  • To investigate the effect of sourdough on the characteristics of quality of barley bread, sourdough starter with Lactobacillus plantarum, Lactobacillus sanfrancisco and Enterococcus sp. were added to baking after 48 hours incubation at $37^{\circ}C$. 1. The pH of bread using Lactobacillus sanfrancisco starter was the lowest among tested starters. while the titratable acidity(TTA) of the strain was the highest, followed by Enterococcus sp.. 2. In bread, moisture contents of control were 38%. It was decreased during 6 days. The moisture contents of bread using Lactobacillus sanfrancisco starter were the highest among tested starters, followed by Enterococcus sp.. 3. Oneset temperature$(T_o)$, peak temperature$(T_p)$ and enthalphy $({\Delta}H)$ were increased during storage of 6 days. The enthalphy of bread using Lactobacillus sanfrancisco starter was the lowest among tested starters, followed by Enterococcus sp..

Isolation, Identification, and Characterization of a Bacteriocin-Producing Enterococcus sp. from Kimchi and Its Application to Kimchi Fermentation

  • A bacteriocin-producing lactic acid bacterium, which strongly inhibited the Lactobacillus plantarum recognized as an important acid spoilage microorganism in kimchi fermentation, was isolated from kimchi. From morphological, physiological, sugar fermentation, biochemical tests, and l6S rDNA sequencing results, the isolate was identified as an Enterococcus sp. and designated as Enterococcus sp. K25. The bacteriocin produced by Enterococcus sp. K25 inhibited several Gram-positive bacteria, including Lb. plantarum, whereas it did not inhibit Gram-negative bacteria and yeasts. Optimal temperature and pH for the bacteriocin production were $25^\circ{C}$ and 5.5, respectively. Enterococcus sp. K25 was applied to kimchi manufacturing alone and together with other preservatives (i.e., chitosan and fumaric acid). In addition, growth of lactic acid bacteria, pH, and titratable acidity (TA) were measured during aging at $5^\circ{C}$ and $10^\circ{C}$. Inoculation of Enterococcus sp. K25 together with fumaric acid showed the most synergistic effect on extension of kimchi shelf-life. Compared to control (no addition), the treatment prolonged the kimchi shelf-life up to 6 days, whereupon the eight-point TA value recognized as the edible limit was reached.

  • A Lactic acid bacterium for barley bread was isolated from barley powder and was identified as Enterococcus sp. It was used as a starter for barley bread and quality of bread was compared with the bread prepared by conventional starter 1. The pH of bread making process using Lactobacillus sanfrancisco starter was the lowest among tested starters. while the titratable acidity(TTA) of the strain was the highest, followed by Enterococcus sp.. 2. In valorimeter value(v/v), control was 70, but dough using all starter was 60. The peak time and stability of dough using Lactobacillus sanfrancisco were the highest among tested starters. 3. Extensibility of dough using Enterococcus sp. was the highest among tested starters, followed by Lactobacillus sanfrancisco. Proper extensibility and maximum resistance were observed in Lactobacillus sanfrancisco.

The Pathology of Infection in the Department of Radiology (영상의학과 검사실의 감염 실태)

  • This study was performed to understand the bacteriologic contamination level of radiological equipments which have frequent contacts with patients in the Department of Radiology of an university hospital in Busan area. Before sterilizing in-patient of the radiology rooms, MRSA, VRE, acinetobacter baumannii, candida albicans, and enterococcus sp. were detected. After sterilization, all the bacteria were not found. As examine times become longer, more bacteria were detected and after 7 hours, bacillus sp.(GPR), CNS, acinetobacter baumannii, and Enterococcus sp. were detected. After examining infected patients, bacillus sp.(GPR), VRE, enterococcus sp. CNS, and micrococcus sp. were detected and on the hands of radiological technologists, CNS, enterococcus sp. escherichia coli, and enterobacter sp. were detected. Similar species of bacteria were detected from each radiology room, but pseudomonas aeruginosa was detected on the handles of portable radiological equipments and the chair in the waiting room. Therefore, it is the most important to regularly sterilize radiological equipments and devices which have frequent contacts with patients and to sterilize them right after the use of infected patients in order to prevent the spread of infection. Also, thorough hand washing, education on infection and management for the characteristics of Department of Radiology should be performed for the systematic prevention of infection.

Production of L-Lactic Acid from Soluble Starch by Enterococcus sp. JA-27. (Enterococcus sp. JA-27에 의한 가용성 전분으로부터 L형 젖산의 생산)

  • Lactic acid bacteria with amylolytic and acid producing activities can ferment starch directly to lactic acid thereby producing a monomer for the production of biodegradable poly lactic acid (PLA). In this study, the strain producing L-lactic acid from soluble starch was isolated from Nuruk. The isolated strain was identified as Enterococcus sp. through its morphological, cultural, biochemical characteristics as well as the 16S rDNA sequence analysis, and named Enterococcus sp. JA-27. Enterococcus sp. JA-27 produced exclusively L-lactic acid from soluble starch as a carbon source. The optimal conditions for the maximum production of L-lactic acid from Enterococcus sp. JA-27 were 30 C, pH 8, 1.5 % soluble starch as a substrate and 3.5 % tryptone as a nitrogen source, 0.1 % $K_2$$HPO_4$, 0.04 % $MgSO_4$. $7H_2$O, 0.014 % $MnSO_4$$.$4$H_2O$, 0.004% $FeSO_4$$.$$7H_2$O. Batch and fed batch culture were carried out and the former was more effective. L-Lactic acid production in the optimum medium was significantly increased in a 7 L jar fermenter, where the maximum L-lactic acid concentration was 3 g/L. For the purification of lactic acid in fermented broth, two stage ionexchange column chromatographies were employed and finally identified by HPLC.

Characterization of a Bacteriocin Produced by Enterococcus sp. T7 Isolated from Humans

  • A bacteriocin-producing organism, Enterococcus sp. T7, was isolated from human fecal samples. Bacteriocin T7, named tentatively as the bacteriocin, was produced by Enterococcus sp. T7 and it inhibited some strains of Lactobacillus. Staphylococcus, Enterococcus, and Streptococcus, but not all the lactococci and gram-negative bacteria tested. Bacteriocin T7 inhibited the growth of Listeria monocytogenes Scott A, but the degree of inhibition was less than those for other sensitive gram-positive vacteria. Bacteriocin T7 in MRS broth started to produce at the middle of the exponential growth phase and the inhibitory activity reached its maximum level during the stationary growth phase. Bacteriocin T7 was stable against heat treatments, pH variations (pH 2-10), and exposure to organic solvents. The molecular weight of bacteriocin T7 was estimated to be 6.500 Da by SDS-PAGe. All these facts, including physico-chemical stabilities, small molecular size, and inhibition of Kisteria monocytogenes, indicate that bacteriocin T7 is likely to be a member of the class IIa bacteriocins.

Fermentation Property by Novel Cellulolytic Lactic Acid Bacteria Enterococcus sp. TO-94 on Omija (Schizandra chinensis Baillon) (신규 섬유질 분해성 젖산균 Enterococcus sp. TO-94를 이용한 오미자의 발효 특성)

  • The use of cellulolytic lactic acid bacteria in new method to prepare high nutrition complementary foods was investigated. For the screening of cellulolytic lactic acid bacteria, more than 1,150 bacterial colony were isolated from diluted infant feces samples. A typical strain which appeared the most excellent cellulolytic activities was identified novel acidophilic Enterococcus sp. TO-94 through the results of morphological, biochemical and chemotaxonomic characteristics and 16S rDNA sequencing. The optimal lactic acid fermentation conditions of Omija(Schizandra chinensis Baillon) by Enterococcus sp. TO-94 were as follows: pH and temperature were 3.0 and $37^{\circ}C$, respectively, and fermentation time was 20hrs. The fructose and glucose were major free sugar and the contents were 5.83 and 4.30 mg/g after fermentation, respectively. The contents of lactic acid and acetic acid were 9.84 mg/g and 2.08 mg/g after fermentation, respectively. The vitamin $B_1$, $B_2$, niacin, folic acid and C were major vitamin in the fermented broth, the contents were 1.5~3 times higher than those of initial fermentation time. Also, the contents of polyphenol and anthocyanine were 3.8 and 1.2 times higher than those of initial fermentation time.

Effects of Complex Direct-Fed Microbial Supplementation on Growth Performance and Nutrient Digestibility for Broilers (복합생균제의 급여가 육계의 성장능력과 영양소 소화율에 미치는 영향)

  • This study was conducted to investigate the effects of dietary direct-fed microbials(DFM) on the growth performance and nutrient digestibility in broiler chickens. A total of two hundred eighty eight broiler chickens were randomly allocated into six treaments with four replications and fed for five weeks. Dietary treatments included 1) CON (basal diet), 2) DFM-1(basal diet + 0.2% Enterococcus sp. & Lactobacillus reuteri), 3) DFM-2(basal diet + 0.2% Enterococcus sp. & Lactobacillus plantarum), 4) DFM-3(basal diet + 0.2% Enterococcus sp. & Lactobacillus reuteri & Lactobacillus plantarum), 5) DFM-4(basal diet + 0.2% Enterococcus sp. & Lactobacillus reuteri & Lactobacillus plantarum & Yeast), 6) DFM-5(basal diet + 0.2% Enterococcus sp. & Lactobacillus reuteri & Lactobacillus plantarum & Bacillus subtilis). During the period of 1∼3weeks, average daily gain (ADG) and Feed/Gain were not significant different among treatments. In the later(3∼5weeks) and overall period(l∼ 5weeks), ADG and Feed/Gain tended to be improved in DFM treatments compared to the control. However, there were no significant differences among DFM complexes and between control and DFM treatments. The broilers fed DFM-3 diet was showd poor growth performance compared to the broilers fed DFM-l diet and similar to the broilers fed DFM-2 diet but there was no significant differences. The treatments of DFM-4 diet added Yeast and DFM-5 added Bacillus subtilis were improved in ADG and Feed/gain but no significant differences were found and also there was no specific DFM treatments in experiment period. In digestibility, the broiler chicks fed DFM treatments tended to improve N digestibility compared to control treatment. However, they were not significantly different among the treatments. In conclusion, DFM tended to improve growth performance in later stage of broiler.

Partial Characterization and Purification of Enterocin K25 Linked to the Plasmid in Enterococcus sp. K25

  • The antimicrobial activity of partially purified enterocin K25, produced by Enterococcus sp. K25, was abolished by proteases such as pepsin and proteinase K. The bacteriocin was resistant to heat treatment at $75^{\circ}C$ for 15 min and lost 75% of its activity at $100^{\circ}C$ for 30 min. Enterocin K25 showed bactericidal mode of action against an indicator strain, Lactobacillus plantarum NCDO 955. Enterocin K25 was purified to 112.6-fold purity via conventional steps of ammonium sulfate precipitation, ion exchange chromatography, and reversed phase high performance liquid chromatography (RP-HPLC). The molecular mass of the purified enterocin K25 was estimated as 4.3 kDa on an electrophoresis gel. Plasmid (${\sim}6.5\;kb$) linkage of production of enterocin K25 was confirmed by plasmid curing.

  • Succinic acid, valuable $C_4$-dicarboxylic acid as a renewable alternative feedstock, is currently produced commercially by the petrochemical process, but extensive efforts have been devoted to establish the biological process for mass production of succinic acid. In this study, the bioconversion of fumaric acid to succinic acid was investigated. We isolated an Enterococcus sp. RKY1 KCTC 8890P, facultative bacterium, capable of the bioconversion of fumaric acid to soccinic acid very rapidly and efficiently. At batch fermentation, the amount of succinic acid production increased with increase in initial fumaric acid from 40 to 100 g/L. With fumaric acid of 70 g/L, the average specific and volumetric production rate, molar yield were reached up to 0.64 g/g.h, 4.87 g/g.h, and 96.5%, respectively. Maximum concentration of succinic acid of 88.9 g/L was achieved with molar yield of 89% with fumaric acid of 100 g/L in less than 20 hours.
