Efficacy of New 6-Phytase from Buttiauxella spp. on Growth Performance and Nutrient Retention in Broiler Chickens Fed Corn Soybean Meal-based Diets

  • A total of 420 day-old male Ross chicks were weighed at d 1 of life and assigned to test diets to assess the efficacy of a new Buttiauxella spp. phytase expressed in Trichoderma reesei. Diets were: positive control (PC) adequate in nutrients and negative control (NC) diet (40% and 17% less available phosphorous (P) and calcium (Ca), respectively) supplemented with 6 levels of phytase 0, 250, 500, 750, 1,000, and 2,000 phytase units (FTU)/kg of diet. All diets had titanium dioxide as digestibility marker and each diet was allocated to ten cages (6 birds/cage). Diets were fed for 3 wk to measure growth performance, apparent retention (AR) on d 17 to 21 and bone ash and ileal digestibility (AID) on d 22. Growth performance and nutrient utilization was lower (p<0.05) for NC vs PC birds. Phytase response in NC birds was linear (p<0.05) with 2,000 FTU showing the greatest improvement on body weight gain (20%), feed conversion (7.4%), tibia ash (18%), AR of Ca (38%), AR of P (51%) and apparent metabolizable energy corrected for nitrogen (5.1%) relative to NC. Furthermore, phytase at ${\geq}750FTU$ resulted in AID of total AA commensurate to that of PC fed birds and at ${\geq}1,000FTU$ improved (p<0.05) AR of P, dry matter, and N beyond that of the lower doses of phytase and PC diet. In conclusion, the result from this study showed that in addition to increased P and Ca utilization, the new Buttiauxella phytase enhanced growth performance and utilization of other nutrients in broiler chickens in a dose-dependent manner.

Preparation and Nutritional Evaluation of Hatchery Waste Meal for Broilers

  • Hatchery waste including infertile eggs, dead embryos in shell, dead or low grade chicks was cooked at $100^{\circ}C$ for 15 minutes and then oven dried and ground. Hatchery waste meal (HWM) thus prepared contained 44.25% protein, 4,573 kcal/kg gross energy, 3,600 kcal/kg metabolizable energy, 30% ether extract, 1.9% fibre, 14% ash, 9.8% nitrogen free extract, 7.26% Ca, 0.84%P, 1.86% lysine and 0.66% methionine with no Salmonella and E. coli. In biological evaluation trial, significantly higher weight gain was observed in ration containing 12% HWM compared with that containing similar amount of fish meal. Protein efficiency ratio on the two rations was 3.96 vs 2.85; protein digestibility, 86.02 vs 71.9; net protein utilization, 64.9 vs 42.37 and biological value, 75.37 vs 58.84, respectively, indicating better balance of amino acids in HWM compared with fish meal. Growth performance trial on broiler chicks also revealed better weight gain and feed efficiency on ration containing 12% HWM than that containing similar level of fish meal.

  • Small size privately owned wastewater treatment plants have been difficult to treat their wasted sludge and maintain steady effluent quality compared with publicly owned wastewater treatment plants. Therefore, this study has focused on treatment efficiency enhancement, specially nitrogen removal efficiency by recycling dewatering filtrate as an alkalinity additive from filter press using $CaCO_3$. As the result, it was found that the optimal mixing ratio between the excess sludge and $CaCO_3$ was 1:2. The major operation parameters such as specific substrate utilization rate, specific nitrification rate, and specific denitrification rate were also improved 64% ($0.048-0.079mg\;BOD_5/mg\;MLVSS{\cdot}day$), 35% ($0.020-0.027mg\;NH_3-N/mg\;MLVSS{\cdot}day$) and 68% ($0.051-0.086mg\;NO_3{^-}-N/mg\;MLVSS{\cdot}day$), respectively, after the adoption of new methods. Therefore, both the problem of sludge treatment at small scale plants and the need for efficiency improvement could be solved.

Effects of Non-protein Energy Intake on Whole Body Protein Synthesis, Nitrogen Retention and Glucose Turnover in Goats

  • The responses of whole body protein and glucose kinetics and of nitrogen (N) metabolism to non-protein energy intake (NPEI) were determined using an isotope dilution approach and measurement of N balance in three adult male goats. The diets containing 1.0, 1.5 and 2.0 times ME maintenance requirement, with fixed intake of CP (1.5 times maintenance) and percentage of hay (33%), were fed twice daily for each 21 d experimental period. After an adaptation period of 11 d, N balance was determined over 3 d. On day 17, whole body protein synthesis (WBPS) and glucose irreversible loss rate (ILR) were determined during the absorptive state by a primed-continuous infusion of [$^2H_5$]phenylalanine, [$^2H_2$]tyrosine, [$^2H_4$]tyrosine and [$^{13}C_6$]glucose, with simultaneous measurements of plasma concentrations of metabolites and insulin. Ruminal characteristics were also measured at 6 h after feeding over 3 d. Nitrogen retention tended to increase (p<0.10) with increasing NPEI, although digestible N decreased linearly (p<0.05). Increasing NPEI decreased (p<0.01) ammonia N concentration, but increased acetate (p<0.05) and propionate (p<0.05) concentrations in the rumen. Despite decreased plasma urea N concentration (p<0.01), increased plasma tyrosine concentration (p<0.05), and trends toward increased plasma total amino N (p<0.10) and phenylalanine concentrations (p<0.10) were found in response to increasing NPEI. Increasing NPEI increased ILR of both glucose (p<0.01) and phenylalanine (p<0.05), but did not affect ($p{\geq}0.10$) that of tyrosine. Whole body protein synthesis increased (p<0.05) in response to increasing NPEI, resulting from increased utilization rate for protein synthesis (p<0.05) and unchanged hydroxylation rate of phenylalanine ($p{\geq}0.10$). These results suggest that increasing NPEI may enhance WBPS and glucose turnover at the absorptive state and improve the efficiency of digestible N retention in goats, with possibly decreased ammonia and increased amino acid absorption. In addition, simultaneous increases in WBPS and glucose ILR suggest stimulatory effect of glucose availability on WBPS, especially when sufficient amino acid is supplied.

Effects of Functional Oils on Coccidiosis and Apparent Metabolizable Energy in Broiler Chickens

  • The objective of the present study was to investigate the effects of a mixture of functional oils (Essential, Oligo Basics Agroind. Ltda) on performance response of chickens challenged with coccidiosis and the determination of apparent metabolizable energy (AME), nitrogen-corrected apparent metabolizable energy (AMEn), the coefficients of protein and ether extract digestibility and intestinal morphology of broilers fed with diets containing Essential. In Exp. 1, a completely randomized design (CRD) was used, with one control diet without Essential inclusion with coccidiosis (Eimeria acervulina, Eimeria maxima, and Eimeria tenella) challenged birds and two different inclusion rates of Essential (1.5 kg/ton and 2 kg/ton) with coccidiosis-challenged and non-challenged birds for each inclusion rate, using 10 replicates and 50 birds per experimental unit. After 7 d of coccidiosis challenge, the livability was approximately 10% lower (p<0.05) for the control group. Intestinal lesion scores were lower (p<0.05) in the anterior intestine and the cecum for the chickens supplemented. Feed efficiency and growth rate were improved in birds supplemented with Essential (p<0.05) before the coccidiosis challenge and during the first 7 d post infection. In Exp. 2, a CRD was used, with one control diet without Essential inclusion and one diet with inclusion of Essential (1.5 kg/ton), using nine replications and 33 chicks per pen. The diets with Essential yielded approximately 4% higher AME (p = 0.003) and $AME_n$ (p = 0.001). Essential supplementation increased villus height in the jejunum on d 14 (p<0.05). Villus height:crypt depth ratio for the supplemented birds was larger (p<0.05) in the jejunum on d 7, larger (p<0.05) in the jejunum and ileum on d 14. In conclusion, these functional oils improved the energy utilization and the livability and decreased lesions caused by coccidiosis in supplemented birds.

Intake, Digestibility In vivo, N Utilization and In sacco Dry Matter Degradability of Grass Silage Harvested at Three Stages of Maturity

  • The objective of this experiment was to study the effects of grass maturity at harvest on the nutritive value of grass silage (GS) in relation to voluntary intake, digestibility, nitrogen (N) utilization and in sacco dry matter (DM) degradability. Silage was cut from a sward dominated by orchardgrass (Dactylis glomerata L.) at the late-vegetative (early-cut), internode elongation (mediumcut) and flowering (late-cut) stages of growth. The DM yield at harvest was the lowest for early-cut silage (5.4 t/ha) and increased to 6.5 and 7.0 t/ha for the medium and late-cut silage respectively. As the crop matured, the crude protein (CP) concentration decreased significantly (p<0.05) and there was a marked increase in acid detergent fiber (ADF) concentration (p<0.001). The three different silages were offered to four 18-month old Charolais wether sheep to measure the voluntary intake, in vivo digestibility and N retention over four 21-day periods in an incomplete changeover design. Silage degradability characteristics were determined using four fistulated sheep to measure DM degradability over 3, 6, 12, 24, 48 and 72 h. There was a linear decrease in the voluntary intake of silage fresh matter, DM, organic matter (OM) and neutral detergent fiber (NDF), digestibility of DM, OM, NDF, ADF and CP, and digestibility of OM in DM (Dvalue) ($P_L<0.01$) as harvesting of grass was delayed. Nitrogen intake, N output in urine, N output in faeces and N balance also linearly decreased ($P_L<0.01$) with postponed harvesting of grass for silage. DM degradability and effective degradability (ED) significantly decreased with increasing maturity of grass at harvest. The results suggest that harvesting date has a significant influence on the nutritive value of GS in terms of intake, digestibility, N balance and in sacco degradability in the rumen. It was concluded that early harvest GS ensured higher intake, digestibility, N intake and DM degradability in comparison with the medium and the late cut GS as a result of improved rumen N efficiency and utilization probably due to a better balance of available energy and protein.

혐기성 암모늄 산화균의 활성에 대한 식종미생물, 히드라진 및 아질산성 질소 농도의 영향 (Effects of Seeding Microorganisms, Hydrazine, and Nitrite Concentration on the Anammox Activity)

  • Anammox (Anaerobic Ammonium Oxidation) bacteria is recently discovered microorganism which can oxidize ammonium to nitrogen gas in the presence of nitrite under anaerobic conditions. The anammox process can save an energy for nitrification and need not require a carbon source for denitrification, however, the start-up periods takes a long time more than several months due to the long doubling time (approximately 11 days). In order to find the effects of seeding microorganisms, hydrazine, and nitrite concentration on the enhancement of the anammox activity, five kinds of microorganisms were selected. Among the several kinds of seeding microorganisms, the granule from acclimated microorganisms treating high concentration of ammonia nitrogen (A-1) and sludge from piggery wastewater treatment plant (A-2) were found to have a high anammox activity. In the case of A-1, the maximum nitrogen conversion rate represented 0.4 mg N/L-hr, and the amount of nitrite utilization was high compared to those of other seeding microorganisms. The A-4 represented a higher nitrogen conversion rate to 0.7 mg N/L-hr although the ammonium concentration in the serum bottle was high as 200 mg/L. Meanwhile, the anaerobic granule from UASB reactor treating distillery wastewater showed a low anammox activity due to the denitrification by the remained carbon sources in the granule. Hydrazine, intermediate product in anammox reaction, enhanced the anammox activity by representing 1.4 times of nitrogen gas was produced in the test bottle than that of control, when 0.4 mM of $N_2H_4$ was added to serum bottle which contains 5 mM of nitrite. The high concentration of nitrite (10 mM) resulted in the decrease of the anammox activity by showing lower production of nitrogen gas compared to that of 5 mM addition of nitrite concentration. As a result of FISH (Florescence In-Situ Hybridization) experiment, the Amx820 probe showed a more than 13% of anammox bacteria in a granule (A-1).

Effects of Protein Supplement Sources on Digestibility of Nutrients, Balance of Nitrogen and Energy in Goats and Their In Situ Degradability in Cattle

  • The experiment was conducted to determine in situ rumen degradability of soybean meal (SM), fish meal (FM), sesame cake (SC) and Italian ryegrass hay (IRGH) and the effect of supplementing the above protein meals to IRGH on digestibility and balance of nutrients in three Saanen goats. For measuring the degradability, nylon bags containing each meal were incubated in the rumen of one fistulated dry cow for 3, 6, 12, 24, 36, and 48 hrs. Degradability revealed that SM protein was highly degradable in the rumen (99.1%), while FM protein was less degradable (76.8%) with SC protein being intermediate (91.2%) at 48 hrs of incubation (p < 0.01). Degradation rate of the potentially degradable fraction was estimated to be 12.12, 5.88 and 5.88%/hr for SM, FM and SC, respectively. In the metabolism trial, all goats were offered daily 900 g IRGH and one of the supplements, SM (100 g), FM (75 g) or SC (100 g). Intake of DM, OM, CP and GE was similar among diets. However, digestibility of OM for SC diet was significantly lower than that for diets supplemented with FM and SM (p < 0.10). Nitrogen (N) excreted in faeces (p < 0.05) and in urine (p < 0.10) was, respectively, higher and lower for SC diet than that for the other two diets. The same tendency was observed in energy losses in faeces (p < 0.10) and in urine (p < 0.05). There was no difference in energy loss in methane or in heat production among diets. Consequently, no significant difference was observed in N retention (2.13, 0.42 and -0.11 g/day for FM, SC and SM diet, respectively) or in energy retention (-1.49, -2.14 and -2.70 MJ/day for FM, SM and SC diet, respectively). These results showed that protein supplements affected the digestion of diets based on grass hay with 7.45% CP in DM in goats, although there was no significant influence on N or energy retention.

Effects of Coated Compound Proteases on Apparent Total Tract Digestibility of Nutrients and Apparent Ileal Digestibility of Amino Acids for Pigs

  • Two experiments were conducted to evaluate effects of coated compound proteases (CC protease) on apparent total tract digestibility (ATTD) of nitrogen (N) and energy, and apparent ileal digestibility (AID) of amino acids (AA) and nutrients in diets for pigs. In Exp. 1, 12 crossbred barrows (initial body weight: $20.79{\pm}1.94kg$) were housed in individual metabolism crates and allotted into 2 treatments with 6 piglets per treatment according to weight in a randomized complete block design. The 2 diets were corn-soybean meal basal diets with (0.2 g/kg) or without CC protease supplementation. The CC protease supplementation increased (p<0.05) the digestible and metabolizable N and energy values and the digestibility and retention rate of N in the diet. The ATTD of energy and nutrients had been improved (p<0.05) in the diet supplemented with CC protease. In Exp. 2, 12 crossbred barrows (initial body weight: $20.79{\pm}1.94kg$), fitted with T-cannulas at the distal ileum, were blocked by body weight into 2 groups with 6 pigs each. The diets were the same as those in Exp. 1. The CC protease increased (p<0.05) the AID of crude protein and some essential AA including arginine, isoleucine and leucine. The AID and ATTD of energy and nutrients had been improved (p<0.05) by supplemental CC protease, but the hindgut digestibility of nutrients was unaffected. Overall, the CC protease improved the ATTD of N and energy and AID of some indispensible AA and nutrients in the corn-soybean meal diet for pigs. Therefore, the CC protease supplement could improve the utilization of protein in the corn-soybean meal diet and thus contribute to lower N excretion to the environment.

단백질과 에너지 수준이 흰쥐의 질소와 에너지 이용에 미치는 영향 (Effect of Dietary Protein and Energy on the Nitrogen and Energy Utilization in Growing Rats)

  • 실험식이의 에너지수준은 저에너지(3,200kcal/kg) 중에너지(3,600kcal/kg), 고에너지(4,000kcal/kg)로 달리했으며, 저단백질(15%), 중단백질(25%), 고단백질(35%)의 3수준으로 하고, 지방수준은 ME 3,600kcal/kg에 대해서 저지방(10%), 중지방(20%), 고지방(40%)의 3수준으로 정하고 나머지 에너지는 전분과 포도당으로 보충한 9종의 식이에 대한 대사실험 결과를 요약하면 다음과 같다. 1. 고형물 조단백질 조지방, 탄수화물의 소화율은 저에너지 수준에서 중 고에너지 수준에서보다 훨씬 낮았으며, 중 고에너지 수준사이에 큰 차이가 없었다. 단백질 수준이 높아질수록 고형물과 탄수화물의 소화율은 약간씩 감소되는 경향이었으며, 단백질의 소화율은 증가되는 경향이었다. 조지방의 소화율은 식이 단백질의 영향을 받지 않았으나 에너지 수준이 높을 때 즉 식이에 포함된 지방함량이 높을 때 증가되었으며 암수 모두 같은 경향을 보였다. 각 영양소의 소화율은 고형물 약 83%, 조단백질 90%, 조지방 96%, 탄수화물 93%로서 단백질과 에너지 수준에 따라 거의 차이가 없었으며 암수 간에는 차이가 없었다. 2. 질소 축적율은 암수 모두 식이 단백질 수준이 증가할수록 감소되는 반면 식이 에너지 수준이 증가할수록 증가되는 것을 볼 수 있었다. 따라서 단백질 수준은 NRC 사양 표준치에서, 에너지 수준은 그 이상에서 암수 모두 질소 축적율이 가장 높았다. 3. 에너지 이용율은 암쥐의 경우 식이 단백질 수준이 증가할수록 감소되었으며 식이 에너지 수준이 증가할수록 증가되었다. 숫쥐의 경우 식이 단백질 수준이 증가함에 따라 감소되었으나, 암쥐와는 달리 중에너지 수준에서 가장 높았다. 즉 단백질 수준은 암수 모두 NRC 사양 표준치에서 에너지 대사율이 가장 높았으나 에너지 수준은 암쥐의 경우 NRC 사양 표준치 이상에서, 숫쥐의 경우 NRC 사양 표준치에서 각각 가장 높았다.
