Protective Effects of Ursolic Acid on Osteoblastic Differentiation via Activation of IER3/Nrf2

  • Background: Oxidative stress is a known to be associated with in the pathogenesis of many inflammatory diseases, including periodontitis. Ursolic acid is a pentacyclic triterpenoid with has antimicrobial, antioxidative, and anticancer properties. However, the role of ursolic acid in the regulating of osteogenesis remains undetermined. This study was aimed to elucidate the crucial osteogenic effects of ursolic acid and its ability to inhibit oxidative stress by targeting the immediate early response 3 (IER3)/nuclear factor erythroid 2-related factor 2 (Nrf2) pathway. Methods: Cell proliferation was determined using water-soluble tetrazolium salt assay, cell differentiation was evaluated by alkaline phosphatase (ALP) activity, and formation of calcium nodules was detected using alizarin red S stain. Generation of reactive oxygen species (ROS) was determined using by DCFH-DA fluorescence dye in hydrogen peroxide ($H_2O_2$)-treated MG-63 cells. Expression levels of IER3, Nrf2, and heme oxygenase-1 (HO-1) were analyzed using western blot analysis. Results: Our results showed that ursolic acid up-regulated the proliferation of osteoblasts without any cytotoxic effects, and promoted ALP activity and mineralization. $H_2O_2$-induced ROS generation was found to be significantly inhibited on treatment with ursolic acid. Furthermore, in $H_2O_2$-treated cells, the expression of the early response genes: IER3, Nrf2, and Nrf2-related phase II enzyme (HO-1) was enhanced in the presence of ursolic acid. Conclusion: The key findings of the present study elucidate the protective effects of ursolic acid against oxidative stress conditions in osteoblasts via the IER3/Nrf2 pathway. Thus, ursolic acid may be developed as a preventative and therapeutic agent for mineral homeostasis and inflammatory diseases caused due to oxidative injury.

  • With evolution in the production environment of the textile industry, the need for non-water-based dyeing technologies and eco-friendly process facilities in the dyeing and processing stages has increased. In recent years, supercritical fluid dyes have been developed and commercialized in Europe, centering on this demand. However, so far, such dyes have been mainly applied in the processing of PET fibers. Basic research has mainly involved investigation of dyeing by supercritical carbon dioxide or solubility of such dyes, and more in-depth research should be continuously carried out. In this review, we describe the types and characteristics of supercritical fluids that exhibit specific properties at pressures and temperatures over the critical point. In addition, the state of the art in the dyeing and processing technology using supercritical fluids and associated, processing problems, environmental regulation, and wastewater treatment issues are described in detail. We hope this review can contribute to the supercritical fluid technology being further developed as an environment friendly dyeing processing method. Furthermore, we expect that the technique can be used as a means of ensuring different, high-quality dyed products.

Study on Isotherm, Kinetic and Thermodynamic Parameters for Adsorption of Methyl Green Using Activated Carbon (활성탄을 이용한 메틸 그린 흡착에 있어서 등온선, 동력학 및 열역학 파라미터에 대한 연구)

  • The adsorption of methyl green dye using an activated carbon from an aqueous solution was investigated. Adsorption experiments were carried out as a function of the adsorbent dose, initial concentration, contact time and temperature. The Langmuir isotherm model showed a good fit to the equilibrium adsorption data. Based on the estimated Langmuir separation factor, ($R_L=0.02{\sim}0.106$), this process could be employed as the effective treatment (0 < $R_L$ < 1). It was found that the adsorption was a physical process with the adsorption energy (E) value range between 316.869 and 340.049 J/mol obtained using Dubinin-Radushkevich equation. The isothermal saturation capacity obtained from brunauer emmett teller (BET) model increased with increasing the temperature. The kinetics of adsorption followed a pseudo second order model. The free energy and enthalphy values of -5.421~-7.889 and 31.915 kJ/mol, respectively indicated that the adsorption process follows spontaneous endothermic reaction. The isosteric heat of adsorption increased with the increase of equilibrium adsorption amounts, and the total interaction of the adsorbent - adsorbate increased as the surface coverage increased.

A study on dyeing of dyed fabrics and its antimicrobial activity on antibiotic-resistant bacteria using Saururus chinensis extract (삼백초 추출액을 이용한 염색포의 염색성과 항생제 내성균에 대한 항균성 연구)

  • The purpose of this study is to investigate the antibacterial efficacy of cotton and silk/rayon fabrics dyed with Saururus chinensis extract against antibiotic-resistant strains. The concentration of the concentrated dye in the Saururus chinensis extracts was 1.1% (o.w.f), and the liquor ratio was 1:10 at 30-70℃. The mordanting method was a post mordanting method. The concentration of Al2(SO4)3, CuSO4 5H2O and FeSO4 and7H2O mordant was 5% (o.w.f), and the liquor ratio was 1:40. In order to assess the antimicrobial activity of naturally dyed fabrics, Methicillin-resistant Staphylococcus aureus (MRSA) ATCC 33591, was used by incubating it in Brain Heart Infusion Agar (BHA) including Oxacillin (2㎍/ml) and Fungizone (2.5㎍/ml) and Brain Heart Infusion broth (BHI; Detroit, MI, USA.) The investigation of the reduction of the rate of antibiotic-resistant strains to dyed cotton fabrics and silk/rayon fabrics revealed that Cu mordanting fabric has the highest antimicrobial effects, with the rate of 99.7%, and Fe mordanting fabric has the lowest, with 77.7%. Non-mordant cotton fabrics also show a high reduction rate of strains (94.6%). In the case of dyed silk/rayon fabrics, it indicates a high reduction in the rate of strains in all fabrics with non-mordant treatment (94.2%), Al mordanting (99.6%), and Cu and Fe mordanting(99.9%).

In Vivo Enhanced Indocyanine Green-Photothermal Therapy for a Subconjunctival Tumor

  • Indocyanine green (ICG) is a dye approved for use in clinical diagnostics. ICG remains in the intravascular space following intravenous administration, due to its ability to rapidly bind to the plasma proteins, and its therapeutic potential has been studied in well-vascularized cutaneous tumors. Here we have evaluated the clinical response of a subconjunctival tumor to photothermal therapy (PTT) using an ICG-enhanced near-infrared diode laser and its adverse effects, in a rabbit. 22 male New Zealand white rabbits with subconjunctival tumors were enrolled (control group 6, laser-only group 8, laser-with-ICG group 8). Rabbits in the laser-with-ICG group received ICG (twice, 2 mg/kg each time, intravenously) directly followed by irradiation with a diode laser (λ = 810 nm). Rabbits in the laser-only group were irradiated with the diode laser. ICG angiography, ultrasonography, and pathologic examination were performed to evaluate PTT response at specific time points (0, 2, and 4 weeks after PTT). Two weeks after initial treatment, the eight rabbits treated by laser with ICG showed a 100% response rate. There was no clinical response in both laser-only and control groups. ICG-PTT is a potential and effective palliative therapeutic modality for subconjunctival tumors.

Anti-Endotoxin 9-Meric Peptide with Therapeutic Potential for the Treatment of Endotoxemia

  • Inflammatory reactions activated by lipopolysaccharide (LPS) of gram-negative bacteria can lead to severe septic shock. With the recent emergence of multidrug-resistant gram-negative bacteria and a lack of efficient ways to treat resulting infections, there is a need to develop novel anti-endotoxin agents. Antimicrobial peptides have been noticed as potential therapeutic molecules for bacterial infection and as candidates for new antibiotic drugs. We previously designed the 9-meric antimicrobial peptide Pro9-3 and it showed high antimicrobial activity against gram-negative bacteria. Here, to further examine its potency as an anti-endotoxin agent, we examined the anti-endotoxin activities of Pro9-3 and elucidated its mechanism of action. We performed a dye-leakage experiment and BODIPY-TR cadaverine and limulus amebocyte lysate assays for Pro9-3 as well as its lysine-substituted analogue and their enantiomers. The results confirmed that Pro9-3 targets the bacterial membrane and the arginine residues play key roles in its antimicrobial activity. Pro9-3 showed excellent LPS-neutralizing activity and LPS-binding properties, which were superior to those of other peptides. Saturation transfer difference-nuclear magnetic resonance experiments to explore the interaction between LPS and Pro9-3 revealed that Trp3 and Tlr7 in Pro9-3 are critical for attracting Pro9-3 to the LPS in the gram-negative bacterial membrane. Moreover, the anti-septic effect of Pro9-3 in vivo was investigated using an LPS-induced endotoxemia mouse model, demonstrating its dual activities: antibacterial activity against gram-negative bacteria and immunosuppressive effect preventing LPS-induced endotoxemia. Collectively, these results confirmed the therapeutic potential of Pro9-3 against infection of gram-negative bacteria.

Cytoprotective effect of the processed Gardeniae Fructus on oxidative stress-induced gastric epithelial cells (산화스트레스를 유발한 위 상피세포에서 수치 치자의 세포 보호 효과)

  • Objective : Gardeniae Fructus (GF) has bitter and cold nature. Thus, it has been traditionally prescribed in processed form roasted with ginger juice for patients with a weak stomach. This study investigated the effects of processed GF in tert-butyl hydroperoxide (tBHP)-treated gastric epithelial cells. Methods : Processed GF was made by applying 40% ginger juice or 10% ethanol for 24 h and then roasting at 150℃ for 5 minutes. Apoptosis was determined by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. Mitochondrial membrane potential (MMP) was monitored by flow cytometry using the membrane permeable fluorescent dye Rh123. Protein expression was measured by Western blot analysis. Results : Cell viability was reduced by tBHP and restored by ethanol extract of GF (GFE). In the TUNEL assay, it was found that cell death by tBHP was due to apoptosis, and GFE had an anti-apoptotic effect. Processed GF roasted with ginger juice showed the best anti-apoptotic effect. Processed GF also inhibited MMP loss and restored tBHP-induced changes in expression levels of apoptosis-related proteins. Increased ROS production and GSH depletion after tBHP treatment were significantly reduced by processed GF. In addition, tBHP-induced activation of mitogen-activated protein kinase (MAPK) was inhibited by processed GF. Conclusion : These results demonstrate that the processed GF is able to protect gastric epithelial cells from oxidative stress-induced cell death with antiapoptotic and antioxidant activity. In addition, it shows that the processing of GF, which have been traditionally used for gastrointestinal protection, partially have scientific validity.

A Study on the Photocatalytic Decomposition of Organic Dyes by Porous Polyethersulfone/TiO2 Composite Membrane (기공형 polyethersulfone/TiO2 복합체 멤브레인의 유기염료분해 반응에 대한 광촉매 특성 연구)

  • Composite membranes consisting of TiO2 nanoparticles (NPs) and porous polymers have been widely utilized in photocatalytic water treatment because the composite membranes can allow an easy recovery of NPs after the photocatalytic reaction as well as the reduction of fouling in the membrane. However, the photocatalytic efficiency of the immobilized TiO2 NPs in the composite membranes has been discussed to a limited degree. In this study, we prepared polyethersulfone (PES)/TiO2 composite membranes to study the photocatalytic decomposition of organic dyes under light illumination. The decomposition kinetics of dye molecules by the PES/TiO2 composite membranes and colloidal TiO2 NPs have been compared to discuss the photocatalytic efficiency of NPs before and after their immobilization on the polymer membrane.

Carbon-Reduced Titanium Dioxide Production and Characterization Using Dyeing Wastewater Sludge (염색 폐수 슬러지를 활용한 탄소저감형 이산화티타늄 제조 및 특성 분석)

  • This study is to manufacture a titanium dioxide (TiO2) photocatalyst by recycling sludge generated using titanium tetrachloride (TiCl4) as a coagulant. Compared to general sewage, a TiCl4 coagulant was applied to dyeing wastewater containing a large amount of non-degradable organic compounds to evaluate its performance. Then the generated sludge was dried and fired to prepare a photocatalyst (TFS). Scanning electron microscope-energy dispersive X-ray spectroscopy (SEM-EDX), X-ray diffraction (XRD), and nitrogen oxide reduction experiments were conducted to analyze the surface properties and evaluate the photoactive ability of the prepared TFS. After using titanium tetrachloride (TiCl4) as a coagulant in the dyeing wastewater, the water quality characteristics were measured at 84 mg/L of chemical oxygen demand (COD), 10 mg/L of T-N, and 0.9 mg/L of T-P to satisfy the discharge water quality standards. The surface properties of the TFS were investigated and the anatase crystal structure was observed. It was confirmed that the ratio of Ti and O, the main components of TiO2, accounted for more than 90 %. As a result of the nitric oxide (NO) reduction experiment, 1.56 uMol of NO was reduced to confirm a removal rate of 20.60 %. This is judged to be a photocatalytic performance similar to that of the existing P-25. Therefore, by applying TiCl4 to the dyeing wastewater, it is possible to solve the problems of the existing coagulant and to reduce the amount of carbon dioxide generated, using an eco-friendly sludge treatment method. In addition, it is believed that environmental and economic advantages can be obtained by manufacturing TiO2 at an eco-friendly and lower cost than before.

Preparationand Characterization of Rutile-anatase Hybrid TiO2 Thin Film by Hydrothermal Synthesis

  • Nanoporous $TiO_2$ films are commonly used as working electrodes in dye-sensitized solar cells (DSSCs). So far, there have been attempts to synthesize films with various $TiO_2$ nanostructures to increase the power-conversion efficiency. In this work, vertically aligned rutile $TiO_2$ nanorods were grown on fluorinated tin oxide (FTO) glass by hydrothermal synthesis, followed by deposition of an anatase $TiO_2$ film. This new method of anatase $TiO_2$ growth avoided the use of a seed layer that is usually required in hydrothermal synthesis of $TiO_2$ electrodes. The dense anatase $TiO_2$ layer was designed to behave as the electron-generating layer, while the less dense rutile nanorods acted as electron-transfer pathwaysto the FTO glass. In order to facilitate the electron transfer, the rutile phase nanorods were treated with a $TiCl_4$ solution so that the nanorods were coated with the anatase $TiO_2$ film after heat treatment. Compared to the electrode consisting of only rutile $TiO_2$, the power-conversion efficiency of the rutile-anatase hybrid $TiO_2$ electrode was found to be much higher. The total thickness of the rutile-anatase hybrid $TiO_2$ structures were around $4.5-5.0{\mu}m$, and the highest power efficiency of the cell assembled with the structured $TiO_2$ electrode was around 3.94%.
