Functional Study of Lysine Decarboxylases from Klebsiella pneumoniae in Escherichia coli and Application of Whole Cell Bioconversion for Cadaverine Production

  • Klebsiella pneumoniae is a gram-negative, non-motile, rod-shaped, and encapsulated bacterium in the normal flora of the intestines, mouth, skin, and food, and has decarboxylation activity, which results in generation of diamines (cadaverine, agmatine, and putrescine). However, there is no specific information on the exact mechanism of decarboxylation in K. pnuemoniae. Specifically lysine decarboxylases that generate cadaverine with a wide range of applications has not been shown. Therefore, we performed a functional study of lysine decarboxylases. Enzymatic characteristics such as optimal pH, temperature, and substrates were examined by overexpressing and purifying CadA and LdcC. CadA and LdcC from K. pneumoniae had a preference for L-lysine, and an optimal reaction temperature of 37℃ and an optimal pH of 7. Although the activity of purified CadA from K. pneumoniae was lower than that of CadA from E. coli, the activity of K. pneumoniae CadA in whole cell bioconversion was comparable to that of E. coli CadA, resulting in 90% lysine conversion to cadaverine with pyridoxal 5'-phosphate L-lysine.

  • 인터넷과 IT 인프라의 증가는 인터넷 기반의 전자상거래 시스템의 성장을 주도하였고, 이와 관련된 새로운 형태의 서비스가 나타나는 계기가 되었다. 그러나, 현재 전자상거래 서비스는 구매자가 웹에 접속하여 인터넷 쇼핑몰에 보이는 상품을 선택하여 결제 및 상품을 수신하는 방식으로 이루어진다. 또한 구매 판단을 결정할 때 최저가격 외에 신속한 배송, 부가 서비스 등의 다양한 의사결정 요소가 있음에도 불구하고, 인터넷 쇼핑몰에서 제시한 최저가의 판단요소에 의존함으로 구매자가 수동적인 입장이 된다. 따라서, 본 논문에서는 이러한 문제점을 해결하기 위해 상호간에 의견교환 및 사용자가 정한 우선순위에 따른 판단을 효과적으로 할 수 있는 서비스인 게임이론 기반의 지능모델을 제시한다 게임이론은 전통적으로 경제문제 협상과 정치문제 협상, 워 게임 등에 적용되었다. 본 연구에서는 이 점에 착안하여 게임이론 알고리듬을 이용하여 e-Commerce를 위한 지능모델에 적용하여, 기존의 수동적이고 사용자의 취향에 따라 다양한 의사결정 요소를 선택 할 수 없었던 문제를 해결한다.

Simulation of Vacuum Characteristics of High Vacuum System Modelled by VacCAD

  • In this paper, we simulated three different HV systems and analyzed of each vacuum characteristics by VacCAD modelling. In each of modelled vacuum systems, selection of chamber materials, combination of rough pump with high vacuum pump and conductance of roughing line (diameter and length) were proposed as system variables. In the modelling of chamber materials, the pumping times to ultimate pressures of different chamber materials (stainless steel, aluminum) were compared by the variations of chamber volume. In this model, the effects of outgassing dependent on the chamber materials was also simulated and aluminum was estimated to optimum chamber materials. It was also obtained that modelling of vane and roots pump with diffusion pump and diameter, length of $50{\times}250$ [mm]roughing line were characterized as optimum variables to reach the ultimate pressure of 10E-7 [mbar] most effectively. Optimum design factors for vacuum characteristics of modelled vacuum system were achieved by VacCAD simulations. Feasibility of VacCAD as vacuum simulator was verified and applications of VacCAD expected to be increased to fields in vacuum needed.

Effect of translucency on CAD/CAM ceramic according to different core/veneer thickness (캐드캠 세라믹의 코어와 베니어의 서로 다른 두께가 반투명도에 미치는 영향)

  • Objectives: The purpose of this study was to evaluation the effect of translucency on CAD/CAM ceramic according to different core/veneer thicknesses. Methods: A total of 42 samples from 2 groups of 7 ceramic cores, each with 3 thickness values (0.8, 1.0, 1.2 mm) were manufactured. The veneers were also manufactured in 3 thicknesses (0.3, 0.5, 0.7 mm). The group names were based on the name of the ceramic core (IPS e.Max CAD; LD, and IPS Empress CAD; LR). The associated number was determined by the combined thickness of the core and veneer: 1 = (0.8 + 0.7); 2 = (1.0 + 0.5); 3 = (1.2 + 0.3). The translucency was measured using a spectrophotometer and defined via the contrast ratio (CR) and translucency parameter (TP). Two-way ANOVA was performed to compare the 2 groups (material and thickness). Results: As the core thickness increased, the TP value also increased, and the CR value (for LR group) decreased. The results of 2-way ANOVA demonstrated that the thicknesses of different core/veneer combinations significantly affected the translucency (p<0.05). Conclusions: The different core and veneer thicknesses affected the translucency of CAD/CAM ceramic.

  • In this paper, we develop a six-axes machining center tool(MCT) and CAD/CAM system based RAD Tool Program. The MCT consists of two mechanical parts, i.e., a X-Y-Z Cartesian coordinate typed MCT and a parallel-typed tilting table. Kinematics and singularity are accomplished to design the parallel-typed tilting table, and RAD Tool Program Is developed for the six-axes MCT, which requires the commands of position as well as orientation for machining of complex shape. In RAD Tool, the CAD/CAM system has a tool path generator, NC code generator and a graphic simulator. This paper designs the parallel-typed tilting table to meet the desired specification and presents the results of CAD/CAM system based RAD Tool Program.

  • The exchange of model design date among heterogeneous CAD systems is a difficult task because each system has different data structures suitable for its own functions. STEP has been able to represent product information as a common computer-interpretable form that is required to remain complete and consistent when the product informant is needed to be exchanged among different computer system. However, STEP has difficult architecture in is representing point, line, curve and vectors of element, more over it can't represent geometry data of feature based models. In this study, a structure of XML document that represents geometry data of feature based models as neutral format has been developed. To use the developed XML document, a Converter has also been developed to exchange modules so that it can exchange feature based data models among heterogeneous CAD systems. Aa for evaluation of the developed XML document and Converter, Solidworks and SolidEdge are selected.

  • This study described computer aided die design system for cold forging of non-axisymmetric parts. To design the die of cold forging. an integrated approach based on a rule-base system and commercial F. E. code were adopted. This system is implemented on the personal computer and its environment is a commercial CAD package named as AutoCAD. The system includes four modules. In the initial data input module, the variables which are necessary to design of die are inputted by user and die material are selected from the database. In the analysis and redesign module, stress distrubution action on the designed die is analyzed by commercial FEM code NISA II. The designed die is modified to prevent failure in both states of stress free and pressurizing. The developed system provides powerful capabilities for die design of non-axisymmetric parts.

  • A new CAD program for double E-plane bandpass filters is developed based on the variational analysis method and the passband correction method combined with the synthesis procedures. By taking the widths of three narrow waveguide equally, we can design bandpass filters with best stopband characteristics.

Immunogenicity of a novel inactivated canine adenovirus type 2 variant vaccine for dogs

  • Purpose: The immunogenicity of vaccines containing the canine adenovirus (CAdV) type 2 (CAdV-2) variant has not yet been reported. We prepared a novel inactivated CAdV-2 variant vaccine using the CAV2232-41 strain, and evaluated its safety and immunogenicity in raccoon dogs. Materials and Methods: The growth kinetics of CAV2232-41 were determined using Madin-Darby Canine Kidney (MDCK) cells. The nucleotide sequences of CAV2232 and CAV2232-41 were determined by next-generation sequencing. To generate the CAdV-2 variant vaccine, CAV2232-41 propagated in the MDCK cells was inactivated with 0.1% formaldehyde. Two vaccines were prepared by blending inactivated CAV2232-41 with Cabopol and Rehydragel adjuvants. Safety and immunogenicity of the CAV2232C and CAV2232R vaccines were evaluated in guinea pigs. Safety and immunogenicity of the CAV2232C vaccine were also evaluated in raccoon dogs. The virus neutralizing antibody (VNA) titer against CAV2232-41 was measured in sera collected from immunized guinea pigs and raccoon dogs. Results: CAV2232-41 showed the highest viral titer on days 4-6 post-inoculation and had a deletion in the E3 gene, which was confirmed as a CAdV-2 variant. Guinea pigs inoculated with CAV2232C showed slightly higher VNA titers than those inoculated with CAV2232R 2 weeks after booster vaccination. Raccoon dogs immunized with the CAV2232C vaccine developed high mean VNA titers, while non-vaccinated raccoon dogs were antibody-negative. Conclusion: The CAV2232C vaccine is safe and induces a protective VNA titer in raccoon dogs.
