스트레스의 신경생물학적 이해 (Neural Circuits Mediating Stress)

  • 스트레스는 여러 가지 정신질환의 병태생리와 관련되는 것으로 알려졌다. 최근 여러 가지 동물 모델이 제시되고 뇌에 대한 연구가 활발해지면서 스트레스의 선경생물학적 기전에 대해 많은 사실이 밝혀지고 있다. 저자들은 동물과 사람을 대상으로 스트레스가 지각되고 대뇌에서 처리되고 신경내분비적 반응으로 전환되는 경로를 밝히고자 했던 최근의 연구들을 고찰하였다. 과거 변연계-시상하부-뇌하수체-부신 축(LHPA axis)과 자율신경계가 스트레스반응의 신경생물학적 담당자로 가장 많이 연구되어 왔으나, 최근에는 노르에피네프린(NE), 세로토닌, GABA/Glutamate, 도파민, 아세틸콜린 등의 신경전달물질과 부신피질자극호르몬방출인자(CRF), arginine vasopressin. glucocorticoid 등의 신경호르몬이 상호작용을 하면서 스트레스반응에 관계되는 것으로 알려지고 있다. 이러한 대뇌의 신경전달체계는 LHPA축과 유기적으로 연관되면서 스트레스반응을 매개하며, 구조적으로도 LHPA축은 해마, 편도 등 다양한 대뇌 부위와 연결된다. LHPA축은 이렇게 중층적으로 조절되는데, 여기에 생기는 이상은 만성 스트레스나 우울증 등 병적 상태와 관련된다. CRF는 LHPA축의 호르몬 역할 이외에 대뇌의 광범위한 부위에 분포하면서 신경전달물질로서 기능하며 다양한 스트레스반응을 매개한다. 스트레스를 주변 자율신경계가 활성화되는데, 청색반점에서 기시하는 NE계가 직접 자극되어 카테콜아민을 분비하기도 하지만, CRF나 다른 신경전달계가 먼저 자극되면서 간접적으로 활성화되기도 한다. 특히 CRF와 NE계는 서로 자극시키는 feed-forward 상호작용을 하며, 이것이 생체가 외부환경의 도전에 맞서 내분비계 뿐만 아니라 중추신경계를 동원하는 데 중요한 역할을 할것으로 보인다. 또한 CRF-NE 상호작용은 불안이나 우울 등 비정상적 스트레스반응의 병태생리를 이해하는 데 중요한 역할을 할 것으로 시사된다. 스트레스반응은 구조적, 신경화학적, 유전적 수준의 다양한 신경생물학적 작용을 통해 일어나며, 이에 대한 연구는 스트레스반응의 병태생리를 밝히고 불안장애, 기분장애 등 정신질환의 원인 규명과 치료에도 크게 기여할 것으로 보인다.

  • 본 연구는 훼손된 하천 수생태계의 훼손원인을 식별하고 그 전이 과정을 규명하기 위해 수생태계 훼손원인 진단체계를 구축하고 정책적 활용 가능성을 보고자 하였다. 수생태계 현황 조사 및 건강성 평가결과는 하천의 훼손에 대한 전반적인 상황을 알 수 있어 하천 복원 및 관리의 판단 근거로 사용된다. 그러나 하천이 훼손된 원인이나 훼손이 발생하게 된 전이 과정을 고려하지 못한다는 한계를 가진다. 따라서 본 연구에서는 과학적이고 체계적인 진단체계를 제안하여 훼손원인과 그 전이 과정을 규명하였다. 미국, 호주, 유럽의 사례를 분석하여 우리나라에 맞는 진단체계를 제안하였다. 총 8단계로 구성되며, 기초자료 조사, 하천 수생태계 훼손 판단, 훼손 구간 설정, 훼손 유형화 및 잠재적 훼손원인 도출, 본 조사, 주훼손원인 진단 및 최적 훼손 모델 도출, 종합분석 및 하천 복원방안으로 이루어져 있다. 이러한 진단체계는 하천 생태계의 복잡하고 다양한 특성을 고려하여 훼손원인을 파악할 수 있으며 하천 복원 및 관리 방안을 제시하여 효율적인 의사결정을 지원할 수 있다.

국내 수의과대학생의 스트레스 경험 연구 (A study on stress experiences of Korean veterinary students)

  • The quality of a veterinary service depends on the veterinarian's physical and mental health. However, a veterinarian's mental health is generally considered to be more vulnerable than that of other health care professionals. As many veterinary students will enter the veterinary profession in near future, it is necessary to investigate their psychological status. This study examined stressors affecting student life in one veterinary school. Anxiety, depression, and stress levels were assessed using questionnaires and qualitative research techniques. The results obtained from questionnaires showed that the proportion of veterinary students with psychological distress was higher than expected. The major stressors experienced by the veterinary students were academic, economic, and environmental. Qualitative study using a photovoice method revealed that the students in this study felt relatively deprived compared with those in other veterinary schools. In addition, they were distressed by the school environment and limited human relationship. Our results suggest that veterinary school administrators should be aware of students' psychological distress and should undertake systematic improvement in their educational system by altering counseling programs, curricula, and school culture principles.

미량오염물질에 의한 항생제 내성 유전자 전이 특성에 대한 실험모델 개발 (The Experimental Model Development of Antibiotic Resistance Gene Transfer Characteristics with Various Micropollutants)

  • Recently, antibiotic resistant genes (ARGs) in the environment are emerging as pollutants, since these genetic contaminants can eventually be transferred to human pathogens. The aim of this study was to develop the experimental model of antibiotic resistant gene (ARG) plasmid transfer as a function of various environmental conditions. For this purpose, the multi drug resistant plasmid pB10, which is known to be originally isolated from a wastewater treatment plant, was selected as a model transfer plasmid and Escherichia coli $DH5{\alpha}$ containing pB10 was used as a model donor. Pseudomonas aeruginosa, an opportunistic pathogen, was selected as the recipient for the conjugation experiment. When the donor and recipient were exposed to various stressors including antibiotics and heavy metal as a function of the concentrations (10, 100 and, 1000 ppb), statistically increased plasmid transfer rate was observed at a concentration of 10 ppb of tetracycline and sulfamethoxazole compared to control (no antibiotic exposure). Accordingly, the developed experimental ARG model by various stressor is a promising tool for evaluating the dissemination of ARGs by micro-contaminants in aquatic environment.

칠게 외골격 표면 거칠기를 이용한 노출 독성 평가: 새로운 융합적 연구 (Toxicological Assessment to Environmental Stressors Using Exoskeleton Surface Roughness in Macrophthalmus japonicus: New Approach for an Integrated End-point Development)

  • 갑각류 외골격은 키틴질로 이루어져 있고 외부자극에 보호와 지지를 위해 주요한 기능을 한다. 수환경은 물리적 변화뿐만 아니라 다양한 해양 오염에 끊임없이 영향을 받고 그 환경에 서식하는 생물은 생존을 위해 생물학적으로 반응한다. 본 연구에서는 이르가롤 독성노출이나 퇴적환경 중금속 분포에 따라 서식하는 칠게의 외골격 표면조도 분석을 통한 생태독성 종말점이 될 수 있음을 제시하였다. 칠게의 외골격 표면 조도는 이르가롤 노출에 대해 거칠기(Ra)도 감소하고 조도 변이(variation)도 줄어들었다. 또한 해양 연안 퇴적물 내 중금속(Cd, Pb, Hg) 분포에 따라 서식하는 칠게에서 표면 거칠기 및 변이가 변화하는 것을 확인하였다. 이러한 외골격 표면 거칠기의 변화는 외부환경 스트레스 요인에 따른 칠게의 생물학적인 반응 종말점(end-point)을 나타내며 나아가 외골격의 보호, 지지, 수송 등의 기능적 면에 영향을 미칠 수 있음을 시사한다. 이러한 접근방법은 공학과 생물의 융합적 기술로 수환경 모니터링을 위한 유용한 방법으로 활용될 수 있을 것이다.

A Study on the New Aptitude Test Items for the Safety Critical Workers: Focused on Stress Tolerance under Emergency and Solitary Driving Situation

  • The main purpose of this study is to highlight the importance and validity of introduction of stress-concerned aptitude items for the safety critical workers, corresponding to the radical environmental change in railway industry. To attain this purpose, the author performed a brief literature survey, and scrutinized the western practice in detail. Then, the necessity of introduction of stress-related aptitude items was suggested, based on the statistics resulted from a survey which has been once performed for experts and drivers. The final conclusion is that the present selection system through which the safety critical workers are being staffed, without any rigorous screening mechanisms cannot but have some limitations. So, an overall restructuring for aptitude management system needs to be done in order for us to advance to the position of upper-class railway country.

Advanced Technologies and Mechanisms for Yeast Evolutionary Engineering

  • In vitro evolution is a powerful technique for the engineering of yeast strains to study cellular mechanisms associated with evolutionary adaptation; strains with desirable traits for industrial processes can also be generated. There are two distinct approaches to generate evolved strains in vitro: the sequential transfer of cells in the stationary phase into fresh medium or the continuous growth of cells in a chemostat bioreactor via the constant supply of fresh medium. In culture, evolutionary forces drive diverse adaptive mechanisms within the cell to overcome environmental or intracellular stressors. Especially, this engineering strategy has expanded to the field of human cell lines; the understanding of such adaptive mechanisms provides promising targets for the treatment of human genetic diseases and cancer. Therefore, this technology has the potential to generate numerous industrial, medical, and academic applications.

The Relationship Between Psychosocial Stress and Allergic Disease Among Children and Adolescents in Gwangyang Bay, Korea

  • Objectives: Stress is considered a causal factor in many diseases, allergic disease being one of them. The prevalence of allergic disease is increasing in Korea, but the relationship between allergic symptoms and stress is not empirically well known. We aimed to evaluate the relationship between allergy-related symptoms and stress in children and adolescents. Methods: We investigated 698 children and adolescents living in Gwangyang Bay, Korea, using a multi-stage cluster sampling method. Using the International Study of Asthma and Allergies in Childhood and the Psychosocial Well-being Index, these subjects were surveyed on allergy-related symptoms and psychosocial stressors in their lives, respectively. We used a multivariate logistic analysis for odds ratios for the complaint rate of allergic symptoms, after adjusting for age, gender, household income, body mass index, and residence. Results: After adjustments, lifetime rhinitis (odds ratio [OR], 1.024), rhinoconjunctivitis (OR, 1.090), diagnosis of itchy eczema (OR, 1.040), treatment of itchy eczema (OR, 1.049), 12-month allergic conjunctivitis (OR, 1.026), diagnosis of allergic conjunctivitis (OR, 1.031), and treatment of allergic conjunctivitis (OR, 1.034) were found to be significantly associated with stress. Conclusions: Our results support the notion that there is a relationship between stress and allergic symptoms in children and adolescents. Further research into any causal relationship between stress and allergies, as well as preventative public health plans for decreasing stress in children and adolescents are needed.

Protoplast Production from Sphacelaria fusca (Sphacelariales, Phaeophyceae) Using Commercial Enzymes

  • Sphacelaria is a filamentous brown algal genus that can be epibiotic on macroalgae, marine plants, and sea turtles. Its important role in benthic ecosystems, exposure to different stressors (e.g., grazing), and use as a model organism make Sphacelaria ideal for assessing physiological responses of organisms to environmental inputs. Single-cell RNA sequencing is a powerful new probe for understanding environmental responses of organisms at the molecular (transcriptome) level, capable of delineating gene regulation in different cell types. In the case of plants, this technique requires protoplasts ("naked" plant cells). The existing protoplast isolation protocols for Sphacelaria use non-commercial enzymes and are low-yielding. This study is the first to report the production of protoplasts from Sphacelaria fusca (Hudson) S.F. Gray, using a combination of commercial enzymes, chelation, and osmolarity treatment. A simple combination of commercial enzymes (cellulase Onozuka RS, alginate lyase, and driselase) with chelation pretreatment and an increased osmolarity (2512 mOsm/L H2O) gave a protoplast yield of 15.08 ± 5.31 × 104 protoplasts/g fresh weight, with all the Sphacelaria cell types represented. Driselase had no crucial effect on the protoplast isolation. However, the increased osmolarity had a highly significant and positive effect on the protoplast isolation, and chelation pretreatment was essential for optimal protoplast yield. The protocol represents a significant step forward for studies on Sphacelaria by efficiently generating protoplasts suitable for cellular studies, including single-cell RNA sequencing and expression profiling.

연안해역 생태계 건강성 평가의 의미와 국내 적용 방향 (Implications for Coastal Ecosystem Health Assessments and Their Applications in Korea)

  • Coastal marine ecosystems continue to suffer unrelenting pressures from human population growth, increased development, and climate change. Moreover, these systems' capacity for self-repair is declining with such increases in anthropogenic production of various pollutants. What is the present health status or condition of the coastal ecosystem? If our coastal areas are unhealthy, which conditions are considered serious? To answer such questions, the United States, Canada, and Australia are currently assessing coastal ecosystem health using systematic monitoring programs as well as identifying and implementing management plans to improve the health of degraded coastal ecosystems. To evaluate marine environments, Korea is currently using a limited number of factors to estimate water quality. In fact, we are ill-prepared for assessing coastal ecosystem health because no biologically specific criteria are in place to measure the responses to various pollutants. We should select ecosystem-specific indicators from physicochemical stressors and evaluate the subsequent biological responses within each ecosystem. Furthermore, a set of practical indicators should be generated by considering the characteristics and uses of a local coastal area and the key issues at hand. The values of indicators should be presented as indices that allow understanding by the general public as well as by practitioners, policy makers, environmental managers and other stakeholders.
