An Animal-Industrial Review on Phytic Acid and Phytase (Phytic Acid와 Phytase에 관한 동물산업적 고찰)

  • Phytic acid (myo-inositol hexaphosphate or IP6) is the major storage form of phosphorus in cereals and legumes, representing 18 to 88% of the total phosphorus. Phytate form of phosphorus is not readily utilized by monogastric animals and this result causes pollution problem by phosporus released in areas of intensive livestock production. The interaction between phytic acid and essential dietary minerals, protein, or vitamins is considered to be one of the primary factors limiting the nutritional values of cereals and legunes in monogastric animals. Attempts have been made to hydrolyze dietary phytic acid by phytases to improve the feed quality and to decrease the amount of phosphorus excreted by animals. Phytase(myo-inositol hexakisphosphate phosphohydrolase) hydrolyzes phytic acid to myo-inositol and phosphoric acid. Two types of phytases are known: 3-phytase (EC 3.1.3.8) and 6-phytase (EC 3.1.3.26), indicating the intial attack to the susceptable phosphoester bond. Because of its great industrial importance, there is ongoing interest in isolating new bacterial strains producing novel and efficient phytases.

Comparison of Cytokine Gene Induction in RAW 264.7 Cells by Porphyromonas gingivalis and Escherichia coli Lipopolysaccharide

  • Porphyromonas gingivalis lipopolysaccharide (Pg LPS) is an important virulence factor in chronic periodontitis. The aim of this study was to compare the expression of inflammatory cytokine genes in Escherichia coli LPS (Ec LPS) and Pg LPS-stimulated mouse macrophage RAW 264.7 cells. Cells were treated with Ec LPS and Pg LPS for 18 hours, and the cytokine gene expression profile was assessed using microarrays and confirmed by real-time PCR. Microarray analysis showed that both types of LPS induced a significant increase in the expression of IL-$17{\beta}$, IL-2, Ccl4, Cxcl2 and $TNF{\alpha}$ compared with the control. However, LT-b was up-regulated by Pg LPS but not by Ec LPS. Real-time PCR analysis of these genes showed similar results for LT-b, Ccl4, Cxcl2, and TNF-$\alpha$ but found that IL-$17{\beta}$ and IL-2 were upregulated by Pg LPS but not by Ec LPS. These data indicate that Pg LPS stimulates the transcription of IL-$17{\beta}$, IL-2, Ccl4, Cxcl2, LT-b, and $TNF{\alpha}$, all of which may be involved in the pathogenesis of chronic periodontitis.

Concentrations of Air Pollutants Measured at Kosan during ACE-Asia Intensive Observation Period (ACE-Asia 집중관측기간에 제주고산에서 측정한 대기오염물질의 농도 분포특성)

  • The concentrations of gaseous (NO$_{x}$, SO$_2$, and $O_3$) and particulate (Elemental Carbon, EC and Organic Carbon, OC) pollutants were measured to evaluate the air quality of Kosan. Samples were taken at Kosan during ACE-Asia (Asian Pacific Regional Aerosol Characterization Experiment) IOP (Intensive Observation Period) (2001. 3. 21~2001. 5. 5). The mean concentrations of $O_3$(46.3$\pm$10.4 ppb) is higher than those at urban area such as Seoul and Busan in Korea. On the other hand, the mean concentrations of other gaseous species, NO$_{x}$(4.73$\pm$3.42 ppb) and SO$_2$(0.62$\pm$0.63 ppb) are lower than those at great cities. So we concluded that there are a few primary sources emitting atmospheric pollutants. The concentration of EC is higher and the concentration of OC is similar with or higher than those at other background sites. The recent EC concentration is higher than those measured before at Kosan. We concluded that there are more primary sources than other background sites and the amount of primary source have increased recently in Jeju. Backward trajectory and co..elation analysis were used to study where the air masses originated and distinguish the source of pollutants. While NO$_{x}$ and $O_3$ were mainly emitted and formed from Jeju inland area, concentrations of SO$_2$, OC and EC were affected by Asian Dust from China. Using the mean relative standard deviation of ozone, cleanness coefficient was obtained. The cleanness coefficient value, is 1.6 times larger than the value in 1992. Recently, the air quality of Kosan has been contaminated because of the Asian Dust events since spring and the rapid industrialization development.pment.

Concentrations and Characteristics of Carbonaceous Compounds in PM10 over Seoul: Measurement between 2006 and 2007 (서울시 대기 중 미세먼지 내 탄소성 입자의 농도 및 특성 : 2006~2007년 측정결과)

  • Carbonaceous compounds in the atmospheric particulate matter with an aerodynamic diameter of less than or equal to a nominal $10{\mu}m$ ($PM_{10}$) were analyzed for the samples collected during the period of August 2006 to August 2007 at Jongro in Seoul. A total 18 dicarboxylic acids (DCAs) and levoglucosan, as well as organic carbon (OC), elemental carbon (EC), and water soluble organic carbon (WSOC), were analyzed. Distinctive seasonal patterns of the concentrations of OC, EC, and WSOC including levoglucosan were observed with the highest concentrations in winter and the lowest concentrations in summer. In addition, OC, WSOC, and most of DCAs showed also higher concentration in summer than in winter. Using the seasonal patterns and relevant indicative ratios (WSOC/EC and $OC_{sec}/OC_{tot}$) of the carbonaceous compounds, it was verified that (1) primary emission sources were elevated in winter, and (2) the formation of secondary OC increased due to the prompted photochemical reaction in summer. Results from this study also suggest that some organic compounds were likely attributed to longrange transport.

  • The purpose of thisanalytical study of an iTECH composite beam subjected to fire conditions is to determine the beam's fire resistance performance using its load ratio and fire protection as parameters. A composite structural system is expected to have a safer and more economical fire safety design than a mere collection of isolated members.heat transfer analysis was performed on the basis of the finite element program ANSYS 10.0 using an ISO834 standard fire, following the main guidelines proposed by EC1 Part 2.2 and EC4 Part 1.2. To validate the analytical simulation of the iTECH composite beam, comparison of the experimental tests was proposed.

The Characteristics of Secondary Carbonaceous Species within PM10 and PM2.5 in Seoul and Incheon Area (서울과 인천지역 PM10 과 PM2.5 중 2차생성 탄소성분 추정)

  • To investigate secondary carbonaceous species within PM$_{10}$ and PM$_{2.5}$ in Seoul urban Metropolitan Area (SMA), Korea. atmospheric particulate matters samples were collected at two sites of SMA at UOS (The University Of Seoul station) sites and IHU (InHa University of Incheon station) during the period of 4 to 14 January and 12 to 22 May, 11 to 15 August 2004, and their characteristics were qualitatively discussed. during January and May and August of 2004. Daily average mass concentration 0.095 mg/㎥ in PM$_{10}$ and 0.053 mg/㎥ in PM$_{2.5}$ for mass respectively. were observed in SMA. The concentrations of carbonaceous species contributed 18.4% and 16.4% of PM$_{2.5}$ and PM$_{10}$ during the sampling period, respectively, of which OC accounted for 68% and 52% more of the total carbon (TC). OC and EC concentrations and their mass percentages were higher in PM$_{2.5}$ than in PM$_{10}$ which could be attributed to generation process. Organic aerosols would constitute up to 38% of PM$_{2.5}$ based on the evaluation of 1.6 for the ratio of OC to organic particulate. Secondary organic carbon (SOC) were estimated to be more than 13% and up to 68% of total OC based on the minimum OC/EC ratio of 1.06/1.11 using least square method. Comparisons of OC and EC with trace elements. As results of carbonaceous species analysis, the dominant factor in view of fine particle (PM$_{10}$/PM$_{2.5}$) is primary emission source such as mobile, fossil fuel combustion etc. during winter time in SMA. But in summer periods, remarkable fine particle increasing factor was secondary organic carbon dependent to photochemical reaction. reaction.n. reaction.

The Investigation of Treatment of Pistachio Processing Industrial Wastewater by Electrochemical Methods in Terms of Chemical Oxygen Demand and Total Phenol Removal

  • This work aims to investigate the efficiency of electrocoagulation (EC) of pistachio processing industrial wastewater (PPIW) using the continuous EC process. The tubular reactor made of stainless steel with an internal diameter of 60 mm was used as a cathode electrode. The effect of some parameters was examined on the removal of chemical oxygen demand (COD) and total phenols (TP) removal efficiency. The influences of the initial pH of wastewater (from 4 to 8), flow rate (from 25 to 125 mL/min), current density (from 7 to 21 mA/cm2), and supporting electrolyte type (NaCl, NaNO3, and Na2SO4), supporting electrolyte concentration (from 10 to 100 mg/L NaCl) on removal efficiency were investigated to determine the best experimental conditions. The examination of the physico-chemical parameters during the EC treatment showed that the best removal efficiency was obtained under conditions where the flow rate was 25 mL/min (20 min reaction time), the pH value was 5.2, and the current density was 21 mA/cm2 has set. Under these experimental conditions, COD and TP removal efficiency were found to be 75% and 97%, respectively, while energy consumption was 18.5 kW h/m3. The study results show that the EC can be applied to PPIW pre-treatment.

Determination of Residue Levels of Ethyl Carbamate in Alcoholic Beverages by Gas Chromatography/Tandem Mass Spectrometry (GC/MS/MS) (가스크로마토그래피/질량분석기(GC/MS/MS)를 이용한 주류 중 에틸카바메이트 잔류량 조사)

  • Ethyl carbamate (EC) is a contaminant generated in the fermentation processes of various fermented foods. In this study, residue levels of EC in 95 alcoholic beverage samples were determined by using Gas Chromatography/Tandem Mass Spectrometry (GC/MS/MS). All the samples were purified by a liquid-liquid extraction (LLE) method using dichloromethane. The LLE method enables an improvement in time and cost to detection and specificity over the conventional extraction methods. The limits of detection and quantification (LOD and LOQ) to analyze EC were 1.3 and 4.0 ng/mL, respectively. The recovery rates of EC were ranged from 90.0 to 97.5% at the levels of 50, 100, and 500 ug/L. Among traditional grain-based alcoholic beverage samples (n = 34), the average residue levels of EC in takju, yakju, and cheongju were 0.63, 7.01, and 14.11 ug/L, respectively. Among fruit-based alcoholic beverage samples (n = 48), those of EC in japanese apricot spirits, bokbunjaju, grape wines, and other fruit wines were 79.18, 1.66, 2.64, and 2.39 ug/L, respectively. Among distilled or diluted alcoholic beverage samples (n = 13), those of EC in soju (distilled or diluted), general distillates, liquors, and brandies were 0, 3.30, 8.20, and 8.52 ug/L, respectively. Therefore, this study reports that the residue levels of EC in the alcoholic beverages, distributed in the current domestic markets, did not reach its maximum allowed levels of 30 and 400 ug/L established for grape and fruit wines in Canada, respectively.

Reduction of Stem Cavity and Improvement of Flower Quality in Chrysanthemum 'Baekma' by Hydroponic Culture (양액재배에 의한 국화 '백마'의 줄기공동 경감과 절화품질 향상)

  • This study was conducted to investigate effect of ionic strength and feeding times of a nutrient solution on reduction of stem cavity size and improvement of flower quality in chrysanthemum 'Baekma'. A nutrient solution was applied with different strengths at three stages, namely, transplanting, budding, and flowering. The solution EC was adjusted as 1.61.82.0, 1.81.81.8, 1.82.01.8, and $2.02.02.0dS{\cdot}m^{-1}$ in four treatments. Feeding frequency per a day were 4 times for 12 min., 8 times for 6 min., 12 times for 4 min., and 18 times for 2.7 min. each. Cut flower length as affected by different strengths of a nutrient solution was the greatest in the plot of EC $2.02.02.0dS{\cdot}m^{-1}$. However number of leaves, stem diameter, and leaf size were greater in EC $1.82.01.8dS{\cdot}m^{-1}$ than in other treatments. Also, petal number of petals was the greatest and stem cavity size was the smallest in the plot of EC $1.81.81.8dS{\cdot}m^{-1}$. Plant height, number of leaves, stem diameter, leaf size were greater in the plot with 12 times feed ing per a day. number of petal was most in the plot with 8 times feeding per day, while stem cavity size was the smallest in the plot with 12 times feed ing per a day. Therefore, the better plant growth, the smaller stem cavity size.

Sensitivity of Botrytis cinerea Isolated from Infected Leaves of Ginseng to Tolyfluanid (Tolyfluanid에 대한 인삼 잿빛곰팡이병균의 감수성)

  • Resistance monitoring for tolyfluanid was conducted over 2 years from 2005 to 2006 with 218 isolates of Botrytis cinerea obtained from infected plants of ginseng. All isolates of B. cinerea were divided into 3 groups such as highly sensitive (HS) group, sensitive (S) group and less sensitive (LS) group to tolyfluanid. HS group was defined as one showing $EC_{50}$ value less than $2.0\;{\mu}g\;mL^{-1}$. $EC_{50}$ value of S group ranged between $2.0\;{\mu}g\;mL^{-1}$ and $18.0\;{\mu}g\;mL^{-1}$, while that of LS group was more than $18.0\;{\mu}g\;mL^{-1}$ Among B. cinerea isolates tested in this study, 174 isolates were included into S group, while 22 isolates were done into HS group. The other isolates belonged to LS group. Botrytis cinerea isolates showed the high correlation of $EC_{50}$ value of mycelial growth with that of spore germination to tolyfluanid. This results could be used for the determination of fungicide resistance and the establishment of strategy for fungicide resistance management.
