Protein -surfactant interactions have been investigate by measuring ζ-potential of $\beta$-lactoglobulin-coated emulsion droplets and $\beta$-lactoglobulin in solution in the rpesenceof surfactant, with particular emphasis on the effect of protein heat treatment(7$0^{\circ}C$, 30min). When ionic surfactant (SDS or DATEM) is added to the protein solution, the ζ-potential of the mixture is found to increase with increasing surfactant concentration, indicating surfactant binding to the protein molecules. For heat-denatured protein,it has been observed that the ζ-potential tends to be lower than that of the native protein. The effect of surfactant on emulsions is rather complicated .With SDS, small amounts of surfactant addition induce a sharp increase in zeta potential arising from the specific interaction of surfactant with protein. With further surfacant addition, there is a gradual reductio in the ζ-potential, presumably caused by the displacement of adsorped protein (and protein-surfactant complex) from the emulsion droplet surfac by the excess of SDS molecules. At even higher surfactant concentrations, the measured zeta potential appears to increase slightly, possibly due to the formation of a surfactant measured zeta potential appears to increase slightly, possibly due to the formation of surfactant micellar structure at the oil droplet surface. This behaviour contrastswith the results of the corresponding systems containing the anionic emulsifier DATEM, in which the ζ-potential of the system is found to increase continuously with R, particularly at very low surfactant concentration. Overall, such behaviour is consisten with a combination of complexation and competitive displacement between surfactant and protein occurring at the oil-water interface. In addition, it has also been found that above the CMC, there is a time-dependent increase in the negative ζ-potential of emulsion droplets in solutions of SDS, possibly due to the solublization of oil droplets into surfactant micelles in the aqueous bulk phase.
In this paper, we proposed an emulsification method without using an emulsifier and investigated the effects of particle size distribution in fluids on dispersion stability. Surfactant-free oil in water emulsion was prepared with 1 % (w/w) of olive oil by using high speed agitation, high pressure and ultrasonic dispersion methods. The particle size, microscopic observation, and dispersion stability of each sample were evaluated and dispersion stability according to various dispersion methods was compared. As a result, the emulsion dispersed by the ultrasonic dispersion method showed the smallest particle size and uniform distribution of $0.07{\sim} 0.3{\mu}m$ and was the most stable in a 7 days stability evaluation. In the above experiment, four olive oil emulsions having different particle sizes were prepared using ultrasonic dispersion technology that was capable of producing stable emulsions. The dispersion stability of each samples with oil droplet sizes of (A) 0.1 to $0.5{\mu}m$, (B) 0.3 to $4{\mu}m$, (C) 1 to $10.5{\mu}m$ and (D) 2 to $120{\mu}m$, was observed for 7 days, and the relationship between the stability and performance was studied. Emulsion (A) with particle size less than $0.5{\mu}m$ displayed the dispersion stability showing below 5 % change in a 7 days stability evaluation. In the case of (B), (C), and (D) that had larger particle than $0.5{\mu}m$, the changes of dispersion stability were 10 %, 13 % and 35 % respectively. From these results, it was proved that dispersion stability of emulsion with uniform particle size of $0.5{\mu}m$ or less was confirmed to be very stable.
본 연구는 액적기반 미세유체 장치를 이용하여 단분산성 마이크로캡슐의 간단한 제조방법에 관한 것이다. 본 연구에서 제시한 제조 방법은 이중액적을 생성시키기 위해 기존의 복잡한 표면처리가 필요한 이중 유화과정을 대신하여 하나의 교차점을 가진 단일공정을 사용하고자 한다. 먼저, 분산상은 광중합이 가능한 ethoxylated trimethylolpropane triacrylate (ETPTA) 단량체와 fluorocarbon (FC-77) 오일을 사용하고 연속상은 poly(vinyl alcohol) (PVA) 수용액을 사용하였으며, 미세유체 채널 내부로 흘려 주면 하나의 교차점에 흐름이 집중되어 균일한 이중액적을 생성한다. 생성된 이중액적은 광중합을 통해 마이크로캡슐을 제조한다. 상기 방법은 ETPTA 유체의 부피유속을 조절하여 이중액적의 껍질두께 제어가 가능하고 연속상인 물의 부피유속을 조절하여 전체 직경을 제어할 수 있다. 더 나아가, 본 시스템을 사용하여 다양한 물질들을 함입한 마이크로캡슐을 제작할 수 있으며, 약물전달시스템의 응용 기술에 활용될 것으로 예측된다.
We have investigated the influence of system composition and preparation conditions on the particle size of vitamin E acetate (VE)-loaded nanoemulsions prepared by PIC(phase inversion composition) emulsification. This method relies on the formation of very fine oil droplets when water is added to oil/surfactant mixture. The oil-to-emulsion ratio content was kept constant (5 wt.%) while the surfactant-to-oil ratio (%SOR) was varied from 50 to 200 %. Oil phase composition (vitamin E to medium chain ester ratio, %VOR) had an effect on particle size, with the smallest droplets being formed below 60 % of VOR. Food-grade non-ionic surfactants (Tween 80 and Span 80) were used as an emulsifier. The effect of f on the droplet size distribution has been studied. In our system, the droplet volume fraction, given by the oil volume fraction plus the surfactant volume fraction, was varied from 0.1 to 0.3. The droplet diameter remains less than 350 nm when O/S is fixed at 1:1. The droplet size increases gradually as the increasing the volume fraction. Particle size could also be reduced by increasing the temperature when water was added to oil/surfactant mixture. By optimizing system composition and homogenization conditions we were able to form VE-loaded nanoemulsions with small mean droplet diameters (d < 50 nm). The PIC emulsification method therefore has great potential for forming nanoemulsion-based delivery systems for food, personal care, and pharmaceutical applications.
금속 에멀젼(metal emulsion)은 제강 공정의 효율성을 높이는 방법으로 수십 년 동안 연구되어 왔습니다. 본 연구는 육안으로 관찰하기 어려운 고온 실험의 단점을 보완하기 위해 상온에서 관찰 가능한 수모델을 이용하여 수행하였다. 슬래그 내 금속 에멀젼의 대신하여 증류수를 실리콘 오일에 적하하여 운동량 균형 방정식에 의한 계산 결과와 비교하는 실험을 하였다. 물방울의 하강 속도는 물방울의 직경과 유체(실리콘 오일)의 점도가 증가함에 따라 감소하였다. 교반 조건에서 실리콘 오일에서 물방울의 하강 속도를 시뮬레이션하기 위해 유체(실리콘 오일)의 유속을 입자 이미지 속도계(PIV) 방법으로 측정하였다. 물방울의 하강 속도 계산은 점성 실리콘 오일을 교반하거나 교반하지 않고 측정된 값과 잘 일치하였다.
Urushiol, a natural monomeric oil, was used to prepare a detergentless micro-emulsion with water and 2-propanol The formation of micro-emulsion was verified by conductivity measurements and dynamic light scattering. The conductivity data showed phase change dynamics, a characteristics of micro-emulsions, and subsequent dynamic light scattering study further confirmed the phenomenon. Average water droplet diameter was 10 nm to 500 nm when the molar ratio of 2-propanol ranged from 0.40 to 0.44 . Earlier studies were performed on toluene and hexane, in which the insoluble substrate in water phase was added to the solvents to be reacted on by enzymes. However, in the present urushiol system, urushiol was used as both solvent and substrate in the laccase polymerization of urushiol. The laccase activity in the system was examined using polymerization of urushiol. The laccase activity in the system was examined using syringaldezine as a substrate, and the activity increased rapidly near the molar ratio of 2-propanol at 0.4, where micro-emulsion started. The activity rose until 0.46 and fell dramatically thereafter. The study of laccase activity in differing mole fractions of 2-propanol showed the existence of an ‘optimal zone’, where the activity of laccase was significantly higher. In order to analyze urushiol polymerization by laccase, a bubble column reactor using a detergentless micro-emulsion system was constructed. Comparative study using other organic solvents systems were conducted and the 2-propanol system was shown to yield the highest polymerization level. The study of laccase activity at a differing mole fraction of 2-propanol showed the existence of an ‘optimal zone’ where the activity was significantly higher. Also, 3,000 cP viscosity was achieved in actual urushi processing, using only 1/100 level of laccase present in urushi.
5% corn oil과 17 mM Brij 700으로 제조한 corn oil-in-water emulsion(O/W)의 droplet의 크기는 첨가되는 phenol의 양이 많아질수록 지방구가 다소 커지는 경향을 보였으며, continuous phase의 surfactant 양은 대조구에 비해 다소 낮아졌다. O/W에 녹차로부터 분리한 phenol류를 100-200 ppm의 농도로 첨가하고 30일간 저장하며 hydroperoxide의 생성량을 측정한 결과 대조구에 비해 낮은 hydroperoxide 함량을 보여 유도기간이 길어지는 것으로 나타났으며, phenol 종류별 hydroperoxide 생성억제 효과는 BHT > procyanidin B3-3-O-gallate>(+)-gallocatechin>(+)-catechin의 순서로 나타났다. 지방 산화의 결과 발생하는 aldehyde의 함량을 측정함으로서 산화정도를 측정하기 위하여 headspace hexanal 양을 측정한 결과, 대조구는 15일까지 hexanal이 검출되지 않았으나 phenol 첨가구는 20-30일로 headspace aldehyde의 불검출기간이 길어졌으며, 농도가 증가할수록 낮은 함량을 나타내었다. Phenol 종류별 headspacealdehyde 생성 억제 효과는 BHT > procyanidin B3-3-O-gallate > (+)-gallocatechin >(+)-catechin의 순서로 나타났다.
나노에멀젼은 서브마이크론 크기의 에멀젼으로 치료 약물의 전달을 향상시키는 약물 전달체로서 많은 연구가 진행되고 있다. 본 연구의 목표는 폴리머를 함유한 나노에멀젼의 안정성에 대한 연구이다. 고농도의 Carbopol 941, Aristoflex AVC, Aronbis M, Permulen TR 2 및 Aculyn 44를 함유한 나노에멀젼의 안정성은 마크로에멀젼과는 다르게 불안정하였다. 폴리머를 유화 전에 첨가하여 제조한 에멀젼이 유화 후 폴리머를 첨가하여 제조한 에멀젼에 비해 초기 입자 크기가 더 크게 형성되었다. 저 농도의 폴리머를 함유한 경우에도 폴리머를 함유하지 않은 나노에멀젼에 비해 안정성이 감소하였으나 그 정도는 고농도에 비해 적었다. 점도가 유사한 경우 안정성에 미치는 순서는 Aristoflex AVC < Carbopol 941 < Permulen TR2 < Carbopol 941 + Aculyn 44 < Aronbis M의 순서로 불안정하였다.
천연계면활성제인 lysolecithin과 오일성분인 squalane(SQ.), liquid paraffin(LP), octylpalminate(OP), octylstearate(OS), alkylbenzoate(AB), isostearylbenzoate (ISB)를 사용하여 제조된 에멀젼의 입자의 크기, 형상을 광산란 장치를 사용하여 다음과 같은 결과를 얻었다. 동적광산란실험을 통하여 에멀젼입자는 크기가 150nm~250nm로서 subemulsion 임을 확인하였고, 오일상의 농도가 0.25wt%에서 입자의 크기가 감소하다가 증가하는 오일의 임계농도가 있음을 알 수 있었다. 그리고 SQ에서 ISB의 순으로 오일의 극성이 증가할수록 입자의 크기가 감소함을 알 수 있었다. 정적광산란을 통하여 산란강도의 각도의존성으로부터 관성회전반경(R$_{g}$)를 구할 수 있었고 R$_{g}$/R$_{h}$로부터 극성류인 ISB, AB의 입자형태는 구형, 반극성인 LP, SQ는 타원형 그리고 비극성류인 LP, SQ는 막대형에 가까운 형태임을 알 수 있었다. 제조된 에멀젼의 점도는 구형입자에 비하여 막대형 입자의 점도가 높음을 알 수 있었다.다.다.
본 실험에서는 화장품에 널리 사용되는 지방산과 지방알코올을 Tween 80과 Span 80을 함께 사용하여 조성 상전이 유화법으로 O/W (oil-in-water) 저점도 나노에멀젼을 제조하였다. 지방알코올의 오일상에서의 농도가 증가할수록 나노에멀젼의 입자 크기가 증가하였다. 혼합계면활성제의 HLB를 조절하여 입도 분포가 좁고 안정한 나노에멀젼의 제조가 가능하였다. 전상점 부근에서 지방산 및 지방알코올을 함유한 계 모두에서 유사한 점도와 전기전도도의 변화를 보였으나 오일과 계면활성제만으로 구성된 계와는 다르게 넓은 수용액상 분율범위에서 높은 점도를 나타냈다. 입자 크기가 100 nm 미만의 저점도 나노에멀젼은 실온에서 한 달 이상 안정하였다. 지방산 또는 지방알코올을 함유하고, 저에너지 유화법으로 제조된 저점도 O/W 나노에멀젼 제형은 화장품의 기재로서 널리 사용될 수 있다.
