지발형 오르니틴 트랜스카바미라제 결핍증 환자들의 신경학적 예후 (Neurological Outcome of Patients with Late-onset Ornithine Transcarbamylase Deficiency)

  • The most common urea cycle disorder is ornithine transcarbamylase deficiency. More than 80 percent of patients with symptomatic ornithine transcarbamylase deficiency are late-onset, which can present various phenotypes from infancy to adulthood. With no regards to the severity of the disease, characteristic fluctuating courses due to hyperammonemia may develop unexpectedly, and can be precipitated by various metabolic stressors. Late-onset ornithine transcarbamylase deficiency is not merely related to a type of genetic variation, but also to the complex relationship between genetic and environmental factors that result in hyperammonemia; therefore, it is difficult to predict the prevalence of neurological symptoms in late-onset ornithine transcarbamylase deficiency. Most common acute neurological manifestations include psychological changes, seizures, cerebral edema, and death; subacute neurological manifestations include developmental delays, learning disabilities, intellectual disabilities, attention-deficit/hyperactivity disorder, executive function deficits, and emotional and behavioral problems. This review aims to increase awareness of late-onset ornithine transcarbamylase deficiency, allowing for an efficient use of biochemical and genetic tests available for diagnosis, ultimately leading to earlier treatment of patients.

External mechanisms driving ecosystem changes in a coastal wetland, the Mississippi Delta, USA

  • The world's most extensive and active deltas, Louisiana's wetlands, are deteriorating rapidly due to multiple stressors such as the discharge of the Mississippi River, sea-level rise, and coastal retreat, the substantial but spatially and temporally variable impacts. However, the ecological and anthropogenic histories, the mode of environmental changes on a multi-millennial timescale have not been thoroughly documented. This study, a palynology-based multiproxy analysis, investigates hydrological, geological, geochemical, and anthropogenic impacts on southern Louisiana wetlands and a variety of external forcing agents influencing ecological succession. Sediment cores extracted from a small pond on a mangrove-dominate island near Port Fourchon, Louisiana, USA yielded a 4,000-year record. The site has been transformed from freshwater to saline water environments, to a mangrove dominant island over the late Holocene. The multivariate principal component analysis identified the relative strength of external drivers responsible for each ecological shift. The Mississippi River delta cycle (lobe switching) was the dominant driver of ecosystem changes during the late Holocene, while relative sea-level rise, tropical cyclones, climate, and anthropogenic effects have been the main drivers late in the site's history.

Characterization and gene expression of heat shock protein 90 in marine crab Charybdis japonica following bisphenol A and 4-nonylphenol exposures

  • Objectives Heat shock protein 90 (HSP90) is a highly conserved molecular chaperone important in the maturation of a broad spectrum of protein. In this study, an HSP90 gene was isolated from Asian paddle crab, Charybdis japonica, as a bio-indicator to monitor the marine ecosystem. Methods This work reports the responses of C. japonica HSP90 mRNA expression to cellular stress by endocrine disrupting chemicals (EDCs), such as bisphenol A (BPA) and 4-nonylphenol (NP) using real-time. reverse transcription polymerase chain reaction. Results The deduced amino acid sequence of HSP90 from C. japonica shared a high degree of homology with their homologues in other species. In a phylogenetic analysis, C. japonica HSP90 is evolutionally related with an ortholog of the other crustacean species. The expression of HSP90 gene was almost distributed in all the examined tissues of the C. japonica crab but expression levels varied among the different body parts of the crabs. We examined HSP90 mRNA expression pattern in C. japonica crabs exposed to EDCs for various exposure times. The expression of HSP90 transcripts was significantly increased in C. japonica crabs exposed to BPA and NP at different concentrations for 12, 24, 48 and 96 hours. The mRNA expression of HSP90 gene was significantly induced in a concentration- and time-dependent manner after BPA or NP exposures for 96 hours. Conclusions Taken together, expression analysis of Asian paddle crab HSP90 gene provided useful molecular information about crab responses in stress conditions and potential ways to monitor the EDCs stressors in marine environments.

Saccharomyces cerevisiae Hsp30 is Necessary for Homeostasis of a Set of Thermal Stress Response Functions

  • Saccharomyces cerevisiae Hsp30 is a plasma membrane heat shock protein that is induced by various environmental stress conditions. However, the functional role of Hsp30 during diverse environmental stressors is not presently known. To gain insight into its function during thermal stress, we have constructed and characterized a ${\Delta}hsp30$ strain during heat stress. $BY4741{\Delta}hsp30$ cells were found to be more sensitive compared with BY4741 cells, when exposed to a lethal heat stress at $50^{\circ}C$. When budding yeast is exposed to either heat shock or weak organic acid, it inhibits Pma1p activity. In this study, we measured the levels of Pma1p in mutant and Wt cells both during optimal temperature and heat shock temperature. We observed that $BY4741{\Delta}hsp30$ cells showed constitutive reduction of Pma1p. To gain further insights into the role of Hsp30 during heat stress, we compared the total protein profile by 2D gel electrophoresis followed by identification of differentially expressed spots by LC-MS. We observed that contrary to that expected from thermal-stress-induced changes in gene expression, the ${\Delta}hsp30$ mutant maintained elevated levels of Pdc1p, Trx1p, and Nbp35p and reduced levels of Atp2p and Sod1p during heat shock. In conclusion, Hsp30 is necessary during lethal heat stress, for the maintenance of Pma1p and a set of thermal stress response functions.

Anti-oxidant and Anti-pollution Composition Containing the Extract of Nypa fruticans Wurmb, Saussurea neoserrata, Codium fragile and Enteromorpha compressa

  • Long-term exposure to environmental pollutants can impair the human skin's barrier function and promote skin aging mechanisms. The visible consequences of these effects are dryness, wrinkles, black spots and worsening skin sensitivity. As awareness of the effects of environmental stressors on the skin has recently increased, consumers' demand for cosmetics that can provide anti-pollution effects is increasing. In this study, the possibility of anti-pollution cosmetic material was investigated by measuring the inhibitory effect of free radicals using a mixture of Nypa fruticans Wurmb (NF), Saussurea neoserrata (SN), Codium fragile (CF) and Enteromorpha compressa (EC), which has anti-inflammatory and antioxidant effects against irritation caused by dust. Combining the experimental results of a mixture of NF, SN, CF and EC by induction of fine dust stimulation, inhibition of inflammatory factors (PGE2) and intracellular free radicals (ROS) by inhibiting effects were significant, indicating the possibility of use as anti-pollution cosmetics. The most plants used as sources of anti-pollution cosmetic ingredients contain antioxidants as active substances. In our highly industrialized and chemically polluted world, it is not surprising that most plants used as sources of anti-pollution cosmetic ingredients contain antioxidants as active substances. Considering our results, it can be suggested that this anti-pollutant consisting of NF, SN, CF and EC may be a good ingredient for skincare products for the cosmetic industry due to their antioxidant properties, which may especially alter skin aging.

제주도 육상 넙치양식장의 해양환경현황 - 쿠도아 양성양식장과 음성양식장을 중심으로 - (Status of Marine Environment of Olive Flounder, Paralichythys olivaceus, Culture Ground in Jeju-do - Focus on Kudoa septempunctata positive and negative farm -)

  • In this study, we monitored the status of marine environment of olive flounder, Paralichythys olivaceus, culture ground in Jeju-do, Republic of Korea. It reported the consumption of raw olive flounder meat containing of Kudoa, Kudoa septempunctata, could induce vomitting and diarrhea in Japan. The Kudoa is a new mycosporean species, researchers found Kudoa from the muscles of olive flounder cultured at western Japan and imported from Jeju-do. We choose two Kudoa positive farms and two negative farms in the mid of concern about the relation between Kudoa and habitat. We found two marine invasive species at the water-pumping seabed at one of the Kudoa positive farm. The concentration of pH and DO at on-growing flounder farms showed the decreasing pattern along side the raceway (influent sea waters > on growing sea waters > outfluent sea waters). The TN and TP values increased gradually following to the raceway (influent sea waters < on growing sea waters < outfluent sea waters). The concentration of COD and SS were in the range of $0.100-2.581mg{\cdot}L^{-1}$, $1.00-12.70mg{\cdot}L^{-1}$, respectively. The calculated residence time was 4hr 32minutes at F1, 11hr 21minutes at F2, and 9hr 50minutes at F3, respectively. It was calculated same distance of 4 km away from effluent pipes. Although direct relation between Kudoa and marine environment could not define well based on this study result, the more studies on marine environmental stressors for olive flounder are required to conduct as a reliable method including socio-economic group and environmental group.

Bacterial Exopolysaccharides: Insight into Their Role in Plant Abiotic Stress Tolerance

  • Various abiotic stressors like drought, salinity, temperature, and heavy metals are major environmental stresses that affect agricultural productivity and crop yields all over the world. Continuous changes in climatic conditions put selective pressure on the microbial ecosystem to produce exopolysaccharides. Apart from soil aggregation, exopolysaccharide (EPS) production also helps in increasing water permeability, nutrient uptake by roots, soil stability, soil fertility, plant biomass, chlorophyll content, root and shoot length, and surface area of leaves while also helping maintain metabolic and physiological activities during drought stress. EPS-producing microbes can impart salt tolerance to plants by binding to sodium ions in the soil and preventing these ions from reaching the stem, thereby decreasing sodium absorption from the soil and increasing nutrient uptake by the roots. Biofilm formation in high-salinity soils increases cell viability, enhances soil fertility, and promotes plant growth and development. The third environmental stressor is presence of heavy metals in the soil due to improper industrial waste disposal practices that are toxic for plants. EPS production by soil bacteria can result in the biomineralization of metal ions, thereby imparting metal stress tolerance to plants. Finally, high temperatures can also affect agricultural productivity by decreasing plant metabolism, seedling growth, and seed germination. The present review discusses the role of exopolysaccharide-producing plant growth-promoting bacteria in modulating plant growth and development in plants and alleviating extreme abiotic stress condition. The review suggests exploring the potential of EPS-producing bacteria for multiple abiotic stress management strategies.

Analysis of gene expression in red maple (Acer rubrum) and trembling aspen (Populus tremuloides) populations from a mining region

  • The Greater Sudbury Region has been known as one of the most ecologically disturbed areas in Canada for the past century. Plant adaptation to environmental stressors often results in modifications in gene expression at the transcriptional level. The main objective of the present study was to compare the expression of genes associated with nickel resistance in Acer rubrum and Populus tremuloides growing in areas contaminated and uncontaminated with metals. Primers targeting Nramps4, Nas 3, At2G, MRP4 and alpha-tubulin genes were used to amplify cDNA of both species. The expression of the At2G gene, was $2{\times}$ and $9{\times}$ higher in P. tremuloides than in A. rubrum for St. Charles (uncontaminated site) and Kelly Lake (metal contaminated site), respectively. There was a much smaller difference between the two species for the Nramps 4 gene as its expression was $2.5{\times}$ and $3{\times}$ higher in P. tremuloides compared to A. rubrum from St. Charles and Kelly Lake, respectively. The same trend was observed for the MRP4 gene whose expression was $2{\times}$ and $14{\times}$ higher in P. tremuloides than in A. rubrum from St. Charles and Kelly Lake, respectively. For the Nas 3 gene, the expression was similar in both sites. This gene was upregulated $11{\times}$ and $10{\times}$ in P. tremuloides compared to A. rubrum in samples from St. Charles and Kelly Lake, respectively. In general, no significant difference was observed between the metal contaminated and uncontaminated sites for gene expression. In depth analysis revealed that AT2G and MRP4 genes were significantly down regulated in A. rubrum from the metal contaminated sites compared to those from uncontaminated areas, but environmental factors driving this differential gene expression couldn't be established.

토양오염 위해성평가를 위한 국가별 노출인자 비교분석 및 국내 노출인자 연구 (Comparative Study on Exposure Factors for Risk Assessment in Contaminated Lands and Proposed Exposure Factors in Korea)

  • 토양매체중의 오염물질들은 다양한 경로를 통해 인체나 생태계에 노출되어 위해성 문제를 유발하고 있다. 토양위해성평가(Soil Risk Assessment)는 토양오염물질로 인한 악영향의 개연성을 정량적으로 평가하는 과정으로, 유해성 확인(Hazard Identification), 용량-반응평가(Dose-Response Assessment), 노출평가(Exposure Assessment), 그리고 위해도 결정(Risk Characterization)의 4가지 단계적 과정으로 이루어진다. 이중 노출평가과정에서 매우 중요하게 작용하는 노출인자(Exposure Factor)는 토양오염물질의 인체노출량을 산정하는데 필수적인 요소로, 미국, 유럽국가 등 선진국에서는 나라별 상황에 적절한 노출인자를 사용하고 있으나, 우리나라는 아직 국내에 적용하기 적합한 노출 인자에 대한 연구가 매우 부족한 실정이다. 본 연구에서는 토양오염 위해성평가를 위한 국가별 노출인자를 비교분석하고, 이를 토대로 국내상황에 적용가능한 노출인자를 제시하였다. 국가별 노출인자 연구는 미국 Environmental Protection Agency, 영국 Environmental Agency, 네덜란드 RIVM, 호주, 그리고 독일에서 적용되고 있는 노출인자값에 대해 노출인자별로 총괄적인 비교분석을 수행하였다. 또한 국가별 비교분석연구와 국내에서 제시된 노출인자 자료를 종합하여, 국내자료가 없는 경우 외국에서 보편적으로 적용하고 있는 인자값을 국내 적용 타당성을 고려하여 제시하였다. 본 연구에서 고려된 노출인자 조사항목은 평균수명, 노출기간, 노출빈도, 체중, 체표면적(또는 노출체표면적), 피부흡수계수(Skin Absorption Factor), 토양-피부간 흡착계수(Soil-Skin Adherence Factor), 음용수 섭취량, 호흡률, 토양 섭취량 그리고 농작물 섭취량 등이다. 본 연구는 오염토양의 위해성평가를 수행할 때 국내 상황을 고려한 노출평가에 필요한 기반연구가 될 것이다.

근로환경에 따른 우리나라 근로자의 고혈압 분포 (Distribution of Hypertension According to Working Conditions among Korean Worker)

  • Objectives: This study is aimed at inquiring into the grasp of real condition of working environment and improvements by calculating the hypertension distribution consequent on job stress risk and exposure level of alleviating factors. In addition, this study is intending to estimate the hypertension distribution through socio-demographic factors and level of stressors occurring at working environment, such as high workload, low control, low support, job insecurity, long working hours, low income. Methods: This study estimated the hypertension distribution using the tertiary Korean Working Conditions Survey data, and conducted comparative analysis according to the category of individual questionnaire items using odds ratio. Result: As a result of study, it was found that in the event that working environment satisfaction is low and business & an immediate superior's attitudes are negative, the hypertension distribution was high. Particularly, it was found that physical risk factor musculoskeletal risk factor, and mental risk factor in a workplace were all increasing the hypertension distribution. Conclusions: With the aged workers' labor market participation ratio increasing, hypertension could be a major issue in the field of Occupational Safety and Health. Thus, it's necessary that the relevant employer should lower the hypertension distribution through creation of pleasant working environment and inducement of workers to improve in the relations with their superiors. As for the uppermost limit of this study, there is a limit to clarifying the mechanism of hypertension through multivariate statistics analysis because it's difficult to establish causal relationship by individual questionnaire item as the working conditions survey is made by cross-sectional study. In the follow-up research, this study is going to do research on the mechanism of hypertension through questionnaire supplementation and in-depth analysis.
