감태(甘苔)에서 추출한 Dieckol 성분이 쥐의 인지능력에 미치는 영향 (Effect of Ecklonia cava Extracts Supplementation on cognitive ability in mice)

  • Objectives : The purpose of this study aimed to investigate that dieckol - isolated from Ecklonia cava - supplementation can improve cognitive ability in mice. Methods : 48-male mice(6 weeks old) were divided into four groups; High-Dieckol group(n=12), Low-Dieckol group(n=12), Placebo group(n=12), Control group(n=12) and they were administered orally 5 days per week for 4 weeks at the same time. We performed Morris water maze test, Passive avoidance test, Blood serotonin analysis. And there was examined on neurogenesis in dentate gyrus of hippocampus using 5-bromo-2'-deoxyuridine (BrdU) to label proliferating cells. Results : The results are as follows; As a Morris water maze results, Trial duration was significantly decreased in high dieckol group comparing to placebo group and control group. Distance to target was significantly decreased in high dieckol group and low dieckol group comparing to placebo group and control group. Mean speed was significantly low in high dieckol group comparing to low dieckol group, placebo group and control group. As a Passive avoidance test results, latency time was significantly long in high dieckol group comparing to low dieckol group, placebo group and control group. BrdU cell count was significantly high in high dieckol group comparing to low dieckol group, placebo group and control group. Conclusions : As a conclusion, it is considered that dieckol supplementation might improve learning and cognitive ability.

Protective Effect of Fucoidan Extract from Ecklonia cava on Hydrogen Peroxide-Induced Neurotoxicity

  • We evaluated the antioxidant activity and neuronal cell-protective effect of fucoidan extract from Ecklonia cava (FEC) on hydrogen peroxide ($H_2O_2$)-induced cytotoxicity in PC-12 and MC-IXC cells to assess its protective effect against oxidative stress. Antioxidant activities were examined using the ABTS radical scavenging activity and malondialdehyde-inhibitory effect, and the results showed that FEC had significant antioxidant activity. Intracellular ROS contents and neuronal cell viability were investigated using the DCF-DA assay and MTT reduction assay. FEC also showed remarkable neuronal cell-protective effect compared with vitamin C as a positive control for both $H_2O_2$-treated PC-12 and MC-IXC cells. Based on the neuronal cell-protective effects, mitochondrial function was analyzed in PC-12 cells, and FEC significantly restored mitochondrial damage by increasing the mitochondrial membrane potential (${\Delta}{\Psi}m$) and ATP levels and regulating mitochondrial-mediated proteins (p-AMPK and BAX). Finally, the inhibitory effects against acetylcholinesterase (AChE), which is a critical hydrolyzing enzyme of the neurotransmitter acetylcholine in the cholinergic system, were investigated ($IC_{50}$ value = 1.3 mg/ml) and showed a mixed (competitive and noncompetitive) pattern of inhibition. Our findings suggest that FEC may be used as a potential material for alleviating oxidative stress-induced neuronal damage by regulating mitochondrial function and AChE inhibition.

  • This study was conducted to develop and characterize of a high quality salted mackerel using enzymatic extracts of Ecklonia cava (EEC). In this study, potential antioxidative properties of EEC were evaluated by DPPH free radical scavenging activity, hydrogen peroxide scavenging activity, peroxide value, and fatty acid composition, and the antimicrobial properties were also measured by analysis for volatile basic nitrogen, pH, viable cells, Eschericia coli and biogenic amine. Compared to EEC-untreated salted mackerel, the salted mackerel with EEC was superior in antioxidative properties, while was negligible in the difference of antimicrobial properties. These results suggested that the high quality salted mackerel with antioxidative activity could be developed by treatment of EEC.

Evaluation of phlorofucofuroeckol-A isolated from Ecklonia cava (Phaeophyta) on anti-lipid peroxidation in vitro and in vivo

  • Lipid peroxidation means the oxidative degradation of lipids. The process from the cell membrane lipids in an organism is generated by free radicals, and result in cell damage. Phlorotannins, well-known marine brown algal polyphenols, have been utilized in functional food supplements as well as in medicine supplements to serve a variety of purposes. In this study, we assessed the potential anti-lipid peroxidation activity of phlorofucofuroeckol-A (PFF-A), one of the phlorotannins, isolated from Ecklonia cava by centrifugal partition chromatography in 2,2-azobis (2-amidinopropane) dihydrochloride (AAPH)-stimulated Vero cells and zebrafish system. PFF-A showed the strongest scavenging activity against alkyl radicals of all other reactive oxygen species (ROS) and exhibited a strong protective effect against ROS and a significantly strong inhibited of malondialdehyde in AAPH-stimulated Vero cells. The apoptotic bodies and pro-apoptotic proteins Bax and caspase-3, which were induced by AAPH, were strongly inhibited by PFF-A in a dose-dependent manner and expression of Bcl-xL, an anti-apoptotic protein, was induced. In the AAPH-stimulated zebrafish model, additionally PFF-A significantly inhibited ROS and cell death, as well as exhibited a strong protective effect against lipid peroxidation. Therefore, these results suggest that PFF-A has excellent protective effects against ROS and lipid peroxidation induced by AAPH in both an in vitro Vero cell model and an in vivo zebrafish model.

감태 에탄올 추출물의 Trypsin 저해활성에 대한 열 및 pH의 영향 (Effect of Temperature and pH on Trypsin Inhibitory Activity of Ethanol Extracts from Ecklonia cava)

  • This study was performed to investigate the inhibitory activity of ethanol extract from Ecklonia cava (EE-EC) against trypsin and the stability of this activity under various heat and pH conditions. As a results, The EE-EC showed trypsin inhibitory activity of 77, 54, and 32% at concentrations of 5, 2.5, and 1 mg/mL and was not affected by the heat treatment conditions used in this study. Whereas trypsin inhibitory activity of EE-EC was stable in the pH range of 2-8, but decreased with pH treatment of pH 10 compared with the control. Therefore, the EE-EC could be useful as a natural and functional agent.

강황, 상백피 및 감태 추출 혼합물을 첨가한 천연 주방세제 개발 (Development of Natural Dishwashing Liquid containing the Curcuma Longa L., Morus alba and Ecklonia cava extracts)

  • This study was conducted to investigate the quality characteristics of the natural dishwashing liquid added with the mixture extracts of Curcuma Longa L. (CL), Morus alba (MA) and Ecklonia cava (EC) at various concentrations. The pH and lightness of the natural dishwashing liquid were reduced by adding the mixture extracts, while the turbidity was increased. The natural dish washing liquid added with mixture extracts was shown to display strong antimicrobial activities against L. monocytogenes compared to that of control. Also, it revealed that antioxidant activity was increased depending on concentrations. However, natural dishwashing liquid added with mixture extracts showed the low detergency efficiency. In sensory evaluation, the natural dishwashing liquid containing CL 0.5%, MA 0.25% and EC 0.25% was preferred than the control and it showed negative result in skin patch test. These results suggest that the addition of CL 0.5%, MA 0.25% and EC 0.25% positively improved the qualities characteristics in the natural dish washing liquid.

  • The Ecklonia cava Kjellman by-product (ECBP) as a feed additive was evaluated in improvement of productivity and immune enhancement against Salmonella Gallinarum (SG). Lohmann Brown chickens proved SG-free were randomly divided into 3 groups of 8 chickens each. Chickens were fed with the experimental diet treatment: T0, Non treatment-commercial feed; T1, commercial feed with 0.5% ECBP; T2, commercial feed with 0.1% Lactobacillus plantarum. In this study, we evaluated the effect of T1 and T2 groups on the body weight and protective efficacy against SG in chickens. The results demonstrated that treatment of T1 group as a feed additive affected significantly body weight gaining in chickens. In addition, T1 group showed a significant different colonization of SG when compared to T2 and T0 groups. We also studied that serum IgG and $interferon-{\gamma}$ levels were significantly different compared with other treatment groups. Therefore, we suggest that ECBP can be used as a good candidate of feed additives in chicken industry.

감귤박 및 감태추출물의 사료첨가제 급여에 따른 둥근전복 (Haliotis discus discus)의 성장 및 생리적 변화 (Effects of dietary supplementation with citrus pomace and Ecklonia cava residue on the physiological changes and growth of disk abalone, Haliotis discus discus)

  • Here, we report the physiological changes and growth in disk abalone, Haliotis discus discus, in relation to dietary supplementation with citrus pomace (CP) 6%, Ecklonia cava residue (ECR) 6%, and CP + ECR (3% + 3%). The composition and nutrient content, survival rate and growth rate were measured 0, 4, 8 and 12 weeks after feeding the supplemented diets of CP and/or ECR. Moreover, the experiment of low salinity stress (25psu) for environmental resistance was examined for a period of 48 hours after feeding the supplemented diets for 12 weeks. The activities of superoxide dismutase (SOD), catalase (CAT), lysozymes, respiratory burst, and phenoloxidase were measured. The moisture content and crude protein condition of the body were increased with the addition of ECR only (P<0.05). We observed higher levels of survival in the experimental group compared with the control group. Moreover, the growth disk abalone that were fed a diet containing ECR was higher compared with the control group. However, the growth of abalone fed a diet containing CP was similar to the control group. With a rearing condition of low salinity stress, survival rate and lysozyme activity were increased in the ECR group compared with the control group. Dietary ECR reduced the level of CAT activity to approximately 30% of the control, however the level of CAT activity in the ECR group was similar to the start level of the previous stress. These results suggest that dietary ECR gives rise to an enhanced immunity in disk abalone, as a result of the decrease in CAT and lysozyme activity in particular. Accordingly, the growth and survival rate were increased by feeding an ECR-supplemented diet in the rearing of disk abalone, Haliotis discus discus.

Therapeutic Potential of Ethanolic Extract of Ecklonia cava on Inhibition of Inflammation

  • 만성염증은 치주염, 대장염, 간염 및 관절염과 밀접한 관련성이 있다고 알려져 있다. 최근에 항염증제가 해양자원으로부터 개발되고 있다. 본 연구에서는 감태 (EC)가 항염증효과가 있다는 것이 발견되었다. 갈조류에 속하는 감태의 에탄올 추출물이 RAW 264.7 세포에서 tumor necrosis factor-${\alpha}$ interleukin-$1{\beta}$, interleukin-6 and prostaglandin $E_2$와 같은 염증매개체의 생성에 탁월한 효과를 나타내었다. 더욱이 reporter gene assay 및 western blot 분석 에 서 감태추출물은 대식세포에서 염증매개체의 발현을 조절하는 NF-${\kappa}B$ 전사인자의 불활성화를 통하여 항염증효과를 나타내었다. 뿐만 아니라 감태추출물은 만성염증에 중요한 역할을 하는 기질금속단백질의 활성을 억제 하였다. 이러한 결과는 갑태추출물이 만성염증을 억제하는데 잠재적으로 이용될 수 있다는 것을 암시하고 있다.
