Essential Cysteine Residues of Yeast Thioredoxin 2 for an electron donor to Thioredoxin Peroxidases

  • Thioredoxin (Trx) is a redox protein possessing conserved sequence Cys-Gly-Pro-Cys in ail organisms. Trx acts as an electron donor of many proteins including thioredoxin peroxidase (TPx). Yeast Trx 2 has two redox active cysteine residues at positions 31 and 34. To investigate the redox activity of each cysteine, we generated mutants C31S, C34S, and C31S/C34S using site directed mutagenesis and examined the redox activity of Trx variants as an electron donor for yeast TPx enzymes. None of the three Cysmutated Trx proteins was active as a redox protein in the 5', 5'-dithiobis-(2-dinitrobenzoic acid) reduction under the condition of the presence of NADPH and thioredoxin reductase, and in the thioredoxin dependent peroxidase activity of yeast TPx II. C34S enhanced the glutamine synthetase protection activity of yeast TPx I, even though 100 times more protein was needed to exhibit the same activity to WT. The formation of a mixed disulfide intermediate between Trx and TPx II subunits was analyzed by SDS-PAGE. The mixed dieter form of TPx II was found only for C34S. These results suggest that Cys-31 more effectively acts as an electron donor for TPx enzymes.

  • 各 種 아민(triethylamine, diethylamine, pyridine, diphenylamine, dimethylaniline)과 各種 Electron Acceptor($CCl_4,\;ICl,\;I_2$)를 Hexane 溶媒 存在下에서 作用시킨 結果 各各 Charge Transfer Complex를 形成하였으나 그 形成能은 아민의 Basicity에 比例하였고 Electron Donor의 Polarity의 差가 클 때에는 다른 形式의 Complex를 形成함을 紫外線 吸收 分光法으로 確認하였다. Complex 形成能과 아민의 Basicity 및 Electron Acceptor의 Polarity의 關係를 論하였다.

전하 이동을 이용한 실세스퀴옥산/폴리스티렌 하이브리드 (Silsesquioxane/Polystyrene Hybrid Materials via Charge Transfer Interactions)

  • Carbazole(electron donor)그룹과 dinitrobenzene(electron acceptor)그룹을 이용하여 전하 이동 작용이 실세스퀴옥산/고분자 하이브리드의 형성 메커니즘으로서 작용할 수 있는지 살펴보는 연구를 진행하였다. 하이브리드 실험은 새롭게 합성된 Poly(carbazole-styrene) (PS/D)와 dinitrobenzyl silsesquioxane (Cube/A)의 톨루엔 용액을 혼합/캐스팅을 하여 만들어진 필름을 이용하였으며 상분리가 없는 투명한 하이브리드 필름이 일부 조건에서 얻어졌다. PS/D및 Cube/A의 $^1H-NMR$분석, 그리고 하이브리드 필름들의 UV 흡수 실험은 실세스퀴옥산에 의한 입체 장애 효과가 없는 조건에서는 acceptor와 donor가 1:1로 전하 이동 착물을 형성할 수 있지만, 상분리가 없는 투명한 실세스퀴옥산 하이브리드는 acceptor/donor의 비율이 0.7 : 1 이하에서 형성된다는 것을 보여주었다. 이 결과들은 또 실세스퀴옥산 한 분자 당 평균 4개의 전하 이동 착물이 형성된다는 하이브리드 나노 구조에 대한 정보도 제공하였다.

염화에텐류 화합물 및 전자공여체가 VC 탈염소화 속도에 미치는 영향 (The Effect of Chlorinated Ethenes and Electron Donor on VC Dehalogenation Rate)

  • Anaerobic reductive dehalogenation of perchloroethene (PCE) was studied with lactate as the electron donor in a continuously stirred tank reactor (CSTR) inoculated with a mixed culture previously shown to dehalogenate vinyl chloride (VC). cis-1,2- dichloroethene (cDCE) was the dominant intermediate at relatively long cell retention times (>56 days) and the electron acceptor to electron donor molar ratio (PCE:lactate) of 1:2. cDCE was transformed to VC completely at the PCE to lactate molar ratio of 1:4, and the final products of PCE dehalogenation were VC (80%) and ethene (20%). VC dehalogenation was inhibited by cDCE dehalogenation. Propionate produced from the fermentation of lactate might be used as electron donor for the dehalogenation. Batch experiments were performed to evaluate the effects of increased hydrogen, VC, and trichloroethene (TCE) on VC dehalogenation which is the rate-limiting step in PCE dehalogenation The addition of TCE increased the VC dehalogenaiton rate more than an increase in the $H_2$ concentration, which suggests that the introduction of TCE induces the production of an enzyme that can comtabolize VC.

Effects of Energetic Disorder and Mobility Anisotropy on Geminate Electron-hole Recombination in the Presence of a Donor-Acceptor Heterojunction

  • Geminate electron-hole recombination in organic solids in the presence of a donor-acceptor heterojunction is studied by computer simulations. We analyze how the charge-pair separation probability in such systems is affected by energetic disorder of the media, anisotropy of charge-carrier mobilities, and other factors. We show that in energetically disordered systems the effect of heterojunction on the charge-pair separation probability is stronger than that in idealized systems without disorder. We also show that a mismatch between electron and hole mobilities reduces the separation probability, although in energetically disordered systems this effect is weaker compared to the case of no energetic disorder. We demonstrate that the most important factor that determines the charge-pair separation probability is the ratio of the sum of electron and hole mobilities to the rate constant of recombination reaction. We also consider systems with mobility anisotropy and calculate the electric field dependence of the charge-pair separation probability for all possible orientations of high-mobility axes in the donor and acceptor phases. We theoretically show that it is possible to increase the charge-pair separation probability by controlling the mobility anisotropy in heterojunction systems and in consequence to achieve higher efficiencies of organic photovoltaic devices.

A Novel Donor-Acceptor-Acceptor-Acceptor Polymer Containing Benzodithiophene and Benzimidazole-Benzothiadiazole-Benzimidazole for PSCs

  • New electron deficient acceptor-acceptor-acceptor type of monomer unit composed of weak electron accepting benzimidazole and relatively strong electron accepting benzothiadiazole derivatives namely 4,7-bis(6-bromo-1-(2-ethylhexyl)-1H-benzo[d]imidazol-2-yl)benzo[c][1,2,5]thiadiazole (BBB) was synthesized. The Stille polycondensation of the newly synthesized BBB monomer with electron donating 2,6-bis(trimethyltin)-4,8-bis(2-ethylhexyloxy)benzo[1,2-b:4,5-b']dithiophene (BDT) afforded donor-acceptor-acceptor-acceptor type of polymer namely 2,6-(4,8-bis(2-ethylhexyloxy)benzo[1,2-b:4,5-b']dithiophene)-alt-4,7-bis(1-(2-ethylhexyl)-1H-benzo[d]imidazol-2-yl)benzo[c][1,2,5]thiadiazole (PBDTBBB). The opto-electrical studies revealed that the absorption band of PBDTBBB appeared in the range of 300 nm-525 nm and its highest occupied molecular orbital (HOMO) and lowest unoccupied molecular orbital (LUMO) energy levels were positioned at -5.18 eV and -2.84 eV, respectively. The power conversion efficiency (PCE) of the polymer solar cell (PSC) prepared from PBDTBBB:PC71BM (1:2 wt %) blend was 1.90%.

  • In place of $Alq_3$ for EL, various Zn-Complex with fluorenscent chromophores were synthesized. PL of Zn-Complex substituted with electron donor group at 2 or 4 position occurred to bathochromic shift of emission$(\lambda_{max})$ and PL intensity was weaker than Zn-Complex non-substituted with electron donor group.

  • The charge transfer (CT) complex dyes between electron donor phenothiazine and electron acceptors namely, 2, 3-dichloro-1,4-naphtoquinone and 2,3-dichloro-5-nitro-1,4-naphtoquinone, were formed in the dichloromethane solution and electrochromic properties were studied using Bu4NC1O4 as supporting electrolyte. A 1 : 1 correspondence between the donor and acceptor molecules in the CT complex was found.

인삼 틸라코이드에서 Singlet Oxygen($^1$O$_2$) 생성에 미치는 전자전달계의 영향 (Effects of Light and Photosynthetic Electron Transport System on the Generation of Singlet Oxygen ($^1$O$_2$) in Ginseng Thylakoid Membrane)

  • In order to Investigate the mechanism of the leaf-burning disease of ginseng (Panax ginseng C.A. Meyer), studies on the generation of singlet oxygen (1O2) and the photooxidation of the pigments were carried out in comparison with the ones of soybean (G1ycine max L). The studies were mainly focalized on the effects of light intensity, light intensity, inhibitor and electron donor/acceptor of the Photosynthetic electron transport system. When we measured the amounts of 1O2 generated in the thylakoids of ginseng and soybean by the irradiation of light (300 w/m2) as a function its time. It was identified that a higher amount of 1O2 was formed in the ginseng thylakoid than the case of soybean. A generation ratio of lO2 between ginseng and soybean sltbstantially identical in the range of light intensities 50∼150w/m2 However much higher amount of 1O2 was generated in ginseng by irradiation of strong intensity of light (200 500w/m2). Wave length dependency on the generation of 1O2 and the pigment photooxidation was observed on ginseng thylakoids; red light (600-700 nm) gave a maximum effect in the contrast with blur green light (400-60 nm). When the ginseng thylalioid was treated with the electron donor (Mn2+) and acceptors (DCPIP, FeCy) of the photosynthetic electron transport system. a drastic inhibition of 1O2 generation was observed. However, treatment with its inhibitors (DCMU, KCW) activated 1O2 generation. An interesting fact that an electron donor or acceptor of the photosystem II(P680) Inhibited 1O2 generation, suggests an intimate relationship between 1O2 generation and photosystem II.

Self-Consistent Subband Calculations of AlGaN/GaN Single Heterojunctions

  • We present a self-consistent numerical method for calculating the conduction-band profile and subband structure of AlGaN/GaN single heterojunctions. The subband calculations take into account the piezoelectric and spontaneous polarization effect and the Hartree and exchange-correlation interaction. We calculate the dependence of electron sheet concentration and subband energies on various structural parameters, such as the width and Al mole fraction of AlGaN, the density of donor impurities in AlGaN, and the density of acceptor impurities in GaN, as well as the electron temperature. The electron sheet concentration was sensitively dependent on the Al mole fraction and width of the AlGaN layer and the doping density of donor impurities in the AlGaN. The calculated results of electron sheet concentration as a function of the Al mole fraction are in excellent agreement with some experimental data available in the literature.
