Fermentation Process Development of Recombinant Hansenula polymorpha for Gamma-Linolenic Acid Production

  • Development of the strain and the fermentation process of Hansenula polymorpha was implemented for the production of ${\gamma}$-linolenic acid ($GLA,\;C18:3{\Delta}^{6,9,12}$), an n-6 polyunsaturated fatty acid (PUFA) that has been reported to possess a number of health benefits. The mutated ${\Delta}^6$-desaturase (S213A) gene of Mucor rouxii was expressed in H. polymorpha under the control of the methanol oxidase (MOX) promoter. Without the utilization of methanol, a high-cell-density culture of the yeast recombinant carrying the ${\Delta}^6$-desaturase gene was then achieved by fed-batch fermentation under glycerol-limited conditions. As a result, high levels of the ${\Delta}^6$-desaturated products, octadecadienoic acid ($C18:2{\Delta}^{6,9}$), GLA, and stearidonic acid ($C18:4{\Delta}^{6,9,12,15}$), were accumulated under the derepression conditions. The GLA production was also optimized by adjusting the specific growth rate. The results show that the specific growth rate affected both the lipid content and the fatty acid composition of the GLA-producing recombinant. Among the various specific growth rates tested, the highest GLA concentration of 697 mg/l was obtained in the culture with a specific growth rate of 0.08 /h. Interestingly, the fatty acid profile of the yeast recombinant bearing the Mucor ${\Delta}^6$-desaturase gene was similar to that of blackcurrant oil, with both containing similar proportions of n-3 and n-6 essential fatty acids.

Meat Quality and Physicochemical Trait Assessments of Berkshire and Commercial 3-way Crossbred Pigs

  • In this study, we compared qualities and physiochemical traits of meat from Berkshire (black color) pigs with those of meat from 3-way Landrace (white color) × Yorkshire (white color) × Duroc (red color) crossbred pigs (LYD). Meat quality characteristics, including pH, color, drip loss, cooking loss, and free amino acid, fatty acid, vitamin, and mineral contents of longissimus dorsi muscles, were compared. Meat from Berkshire pigs had deeper meat color (redness), higher pH, and lower drip loss and cooking loss than meat from LYD pigs. Moreover, meat from Berkshire pigs had higher levels of phosphoserine, aspartic acid, threonine, serine, asparagine, α-aminoadipic acid, valine, methionine, isoleucine, leucine, tyrosine, histidine, tryptophan, and carnosine and lower levels of glutamic acid, glycine, alanine, and ammonia than did meat from LYD pigs. The fatty acids oleic acid, docosahexaenoic acid (DHA), and monounsaturated fatty acids (MUFA) were present in significantly higher concentrations in Berkshire muscles than they were in LYD muscles. Additionally, Berkshire muscles were significantly enriched with nucleotide components (inosine), minerals (Mg and K), and antioxidant vitamins such as ascorbic acid (C) in comparison with LYD muscles. In conclusion, our results show that in comparison with LYD meat, Berkshire meat has better meat quality traits and is a superior nutritional source of all essential amino acids, monounsaturated fatty acids, vitamin C, and minerals (Mg and K).

Changes in Body Composition of Protaetia brevitarsis seulensis Larvae Fed Sulfur and Selenium (황 및 유황 급이에 따른 흰점박이꽃무지 유충의 체성분 변화)

  • This study examined changes in the body composition of Protaetia brevitarsis larvae fed sulfur and selenium. Among the general chemical components, an increase in protein was observed in the sulfur-fed group, whereas an increase in fatty acid was found in the groups fed selenium and sulfur/selenium. The structural amino acid showed some tendency to be increased compared to the control group, but this was especially true in the sulfur-fed group. The free amino acid were increased in the groups fed selenium and sulfur/selenium, with GABA, Glu, and Pro, in particular, being increased in the group fed sulfur/selenium. Among the essential amino acids, Met, Trp, and Val tended to be increased in selenium and sulfur/selenium groups. Saturated and polyunsaturated fatty acids showed a tendency to increase in the group fed sulfur/selenium, but monounsaturated fatty acid tended to be decreased in the group. In particular, no oleic acid was detected in the sulfur/selenium group, whereas palmitic acid, palmitoleic acid, and linoleic acid were increased. From observations of these body composition changes associated with the feeding of sulfur and selenium, it is estimated that the group fed sulfur/selenium would likely undergo a pronounced change in body composition. Therefore, we suggest that it is possible to manufacture a food that has enhanced functionality by appropriately adjusting the diet of Protaetia brevitarsis larvae.

Physicochemical Compositions of Pimpinella brachycarpa (참나물의 이화학적 성분)

  • This study was carried out to analyze major chemical components of dried Pimpinella brachycarpa leaves. Proximate compositions of the dried Pimpinella brachycarpa were 8.96% moisture content, 28.73% crude protein, 2.66% lipids, 19.14% ash, and 40.50% carbohydrates. Major components of the free sugars and disaccharides were glucose and maltose. A total of 15 kinds of amino acids were isolated from Pimpinella brachycarpa. Essential amino acids accounted for 49.13% of the total amino acids and non-essential amino acids accounted for 50.87%. A major fatty acid was linolenic acid. A ratio of polyunsaturated fatty acids to saturated fatty acids (P/S ratio) was 2.18. Oxalic acid was a major organic acid. The contents of the vitamins A and E were 9.23 mg% and 0.26 mg%, respectively. Among the minerals in the dried Pimpinella brachycarpa, the content of calcium was the highest (765.13 mg%) and those of magnesium and sodium were also comparatively high (303.00 mg%, 96.21 mg%).

  • This paper was developed for production of the humanized milk, comprising similarly to the composition and characteristic of human milk. Humanized milk of superior quality can be made directly from the fresh raw milk mixed vegetable oil, corn syrup, whey powder, ${\beta}-lactose$, sugar, vitamin, ${\beta}-carotene$ and minerals showing formulation of the humanized milk at table 2. The improving effects of adding vegetable oil and corn syrup are both more reformed the chemical and physical properties of humanized milk. The former enhanced the essential fatty acid and energy source in this product, the latter has the most solving function in water and induced amount of emulsion and stabilizer. The products contain about 13% protein, 23% fat, 58.3% carbohydrate, 2% ash and ensue reasonably balance of essential amino acid, poly-unsaturated fatty acid for the requirement of infants and controlled component of the humanized milk such as human milk.

Effects of Gender on Sensory Scores, Fatty Acid and Amino Acid of Berkshire Pork (버크셔의 성별이 돈육의 관능 평가, 지방산 및 아미노산 조성에 미치는 영향)

  • This study was conducted to investigate the effects of gender on sensory scores, fatty acid and amino acid of Berkshire pork A total of 40 pigs (60 kg) were divided into 2 group (gilt and barrow) and raised up to 110 kg live weight. Pigs were conventionally slaughtered, and then chilled overnight. Pork loin (longissimus dorsi) muscle was removed from each left side and meat qualities were evaluated. The results obtained were as follows; In sensory evaluation, color, marbling score, aroma and overall acceptability of brrow groups were significantly higher than that of gilt groups (p<0.05). In fatty acid analysis, linoleic acid and arachidonic acid of gilt groups were higher than those of barrow groups, EFA (essential fatty acid) and UFA (unsaturated fatty acid) were lower in barrow groups compared to the gilt groups. In amino acid compositions, alanine, tyrosine and arginine of barrow groups were significantly higher and threonine, proline and histidine lower than those of gilt groups (p<0.05).

Fatty Acid, Amino Acid Composition and Sensory Traits of Pork from Pigs Fed Artificial Culture Medium of Wild Ginseng (산삼 배양액 급여 돈육의 지방산, 아미노산 조성 및 관능적 특성)

  • A total of 120 pigs (Berkshire) were used to investigate the effect of dietary supplementation with artificial culture medium ot wild ginseng (CMWG) on the fatty acid composition, amino acid composition and sensory characteristics or pork About $60{\pm}3kg$ pigs were randomly assigned to one of four experimental diet groups[(both sexes)${\times}$(C: commercial diet feed; T: commercial diet+1 L CMWG per day for 70 days)]. Pigs were slaughtered at approximately 110 kg live weight, and fatty acid composition, amino acid composition and sensory characteristics were measured in pork loin. The monounsaturated and saturated fatty acid contents was greater in barrow fed a diet containing CMWG than those of the gilt pork and control groups, however the polyunsaturated fatty acid composition decreased. The EAA (essential amino acid) content was lower in pigs fed diets containing CMWG than that of the control groups, whereas the SAA (amino acid with sulfide) and FRAA (fragrant amino acid) contents were higher. The EAA and FAA (amino acid in relation to flavor) contents were lower for harrow than for gilt, however the SAAA (amino acid in relation to saccarinity), SAA and FRAA the contents were higher. Regarding the sensory evaluation of fresh meat, the color of gilt pork increased with diets containing CMWG relative to the control group. Drip loss and the marbling score for pigs fed with diets containing CMWG were higher in barrow than in gilt. The flavor and overall acceptability of cooked meat from the control group was higher for barrow than for silt.

Preparation of Rockfish (Sebastes schlegelii) Feed Fortification with Mideodeok (Styela clava) Extracts (미더덕 추출물을 첨가한 조피볼락용 사료의 제조)

  • The nutritional contribution of mideodeok extracts (ME) on rockfish (Sebastes schlegelii) feed and fish muscle was investigated. Different concentrations of the ME mixed with commercial diet were fed to mature rockfish for 8 weeks. The lipid and ash contents of the formulated diets were relatively similar to the control diet, while increasing the extract concentration increased the moisture content and decreased the protein contents. Major fatty acid components (C18:1n-9, 16:0, C20:5n-3, C22:6n-3) were of comparable quantity. High presence of C18:2n-6 was attributed to soybean oil incorporated in the diets, while the essential fatty acids were within limits (0.9-1.0%). The diet fortified with 6% ME produced the highest feed efficiency, with increased protein content in the muscle as well as lipid content for both muscle and liver. Hepato- and visceral-somatic index values were elevated with increasing ME concentration Muscle fatty acid contents were mostly C18:1n-9 and C16:0, with low absorption of C18:2n-6 in both the muscle and liver. Total highly unsaturated fatty acid content was significantly reduced in the fish muscle, but the values were higher for fish fed with a ME-fortified diet. An increasing trend for eicosapentaenoic acid and docosahexaenoic acid was also observed with increased ME fortification, with liver levels of these compounds remaining within range throughout the duration of the experiment.

Effect of DHA-Rich Fish Oil on Brain Development and Learing Ability in Rats (DHA가 풍부한 어유가 새끼쥐의 뇌발달과 학습능력에 미치는 영향)

  • Effect of DHA-rich fish oil on brain development and learning ability has been studied in Sprague Dawley rats. Female rats were fed experimental diets containing either corn oil fish oil at 10%(w/w) level throughout the gestation and lactation. Corn oil was added in fish oil diet to supply essential fatty acid at 2.3% of the calories. All male pups were weaned to the same diets of dams at 21-days after birth. Plasma fatty acid composition was analyzed for dams and pups at 21-days, 28-days and 22-weeks after birth. The analysis of DNA and fatty acid profile in the brain were undertaken at birth, 3, 7, 14, 21, 28 days and 22 weeks after birth and learning ability was tested at 18-20 weeks of age. Regardless of dietary fats, arachidonic acid(AA) and docosahexaenoic acid(DHA) were the principal polyunsaturated fatty acids in the brain. Rats fed CO diet showed a continouus increase of AA content in the brain from 10.9%(at birth) to maximum 15.3% level (14-days old), while the rars fed FO diet showed 78-79% of CO group throughout the period. Rats fed FO diet showed higher incorparation of DHA from 15.2% at birth to a maximum level of 18.5% at 140days, while the rats fed CO diet showed only 7.0% incorporation of DHA at birth and a maximum level of 11.1% at 21-days. Compared to CO group, FO group showed lower ratio of chol/PL and higher content of DHA in brain microsomal membrane, resulting in better membrane fluidity. Total amount of DNA per gram of brain was reached maximum level at 21 days in both groups. This would be a period of the cell proliferation during brain development. Overall, the rats fed fish oil diet showed a higher incorporation of DHA and membrane fluidity in the brain and better learning performances (p<0.05).

Food Quality Characteristics of Instant Gruel Prepared with Peeled Krill Euphausia superba Meat (크릴(Euphausia superba) 육을 이용한 인스턴트 죽의 품질특성 평가)

  • Instant krill gruel was produced using peeled Antarctic krill Euphausia superba as a high nutritional ingredient and the food quality was investigated. The food quality of krill gruel was examined by measuring proximate composition, cholesterol, calorie, Hunter color value, total amino acids, fatty acids, fluoride, viscoelastic properties, and by sensory evaluation. The krill gruel had a moisture content of 87% and a pH of 6.65. The krill gruel contained 51 kcal/100 g, and 0.1% fat and 3.5 mg/100 g cholesterol. Its fatty acid composition exhibited high levels of unsaturated fatty acids. The levels of oleic acid and linolenic acid were high, and n-3, n-6, and n-9 fatty acid contents ranged from 1% to 6%. The total amino acid content was 2132 mg/100 g, and the levels of glutamic acid, aspartic acid, leucine, alanine, and arginine were particularly high. Essential amino acids accounted for over 30% of the total amino acids. Fluoride level in the krill gruel was 3.07 mg/kg. The viscoelastic properties of the krill gruel were determined as 6.28 Pa at shear stress of 2.51 Pa. In the recovery test, the elastic restoring force after deformation was low.
