Expression and pH-dependence of the Photosystem II Subunit S from Arabidopsis thaliana

  • Photosynthesis uses light energy to drive the oxidation of water at an oxygen-evolving catalytic site within photosystem II (PSII). Chlorophyll binding by the photosystem II subunit S protein, PsbS, was found to be necessary for energy-dependent quenching (qE), the major energy-dependent component of non-photochemical quenching (NPQ) in Arabidopsis thaliana. It is proposed that PsbS acts as a trigger of the conformational change that leads to the establishment of nonphotochemical quenching. However, the exact structure and function of PsbS in PSII are still unknown. Here, we clone and express the recombinant PsbS gene from Arabidopsis thaliana in E. coli and purify the resulting homogeneous protein. We used various biochemical and biophysical techniques to elucidate PsbS structure and function, including circular dichroism (CD), fluorescence, and DSC. The protein shows optimal stability at $4^{\circ}C$ and pH 7.5. The CD spectra of PsbS show that the conformational changes of the protein were strongly dependent on pH conditions. The CD curve for PsbS at pH 10.5 curve had the deepest negative peak and the peak of PsbS at pH 4.5 was the least negative. The fluorescence emission spectrum of the purified PsbS protein was also measured, and the ${\lambda}_{max}$ was found to be at 328 nm. PsbS revealed some structural changes under varying temperature and oxygen gas condition.

MHD WAVE ENERGY FLUXES GENERATED FROM CONVECTION ZONES OF LATE TYPE STARS

  • An attempt has been made to examine the characteristics of acoustic and MHD waves generated in stellar convection zones($4000\;K\;{\leq}\;T_{eff}\;{\leq}\;7000\;K$, $3\;{\leq}\;\log\;g\;{\leq}\;4.5$). With the use of wave generation theories formulated for acoustic waves by Stein (1967), for MHD body waves by Musielak and Rosner (1987, 1988) and for MHD tube waves by Musielak et al.(l989a, 1989b), the energy fluxes are calculated and their dependence on effective temperature, surface gravity and megnetic field strength are analyzed by optimization techniques. In computing magneto-convection models, the effect of magnetic fields on the efficiency of convection has been taking into account by extrapolating it from Yun's sunspot models(1968; 1970). Our study shows that acoustic wave fluxes are dominant in F and G stars, while the MHD waves dominant in K and M stars, and that the MHD wave fluxes vary as $T_{eff}^4{\sim}T_{eff}^7$ in contrast to the acoustic fluxes, as $T_{eff}^{10}$. The gravity dependence, on the other hand, is found to be relatively weak; the acoustic wave fluxes ${\varpropto}\;g^{-0.5}$, the longitudinal tube wave fluxes ${\varpropto}\;g^{0.3}$ and the transverse tube wave fluxes ${\varpropto}\;g^{0.3}$. In the case of the MHD body waves their gravity dependence is found to be nearly negligible. Finally we assesed the computed energy fluxes by comparing them with the observed fluxes $F_{ob}$ of CIV(${\lambda}1549$) lines and soft X-rays for selected main sequence stars. When we scaled the corrected wave fluxes down to $F_{ob}$, it is found that these slopes are almost in line with each other.

A Wavepacket Study on Translational Energy Distributions of the Photo-stimulated Desorbed Xe from an Oxidized Si(001) Surface

  • We report a quantum wavepacket study on the characteristic bimodal translational energy distribution of photostimulated desorbed Xe from an oxidized silicon (001) surface observed by Watanabe and Matsumoto, Faraday Discuss. 117 (2000) 203. We have simulated the theoretical translational energy distributions based on wavepacket calculations with a sudden transition and averaging model to reproduce the experiment. We discuss the desorption mechanism and suggest a very strong position dependence of the deexcitation processes for Xe/oxidized Si(001).

  • Orange-yellow phosphor has been synthesized by solid-state method for inorganic EL devices. Zinc sulfide is used as host material for the phosphor and the phosphor consists of copper and manganese as activators. The dependence of the photoluminescence (PL) and electroluminescence(EL) properties on the copper and manganese concentrations has been investigated.

저온에서 La2/3+xTiO3-δ (x = 0, 0.13)세라믹스의 전자전도특성 (Low-Temperature Electron Transport Properties of La2/3+xTiO3-δ (x = 0, 0.13))

  • The thermoelectric power and dc conductivity of $La_{2/3+x}TiO_{3-{\delta}}$ (x = 0, 0.13) were investigated. The thermoelectric power was negative between 80K and 300K. The measured thermoelectric power of x = 0.13 increased linearly with increased temperatures and was represented by $S_0+BT$. The x = 0 sample exhibited insulating behavior, while the x = 0.13 sample showed metallic behavior. The electric resistivity of x = 0.13 had a linear temperature dependence at high temperatures and a T3/2 dependence below about 100K. On the other hand, the electric resistivity of x = 0 has a linear relation between $ln{\rho}/T$ and 1/T in the range of 200 to 300K, and the activation energy for small polaron hopping was 0.23 eV. The temperature dependence of thermoelectric power and the resistivity of x = 0 suggests that the charge carriers responsible for conduction are strongly localized. This temperature dependence indicates that the charge carrier (x = 0) is an adiabatic small polaron. These experimental results are interpreted in terms of spin (x = 0.13) and small polaron (x = 0) hopping of almost localized Ti 3d electrons.

  • Recently, wind power generation is an emerging industry expanding its market rapidly thanks to the increasing need to solve the scarcity of fossil fuels and the risk of potential global warming. Wind power generation has shown to be an effective response plan to global warming, showing the most price competitiveness among the renewable energy sources by its higher efficiency. Therefore wind energy has attracted considerable attention as the industrial growth drive for the next generation. Considering Korea's high dependence of overseas energy resources, the importance of wind power is growing as the most effective alternative energy source to ensure energy security as well as becoming a key strategic industry for exports. In this study, the social and economic effects of the wind power industry is discussed and the current status and the future prospects of the wind energy market is also examined.

난류예혼합화염이 음파의 산란에 미치는 영향에 관한 연구 (The Effect of Turbulent Premixed Flame on the Wave Scattering)

  • Analytical investigation of acoustic wave scattering from turbulent premixed flames was conducted to evaluate the acoustic energy amplification/damping. Such acoustic energy change is attributed to the acoustic velocity jump due to flame's heat release. Small perturbation method up to second order and stochastic analysis were utilized to formulate net acoustic energy and the energy transfer from coherent to incoherent energy. Randomly wrinkled flame surface is responsible for the energy transfer from coherent to incoherent field. Nondimensional parameters that govern net acoustic energy were determined: rms height and correlation length of flame front, incident wave frequency, incidence angle, and temperature ratio. The dependence of net acoustic energy upon these parameters is illustrated by numerical simulations in case of Gaussian statistics of flame front. Total net energy was amplified and the major factors that affect such energy amplification are incidence angle and temperature ratio. Coherent (incoherent) energy is damped (amplified) with rms height and correlation length of flame front.

  • From the dependence the primary energy of about 97% on imported energy from abroad in Korea, 1997, and the pressure by international address to the Global Climate Change and environmental problem, we should do our best efforts to develop and diffuse of energy technologies. Also, the developed countries such USA, EU, Japan, and so on, are promoting and accelerating the development of energy technologies. From those point of views, we thought that Korea now in the time to provide the new and updated strategies to develop the energy technologies more actively for enhancing national capability, The Korea Institute of Energy Research(KIER), the complex research institution for energies except nuclear energy sponsored by government, prepared the mid and long term strategies and primary research projects, must be worked in and with the KIER, for the areas related to the energy conservation and efficiency increase, the energy environment, the alternative energy, and the diffusion of developed energy technologies.

Effects of Doping in Organic Electroluminescent Devices Doped with a Fluorescent Dye

  • The effect of doping on the energy transfer and charge carrier trapping processes has been studied in organic light-emitting diodes (OLEDs) doped with a fluorescent laser dye. The devices consisted of N,N'-diphenyl-N,N'-bis(3-methylphenyl)-1,1-biphenyl-4,4'-diamine (TPD) as a hole transporting layer, tris(8-hydroxyquinoline) aluminum ($Alq_3$) as the host, and a fluorescent dye, 4-dicyanomethylene-2-methyl-6-[2-(2,3,6,7-tetrahydro-1 H,5H-benzo[i,j]quinolizin-8-yl) vinyl]-4H-pyran) (DCM2) as the dopant. Temperature dependence of the current-voltage-luminescence (I-V-L) characteristics, the electroluminescence (EL) and photoluminescence (PL) spectra are studied in the temperature ranging between 15 K and 300 K. The emission from DCM2 was seen to be much stronger compared with the emission from $Alq_3$, indicative of efficient energy transfer from $Alq_3$ to DCM2. In addition, the EL emission from DCM2 increasd with increasing temperature while the emission from the host $Alq_3$ decreased. The result indicates that direct charge carrier trapping becomes efficient with increasing temperature. The EL emission from DCM2 shows a slightly sublinear dependence on the current density, implying the enhanced quenching of excitons at high current densities due to the exciton-exciton annihilation.
