농축 블루베리 퓌레 열처리 시 pH 조절제의 품질 저하 억제 효과 (Quality Deterioration Suppressing Effects of pH Regulators during Heat Treatment of Concentrated Blueberry Puree)

  • Purpose: In this study, we aimed to minimize quality degradation of concentrated blueberry puree by thermal treatment at $90^{\circ}C$ for 5 min, with citric acid (CA) and acidic sodium metaphosphate (ASM) as pH modifiers. Methods: The color values, redness a, anthocyanin contents, DPPH radical scavenging, elastase inhibition and tyrosinase inhibition activities were evaluated for quality parameters of concentrated blueberry puree. Results: The highest values of color value, redness a, anthocyanin contents, DPPH radical scavenging, elastase inhibition and tyrosinase inhibition activities were 1% CA+0.5% ASM followed by 1% CA and control. The quality parameters of concentrated blueberry puree were significantly different between control and 1% CA+0.5% ASM treatment (p<0.05). Conclusion: During thermal treatment, quality degradation of concentrated blueberry puree was reduced by 1% CA+0.5% ASM treatment, which is expected for new acidulants.

오이풀 뿌리 분획물의 화장품 소재로서의 약리활성 연구 (The research of pharmacological activation for Sanguisorbae Radix Fractions as cosmetic material)

  • Objectives : Sanguisorbae Radix(SO) is a plant in the family Rosaceae, which grows widely in open fields Korea. It has been used as traditional medicine for thousands of years, as a treatment for anti-inflammatory and it is widely used for throat infection, tonsilitis, conjuctivitis and lymphadentis. In this study, investigated skin antiaging and anti-bacterial by using SO fractions water, acetone and butanol, chloroform. Methods : The effects of anti-microbial on SO fractions and elastase inhibition activity, collagenase inhibition activity were experimented. Results : 1. The ethyl acetate fraction showed the strongest antimicrobial activity against Staphylococcus epidermidis. 2. The elastase inhibition rate and collagenase inhibition rate of the water fraction of SO was the highest other factions. Conclusions : From the above results, it was confirmed the SO has sufficient potentiality applying itself to industry and also SO can be utilized as antimicrobial natural materials and antiaging cosmetics.

B16 Melanoma 세포에서 Phenylprlopanoid 화합물이 Melanin 생성에 미치는 영향 (Effects of Phenylpropanoid Compounds on Melanin Production in B16 Melanoma Cells)

  • To investigate the relationship between structure and biological activity of phenylpropanoids, we measured effects of phenylpropanoids on anti-oxidant and whitening activity, In DPPH radical scavenging activity, caffeic acid analogues showed the significant anti-oxidant activity. Although phenylpropanoids did not inhibit purified-tyrosinase activity, they significantly inhibited tyrosinase activity and melanin production in MSH-stimulated B16 melanoma cells. However, phenylpropanoids did not affect tyrosinase expression in MSH-stimulated B16 melanoma cells, which suggest that inhibition of MSH-induced melanin production was due to tyrosinase inhibition mediated via other signal pathways but not expression of tyrosinase. Phenylpropanoids also significantly inhibited both hyaluronidase and elastase activity, suggesting that phenylpropanoids may be used as whitening, hydration and anti-wrinkling agents. Hydroxyl residue of aromatic ring in phenylpropanoids plays an important role in anti-oxidant and whitening activity.

지황 추출물 첨가 chitosan 기반 기능성 바이오 소재 제조 및 응용 (Preparation and Application of Rehmannia Glutinosa Extract Incorporated Functional Chitosan Based Biomaterials)

  • 본 연구에서는 지황 추출물[Rehmannia glutinosa extract (RE)]이 함유된 chitosan (CH) 기반 바이오 소재를 제조하고, 물리·화학적 특성, RE 방출 특성, 멜라닌 합성 억제 효과, 항산화 효과, elastase 억제 효과를 조사하였다. RE 함유 CH기반 바이오 소재는 casting method와 UV 경화 공정을 통해 제조되었다. 제조된 바이오 소재의 표면 특성은 FE-SEM으로 분석하였고, 물리적 특성은 인장강도, 신축률을 통하여 조사하였다. 경피 약물 전달 시스템을 적용하기 위해 36.5 ℃에서 pH 4.5, pH 5.5, pH 6.5 용액에서와 인공피부를 이용해 제조된 바이오 소재에서 RE의 방출 특성을 조사하였으며, 그 결과, pH 6.5에서 가소제를 첨가하지 않은 바이오 소재와 가소제[glycerol (GL)와 citric acid (CA)]를 첨가하여 제조한 RE 함유 바이오 소재에서 RE의 방출이 pH 4.5에서 보다 약 1.10배 빠르게 일어남을 알 수 있었다. 또한, 인공 피부에서 RE의 방출은 약 6 h 동안 지속적으로 방출됨을 확인하였다. Tyrosinase assay, 2,2-diphenyl-1-picrylhydrazyl(DPPH) assay, elastase assay를 통해 기능성을 평가하였으며, 가소제로 CA를 첨가하고 RE가 함유된 바이오 소재에서 각각 45.12%의 tyrosinase 활성율, 89.40%의 DPPH 라디칼 소거능, 59.94%의 elastase 활성율을 나타내었다.

황백추출물 및 분획물의 Nitric Oxide 생성 억제 효과 및 Elastase 억제 효과 (Nitric Oxide Production and Elastase Inhibitory Activities of Extract and Its Fraction from Phellodendri Cortex)

  • Background: To date, the anti-wrinkle efficacy of phellodendri cortex has not been defined. In this study, we investigated the nitric oxide (NO) production and elastase inhibitory activities of 80% methanol extract of Phellodendri cortex and its ethyl acetate fraction. Methods and Results: We prepared 80% methanol extract, and its fractions from phellodendri cortex. The treatment of RAW 264.7 cell with $25{\mu}g/m{\ell}$ 80% methanol extract and ethyl acetate fraction resulted in no toxicity. We conducted assays of nitric oxide (NO) production and elastase inhibition. In the NO production assay, the ethyl acetate fraction showed an inhibitory effect approximately 17 times stronger than the 80% methanol extract. In elastase inhibitory assay, the ethyl acetate fraction also showed a stronger effect than the 80% methanol extract. In order to standardize the extract and fraction, we used TLC to separate the extract and observed the plate under UV light. We confirmed that the known pharmacological ingredients berberine, and palmatine in the 80% methanol extract and the ethyl acetate fraction. Conclusions: These results indicated that phellodendri cortex extract and its ethyl acetate fraction produced strong inhibitory effect on elastase and NO production.

도인승기탕 및 그 구성약재의 화장품약리활성 (Cosmeceutical Activities of Doinseunguitang and Its Composition)

  • Objectives : Cosmeceutical activities such as anti-oxidative and anti-aging effects of the Doinseunguitang and its composition, a traditional prescription, were evaluated. Methods : We performed MTT assay, melanin synthesis inhibition assay, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, tyrosinase inhibition assay, elastase and collagenase inhibition assay. Results : The results were obtained as follows : DPPH free radical scavenging of water extract Doinseunguitang (DISG) and ethanol extract DISG was 60% and 50% at 1,000 ppm. Xanthine oxidase inhibition effect of ethanol extract showed more than 80% at 500 ppm. Tyrosinase inhibition and inhibition melanin synthesis activities were measured in 40% and 50% at 1,000 ppm and 100 ppm. Elastase and collagenase inhibition rate of ethanol extracts DISG 40% and 80% at 1,000 ppm. It was concluded that compositive ingredients (Persicae Seman, Glycyrrhizae Radix, Cinnamomi Ramulus, Rhei Rhizoma) influenced the most results. Conclusions : The results indicated that, ethanol extract which is superior in its anti-oxidative and anti-aging effects is useful to be applied in herbal cosmetic industry.

연근 추출물에서 주름개선 소재의 개발 (Development of Anti-Wrinkle Agent from Nelumbo nucifera Root Extract)

  • 본 연구에서는 연근의 추출물에서 기능성화장품 소재를 개발하고자 하였다. 용매는 70~100%의 에탄올을 이용하였으며, 연근의 유효성분으로 nuciferine을 확인하였다. 연근 추출물의 화장품소재로서의 응용가능성을 보기 위해 안전성시험(MTT assay), 주름개선 효과시험(Elastase 활성저해 시험) 및 항산화효과시험(DPPH free radical scavenging assay)을 실시하였다. 100% 에탄올로 연근을 추출할 경우 $100{\mu}g/ml$의 농도에서 세포생존율이 80% 이상이어서 연근 추출물은 화장품소재로서 비교적 안전함을 알 수 있었다. 또한 연근 추출물은 $100{\mu}g/ml$의 농도에서 elastase 활성억제가 40~50%를 보여주어서 비교적 우수한 주름개선 효과를 나타내었다. 50% 항산화 저해능을 보이는 농도($FSC_{50}$)는 $5.0{\sim}38{\mu}g/ml$로 연근 추출물의 항산화효과가 매우 우수하였다. 따라서 연근 에탄올추출물은 주름개선 기능성화장품 소재로서 그 가능성이 높음을 알 수 있었다.

Anti-oxidation and Anti-wrinkling Effects of Jeju Horse Leg Bone Hydrolysates

  • This study focused on the anti-oxidative and collagenase- and elastase inhibition effects of low molecular weight peptides (LMP) from commercial Jeju horse leg bone hydrolysates (JHLB) on pancreatin, via enzymatic hydrolysis. Cell viability of dermal fibroblasts exposed to UVB radiation upon treatment with LMP from JHLB was evaluated. Determination of the antioxidant activity of various concentrations of LMP from JHLB were carried out by assessing 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis-3-ethybenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power (FRAP), and oxygen radical absorbance capacity (ORAC). The DPPH radical scavenging activity of LMP from JHLB (20 mg/mL) was 92.21% and ABTS radical scavenging activity (15 mg/mL) was 99.50%. FRAP activity (30 mg/mL) was $364.72{\mu}M/TE$ and ORAC activity (1 mg/mL) was $101.85{\mu}M/TE$. The anti-wrinkle potential was assessed by evaluating the elastase- and collagenase inhibition potential of these LMP. We found that 200 mg/mL of LMP from JHLB inhibited elastase activity by 41.32%, and 100 mg/mL of LMP from JHLB inhibited collagenase activity by 91.32%. The cell viability of untreated HS68 human dermal fibroblasts was 45% when exposed to a UVB radiation dose of $100mJ/cm^2$. After 24 h of incubation with $500{\mu}g/mL$ LMP from JHLB, the cell viability increased to 60%. These results indicate that LMP from JHLB has potential utility as an anti-oxidant and anti-wrinkle agent in the food and cosmetic industry. Additional in vivo tests should be carried out to further characterize these potential benefits.

감 추출물의 항산화, 항노화 및 미백 기능 규명 (Identification of Anti-oxidant, Anti-aging, and Whitening Effects of Diospyros kaki Extracts)

  • 본 연구에서는 감(Diospyros kaki)의 1,3-butylene glycol 추출물(DBG)과 에탄올 추출물(DET)를 이용하여 항산화, 항노화 및 미백 기능을 규명하였다. 항산화능은 DPPH와 superoxide dismutase (SOD) assay를 통해 규명하였으며, 그 결과 DBG와 DET 모두 현저한 DPPH 소거능 및 SOD-유사능을 보여주었다. 또한 elastase와 hyaluronidase 억제능 그리고 matrix metalloproteinase (MMP-1) 발현을 통해 항노화 기능을 규명하였다. DBG와 DET 모두 농도의존적으로 elastase와 hyaluronidase를 억제하였으며, MMP-1 발현 역시 저해하였다. 마지막으로 미백 기능을 규명하기 위하여 tyronisinase 억제 및 melanin 생성 저해능을 알아보았는데, DBG 추출물만이 tyrosinase를 저해할 수 있었다. 요약하면, 감 추출물은 강한 항산화, 항노화, 미백 기능을 갖고 있으며, 추후 화장품 소재로 활용이 가능할 것으로 사료된다.

New anti-wrinkle cosmetics

  • In the aged skin especially in the face and eyelid, deep and slight wrinkles are one of the remarkable phenomena of aging and the cause of wrinkle is various. Among the cause of wrinkles an oxidative stress plays an important roles in wrinkle formation process. It caused the lipid peroxidation of cell membrane, the increase of the MMPs(MatrixMetalloProteinase) gene expression and cellular DNA damage. These ROS induced materials may cause the degradation of collagen matrix system in the dermis and cause the formation of skin wrinkle. So, it is very important for protecting skin wrinkle formation to regulate ROS activity. In this study, we developed one active ingredient having multi functional activities such as activation of collagen synthesis, inhibition of MMPs activity, lipid peroxidation and free radical scavenging activity and inhibition of free radical induced DNA damage in vitro. Pericarpium castaneae extracts showed collagen synthesis increase in Normal Human Fibroblast and the inhibition of elastase activity (IC$\_$50/ of Elastase: 43.9$\mu\textrm{g}$/㎖). It showed also anti-oxidative activity (IC$\_$50/ : 48$\mu\textrm{g}$/㎖) and free radical scavenging activity(IC$\_$50/: 7.6$\mu\textrm{g}$/㎖). Conclusively, Pericarpium castaneae extracts may be used as an ingredient for new anti-wrinkle cosmetics.
