무증자전분의 분쇄마찰매체에 의한 효소당화촉진 Mechanism의 규명 (Enhancing mechanism of the saccharification of uncooked starch in an agitated bead reaction system)

  • 분쇄마찰매체에 의한 무증자 생전분의 효소당화촉진 mechanism을 전분의 구조적 측면을 중심으로 규명하였다. 당화촉진 효과를 줄 수 있는 수준의 분쇄마찰매체의 기계적 교반운동은 생전분의 미세결정구조(microcrystalline structure) 파괴는 물론 전분입자를 붕괴 (fragmentation) 시키는 효과도 없었다. 생전분 입자구조 변화의 중요한 특징은 입자구조의 팽윤(swelling) 현상으로써 팽윤된 전분은 보수능력이 2.5배 정도까지 증가되었다. 이와 같은 기계적 충격에 의한 전분입자의 팽윤현상은 가열호화에 의한 $\alpha$-전분화에 따른 팽윤현상가는 상이하였다. 생전분을 장시간 bead-milling하여 전처리하며 팽윤시킨 생전분은 당화가 촉진되었으나 bead와 효소를 동시에 첨가시킨 경우의 당화속도와 수율에는 미치지 못하였다. 분쇄마찰 반응계에서 효소를 첨가 무증자 전분을 당화시킬 경우에는 생전분입자 2시간 전후하여 수많은 입자로 fragmentation되었다. 생전분의 당화촉진 mechanism은 분쇄마찰매체에 의하여 전분입자가 균열팽윤되고 이 팽윤된 생전분은 보다 쉽게 효소작용을 받아 침식되며 이 침식된 전분입자는 분쇄마찰매체에 의하여 더욱 가속적으로 fragmentation되어 효소작용이 촉진된다고 판단된다. 옥수수, 감자, 고구마 등 각종 생전분은 그 종류에 따라 분쇄마찰 반응계를 활용한 무증자 당화에 많은 차이가 있었으며 이를 전분입자의 구조와 연결시켜 고찰하였다.

  • This study shows that lignocellulosic biomass saccharification work has been carried out with rice-straw by the extracellular enzyme from KMU001, and the enzymes produced in 5%(w/v) wood biomass were characterized by protein and various enzyme activity measurements. Several cellulases such as Endoglucanase(EG), $\beta$-D-1,4-Glucosidase(BGL), Cellobiohydrolase(CBH), and $\beta$-D-1,4-Xylanase (BXL) were detected. Saccharification of rice-straw by the enzyme yielded about 233mg/g of glucose after 48hrs.

The Application of Thermotolerant Yeast Kluyveromyces marxianus as a Potential Industrial Workhorse for Biofuel Production

  • Kluyveromyces marxianus is a well-known thermotolerant yeast. Although Saccharomyces cerevisiae is the most commonly used yeast species for ethanol production, the thermotolerant K. marxianus is more suitable for simultaneous saccharification and fermentation (SSF) processes. This is because enzymatic saccharification usually requires a higher temperature than that needed for the optimum growth of S. cerevisiae. In this study, we compared the fermentation patterns of S. cerevisiae and K. marxianus under various temperatures of fermentation. The results show that at a fermentation temperature of $45^{\circ}C$, K. marxianus exhibited more than two fold higher growth rate and ethanol production rate in comparison to S. cerevisiae. For SSF using starch or corn stover as the sole carbon source by K. marxianus, the high temperature ($45^{\circ}C$) fermentations showed higher enzymatic activities and ethanol production compared to SSF at $30^{\circ}C$. These results demonstrate the potential of the thermotolerant yeast K. marxianus for SSF in the industrial production of biofuels.

Ethanol Production from Seaweed, Enteromorpha intestinalis, by Separate Hydrolysis and Fermentation (SHF) and Simultaneous Saccharification and Fermentation (SSF) with Saccharomyces cerevisiae

  • Ethanol productions were performed by separate hydrolysis and fermentation (SHF) and simultaneous saccharification and fermentation (SSF) processes using seaweed, Enteromorpha intestinalis (sea lettuce). Pretreatment conditions were optimized by the performing thermal acid hydrolysis and enzymatic hydrolysis for the increase of ethanol yield. The pretreatment by thermal acid hydrolysis was carried out with different sulfuric acid concentrations in the range of 25 mM to 75 mM $H_2SO_4$, pretreatment time from 30 to 90 minutes and solid contents of seaweed powder in the range of 10~16% (w/v). Optimal pretreatment conditions were determined as 75 mM $H_2SO_4$ and 13% (w/v) slurry at $121^{\circ}C$ for 60 min. For the further saccharification, enzymatic hydrolysis was performed by the addition of commercial enzymes, Celluclast 1.5 L and Viscozyme L, after the neutralization. A maximum reducing sugar concentration of 40.4 g/L was obtained with 73% of theoretical yield from total carbohydrate. The ethanol concentration of 8.6 g/L of SHF process and 7.6 g/L of SSF process were obtained by the yeast, Saccharomyces cerevisiae KCTC 1126, with the inoculation cell density of 0.2 g dcw/L.

일본(日本)에 있어서 전분이용(澱粉利用)의 현황(現況)과 전망(展望) (Present Status and Prospect of Starch Utilization in Japan)

  • Since 1950 there has been a dramatic progress in rationalization of the production of sweet potato and potato starch in Japan. This enabled dextrose industry by enzymatic process to develop rapidly due to the success of enzymatic liquefaction and saccharification. Isomerization of glucose to fructose has been studied, and the immobilization of isomerases prompted its products on industrial scale in 1970. Another advance is the development of effective methods of producing high purity maltose. A malto-hexaose forming amylase was discovered in 1971 and attempts are being made for its pharmaceutical utilization. Saccharification of cellulose by cellulase has been studied. Conversion of starch to other polysaccharides is another example for the numerous Japanese activities.

Ethanol Production from the Seaweed Gelidium amansii, Using Specific Sugar Acclimated Yeasts

  • For the production of ethanol from seaweed as the source material, thermal acid hydrolysis and enzymatic saccharification were carried out for monosugars production of 25.5 g/l galactose and 7.6 g/l glucose using Gelidium amansii. The fermentation was performed with Pichia stipitis KCTC 7228 or Saccharomyces cerevisiae KCCM 1129. When wild P. stipitis and S. cerevisiae were used, the ethanol productions of 11.2 g/l and 6.9 g/l were produced, respectively. The ethanol productions of 16.6 g/l and 14.6 g/l were produced using P. stipitis and S. cerevisiae acclimated to high concentration of galactose, respectively. The yields of ethanol fermentation increased to 0.5 and 0.44 from 0.34 and 0.21 using acclimated P. stipitis and S. cerevisiae, respectively. Therefore, acclimation of yeasts to a specific sugar such as galactose reduced the glucose-induced repression on the transport of galactose.

Simultanceous Saccharification and Fermentation of Cellulose for Lactic Acid Production

  • Lactic acid production from ${\alpha}$-cellulose by simultaneous saccharification and fermentation (SSF) was studied. The cellulose was converted in a batch SSF using cellulase enzyme Cytolase CL to produce glucose sugar and Lactobacillus delbrueckii to ferment the glucose to lactic acid. The effects of temperature, PH, yeast extract loading, and lactic acid inhibition were studied to determine the optimum conditions for the batch processing. Cellulose was converted efficiently to lactic acid, and enzymatic hydrolysis was the rate controlling step in the SSF. The highest conversion rate was obtained at 46$^{\circ}C$ and pH 5.0. The observed yield of lactic acid from ${\alpha}$-cellulose was 0.90 at 72 hours. The optimum pH of the SSF was coincident with that of enzymatic hydrolysis. The optimum temperature of the SSF was chosen as the highest temperature the microoraganism could withstand. The optimum yeast extract loading was found to be 2.5g/L. Lactic acid was observed to be inhibitory to the microorganisms' activity.

  • 에너지소모를 최소화하고 최대의 당화효과를 얻을 수 있는 고효율 당화장치의 개발을 목표로 impeller교반형 bioattritor를 설계, 제작하여 불용성 섬유소를 기질로 실험하였다. Bioattrotor의 효용성과 효소당화촉진 mechan-ism을 규명하였으며, 최적운전조건을 검토하였다. 분쇄 마찰매체함유 반응기에 일정한 탄성계수를 지닌 spiral spring coil을 내장한 torzue측정장치를 개발, 부하되는 torque, power, 그리고 소요에너지를 측정하여 이를 당화촉진과 비교함으로서 bioattritor의 경제성을 검토하였다. 분쇄마찰매체의 첨가량 및 종류, impeller형태, 교반속도는 중요한 요소로서 작용하였다. 비록 더 연구가 필요하나 분쇄마찰매체의 교반에 소모되는 동력은 당화 촉진효과의 증대에 따른 당화시간의 단축과 섬유소의 고효율 전환에 따른 고농도당의 확보등에 의해 보상되리라 예상된다.

Biological Pretreatment of Softwood Pinus densiflora by Three White Rot Fungi

  • The effects of biological pretreatment on the Japanese red pine Pinus densiflora, was evaluated after exposure to three white rot fungi Ceriporia lacerata, Stereum hirsutum, and Polyporus brumalis. Change in chemical composition, structural modification, and their susceptibility to enzymatic saccharification in the degraded wood were analyzed. Of the three white rot fungi tested, S. hirsutum selectively degraded the lignin of this sortwood rather than the holocellulose component. After eight weeks of pretreatment with S. hirsutum, total weight loss was 10.7%, while lignin loss was the highest at 14.52% among the tested samples. However, holocellulose loss was lower at 7.81 % compared to those of C. lacerata and P. brumalis. Extracelluar enzymes from S. hirsutum showed higher activity of ligninase and lower activity of cellulase than those from other white rot fungi. Thus, total weight loss and changes in chemical composition of the Japanese red pine was well correlated with the enzyme activities related with lignin- and cellulose degradation in these fungi. Based on the data obtained from analysis of physical characterization of degraded wood by X-ray Diffractometry (XRD) and pore size distribution, S. hirsutum was considered as an effective potential fungus for biological pretreatment. In particular, the increase of available pore size of over 120 nm in pretreated wood powder with S. hirsutum made enzymes accessible for further enzymatic saccharification. When Japanese red pine chips treated with S. hirsutum were enzymatically saccharified using commercial enzymes (Cellulclast 1.5 L and Novozyme 188), sugar yield was greatly increased (21.01 %) compared to non-pre treated control samples, indicating that white rot fungus S. hirsutum provides an effective process in increasing sugar yield from woody biomass.

바이오에탄올 제조를 위한 억새의 암모니아-희황산 복합 전처리 (Combined Aqueous Ammonia-Dilute Sulfuric Acid Pretreatment of Miscanthus for Bioethanol Production)

  • Pretreatment of cellulosic biomass is necessary before enzymatic saccharification and fermentation. The objective of this study was to evaluate the effect of combined aqueous ammonia-dilute sulfuric acid treatment on cellulosic biomass. Miscanthus was pretreated using aqueous ammonia and dilute sulfuric acid solution under high temperature and pressure conditions to be converted into bioethanol. Aqueous ammonia treatment was performed with 15 %(w/w) ammonia solution at $150^{\circ}C$ of reaction temperature and 20 minutes of reaction time. And then, dilute sulfuric acid treatment was performed with 1.0 %(w/w) sulfuric acid solution at $150^{\circ}C$ of reaction temperature and 10 minutes of reaction time. The compositional variations of this combined aqueous ammonia-dilute sulfuric acid treatment resulted in 68.0 % of cellulose recovery and 95.7 % of hemicellulose, 81.3 % of lignin, 89.1 % of ash removal respectively. The enzymatic digestibility of 90.5 % was recorded in the combined pretreated Miscanthus sample and it was 14.7 times higher than the untreated sample. The ethanol yield in the Simultaneous Saccharification and Fermentation was 90.4 % of maximum theoretical yield based on cellulose content of the combined pretreated sample and it was about 98 % compared to the ${\alpha}$-cellulose ethanol yield.
