol concentration used for extraction. Contrary to the extract yields, saponin yields from red ginseng were conspicuously increased with the increase of ethanol concentration and were $3.47{\sim}5.13%$ of crude saponins and $1.28{\sim}1.93%$ of six major ginsenosides. Saponin contents in the red ginseng extracts were $6.9{\sim}24.2%$, of crude saponin and $2.57{\sim}9.22%$, of six major ginsenosides.

  • The purpose of this study was to investigate the nitrite scavenging activity of the ethanol and water extracts obtained from Hizikia fusiforme and Sargassum fulvelum using sodium nitrite under different pH conditions. The nitrite scavenging activity was appeared in all the extracts examined and it showed the highest value at pH 1.2 The ethanol extract of Hizikia fusiforme provided higher activity than the water extract of that. Ohterwise, the nitrite scavenging activity of Sargassum fulvelum ethanol extract was similar to that Sargassum fulvelum water extract. There was significant difference among concentration of extracts Hizikia fusiforme and Sargassum fulvelum. Also, nitrite scavenging activity of the extracts Hizikia fusiforme and Sargassum fulvelum decreased with higher pH condition. In addition, the activity was increaed with higher concentration of extracts Hizikia fusiforme and Sargassum fulvelum in the course of reaction time 30 minute to 3 hour.
