The objective of this study was to examine the effect of in vitro culture media on embryonic development of in vitro-matured (IVM) oocytes after parthenogenetic activation (PA) in pigs. Immature pig oocytes were matured in TCM-199 supplemented with porcine follicular fluid, cysteine, pyruvate, EGF, insulin, and hormones for the first 22 h and then further cultured in hormone-free medium for an additional 22~26 h. IVM oocytes were activated by electric pulses and cultured in porcine zygote medium-3 (PZM-3) and North Carolina State University-23 supplemented with essential and non-essential amino acids (NCSU-23aa). These media were further modified by supplementing 2.77 mM myo-inositol, 0.34 mM trisodium citrate, and $10{\mu}M$${\beta}$-mercaptoethanol (designated as mPZM-3 and mNCSU-23aa, respectively). Culture of PA embryos in mPZM-3 significantly increased development to the blastocyst stage than culture in NCSU-23aa (36.2% vs. 24.8%, p<0.05). Modified PZM-3 showed a significantly higher blastocyst formation than NCSU-23aa in both groups of embryos that were activated at 44 h and 48 h of IVM (51.0% vs. 35.5% and 49.0% vs. 34.2% in oocytes activated at 44 h and 48 h of IVM, respectively). Irrespective of the follicle diameter where oocytes were collected, embryonic development to the blastocyst stage was increased (p<0.05) by the culture in mPZM-3 compared to culture in NCSU-23aa (25.9% vs. 34.2% and 32.9% vs. 44.8% in embryos derived from small and medium size follicles, respectively). Our results demonstrated that culture media had significant effect on preimplantation development PA embryos and that mPZM-3 was superior to mNCSU-23 in supporting development to the blastocyst stage in pigs. This beneficial effect of mPZM-3 on embryonic development was not impaired by other factors such as time of oocyte activation and origin of immature oocytes (small and medium size follicles).
Maize grain quality can be improved by raising lysine content, which is an essential amino acid present in insufficient quantities in normal maize. Maize varieties with such modifications are known as quality protein maize (QPM). To date, no Korean maize cultivars contain high amounts of lysine. To introduce quality protein maize to Korean cultivars, we crossed QPM CIMMYT maize lines (CML) with KS140, an elite inbred line used as a parent of several cultivars such as 'Gangdaok' and 'Pyeonganok'. We analyzed the phenotypic characteristics of F1 plants as well as the protein contents, amino acids, and fatty acids profiles of the self-pollinated seeds of the F1 hybrids, and evaluated the feasibility of CML as a source of QPM. Days to anthesis of the CML ranged from 78 to 90 days after planting (DAP), whereas a range of 81~87 was recorded for F1 hybrids. The average days to anthesis was 85 for KS140, CML, and the F1 hybrids. The protein content of the CML was measured to be between 9.1 and 12.1%, with the highest and lowest values being recorded in CML153 and 191, respectively. The F1 hybrids had protein contents of 9.1~11.1%, and the highest content was observed in KS141/CML188. The fatty acids profiles were very similar across all analyzed maize samples, and linoleic acid (C18:2) composed the greatest proportion. Glutamic acid made up the largest proportion of amino acids in all maize samples. Lysine composition was highest in CML155 (6.92% of all amino acids), with an average composition of 4.83% across the CML. In contrast, KS140 showed a lysine content of 2.51%. In F1 hybrids, the average lysine composition was 3.46%, and KS140/CML164 (4.18%) and KS140/CML163 (4.99%) contained more lysine than either parent. Taken together, these results indicate that CML could become promising QPM sources to improve grain quality in Korean maize cultivars.
The present study provides nutritional characteristics of body composition based on the different collected locations and body sizes in wild eels, Anguilla japonica. Eels were collected in Kyungho river, near the Sachun bay (Gyeongnam) and Inchun river, near Gochang, Jeonnam. Eels were divided by size into the following groups: less than 100 g (Gochang: 75.8$\pm$17.6 g, Sachun: 89.3$\pm$6.42 g) and more than 200 g group (Gochang: 215$\pm$8.95 g, Sachun: 236$\pm$11.7 g). The moisture content of individuals in Gochang area (Gw) was higher than that in Sacheon area (Sw). In the results of proximate analysis, the ranges of moisture, crude protein, crude fat, and crude ash were 63∼72, 16∼20, 6.1∼15.4 and 2.2∼2.8%, respectively. Moisture and crude fat were significantly different (P<0.05) based on the captured locations. In fatty acid composition, the ranges of saturates, monoenes, polyenes, and n-3 PUFA content were 38.0∼48.0, 51.5∼58.5, 1.95∼4.12 and 0.68∼1.53%, respectively. It was significantly different (P<0.05) based on the captured locations. In the amino acid composition, the ranges of essential and non-essential amino acids were 7.11∼12.5 and 8.34∼ 12.9%, respectively. These values were not significantly different (P>0.05) based on the captured locations, however these were significant different (P<0.05) based on their body weight. Therefore, these results indicated that the different body composition depends on the different captured locations and body sizes. Furthermore, it is necessary to examine more varied locations and body sizes for their body composition in eels.

Physiological activities of 70% methanol, fermented ethanol and hot-water extracts of Pleurotus eryngii were investigated. Free radical scavenging activities of P. eryngii extracts were determined according to the elimination of DPPH radicals. Their nitrite scavenging activity and total polyphenol content were also determined. Amino acid analysis showed that phenylalanine (Phe) and glutamic acid (Glu) are most abundant essential and non-essential amino acids in the analyzed extracts. The hot-water extract of ASI 2394 represented the highest antioxidant activity with the DPPH radical scavenging rate value of $40.97{\pm}1.65%$. ASI 2820 displayed the superior capacity to eliminate nitrate regardless of extraction solvents. The hot-water extract of ASI 2887 had the highest content of polyphenol. Our results showed that P. eryngii is well qualified as a functional food.
A drug delivery system (DDS) based on nanoparticles has been used as a mediator to improve the efficacy of a drug by controlling the amount of drug released and delivering it to a target place. Chitosan, which is non-toxic and biodegradable, has good biocompatibility and excellent adsorption, so it can be used as a drug delivery vehicle. Valine, the essential amino acids, helps muscle growth and tissue recovery, and along with other amino acids. It lowers blood sugar levels and increases growth hormone production. In this study, Valine was adsorbed on magnetic chitosan which is capable of drug absorption, and Fe3O4-Valine CNPs was prepared through cross-linking with TPP (Tripolyphosphate). And then absorption and release trends of valine were investigated with the Fe3O4-Valine CNPs. Fe3O4, which has relatively high stability, is used to make the drug carrier magnetic so that the drug can be delivered to a target place. At optimal conditions, the absorption and release tendency of Fe3O4-Valine CNP was confirmed by analyzing by UV-Vis through the Ninhydrin test which is the color reaction of amino acids and by measuring the size of the particles, it was confirmed that it is suitable as a drug carrier.
The nutritional compositions and antioxidative activities of the Kimcheon lowbush and Pyungtaek highbush blueberries cultivated in South Korea were investigated. The approximate compositions, pH, and soluble-solid and mineral contents of P, Ca, Mg, Na, and Zn were determined. Both blueberry cultivars had mainly fructose and glucose as free sugars while disaccharides such as sucrose, maltose, and lactose were not detected in both. Oleic, linoleic, and linolenic fatty acids were the major fatty acids in both types of blueberries. The total of 16 free amino acids, including seven essential and nine non-essential amino acids, were higher in the Pyungtaek highbush blueberry. Among the amino acids, arginine was especially much higher in the Pyungtaek highbush blueberry. The anthocyanin, resveratrol, and polyphenol compounds, which are the important biologically active compounds in blueberries, were found. The anthocyanin contents of the Kimcheon lowbush and Pyungtaek highbush blueberries were 22.0 and 18.1 mg/100 g, respectively; the resveratrol contents by HPLC, 0.12 and 0.11 mg/100 g; and the total polyphenol contents, 141.3 and 138.4 mg/100 g. The electron-donating ability determined based on the DPPH radical scavenging activity was increased in a concentration-dependent manner, and it was higher than that of the Pyungtaek highbush blueberries, which implies that it is highly correlated with the higher amounts of total polyphenol, anthocyanin, and resveratrol in it. In conclusion, the two varieties of Korean blueberries can be suggested as potential sources of high-value-added functional foods.

This work was carried out to evaluate the feces and ileal digestibility of canola meal and wheat bran in ducks. Ninety Pekin ducks (6-week-old, average 3.4 kg) were divided into three treatments (6 replications/treatment, 5 birds/replication) by feedstuffs (basal diet, canola meal, and wheat bran) and were fed on these diets in individual metabolic cages for one week. After one week, the samples for analysis were collected from feces and ileum. On comparison of treatments and collected sites, the feces digestibility of basal diet was the highest among treatments on chemical compositions except crude protein and crude fat and ileal digestibility of canola meal was the lowest (P<0.01). On comparison of treatments, the digestibility of basal diet was higher and that of wheat bran was lower when compared to other treatments (P<0.05). The feces digestibility was lower than the ileal digestibility in crude protein (P<0.05) and crude fat (P<0.01). The digestibility of amino acid was the highest at feces of control group among treatments (P<0.01). No significant difference was detected in terms of digestibility of isoleucine, leucine and phenylalanine among essential amino acids, and alanine, glycine and tyrosine among non-essential amino acids between control and wheat bran, and wheat bran and canola meal. However amino acid digestibility was higher in control group when compared to canola meal group (P<0.05). Other amino acid digestibilities were not significantly differ by treatments. These results may provide the basic data on amino acid digestibility of ducks to evaluate feedstuffs and to formulate optimum diet for commercial Pekin duck.

To investigate the effects of dietary substituent of corn distiller's dries grains with solubles (CDDGS) on the amino acid digestibility of broiler diet, twenty four Ross broilers (6-wk-old) were used in this study. Basal diet (CP 19%, ME 3,150 kcal/kg) was formulated on corn-soybean meal and two levels of CDDGS (6 and 12%) were supplemented to basal diet. CDDDS levels (0, 6 and 12%) and 4 different sites of chicken intestines (distal end of jejunum, the middle part of the ileum, the distal end of ileum and the distal end of the rectum) were evaluated factorial as $3{\times}4$ three arrangement. Statistical analysis did not show any significant interaction between dietary treatments and the sites of the intestines in amino acid digestibility (P>0.05). There was no significant difference among the sites of chicken intestines in the digestibility (P<0.05). The digestibility of lysine (essential amino acid) and glycine (non-essential amino acid) decreased at increasing supplementation of CDDGS (P<0.05). There was no significant difference on the digestibility of other amino acids studied. In conclusion, the result of this work showed that CDDGS can use as alternative resources in broiler diets.
The study was conducted to obtain the basic information on increasing aromatic degrees by shading and organic matter application on Condonopsis lanceolata. The result were as followings : Fresh root wt. in shading 55 % treatment was increased about twices, whereas fresh root weight in non-shading was decreased. But growth of ground parts such as vine length, leaf width, and leaf number was promoted under non-shading treatment. Macroelements such as K, Ca, and Mg, were increased in non-shading and higher organic matter application. The components such as crude protein, fiber, and ash were increased in the shading 55% treatment compared with non-shading. In the sixteen amino acids, arginine contents were the highest in non-shading and organic matter 30M/T. Recovery yield of essential oil was 0.005% in non-shading. The highest was organic matter 30M/T in the non-shading as 0.007%. Major aromatic constituents were 11 kinds of aliphatic alcohols such as I-hexanol, cis-3-hexenol, and trans-2-hexanal. As a result, major aromatic degree was higher in shading 55%, also increased in the much organic matter application. It was considered to be applied dense shading 55% and much organic matters to produce Condonopsis lanceolata plants which have high aroma and good quality.
Supplementation of maturation medium with additional granulosa cells has beneficial effect on in vitro maturation of bovine follicular oocytes and their subsequent cleavage and development in vitro. However, maturation system using granulosa cells have some disadvantages that collection of granulosa cells is cumbersome and metabolic activity of the cells is variable according to ovarian cycle or follicular size. We hypothesized that bovine immsture oocytes matured without granulosa cell coculture can fertilize and develop normally if the medium volume per oocyte is reduced during in vitro maturation. Immature oocytes were matured for 24 hours in a TCM199 containing 10% fetal calf serum, anterior pitultary hormone (0.02 AU /ml Antrinⓡ) and estradiol with or without granulosa cells in vitro. In Group 1, 35 to 40 oocytes were matured in a well of 4-well plastic dish containing 500 $\mu$l of maturation medium and granulosa cells, and 9 to 10 oocytes were matured in a 50-$\mu$l drop of maturation medium without granulosa cells in Group 2. After maturation, oocytes were coincubated with sperm for 30 hours in a modified Tyrode's medium (IVF). Inseminated oocytes were cultured in a microdrop (30 $\mu$l) of a synthetic oviduct fluld medium (SOFM) containing BSA, Minimum Essential Medium essential and non-essential amino acids for 9 days. As a preliminary experiment, we investigated the beneficial effect of granulosa cells during maturation on subsequent cleavage and development using the same type of culturedishes (4-well dish). Granulosa cells could not increase embryo cleavage after fertilization but significantly improved (p<0.05) embryo development to expanding blastocyst (Table1 and 2). In Group 1, 68 and 80% of inseminated oocytes have cleaved at 30 hours and 2 days after IVF, respectively, which is similar (p>0.05) to the result of Group 2 (69% at 30 hours and 78% at 2 days after IVF). The oocytes in Group 2 showed 21 and 11% of developmental rates to expanding and hatching blastocysts, respectively, which was not significantly different (p>0.05) from those (20 and 10%, respectively) of oocytes in Group 1. In conclusion, it has been clarified that a microdrop culture system without granulosa cells for in vitro maturation can support bovine embryonic development to blastocyst in vitro as readily as a granulosa cell coculture system.
