칠장사 오불회 괘불탱 화기에 기록된 채색 재료의 해석 (Interpretation of Coloring Materials Recorded in Ceremonial Writing of the Hanging Painting of Chiljangsa Temple (Five Buddhas))

  • 본 연구는 칠장사 오불회 괘불탱의 과학적 조사 결과를 종합하여 화기에 기록된 채색 재료에 대해 해석하고자 하였다. 연구결과 진분은 연백, 주홍은 진사, 황단은 연단, 황금은 금, 석자황은 석황으로 확인되었다. 녹색의 당하엽과 삼록은 copper trihydroxychloride, 청색의 대청과 중청은 석청으로 확인되며 이들의 입도에 따라 색상의 변화를 준 것으로 보인다. 연지와 청화는 각각 적색과 청색의 유기염료로서 청색은 쪽의 존재가 확인되나 연지는 고문헌과 한자의 명칭에 차이를 보여 통시적으로 사용된 연지인지는 불분명하다. 수토황은 괘불탱에서 그 존재가 확인되지 않았으나 황색 계열의 재료로 보이며 토양추출물이나 등황일 가능성이 높다.

추적자 실험 및 3차원 수치모의를 이용한 저수지 수처리 장치의 영향반경 평가 (Assessing the Influence Radius of a Water Treatment System Installed in a Reservoir Using Tracer Experiment and 3D Numerical Simulation)

  • The objective of this study was to evaluate the radius of influence of effluent of water treatment system developed for the purpose of improvement of reservoir water quality using fluorescent dye (Rhodamine-WT) tracer experiment and 3-D numerical model. The tracer experiment was carried out in a medium-sized agricultural reservoir with a storage capacity of $227,000m^3$ and an average depth of 1.6 m. A guideline with a total length of 160 m was installed at intervals of 10 m in the horizontal direction from the discharge part, and a Rhodamine measurement sensor (YSI 6130, measurement range $0-200{\mu}g/L$) was used to measure concentration changes in time, distance, and depth. Experimental design was established in advance through Jet theory and the diffusion process was simulated using ELCOM, a three dimensional hydraulic dynamics model. As a result of the study, the direct effect radius of the jet emitted from the applied water treatment system was about 50-70 m, and the radius of physical effect by the advection diffusion was judged to be 100-120 m. The numerical simulations of effluent advection-diffusion of the water treatment system using ELCOM showed very similar results to those of the impact radius analysis using the tracer experiment and jet flow empirical equations. The results provide valuable information on the spatial extent of the water quality improvement devices installed in the reservoir and the facility layout design.

카드뮴으로 유발한 간세포 자멸사에서 진피의 보호효과 (Protective effect of Citrus unshiu peel on the cadmium-induced apoptosis in HepG2 cells)

  • Objective : Citrus unshiu peel (Citri Unshius Pericarpium) has been prescribed to suppress coughing and phlegm in Korean medicine. In this study, the effect of ethanol extract of Citrus unshiu peel (CEE) on apoptosis was investigated using cadmium chloride (CdCl2) treated HepG2 cells. Methods : CEE was prepared by extracting 300 g of Citri Unshius Pericarpium in 3 L of ethanol for 72 h. Apoptosis was determined by the TUNEL assay. The mitochondrial membrane potential (MMP) was monitored using the membrane-permeable fluorescent dye Rh123. The expression level of each protein was monitored by Western blot analysis. Results : CEE protected HepG2 cells from apoptosis as determined by the TUNEL assay. A decrease in MMP was observed in cells exposed to cadmium, indicating that mitochondria are involved in the induction of apoptosis. However, CEE recovered the reduction in MMP caused by cadmium. In addition, decreased expression of B-cell lymphoma 2 (Bcl-2), procaspase, and poly(ADP-ribose) polymerase (PARP) by cadmium was increased by CEE. The anti-apoptotic effect of CEE was found to be associated with inhibition of JNK and p38 phosphorylation when examining the expression of phosphorylated MAPK by Western blot. Conclusion : This study showed that CEE exerted anti-apoptotic effects in cadmium-induced HepG2 cells by inhibiting the reduction of MMP and changes in the expression level of apoptotic proteins. These results suggest the potential for CEE to be used for heavy metal-induced liver damage.

산화스트레스를 유발한 위 상피세포에서 수치 치자의 세포 보호 효과 (Cytoprotective effect of the processed Gardeniae Fructus on oxidative stress-induced gastric epithelial cells)

  • Objective : Gardeniae Fructus (GF) has bitter and cold nature. Thus, it has been traditionally prescribed in processed form roasted with ginger juice for patients with a weak stomach. This study investigated the effects of processed GF in tert-butyl hydroperoxide (tBHP)-treated gastric epithelial cells. Methods : Processed GF was made by applying 40% ginger juice or 10% ethanol for 24 h and then roasting at 150℃ for 5 minutes. Apoptosis was determined by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. Mitochondrial membrane potential (MMP) was monitored by flow cytometry using the membrane permeable fluorescent dye Rh123. Protein expression was measured by Western blot analysis. Results : Cell viability was reduced by tBHP and restored by ethanol extract of GF (GFE). In the TUNEL assay, it was found that cell death by tBHP was due to apoptosis, and GFE had an anti-apoptotic effect. Processed GF roasted with ginger juice showed the best anti-apoptotic effect. Processed GF also inhibited MMP loss and restored tBHP-induced changes in expression levels of apoptosis-related proteins. Increased ROS production and GSH depletion after tBHP treatment were significantly reduced by processed GF. In addition, tBHP-induced activation of mitogen-activated protein kinase (MAPK) was inhibited by processed GF. Conclusion : These results demonstrate that the processed GF is able to protect gastric epithelial cells from oxidative stress-induced cell death with antiapoptotic and antioxidant activity. In addition, it shows that the processing of GF, which have been traditionally used for gastrointestinal protection, partially have scientific validity.

Development of transgenic disease-resistance root stock for growth of watermelon.(oral)

  • To protect the plant against several soil-borne pathogens, we are currently constructing disease-resistant transgenic root stock for the growth of cucurbitaceae vegetable plants, watermelon and gourd. We made a watermelon cDNA library from Cladosporium cucumerinum-Infected leaves for substractive hybriazation and differential screening. We isolated the several pathogen inducible cDNA clones, such as caffeoyl-CoA-methyltransferase, LAA induced protein, receptor-like kinase homolog, hydroxyproline-rich glycoprotein, catalase, calmodulin binding protein, mitochondrial ATPase beta subunit, methyl tRNA synthetase and WRKY transcription factors. We previously obtained CaMADS in pepper and galactinol synthase ( CsGolS) in cucumber that were confirmed to be related with disease-resistance. CaMADS and CsGolS2 were transformed into the inbred line 'GO701-2' gourd, the inbred line '6-2-2' watermelon and the Kong-dye watermelon by Agrobacterium tumerfaciens LBA4404. Plant growth regulators (zeatin, BAP and IAA) were used for shoot regeneration and root induction for optimal condition. Putative transgenic plants were selected in medium containing 100mg/L kanamycin and integration of the CaMADS and CsGO/S2 into the genomic DNA were demonstrated by the PCR analysis. We isolated major soil-borne pathogens, such as Monosporascus cannonballus, Didymella bryoniae, Cladosporium cuvumerinum from the cultivation area of watermelon or root stock, and successfully established artificial inoculation method for each pathogen. 리그닌 및 고분자/리그닌 복합소재 탄화 생성물의 흡착 특성 비교 (Comparative Study on Adsorption Properties of Carbons Derived from Lignin and Polymer/Lignin Composite Precursors)

  • 본 연구에서는, 탄화가 가능한 고분자/리그닌 복합소재 필름을 형성한 후, 제어된 온도 프로파일(profile)을 갖는 탄화 공정을 통해 탄화 생성물을 얻는다. 탄화 공정이 리그닌의 탄화 생성물 및 탄소 필름에 미치는 영향을 파악하여 성공적인 탄화물 생산이 가능한지를 탐색하였다. 얻어진 탄소 필름이 다양한 분자들, 비극성 유기 분자, 극성 유기 분자, 염료 분자 등에 보여주는 흡착 특성을 리그닌의 탄화물과 비교하며 자외선/가시광선(UV/Vis) 분광기로 분석하였다. 순환전류법으로 분석한 결과 본 연구에서 활용한 탄화 과정을 통해 탄소 필름이 성공적으로 얻어졌음을 확인하였다. 다양한 흡착 실험 결과 얻어진 탄화 리그닌이 다양한 분자들에 대한 흡착이 가능함을 보여주었고, 특히 염료 분자에 대해 효과가 가장 높게 나타났다. 이는, 분자 크기가 큰 분자에 대해 흡착이 용이함을 암시한다. 복합소재 필름의 경우 도입된 고분자가 리그닌의 흡착 성능을 다소 약화시키는 것으로 나타났다. 이 연구는 금속 양이온에 대한 흡착 거동을 분석한 이전 결과로 함께 향후 연구에 대한 중요한 정보를 제공할 것이다.

Synthesis and Characterization of Magnetic Cellulose Powder from Sawdust Waste

  • Timber industry waste is being examined for cellulose manufacturing to give important benefits. The study's goal is to investigate the properties of cellulose powder derived from sawdust waste after it has been reinforced with ferrous-ferric oxide nanoparticles (Fe3O4-NPs). Sawdust cellulose powder was produced from Sengon (Albizia chinensis) wood waste in this study. The crushed sawdust waste is handled with alkalization and bleaching. Cellulose powder is then reinforced with Fe3O4-NPs at 10 wt.%, 20 wt.%, and 30 wt.%. The magnetic cellulose powder was analysed by X-ray diffraction, Fourier Transform Infrared, scanning electron microscopy morphology, magnetic vibrating sample magnetometer, Brunauer-Emmett teller, and adsorption tests for Methylene Blue and Congo Red dyes. Structure study identifies sawdust as cellulose 1β, with peaks at 14°, 16°, and 22° diffraction angles. The addition of Fe3O4-NPs reduces the crystalline index of sawdust cellulose powder from 68.50% to 63.38%, and functional group bond analysis revealed many peak shifts indicating a change in the chemical bonds of magnetic cellulose powder. Incorporating Fe3O4-NPs into sawdust cellulose powder confers magnetic and superparamagnetic properties to the sawdust cellulose. Similarly, the surface texture of magnetic cellulose seems rougher as the surface area increases. These parameters imply a 31.8% increase in Congo Red adsorption, using adsorption kinetics based on the pseudo-first-order model.

수두 고단백 계통육성을 위한 기초적 연구 I. 계통육성을 위한 조단백질 분석법의 비교 (Basic Studies for the Breeding of High Protein Rice. I. Comparison of the analytical methods for the measurement of the protein content in the brown rice)

  • 수도 고단백 계통선발을 위한 미립내 질소함량 검정 방법을 첨부 검토하기 위하여 예비선발된 36계통 및 대조품종을 5가지 질소수준(0, 7.5, 15.0, 22.5, 30.0, kg/10a)에서 재배하여 그 현미를 Kjeldahl법, Dye binding법 및 Biurett법으로 분석하여 3가지 방법에 의한 분석치간의 상관을 검토하였으며 그 중 6개품종에 대하여 동일시료를 3가지 분석법으로 6회 반복한 분석치의 분산을 검토하였고 3가지 방법으로 시료 100점당 소요된 시약과 노력을 비교하였다. 그 결과를 요약하면 다음과 같다. 1. D.B.C.법에 의한 분석치는 Biurett법에 의한 분석치보다 Kjeldahl법과의 상관이 높았으나 D.B.C. 법에서는 극도로 높은 질소수준에서와 극도로 낮은 질소수준에서의 분석치는 Kjeldahl법에 의한 분석치에 비하여 과대평가되는 경향이 있다. Biurett법은 비교적 오차가 커서 Kjeldahl 법과의 상관이 떨어지지만 모든 질소수준에서 평행적인 경향이었다. 2. 품종에 따라 시용된 질소수주에 대한 반응도 달랐지만 품종과 질소수준에 따라 3가지 분석방법의 반응도 달랐다. 3. 3가지 분석방법별 분석치의 변이계수를 비교하면 Biurett법이 가장 크며 다음은 D.B.C.법인데 Biurett법에서는 각질소수준에서 모두 높았지만 질소수준이 높아짐에 따라 변이계수도 커졌는데 Kjeldahl법이나 D.B.C.에서는 질소수준이 낮아짐에 따라 변이 계수가 커졌다. 4. 3가 분석방법에서 소요되는 약품과 노력을 조사하여 Kjeldahl법에 비하여 D.B.C.법이 현저하게 절약되며 D.B.C.법 보다 Biurett법은 더욱 절약될 수 있음을 알았다.

약용버섯의 계통분류 및 국내유통 Inonotus속내 종간 구별을 위한 신속동정법 개발 (Phylogenetic analysis of the medicinal mushroom and taxonomical positions of their commercial products)

  • 국내유통약용버섯의 분류체계를 확립하고 이들 속 및 종간의 유연관계확립을 위하여 계통학적 정보를 지니고 있는 ITS부위의 염기서열을 밝히고 ITS1과 ITS2부위의 다양한 염기서열을 이용하여 분류학적 위치를 확립하였고, 시판 I. obliquus 종의 진위여부와 계통분류학적인 유연관계 확립 및 종 특이적인 유전자 탐침을 개발하였다. 본 연구의 조사결과에 의하면, 약용버섯으로 주로 시판되고 있는 국내유통균주는 총 6개의 속(Phellinus, Inonotus, Sparassis, Fomes, Ganoderma, Hericium)으로 나누어짐을 알 수 있었고 그 중에서 기존에 잘 알려진 상황버섯과 최근 들어 수입양이 급증하고 있는 차가버섯이 대부분을 차지하고 있는 것으로 확인되었다. 본 연구에 사용되어진 일명 차가버섯으로 유통 중인 15개 제품 중 중국에서 수입되어진 59번 균주가 P. pini로 확인되었으며 일본에서 수입되어진 51, 52번 균주가 P. baumii와 P. linteus와 유사종 혹은 동일종으로 확인되었다. 한편 I. rheades(AY237731)와 I. radiatus(AY354217) 및 F. fomentarius(AY354213)는 NCBI 등록 시 문제점이 있는 것으로 사료되며 본 실험에서 조사되어진 30번 균주 F. fomentarius가 정확한 말굽버섯인 것으로 사료된다.

심도자 검사 환자를 위한 구체적 감각 정보의 개발 (Development of Concrete Sensory Information for Patients Undergoing Cardiac Catheterization)

  • Cardiac catheterization is a diagnostic procedure which is intrusive and anxiety provoking. Patient education and information offer have been suggested as effective nursing interventions to reduce patients' anxiety and stress. Main objectives of this study are : 1) to develop concrete objective information for patients undergoing cardiac catheterization : 2) to analyze the types of information included in the newly developed concrete objective information. The subjects of this study are 11 patients who were admitted to the CCU of a general hospital in Seoul. The subjects were provided with preparatory information about cardiac catheterization by using interview and a booklet. After the procedure, the subjects were asked to describe additional information which they suggest to be added into the booklet and the physical sensations felt during the procedure. The results of the study are summarized as follows : Most subjects were satisfied with the preparatory information that was provided by interview and the booklet before the procedure. But patients suggested several problems related to the content of the booklet. First, they reported difficulty to differentiate the terms -coronary angio gram and cardiac catheterization. Also, some patients expressed that they feared after reading the information about the incision of inguinal area. Subjects responded that the information about the direct process of the test did not reduce their anxiety and the information was not detail enough. Next, most subjects would want to know about the monitor and the sound from monitor. They said that they could not hear instructions from doctor or nurse during the procedure due to tension. Considering above response results, the need for more effective way to provide information, like visual and auditory information through video tape for giving information is suggested. Sensations related to the procedure were the smell coming from sterilization of inguinal area, stinging pain in groin when the doctor inserts a needle into artery, and the sensation of pressure and moving of vessels surrounding neck when the catheter was inserted and visualized on fluoroscopy. Besides, subject reported hot sensation and burning feeling in face and chest area, and nausea when dye is injected by hand. In the analysis of information content, there was 79% agreement on the actual units of analysis that were coded. In the analysis of type of information, procedural information was 60.4%. Concrete objective information was 28.1%. and other information was 11.5%. Agreement of the coders in categorizing the units of information was determined by using Cohen's kappa which corrects for chance agreement. Cohen's kappa was .84.
