Characterization of Hyaluronic Acid Membrane Cross-linked with Lactide (락타이드로 가교시킨 히아루론산 막의 특성)

  • The hyaluronic acid (HA) with excellent biocompatibility has been combined with lactide, the ester dimer of polylactide, with good biodegradability to produce biocompatible materials which can control the period of degradation in a human body. By freeze frying method, HA and lactide were crosslinked with crosslinking agent, 1-ethyl-3-(3-dimethyl aminopropyl) carbodiimide (EDC). Degree of lactide and EDC reaction was determined by the analysis of nuclear magnetic resonance spectroscopy. Both lactyl group and EDC conversion increased as the mole ratio of lactide to HA increased from 5 to 13. The membrane swelled less and became more brittle with the more addition of lactyl group resulting from the higher mole ratio of lactide to HA. Swelling ratio decreased and tensile modulus increased due to the more addition of lactyl group as the EDC concentration increased or reaction temperature decreased. Drug release experiment from various membranes with different degree of crosslinking showed that permeability decreased with increasing degree of crosslinking. The degradation became slower with the more addition of lactyl group. Mechanical property and degradation rate of the synthesized membrane were shown to be controlled through adjusting operation parameters such as mole ratio, temperature, and crosslinking agent concentration.

An Anatomic Study of the Extensor Tendons of the Human Hand (수배부의 신전건에 대한 해부학적 연구)

  • Purpose: Hands are the chief organs for physically manipulating the environment, using anywhere from the roughest motor skills to the finest, and since the fingertips contain some of the densest areas of nerve endings on the human body, they are continuously used organ with complex functions, and therefore, often gets injured. To prevent any functional loss, a detailed anatomical knowledge is required to have a perfect surgical treatment. Also it is necessary to have a thorough understanding of arrangements of the human extensor tendons and intertendinous connections when tenoplasty or tendon transfer is required. We performed a study of the arrangements of the human extensor tendons and the configuration of the intertendinous connections over the dorsum of the wrist and hand. Methods: A total of 58 hands from Korean cadavers were dissected. The arrangements of extensor indicis proprius, extensor digitorum communis, and extensor digiti minimi tendons and intertendinous connections were studied. Results: The most common distribution patterns of the extensor tendons of the fingers were as follows: a single extensor indicis proprius (EIP) tendon which inserted ulnar to the extensor digitorum-index (EDC-index); a single EDC-index; a single EDC-middle; a double EDC-ring; an absent EDC-little; a double extensor digiti minimi (EDM), a single EDC-index (98.3%), a single EDC-middle (62%), a double EDC-ring (50%), and an absent (65.5%) or a single (32.8%) EDC-little. A double (70.6%) EDM tendons were seen. Intertendinous connections were classified into 3 types: type 1 with thin filamentous type, type 2 with a thick filamentous type, and type 3 with a tendinous type subdivided to r shaped 3r type and y shaped 3y type. The most common patterns were type 1 in the 2nd intermetacarpal space, type 2 in the 3rd intermetacarpal space, and type 3r in the 4th intermetacarpal space. And in the present study, we observed one case of the extensor digitorum brevis manus (EDBM) on the boht side. Conclusion: A knowledge of both the usual and possible variations of the extensor tendon and the intertendinous connection is useful in the identification and repair of these structures.

Effect of cultured chondrocyte-seeded chondroitin-sulfate conjugated type I collagen scaffold on cartilage regeneration (콘드로이틴 환산염을 결합한 I형 콜라겐지지체와 연골세포를 이용한 연골재생)

  • Purpose: In this study, porous type I collagen scaffolds were cross-linked using dehydrothermal(DHT) treatment and/or 1-ethyl-3-(3-dimethyl aminopropyl) carbodiimide(EDC), in the presence and absence of chondroitin-6-sulfate(CS) and cultured autologous chondrocytes(Chondro) for cartilage regeneration. Methods: Cartilage defects were created in the proximal part of the ear of New Zealand rabbits. Four prepared types of scaffolds(n=4) were inserted. The groups included Chondro-Collagen-DHT(Group 1), Chondro- Collagen-DHT-EDC(Group 2), Chondro-CS-Collagen- DHT(Group 3), and Chondro-CS-Collagen-DHT-EDC (Group 4). Histomorphometric analysis and cartilage-specific gene expression of the reconstructed tissues were evaluated 4, 8, and 12 weeks after implantation. Results: EDC cross-linked groups 2 and 4 regenerated more cartilage than other groups. However, calcification was observed in the 4th week after implantation. CS did not increase chondrogenesis in all groups. Cartilage-specific type II collagen mRNA expression increased in the course of time in all groups.Conclusion: EDC cross-linking methods maintain the scaffold and promote extracellular matrix production of chondrocytes.

Numerical Simulations of the Pyrolysis of 1,2 Dichloroethane (1,2 Dichloroethane의 열분해에 대한 수치해석)

  • Numerical simulations of 1,2 dichloroethane(EDC) pyrolysis are conducted to understand the process on the production of the vinyl chloride monomer(VCM) and by-products. A chemical kinetic mechanism is developed, the adopted scheme involving 44 gas-phase species and 260 elementary forward and backward reactions. Detailed sensitivity analyses and the rates of production analysis are performed on each of the reactions and the various species, respectively. The concentrations of EDC, VCM, and HCI predicted by this mechanism are in good agreement with those deduced from experiments of commercial and laboratory scale. The mechanism is found to accurately predict the VCM yield and the production of by-products by varying the ranges of pyrolysis temperature, residence time, and pressure which impact on the pyrolysis of 1,2 dichloroethane. The influence of reactions related to H atom on the relative sensitivity of EDC becomes important as the residence time increases. The pyrolysis of EDC mainly occurs through $C_{2}H_{4}Cl_{2}+Cl=CH_{2}ClCHCl$.

  • 서낙동강 하류에 국내 최초로 친수구역 활용에 관한 특별법에 따른 부산 에코델타시티(Eco Delta City, EDC) 조성사업이 시행 중에 있으며, 친수공간 확보 및 친수활동에 대한 관심의 증가로 서낙동강 수질개선에 대한 필요성이 증가하고 있다. 이에 부산 EDC 사업에서는 친수활동을 위한 목표 수질(II급수)을 설정하고 목표수질을 달성하기 위해 EFDC 모형을 활용하여 서낙동강 유역의 수질개선 방안을 검토하였다. 서낙동강 유역의 하수처리 정비 등을 통한 오염부하의 삭감, 부산 EDC 사업의 오염부하 삭감 및 서낙동강, 평강천, 맥도강 물순환 등의 대책을 통하여 목표 수질을 달성할 수 있을 것으로 연구되었다.

Approximation of Multiple Trait Effective Daughter Contribution by Dairy Proven Bulls for MACE (젖소 국제유전능력 평가를 위한 종모우별 다형질 Effective Daughter Contribution 추정)

  • This study was conducted to investigate the basic concept of multiple trait effective daughter contribution (MTEDC) for dairy cattle sires and calculate effective daughter contribution (EDC) by applying a five lactation multiple trait model using milk yield test records of daughters for the Multiple-trait Across Country Evaluation (MACE). Milk yield data and pedigree information of 301,551 cows that were the progeny of 2,046 Korean and imported dairy bulls were collected from the National Agricultural Cooperative Federation and used in this study. For MTEDC approximation, the reliability of the breeding value was separated based on parents average, own yield deviation and mate adjusted progeny contribution. EDC was then calculated by lactation using these reliabilities. The average number of recorded daughters per sire by lactations were 140.57, 94.24, 55.14, 29.20 and 14.06 from the first to fifth lactation, respectively. However, the average EDC per sire by lactation using the five lactation multiple trait model was 113.49, 89.28, 73.56, 54.02 and 35.08 from the first to fifth lactation, respectively, while the decrease of EDC in late lactations was comparably lower than the average number of recorded daughters per sire. These findings indicate that the availability of daughters without late lactation records is increased by genetic correlation using the multiple trait model. Owing to the relatedness between the EDC and reliability of the estimated breeding value for sire, understanding the MTEDC algorithm and continuous monitoring of EDC is required for correct MACE application of the five lactation multiple trait model.

A Study on the Active site of Glucoamylase from Aspergillus shirousamii

  • Glucoamylase was inactivated with 1-ethyl-2-(dimethylaminopropyl)carbodiimide (EDC) at pH 5.0. Time course of inactivation of glucoamylase was at least biphasic. From the results of the titration of SH groups with Ellman's reagent and hydroxylamine treatment at pH 7.0, it was concluded that the crucial sites of modification were carboxyl groups of glucoamylase. The CD spectrum of EDC-modified glucoamylase suggested that the gross conformation of the native enzyme was retained. The inactivation of glucoamylase was reduced remarkably in the presence of maltose. The logarithm of the half-life of the inactivation of glucoamylase by EDC was a linear function of log[EDC] in each stage indicating that one carboxyl group among the modified ones was crucial for inactivation of glucoamylase. The change in the binding affinity due to modification was determined by using an affinity column. It indicates that the carboxyl group of glucoamylase seems to play a role in both, the catalysis and substrate binding in the first stage, but in the second stage the binding affinity is recovered almost up to that of native enzyme.

The Effect of Various Methods of Cross-linking in Type I Collagen Scaffold on Cartilage Regeneration (I형 콜라겐지지체의 다양한 가교처리 방법이 연골막성 연골재생에 미치는 영향)

  • Purpose: Collagen is the principal structural biomolecule in cartilage extracellular matrix, which makes it a logical target for cartilage engineering. Results: The largest quantity of regenerated cartilage was found in DHT cross-linked groups 1 and 3 in the 8th week and then decreased in the 12th week, while calcification increased. Calcification was observed from the 8th week and the area increased in the 12th week. Group 4 was treated with EDC cross-linking and CS, and the matrix did not degrade in the 12th week. Cartilage-specific type II collagen mRNA expression increased with time in all groups. Conclusion: CS did not increase chondrogenesis in all groups. EDC cross-linking may prevent chondrocyte infiltration from the perichondrium into the collagen scaffold.

EDC System Development based on CDISC Standard (CDISC 국제표준을 기반으로한 EDC 시스템 개발)

  • 국내 임상시험기관에서 수행되는 다국가 임상시험은 매년 증가하고 있지만, 국내 임상시험의 대부분은 종이기반 시스템이 주로 사용되면서 데이터 오류와 시간 및 비용의 증가등의 문제점을 내포하고 있었다. 또한 다국적 임상시험의 경우 임상 데이터의 효율적인 관리를 위해 CDISC 표준을 이용한 전자 임상자료 관리 시스템 사용을 요구하고 있다. 본 연구에서는 국제표준에 적합한 효율적인 데이터 관리와 기존 시스템의 한계를 극복하기 위해 CDISC 표준을 기반으로 한 전자데이터관리(EDC) 시스템을 개발하였다. 이를 통해 데이터의 유효성 확보뿐만 아니라 실험실 데이터를 통한 안전성 분석의 효율적 진행, 시간 단축 등의 효과를 거두고 있다.
