Isolation and Characterization of a Rice Mitochondrial Small Heat Shock Protein Gene

  • To understand the role of small heat shock protein (sHSPs) in rice plant response to various stresses such as the heat and oxidative stresses, a cDNA encoding a 24.1 kDa mitochondrial small HSP (Oshsp24.1) was isolated from rice by rapid amplification of cDNA ends (RACE) PCR. The deduced amino acid sequence shows very high similarity with other plant small HSPs. DNA gel blot analysis suggests that the rice genome contains more than one copy of Oshsp24.1. High level of expression of Oshsp24.1 transcript was observed in rice seedlings in response to heat, methyl viologen, hydrogen peroxide, ozone, salt and heavy metal stresses. Recombinant OsHSP24.1 protein was produced in E. coli cells for biochemical assay. The protein formed oligomeric complex when incubated with Sulfo-EGS (ethylene glycol bis (succinimidyl succinate)). Our results shows that Oshsp24.1 has an important role in abiotic stress response and have potential for developing stress-tolerant plants.

A study of detector size effect using Monte Carlo simulation

  • The detector size effect due to the spatial response of defectors is one critical source of inaccuracy in clinical dosimetry and has been a subject of numerous studies. Conventionally, the detector response kernel contains all of the influence that the detector size has on the measured beam profile. Various analytic models for this kernel have been proposed and studied in theoretical and experimental works. Here, we use a method to determine detector response kernel simply by using Monte Carlo simulation and convolution theory. Based on this numerical method and DOSIMETER, an EGS4 Monte Carlo code, the detector response for a Farmer type ion chamber embedded in water phantom is obtained. There exists characteristic difference in the simulated chamber readings between one with carbon graphite wall and the other with Acrylic wail. Using the obtained response and the convolution theory, we are planning to derive the detector response kernel numerically and remove detector size effect from measurements for 6MV, 10${\times}$l0cm2 and 0.5${\times}$10 cm2 photon beam.

  • Using Monte Carlo calculations the effects of longitudinal magnetic fields on the beam profiles produced by clinical electron beam were studied. The Monte Carlo calculations were performed using the EGS4 code system modified to account for external magnetic fields. The beam profiles for a 6 MeV electron beam with longitudinal magnetic fields of 0.5-3.0 T were calculated. As a result of these calculations we found that the penumbra widths can be reduced with increased magnetic fields. This means that the electron therapy benefits from the external magnetic fields.

  • Using the electron microscopical method, this study was attempted to investigate the ultrastructural characteristics of glomeruli in FGS/Kist mice showing high proteinuria. The mucus-like substances were observed in glomerular capillary lumen which were usually narrowed by the widening of mesangial area filled with electron-dense deposits. Some portions of basal lamina in capillary wall were projecting to urinary space. Moreover, many vacuoles were observed in the cytoplasm of visceral epithelial cells, and an irregular-shaped fusion or effacement of foot processes were often shown in areas adjacent to basal lamina of glomeruli.

Beta Dosimetry in Intraperitoneal Administration of 166Ho-chitosan Complex (166Ho-chitosan 복합체의 복강 내 투여를 위한 베타선 흡수선량 평가)

  • Intraperitoneal administration of radioisotopes is suggested to treat the metastatic ovarian cancer in the peritoneal cavity. Administering beta-emitting radioisotopes into the peritoneal cavity allows the maximum energy delivery to the cancerous cells of the peritoneal wall surface while sparing the normal cells located in deep site of the peritoneal wall. In this study, dose estimates of the peritoneal wall are provided to be used for prescribing the amount of $^{166}Ho$-chitosan complex administered. The $^{166}Ho$-chitosan complex diffused in the peritoneal fluid may attach to the peritoneal wall surface. The attachment fraction of $^{166}Ho$-chitosan complex to the peritoneal wall surface is obtained by simulating the ascites with Fischer rats. Both volume source in the peritoneal fluid and the surface source over the peritoneal wall surface are counted for the contribution to the peritoneal wall dose. The Monte Carlo code EGS4 is used to simulate the energy transfer of the beta particles emitted from $^{166}Ho$. A plane geometrical model of semi-infinite volume describes the peritoneal cavity and the peritoneal wall. A semi-infinite plane of $10{\mu}m$ in thickness at every 1 mm of depth in the peritoneal wall is taken as the target in dose estimation. Greater than 98 percents of attachment fraction has been observed from the experiments with Fischer rats. Given $1.3{\mu}Ci/cm^2$ and $2.4{\mu}Ci/ml$ of uniform activity density, absorbed dose is 123 Gy, 8.59 Gy, 3.00 Gy, 1.03 Gy, and .327 Gy at 0 mm, 1 mm, 2 mm, 3 mm, and 4 mm in depth to the peritoneal wall, respectively.

Effects on Storage Property of Fresh Jujube Treated with Grape Fruit Stem Extracts (GFSE) and Fermented Pollen (FP) (포도줄기추출물 및 발효화분 처리가 생대추의 저장기간에 미치는 영향)

  • Treatment effects of grape fruit stem extracts (GFSE) containing trans-resveratrol or fermented pollen (FP), and ethylene gas scavenging material (EGS) on the storage property of fresh jujube (Z izyphus jujuba forma hoonensis C.S. Yook) were investigated. Fresh jujubes were packed in different storage containers (PE film and Lock & Lock vessel), and treated with GFSE containing trans-resveratrol of 30 ppm or 1% FP, and EGS. The storage vessels were stored in refrigerator ($0{\pm}1^{\circ}C$) during 12 weeks, and then quality characteristics during storage period were analysed. Hardness slightly increased until 4 weeks and decreased afterward. Soluble solid ($^{\circ}Bx$) and total titratable acidity of fresh jujube slightly increased in all treatments during storage period. Vitamin C content of fresh jujube slightly decreased in all treatments during storage period. Number of microorganisms decreased until 4 weeks and increased afterward. The decay enzyme activity increased in all treatments during storage period. Storage stability was higher for PE film than Lock & Lock containers. Storage period of fresh jujube in this experiment ranges in $8{\sim}9$ weeks for maturity fruits treated with 30 ppm of GFSE and ethylene gas scavenging material.

A Study on the dose distribution produced by $^{32}$ P source form in treatment for inhibiting restenosis of coronary artery (관상동맥 재협착 방지를 위한 치료에서 $^{32}$ P 핵종의 선원 형태에 따른 선량분포에 관한 연구)

  • In this study, the dose distributions of a $^{32}$ p uniform cylindrical volume source and a surface source, a pure $\beta$emitter, were calculated in order to obtain information relevant to the utilization of a balloon catheter and a radioactive stent. The dose distributions of $^{32}$ p were calculated by means of the EGS4 code system. The sources are considered to be distributed uniformly in the volume and on the surface in the form of a cylinder with a radius of 1.5 mm and length of 20 mm. The energy of $\beta$particles emitted is chosen at random in the $\beta$ energy spectrum evaluated by the solution of the Dirac equation for the Coulomb potential. Liquid water is used to simulate the particle transport in the human body. The dose rates in a target at a 0.5mm radial distance from the surface of cylindrical volume and surface source are 12.133 cGy/s per GBq (0.449 cGy/s per mCi, uncertainty: 1.51%) and 24.732 cGy/s per GBq (0.915 cGy/s per mCi, uncertainty: 1.01%), respectively. The dose rates in the two sources decrease with distance in both radial and axial direction. On the basis of the above results, the determined initial activities were 29.69 mCi and 1.2278 $\mu$Ci for the balloon catheter and the radioactive stent using $^{32}$ P isotope, respectively. The total absorbed dose for optimal therapeutic regimen is considered to be 20 Gy and the treatment time in the case of the balloon catheter is less than 3 min. Absorbed doses in targets placed in a radial direction for the two sources were also calculated when it expressed initial activity in a 1 mCi/ml volume activity density for the cylindrical volume source and a 0.1 mCi/cm$^2$ area activity density for the surface source. The absorbed dose distribution around the $^{32}$ P cylindrical source with different size can be easily calculated using our results when the volume activity density and area activity density for the source are known.

  • Several investigators have presented the effects of external magnetic fields on the dose distributions for clinical electron and photon beams. We focus the low energy electron beam with more lateral scatter In this study we calculated the beam profiles for an clinical electron beam of 6 MeV with longitudinal magnetic fields of 0.5 T-3.0 T using a Monte Carlo code. The principle of dose enhancements in the penumbra region is to deflect the laterally scattered electrons from its initial direction by the skewness of the laterally scattered electrons along the direction of magnetic field lines due to Lorentz force under longitudinal magnetic field. To discuss the dose enhancement effect on the penumbra area from the calculated results, we introduced the simple term of penumbra reduction ratio (PRR), which is defined as the percentage difference between the penumbra with and without magnetic field at the same depth. We found that the average PRR are 33%, and 49% over the depths of 1.5 cm, 2.0 cm, and 2.4 cm for the magnetic fields of 2.0 T and 3.0 T respectively. For the case of 0.5 T and 1.0 T the effects of magnetic filed were not observed significantly. In order to obtain the dose enhancement effects by the external magnetic field, we think that its strength should be more than 2 T approximately. We expect that the PRR would be saturated to 50-60% with magnetic fields of 3 T-5 T As a result of these calculations we found that the penumbra widths can be reduced with increased magnetic fields. This Penumbra reduction is explained as a result of electron lateral spread outside the geometrical edges of the beam in a longitudinal magnetic field. This means that the electron therapy benefits from the external magnetic fields.

Situating the Anthropocene: The Social Construction of the Pohang 'Triggered' Earthquake (인류세 맥락화하기: 포항 '촉발지진'의 사회적 구성)

  • On 15th November 2017, the coastal city of Pohang, located in the Southeastern part of South Korea was shaken by a magnitude 5.4 earthquake. The earthquake displaced more than 1,700 residents and caused more than $ 300 million dollars of economic loss. It was the second most damaging earthquake in the history of Korea. Soon after the earthquake, a group of scientists raised a possible link between the first Enhanced Geothermal System (EGS) project and the earthquake. At the same time, another group of scientists put forward a different hypothesis of the causation of the earthquake claiming that it was caused by the geological movements that were initiated by the Great Tohoku Earthquake in 2011. Since then, there were scientific debates between the two different groups of scientists. The scientific debate on the causation of the earthquake has been concluded temporarily by the Research Investigatory Committee on the Pohang Earthquake in 2019. The research committee concluded that the earthquake was caused by the Pohang EGS system: this means that the earthquake can be defined not as a natural earthquake, but as an artificially triggered earthquake. This article is to examine the Pohang earthquake can be defined as an Anthropocenic event. The newly suggested concept, the Anthropocene is a relatively novel term to classify the earthly strata and their relationship to geological time. The current geological period should be defined by human activities and man-made earthly environment. Although the term is basically related to geological classification, the Anthropocene has been widely debated amongst humanist and social science scholars. The current disastrous situation of our planet also implies with the Anthropocene. This paper is to discuss how to understand anthropogenic events. In particular, the paper pays attention to two different scholarly positions on the Anthropocene: Isabelle Stenger's Gaia theory and Barbara Herrnstein Smith's relativist theory. The former focuses on the earthly inevitable catastrophe of Anthropocene while the latter suggests to situate and contextualise anthropogenic events. On the basis of the theoretical positions, the article is to analyse how the Pohang earthquake can be located and situated.

Beta Dosimetry for Applying $^{166}Ho$-chitosan Complex to Cystic Brain Tumor Treatment : Monte Carlo Simulations Using a Spherical Model ($^{166}Ho$-chitosan 복합체를 이용한 낭성뇌종양 치료를 위한 베타선의 흡수선량 평가 : 구형 모델을 이용한 Monte Cairo 모사계산)

  • $^{166}Ho$-chitosan complex, or $^{166}Ho$-CHICO, is a candidate pharmaceutical for intracavitary radiation therapy of cystic brain tumors because of the desirable nuclear characteristics of $^{166}Ho$ for therapeutic use and the suitable biological and chemical characteristics of chitosan, not to mention its ready producibility The amount of $^{166}Ho$-CHICO to be administered to obtain the goal therapeutic effect can be suggested by predicting the dose to the cyst wall for a varying pharmaceutical dose. When $^{166}Ho$-CHICO is infused into the cyst, the major part of the energy delivery by beta particles emitted from $^{166}Ho$ occurs in the cyst wall within 4mm in depth from the cyst wall surface. Also, realizing the attachment of $^{166}Ho$-CHICO to the cyst wall surface would change the predictions of dose to the cyst wall.
