생쥐에서 가정분진 입자로 인한 항원관련 호산구성 기도 염증과 기도 과민성의 증대 (Enhancement of Allergen-related Eosinophilic Airway Inflammation and Airway Hyperresponsiveness by House Dust Particles in Mice)

  • The number of patient with allergic asthma and atopy have increased in the cities of Korea steadily. In order to elucidate the primary factor, we investigated whether the house dust particles collected from an apartment of the middle classes has promoting effects of allergen-related airway inflammation and airway hyperresponsiveness. Mice were treated with 0.1 mL of 1 mg/mL of house dust particles suspension by intratracheal instillation once weekly for 10 weeks combined with ovalalbumin (OVA) sensitization. Intratracheal instillation of house dust particles and OVA sensitization caused an increase in the level of serum L-lactate dehydrogenase (LDH), immunoglobulun-E (IgE) and histamine, and an elevation in respiratory resistance. It also enhanced infiltration of eosinophils in the bronchoalveolar lavage fluid (BALF) of mice, IgE and eotaxin expression in blood, and T helper type 2 cell derived cytokine levels such as of interleukin (IL)-4, IL-13 and IL-5 in the BALF. However, it did not influence T helper type 1 cytokine such as interferon-gamma in the BALF. These results indicate that house dust particles elevate allergen-related airway inflammation and airway hyperresponsiveness in mice and may play an important role in the aggravation of asthma and atopy in Korea.

Dexamethasone Downregulates Expressions of 14-3-3β and γ-Isoforms in Mice with Eosinophilic Meningitis Caused by Angiostrongylus cantonensis Infection

  • Steroids are commonly used in patients with eosinophilic meningitis caused by A. cantonensis infections. The mechanism steroids act on eosinophilic meningitis remains unclear. In this mouse experiments, expressions of 14-3-3 isoform ${\beta}$ and ${\gamma}$ proteins significantly increased in the CSF 2-3 weeks after the infection, but not increasedin the dexamethasone-treated group. Expression of 14-3-3 ${\beta}$, ${\gamma}$, ${\varepsilon}$, and ${\theta}$ isoforms increased in brain meninges over the 3-week period after infection and decreased due to dexamethasone treatment. In conclusion, administration of dexamethasone in mice with eosinophilic meningitis decreased expressions of 14-3-3 isoform proteins in the CSF and in brain meninges.

Splenocyte, BMMC, eosinophil을 통해 본 단삼(丹蔘)이 천식 관련 염증세포에 미치는 영향 (Effects of Salviae miltiorrhizae on Inflammatory Cells Associated with Asthma via Splenocyte, BMMC, Eosinophil)

  • Objectives : Asthma is a chronic inflammatory disorder of the airways by many cells such as mast cells, Th2 lymphocytes and eosinophile. The present study was aimed to evaluate the effects of Salviae miltiorrhizae (SM) on T cell cytokine production, mast cells. and eosinophils, Methods : We screened 13 herbs to find compounds with potential to control Th cytokine production. using concanavalin A (con A)-activated splenocyte cultures. Con A-activated $IFN-\gamma$ and IL-4 levels in supernatants of splenocyte cultures. Bone marrow derived mast cells (BMMC) were incubated with SM and then the expressions of membrane proteins of BMMC were analyzed by fluorescence activated cell sorter (FACS). BALB/c mice sensitized to ovalbumin (OVA) were challenged with aerosolized OVA for 6 weeks. During the last weeks some mice were treated with SM. Then eosinophils in bronchoalveolar lavage fluid (BALf) were counted and pathologic changes of lung tissue were observed with hematoxylin-eosin stain. Results : SM increased $IFN-\gamma$ level on splenocyte culture significantly. but had no significant effects on expressions of ICAM-1, CD62L, integrin $a_4$. c-kit, IL-3 receptors. CD11a, or IgE receptors of BMMC. SM treatment significantly inhibited eosinophil infiltrates in BALf and peribronchial lung inflammation. Conculusions : The present data suggested that SM may have an effect on Th cytokine secretion and eosinophils associated with asthma responses. Therefore SM might be of therapeutic value in treating asthma.

배(리(梨)) 추출 Phenolic Compound 및 길경(桔梗) 행인(杏仁) 배합 투여가 Ovalbumin으로 유발된 천식 동물 모델에서 기관지 평활근 장력, 호산구 및 IL-4에 미치는 영향 (The effects of pear phenolic compound and herbal drugs on tension of the tracheal smooth muscle, eosinophil and interleukin-4 in mouse model of allergic bronchial asthma induced by ovalbumin)

  • Objectives : Oriental pear was used as treatment of asthma, control of blood pressure, diabetes in oriental medicine. The aim of this study was to observe the effects of Phenolic compound extracted from pear and herbal drugs to treat asthma. Methods : In order to study the effect of oral administration of phenolic compound extracted from pear and herbal drugs(Platycodon grandiflorum, Prunus armeniaca) on allergic asthma, mice were pre-treated by oral administration of the solution before antigen sensitization four times for 8 days. 2 days later, mice were actively sensitized with a subcutaneous injection of ovalbumin and 13 days later, they were provoked with ovalbumin aerosols. The animals were divided into four groups; Saline, orally administered saline. PC-A, orally administered Phenolic compound extracted from pear peel 10mg/kg/ml. PC-B, orally administered Phenolic compound extracted from pear peel and flesh 10mg/kg/ml. PC-C, orally administered pear 10m/kg/ml, Platycodon grandiflorum 24.4 mg/kg/ml and Prunus armeniaca 33.3 mg/kg/ml. Serum level of IgE, IL-4, cell numbers in the bronchoalveolar lavage fluid(BALF), and in vitro isometric contractile responses of the isolated tracheal smooth muscle(TSM) to acetylcholine(ACh, $0.1-1000{\mu}M$), KCl were measured. Results : Contractile responses of TSM to ACh were decreased in PC-A group at Ach 0.1, 0.3, 1 ${\mu}M$, decreased in PC-B at 0.1 ${\mu}M$ and decreased in PC-C at 0.1, 0.3, 1, 10, 30 ${\mu}M$. The maximal contractile response of TSM to KCl was decreased in PC-C group, The cell numbers of eosinophil in BALF were decreased in PC-C group, and those of macrophages in BALF were decreased in PC-A and PC-C group. Interleukin-4 in BALF was decreased in PC-A, PC-B, PC-C group. Conclusion : Based on the above results it is assumed that oral administration of phenolic compound extracted from pear and herbal drugs can help the treatment of deficiency allergic Asthma.

인삼주정추출액이 스트레스에 폭로된 동물의 생체반응에 미치는 영향 (Influence of Panax Ginseng on the Responce of Btressful Stimuli in the Experimental Animal exposed to Various Stress)

  • Three hundred gram of Korean ginseng root was extracted with 95% ethanol on a boiling water bath for about 300 hr. Evaporation of alcohol yieled 50.2g of dark brown residue which was used by dissolving 4 mg of the residue in 1 ml of physiological saline. The ginseng group and the saline group received each day 0.5 ml per 100 g body weight of ginseng extract and physiological saline, respectively. Both the ginseng and saline group with stress were exposed to positive radial acceleration (1∼29g), cold (5$^{\circ}C$, 0$^{\circ}C$ &-10$^{\circ}C$) and heat (35$^{\circ}C$) environment, and surgical stress. After termination of the last stress, the tolerance, body weight, visceral organ weight, basal metabolism rate, rectal temperature, the number of erythrocyte and leucocyte, hemoglobin level, hematocrit ratio, total serum protein content and it's fraction and the content of adrenal ascorbic acid in the experimental animal exposed to stress were measured and at the corresponding periods, the same measurements were also carried out with the ginseng and the saline groups without stress exposure (serving as control). Results obtained were as follows. 1. Administration of ginseng does depressed the decrease of the tolerance, body weight, visceral organ weight, basal metabolism rate, the number of erythrocyte, hemoglobin value, hematocrit ratio and the A/G ratio in the mice and rats exposed to various stress. 2. The change of the rectal temperature, eosinophile counts, total serum protein content and the content of adrenal ascorbic acid of ginseng group that exposured to various stress facilitates the reaction to, and accelerates the recovery from the stress. 3. Even after hypophysectomy which served the link between the central and the peripheral portion of the stress mechanism, the adrenal ascorbic acid content of ginseng group decreased significantly more than that of the saline group 30 min. after administration of ACTH, while the value approached the normal level significantly closer in the ginseng group than in the saline group 1 and 2 hr after ACTH administration. Judging from the above results, it is concluded that administration of ginseng extract tolerated the experimental animals under the environment of stressfu1 stmuli, although the ginseng has no significant influence upon the stress mechanism in the absence of stressful stimuli. The site of action of the ginseng appears to be in the peripheral portion of the stress mechanism.

한국재래오골계의 혈액상 및 혈액화학치에 관한 연구 - 1. 오골계의 혈액상에 관하여 (Studies on the Hematologic Values and Blood Chemistry Values of Korean Native Ogolgye 1. Hematologic Values for Korean Native Ogolgye)

  • 본 시험은 천연기념물 제265호인 182일령 한국재래종 오골계 20수(남 ; 10, 여 ; 10수)를 대상으로 정상혈액상을 구명하고 성별로 비교하여 유의성을 검정했던 바 다음과 같은 성적을 얻었기에 보고하는 바이다. 1. 적혈구치, 혈색소치 및 적혈구용적은 각각평균 $3.23\pm0.29\times10^6/{\mu}{\iota}$, 11.75$\pm$2.27g/100m1 및 33.41$\pm$4.85ml/100m1를 나타냈으며 이들은 공히 수컷이 암컷에 비해 그 수치가 다소 높았으나 성별 간에 유의성은 없었다. 2. 평균적혈구용적, 평균적혈구혈색소양 및 평균적혈구혈색소농도의 평균치는 각각 102.65$\pm$17.45f1, 36.36$\pm$6.20pg, 및 34.44$\pm$0.63g/100m1을 나타냈으며 성별간에 비슷한 경향이었다. 3. 백혈구수는 평균 $16.33\pm4.75\times10^3/{\mu}{\iota}$ 이고 성별간의 유의성은 없었으며 동요범위는 매우 넓었다. 4. 혈소판수의 평균치는 $2.85\pm0.43\times10^5/{\mu}{\iota}$ 이고 남; 2.79$\pm$0.55, 여 $2.95\pm0.37\times10^5/{\mu}{\iota}$ 으로서 남이 여에 비하여 다소 높았으나 성별간에 유의차는 인정되지 않았다. 5. 백혈구감별계수의 평균치는 Neutrophile $25.8\pm5.32\%$, Lymphocyte $59.1\pm6.48\%$, Eosinophile $3.0\pm1.50\%$, Basophile $2.6\pm1.14\%$ 및 Monocyte $9.5\pm2.95\%$로 성별간에 비슷한 수치를 보였다. 6. 적혈구의 크기는 장경$\times$단경$\times$두께가 남 $13.06\pm0.53{\mu}{\times}8.66\pm0.22{\mu}{\times}3.49\pm0.07{\mu}$, 여 $12.71\pm0.51{\mu}{\times}8.55\pm0.24{\mu}{\times}3.44\pm0.06{\mu}$을 나타내었다. 7. 각 측정치에 있어서 성별간에 대체적으로 차이를 보였으나 유의차는 인정되지 않았다.

5-Fluorouracil 및 Mitomycin이 가슴샘의 미세구조에 미치는 영향 (Ultrastructural Study on the Thymus following the Administration of 5-Fluoruracil or Mitomycin)

  • The experiment was performed to study the morphological responses of the thymus of the mice, to antitumour agents (5-Fluorouracil or mitomycin C). Healthy adult mice weighing 25 gm each were divided into normal and experimental groups. 5-Fluorouracil (60 mg/kg) or Mitomycin-C $(400{\mu}g/kg)$ were injected subcutaneously to the animals every other day. Animals were sacrificed at 4 days and 7 days following the first injection. Pieces of the tissue taken from the thymus were prefixed with 2.5% paraformaldehyde-l.5% glutaraldehyde, followed by post-fixation with 1% osmium tetroxide. Ultrathin sections stained with uranyl acetate and lead citrate were observed with a JEM 100 CX-II electron microscope. The observed results were as follow: 1. Apoptoses of T-lymphocytes were observed more frequently in the thymus of the experimental groups than in those of a normal group. 2. In the experimental group, the plasma cells with distended cisternae of the granular endoplasmic reticulum and the eosinophile leukocytes were observed frequently. 3. In the experimental group, newly forming Hassall's corpurscles were observed frequently. 4. In the mitomycin-treated group, the epithelial reticular cells containing distended perinuclear cisternae, distended the granular endoplasmic reticula and pyknotic nuclei were observed in the cortico-medullary junctional area. 5. In the mitomycin-treated group, nuclear bodies with medium electron dense materials were often observed in the T lymphocyte. 6. In the 5-fluorouracil-treated groups, fused and dissolved tonofilament bundles and apoptotic bodies were observed in the some epithelial reticular cells in the medullary area. 7. In the 5-fluorouracil-treated groups, some elongated and bar-shaped lysosomes with electron lucent gap were often observed in the macrophages. 8. In the 5-fluorouracil-treated group, membrane complex of the smooth endoplasmic reticulum were ofen observed in the macrophage. From the above results, it was suggested that 5-fluorouracil or mitomycin could induce rapid involution of the thymus, and disturb maturation and differentiation of T lymphocytes, and, in turn, supress immunity.

황금 추출물이 $CCl_4$ 중독 쥐의 간장기능 회복에 미치는 영향 (Effects of Baikal Skullcap (Scutellaria baicalensis Georgi) extracts on the recovery of liver function in $CCl_4$-exposed rats)

  • 황금추출물의 간장기능 회복능을 알아보기 위하여, $CCl_4$로 간중독을 유도한 쥐에게 황금추출물을 급여한 후, 혈액성상, 혈액 내 효소활성치 및 간장 내 전염증성 cytokines의 농도변화를 검토했다. RBC, Hb 및 PCV의 값은 처리군간에 유의한 차이를 나타내지 않았다. 백혈구 총수는 대조군과 비교하여 황금 처리군 모두가 낮은 수치를 나타내었으며, 황금추출물 첨가량이 증가함에 따라 neutrophils 및 eosinophile의 비율이 감소하고, lymphocytes 및 monocytes의 비율이 증가하였다. basophils의 구성비는 처리군 별 유의한 차이를 나타내지 않았다. Total protein 농도 및 albumin 농도는 처리군 간에 유의한 차이를 나타내지 않았다. 그러나 albumin/globulin 농도 비율은 대조군보다 황금추출물 처리군에서 높았다. GOT, GPT 및 LDH의 효소활성치들은 모두가 대조군보다 황금추출물 처리군에서 낮은 수치를 나타내었으며, 황금추출물 첨가량이 증가함에 따라 하락하는 경향을 보였다. 간장 내 IL-$1{\beta}$, IL-6 및 TNF-$\alpha$의 농도는 대조군보다 황금추출물 처리군 모두가 낮은 값을 나타내었으며, IL-10의 농도는 대조군보다 황금추출물 처리군 모두가 높은 값을 나타내었다. 이상의 결과를 종합해 보면 황금추출물은 간장기능회복에 효과를 나타내는 기능성 물질을 내재하고 있음을 시사해 준다.

Bacillus Calmette-Guerin (BCG) 투여가 Ehrlich 종양세포를 이식한 생쥐 샘창자 상피세포의 DNA합성에 미치는 영향 (Effects of Bacillus Calmette-Guerin (BCG) on the DNA Synthesis of Mouse Duodenal Mucosal Epithelial Cells Inoculated with Ehrlich Carcinoma Cells)

  • 이 실험은 Ehrlich 종양세포를 샅부위에 이식한 후 고농도의 BCG(Bacillus Calmette-Guerin)를 피부밑조직에 반복 투여하였을 때, 샘창자점막의 형태학적 변화와 함께 DNA합성지수를 비교 연구하고자 3H-thymidine을 투여한 후 자기방사법적 연구를 시행하였다. 실험동물로는 체중 25 g 내외의 ICR 흰생쥐를 사용하였으며 정상대조군을 제외한 종양대조군과 BCG투여군의 동물에는 샅부위 피부밑조직에 각각 $1{\times}10^7$의 Ehrlich 종양세포를 이식하였다. BCG투여군은 종양세포이식 다음날부터 농축 건조된 BCG를 생리식염수에 용해시킨 다음, 일정량을 하루건너 한 번씩 7회 피부밑조직에 주사하였다. 종양대조군은 종양세포이식 후에 생리식염수를 하루건너 한 번씩 7회 피부밑조직에 주사하였고, 정상대조군은 종양세포를 이식하지 않은 동물을 사용하였으며 각 군에는 5마리씩의 동물을 배정하였다. 자기방사법적 관찰을 위하여는 약제를 마지막으로 주사한 다음날 3H-thymidine $0.7{\mu}Ci$/gm를 꼬리에 한차례 정맥주사하고, 70분 후 도살하여 샘창자조직을 떼어내어 10% 포르마린으로 고정하였다. 관찰은 샘창자조직이 세로로 잘 절단된 창자움 부위를 택하여 점막근육판을 따라 점막길이 3.5mm의 창자샘상피에 분포하는 3H-thymidine 표지세포의 수를 계수하였으며, 일반조직 관찰을 위해서는 hematoxylineosin (H-E)염색을 시행하였다. 일반조직관찰에서 종양대조군과 BCG투여군은 정상대조군에 비해 샘창자 점막 고유판에 림프구와 호산성백혈구가 다수 관찰된 것 이외에는 형태적으로 큰 변화를 볼 수 없었다. 자기방사법적 연구에서 정상대조군, 종양대조군 및 BCG투여군은 점막길이 3.5 mm 당 출현하는 표지세포수가 각각 632.3 (${\pm}14.47$), 761.7 (${\pm}27.65$) 및 505.0 (${\pm}17.09$)개였다. 이상의 결과를 종합해보면 Ehrlich 종양세포를 이식한 생쥐에 BCG를 반복 투여하면 샘창자 점막상피세포의 DNA합성을 유의하게 억제하면서도 형태적인 변화가 경미하였으므로 BCG는 항암치료시 항암효과를 높일 수 있는 좋은 약제라고 생각된다.
