Cedrela sinensis Leaves Suppress Oxidative Stress and Expressions of iNOS and COX-2 via MAPK Signaling Pathways in RAW 264.7 Cells

  • Overproduction of reactive oxygen species (ROS), including nitric oxide (NO), could be associated with the pathogenesis of various diseases such as cancer and chronic inflammation. Inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) are known to play key roles in the development of these diseases. Cedrela sinensis leaves have been used in Asian countries as a traditional remedy for enteritis, dysentery and itching. In the present study, we investigated the anti-inflammatory effects of Cedrela sinensis leaves in lipopolysaccharide (LPS)- stimulated RAW 264.7 macrophages. Powder of C. sinensis leaves was extracted with 95% ethanol and fractionated with a series of organic solvents including n-hexane, dichloromethane, ethyl acetate, n-butanol, and water. The dichloromethane (DCM) fraction strongly inhibited NO production possibly by down-regulating iNOS and COX-2 expression, as determined by Western blotting. Hydrogen peroxide-induced generation of reactive oxygen species (ROS) was also effectively inhibited by the DCM fraction from C. sinensis leaves. In addition, C. sinensis inhibited LPS-mediated p65 activation via the prevention of IκB-$\alpha$ phosphorylation. Furthermore, mitogen-activated protein kinases (MAPKs) such as ERK 1/2 and p38 were found to affect the expression of iNOS and COX-2 in the cells. Taken together, our data suggest that leaves of C. sinensis could be used as a potential source for anti-inflammatory agents.

Extract of Alnus japonica Induces Apoptosis of Human Colon Adenocarcinoma Cells through the Mitochondria/Caspase Pathway (대장암세포주에서 적양 추출물의 미토콘드리아/Caspases 경로를 통한 Apoptosis 유도 작용)

  • An extract of Alnus japonica (Betulaceae) cortex has been traditionally used for purifying blood, and curing feces containing blood, enteritis, diarrhea, alcoholism and cut wounds. In the present study, we demonstrated that the ethanol extract of Alnus japonica (EAJ) exhibited significantly cytotoxicity in human colon adenocarcinoma HT-29 cells. The results showed that the induction of apoptosis in HT-29 cells by EAJ was characterized by chromatin condensation and activation of caspase-3. EAJ-induced activation of caspase-9 and -3 caused the cleavage of poly ADP-ribose polymerase (PARP) and the release of cytochrome c. The expressions of Bcl-2 and Bid were reduced by EAJ in HT-29 cells, whereas pro-apoptotic protein Bak was increased in the cells. EAJ-induced, dose-dependent induction of apoptosis was accompanied by sustained phosphorylation of MAP kinases (JNK and p38 MAPK), ASK1, and p53. NAC administration, a scavenger of ROS, reversed EAJ-induced cell death. In conclusion, these results indicated that EAJ can cause apoptosis through a ROS-mitochondria-caspases-dependent pathway in human HT-29 cells.

Prevalence and Drug Susceptibility of Campylobacter jejuni and Campylobacter coli in Korean Native Goats (한국 재래산양에서 있어서 Campylobacter jejuni 및 Campylobacter coli의 분포와 약제감수성)

  • This study was conducted to determine the epidemiological characteristics of Campylobacter enteritis. A total of 187 fecal specimens of Korean native goat were examined for the presence of C. jejuni and C. coli by direct plating. Fifty strains isolated were examined for biochemical and serological properties and susceptibility to 19 chemotherapeutic agents. A total of 29(15.5%) C. jejuni and 21 (11.2%) C. coli were isolated from the fecal specimen of 187 Korean native goats. Of the 50 isolates of C. jejuni and C. coli, 29 isolates of C. jejuni grouped as 7 biotypes (1,2,3,4,6,7 and 8) and biotypes 1(34.5%), 2(17.2%) and 3(20.7%) were encountered most frequently. Twenty-one C. coli strains were differentated into biotype I (61.9% of the isolates) and biotype II (38.1%). Of the 29 C. jejuni strains examined, 24(83.0%) were typable by the Lior serotyping scheme and five isolates were non typable. C. jejuni grouped as 8 serotypes, serotype 4(24.1%) and 26(20.7%) were encountered most frequently. In the case of 21 strains of C. coli grouped as 6 serotypes, the most frequent serotypes were 21(28.6%) and 25(23.8%). Total of 50 strains of isolated were all susceptible to amikacin, clindamycin and tobramycine. Overall 85% of isolates were sensitive to erythromycin, doxycycline, chloramphenicol, flume-quine, kanamycin, gentamicin, nalidixic acid, polymyxin B, colistin, tetracycline and ampicillin, but about 65% of isolates were resistant to cefamandole and ethyl hydrocuprein hydrochloride.

  • Purpose. Natural herb has been used for traditional medicine for thousands of years. For this reason, alternative medicine methods affect to domestic animal field. The main purpose of this study was to confirm the regulated proteins after feed additive treatment. Methods. We used total thirty male pigs were used for this experiment. E. coli and Salmonella typhimurium, were used for promote enteritis. Animals were divided into a negative control group, positive control group and test group to determine the effect of an additive mixture on the changes in protein expression in serum. The pigs were given a food supplemented with a natural herbal additive containing immunoglobulin yolksac (IgY) at concentrations 1%. On the 1st week and after eight weeks of feeding, the serums were collected from each group. The changes in the serum proteome as a response to the herbal additive were examined using two-dimensional polyacrylamide gel electrophoresis. Results. A total of 14 differentially expressed protein spots were identified by comparing the protein profiles of the control and additive treated porcine serums. Finally, 7 proteins were detected by MALDI-TOF/MS. Moreover, the proteins detected are involved in a cholesterol control factor, inflammation regulator, Conclusion. These results support of the hypothesis that a natural herbal additive containing IgY can affect the immune regulation system and reduce the pathological process by microbial infections.

  • The purpose of this study was to identify the categories and frequencies of communication of students in nursing simulation using standard patient. Data were collected through questionnaires and video recording of communication process of 41 nursing students in simulation related to pediatric enteritis from March to June, 2017. The data were analyzed the contents and frequencies of completed communication forms by trained researcher based on Interaction Process Analysis. In result, students used communication skills in task area frequently. Communication skills, such as orientation, open question, identification, giving information, opinion, and suggestion, and eye contact, were used frequently. In an uncomfortable atmosphere such as listening to the nursing client's complaints, students showed tension and stiff appearance. Students used high quality communication skills such as empathic understanding and therapeutic touch restrictively. In conclusion, we need education strategies relating to therapeutic communication for nursing professionalism and qualitative improvement of nursing service.

Evaluation of a Double-Pigtail Ureteral Stent Fixation in Cats with Complete Ureteral Obstruction

  • The aim of this study was to evaluate double-pigtail ureteral stent fixation in cats. Medical records of 19 cats (23 ureters) with complete ureteral obstruction that double-pigtail ureteral stent placement were carried out were retrospectively reviewed. The cats were randomly classified into two groups; 13 cats (16 ureters) with double-pigtail ureteral stent fixed to urinary bladder (SF group) and 6 cats (7 ureters) with not fixed to urinary bladder (SNF group). The average age and weight of the cats was 7.4 years and 3.73 kg, respectively. Postoperative complications included chronic renal failure (n = 11), lower urinary track diseases (cystitis, hematuria, pollakiuria) (n = 7), stent migration (n = 6), anemia (n = 5), ascites (n = 2), hyperthermia (n = 1), enteritis (n = 1), oliguria (n = 1), hypotension (n = 1), ureteritis (n = 1), and pyelonephritis (n = 1). Stent migration did not occur in the 16 ureters of the cats in SF group but did occur in 4 out of 7 ureters of the cats in SNF group. The prevention of stent migration by stent fixation was significant (P = 0.04). Among the 13 cats in SF group, only 2 cats developed lower urinary track diseases, while 4 of the 6 cats in SNF group showed symptoms of lower urinary track disease. Thus, the cats that underwent double-pigtail ureteral stent fixation to the urinary bladder developed significantly fewer lower urinary diseases (P = 0.046). In conclusion, double-pigtail ureteral stent fixation to the urinary bladder for treatment of complete ureteral obstruction in cats can effectively prevent stent migration, which is common complication of stent placement.

The American Cockroach Peptide Periplanetasin-2 Blocks Clostridium Difficile Toxin A-Induced Cell Damage and Inflammation in the Gut

  • Clostridium difficile, which causes pseudomembranous colitis, releases toxin A and toxin B. These toxins are considered to be the main causative agents for the disease pathogenesis, and their expression is associated with a marked increase of apoptosis in mucosal epithelial cells. Colonic epithelial cells are believed to form a physical barrier between the lumen and the submucosa, and abnormally increased mucosal epithelial cell apoptosis is considered to be an initial step in gut inflammation responses. Therefore, one approach to treating pseudomembranous colitis would be to develop agents that block the mucosal epithelial cell apoptosis caused by toxin A, thus restoring barrier function and curing inflammatory responses in the gut. We recently isolated an antimicrobial peptide, Periplanetasin-2 (Peri-2, YPCKLNLKLGKVPFH) from the American cockroach, whose extracts have shown great potential for clinical use. Here, we assessed whether Peri-2 could inhibit the cell toxicity and inflammation caused by C. difficile toxin A. Indeed, in human colonocyte HT29 cells, Peri-2 inhibited the toxin A-induced decrease in cell proliferation and ameliorated the cell apoptosis induced by this toxin. Moreover, in the toxin A-induced mouse enteritis model, Peri-2 blocked the mucosal disruption and inflammatory response caused by toxin A. These results suggest that the American cockroach peptide Peri-2 could be a possible drug candidate for addressing the pseudomembranous colitis caused by C. difficile toxin A.

  • Cedrela sinensis is a broadleaf tree that is widely cultivated in Korea and China. It was used for treating enteritis, dysentery, and skin itch in oriental medicine. In this study, three major flavonoids, kaempferol-3-O-rhamnoside (1), quercetin-3-O-rhamnoside (2), and quercetin-3-O-glucoside (3), were isolated from the leaf of Cedrela sinensis. The biological activities of these compounds were tested by inhibitory activity of matrix metalloproteinases-2 (Type IV collagenase) method together with a cytotoxicity and a apoptosis test against human cancer cell lines.

Analysis of Outer Membrane Proteins of Yersinia enterocolitica Isolated from Mountainspring Water and Pig

  • Yersinia enterocolitica causes various diseases in humans, including enteritis. The onset of such diseases is closely related with the expression of important virulence factors, particularly outer membrane proteins (OMPs). The expression of OMPs depends on several factors, including temperature, and origin, biotype and serotype of the bacteria. Recently, concerns over food safety have increased along with the demand for the development of sensitive, rapid, and pathogen-specific detection methods. To develop a suitable detection method for Y. enterocolitica isolated from Korean moutainspring water and pig feces, the OMP expression patterns were analyzed phenotypically and immunologically using 12 representative strains from 51 Y. enterocolitica Korean isolates. A 38-kDa OMP was commonly observed in all strains. However, additional OMPs were also observed in different biotypes and serotypes as well as bacterial origins, by incubating Y. enterocolitica at a low temperature. The specificity of the 38-kDa OMP was confirmed by a Western blot analysis with antisera against Y. enterocolitica and Brucella abortus. The results, therefore, indicate that the 38-kDa OMP could be used as a marker for detecting Y. enterocolitica in the environment or for seromonitoring.

Health Effects and Utilization of the Levator Ani Muscles Training in Health Qigong Exercise - based on Chinese Literatures - (기공운동 제항의 효과 분석 및 활용 방안 - 중국 문헌을 중심으로 -)

  • Objectives : The Levator Ani Muscles Training(LAMT) is one of the well known very effective prevention and treatment method of some specific diseases in the anal organs, digestion system, urinary organs and genital organs, which means LAMT could be considered as an independent exercise. Methods : Chinese databases were searched and found one good source named CNKI(China National Knowledge Infrastructure) where there were downloaded 154 articles, out of which were finally selected 82 used for the introduction of the historical background, health-effectiveness, working mechanism, training methods, etc. Results : LAMT as an independent exercise, is considered very effective for the prevention of diseases in the anal organs, e.g. hemorrhoids, anal fissure, rectal prolapse; urinary organs, e.g. urinary incontinence, frequent micturition; digestion system, e.g. visceroptosis, diarrhea, chronic enteritis; genital organs, e.g. prostatitis, impotence, premature ejaculation, feminine vaginal relaxation, sexual indifference, etc. It is also told LAMT is effective for coronary heart disease, hyperyension and varicose veins of lower extremity to a certain degree. LAMT is also very similar to the Kegel exercise in couple of aspects. They are equally effective for the urinary incontinence and impotence. Conclusions : LAMT as an independent exercise has developed and become more and more health-effective, especially through the combination with the Qigong exercise, and thus many training methods have been introduced. There are not many scientific research and development with very limited accomplishments even in China. It is strongly required to strengthen the research and development activities so that LAMT will become one of the very effective natural healing soonest possible.
