중독예방에 대한 중학교 교과서 내용 분석 (Contents Analysis of Addiction Prevention in Middle School Textbooks)

  • Purpose: The purpose of this study was to analyze addiction prevention related content shown in middle school text books. Methods: Using a combination of the terms "addiction", "drug", "medicine", "personal preference", "smoking", "drinking", "sex", "misuse" or "abuse" as key words, the researchers screened the table of contents of 23 randomly selected middle school textbooks from the 2009 curriculum. Finally 13 textbooks (physical education=10, health=3) were selected for this study, and analyzed using Krippendorff's contents analysis. Results: Through indepth discussion and investigation of the relevant textbooks, content related to addiction prevention included material addiction (77.8%) and behavioral addiction (22.2%). The construction of addiction prevention in middle school textbooks included understanding addiction, rejection of peer temptation, and empowerment of self-control. Conclusion: When developing an intervention for addiction prevention that targets middle school students, education with accurate addiction information must be considered so that the students have the correct knowledge about addiction and addiction prevention. Additionally, the ability to reject peer temptation when invited to act upon related addiction behaviors should be treated seriously. The empowerment of self-control is also important, and can be promoted through strengthening self-regulation competence.

홍삼박으로부터 산성다당체의 최적 추출 조건 분석 (Conditioning of the Extraction of Acidic Polysaccharide from Red Ginseng Marc)

  • This study was carried out to investigate the optimum conditions for extraction of acidic polysaccharides from red ginseng marc produced by manufacturing alcoholic extract from red ginseng. Method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharides in red ginseng marc. The amounts of acidic polysaccharides in water extract of red ginseng marc were increased with increasing extraction temperature. The contents of acidic polysaccharides were not significantly different despite of the extraction time increasing from 6 hours to 48 hours. The contents of starch in water-extract of red ginseng marc were increased with increasing extraction temperature. The starch amounts in water extract of red ginseng marc extracted for 48 hours were increased. The yields of polysaccharide precipitated from water-extract of red ginseng marc were increased with increasing extraction temperature. The hydration rate of acidic polysaccharides and starch from water-extract of red ginseng marc were decreased with increasing extraction temperature. The contents of starch were not significantly different despite of the extraction time increasing from 6 hours to 48 hours at $8^{\circ}C$. However, the rehydration rate of acidic polysaccharide for 48 hours were decreased at $8^{\circ}C$. The rehydration rate of acidic polysaccharide and starch extracted from 6 hours to 24 hours at $25^{\circ}C$ were not significantly different, but those extracted for 48 hours were increased. From the above results, we suggest that by altering the extraction conditions in red ginseng marc it is possible to develop optimum conditions for extraction that modulate the proportions of acidic polysaccharide and starch.

  • 최근 고령화 사회에 진입과 소득수준이 향상됨에 따라 의료 수요가 빠르게 증가되고 있다. 이중 신약 개발은 의료분야의 핵심 분야 중 하나로 전 세계적으로 많은 투자가 연구가 이루어지고 있다. 신약 개발의 중요한 과제 중 하나는 다양한 후보를 빠르고 정확하게 발굴하는 자동화 시스템을 개발하는 것이다. 고속으로 후보물질을 발굴하는 기술 중 대표적 기술이 세포 기반 고집적 스크리닝 시스템(High Contents Screening, 이하 HCS) 기술이다. 이 HCS 시스템에서 다양한 물질을 한꺼번에 스크리닝하기 위해서는 다기능 고정밀 디스팬서 제작이 필수적이다. 본 연구에서는 기존의 HCS 디스팬서 시스템이 가지고 있는 단점을 보완하기 위해 에어건과 압력 구동식 펌프를 이용한 새로운 컨셉의 비접촉식 디스팬서를 개발하고 그 가능성을 확인한다.

재배지역에 따른 농산물의 phytochemical 함량 (Phytochemical contents of agricultural products cultivated by region)

  • 최근 건강 증진을 위해 소비가 증가하고 있는 농산물을 대상으로 phytochemical 성분을 정량하고, 다양한 phytochemical 성분을 동시에 분석하는 방법에 대해 조사하였다. Lycopene, ${\alpha}$, ${\beta}$-carotene, cryptoxanthin과 lutein을 대상으로 동시분석을 실시한 결과 회수율은 각각 lycopene $120.7{\pm}4.1%$, lutein $89.1{\pm}3.5%$, ${\alpha}$-carotene $91.2{\pm}2.9%$, ${\beta}$-carotene $99.1{\pm}4.4%$, cryptoxanthin $100.0{\pm}5.3%$로 나타나 향후 다양한 phytochemical 성분을 동시에 분석하는데 유효하 방법이 될 것으로 판단된다. 농산물을 대상으로 4종의 phytochemical(${\beta}$-carotene, lycopene, lutein 및 cryptoxanthin)을 분석한 결과, lutein은 시금치>참다래>토마토>블루베리>메론 순이었으며 시료별 분석 결과 시금치(나주, 신안산), 참다래(해남, 보성산)와 블루베리(담양산)가 타 시료에 비해 lutein 함량이 더 높은 결과를 보였다. Lycopene은 토마토와 시금치에서 검출되었으며, 분석 시료 중 흑 토마토($56.66{\pm}7.48mg/kg$)와 장성 토마토($50.28{\pm}5.42mg/kg$)의 lycopene 함량이 가장 높았다. ${\beta}$-carotene의 경우 시금치와 토마토 시료에 가장 많이 함유되어 있었으며 특히, 나주 시금치($65.03{\pm}4.83mg/kg$)와 신안 시금치($37.67{\pm}5.49mg/kg$)에 다량 함유되어 있었다. Quercetin 분석 결과 블루베리에서 가장 높게 검출되어 담양산 블루베리가 $1,054.06{\pm}80.54mg/100g$으로 수입산($986.57{\pm}67.85mg/100g$) 보다 높았으며, kaempferol의 경우도 비슷한 경향을 보였다. 총 폴리페놀의 경우 블루베리(213.60~229.96 mg/100 g)가 가장 높은 결과를 보였고, 시금치(112.50~141.67 mg/100 g), 참다래(46.49~70.44 mg/100 g) 에서도 높게 검출되었다. 총 플라보노이드 함량을 분석한 결과 블루베리, 시금치 시료에서 타 시료에 비해 함량이 높게 검출되었으며, 비타민 C 함량은 참다래(39.45~86.79 mg/100 g), 한라봉(38.65~50.96 mg/100 g), 토마토(5.90~15.97 mg/100 g), 블루베리 순으로 나타났고, 블루베리에서 총 안토시아닌 함량을 측정한 결과 담양산 블루베리>수입산 블루베리 순으로 나타났다. 이상의 결과로 보아 유통 중인 농산물에 다양한 phytochemical 성분이 존재하는 것으로 확인되었으며, 이는 재배지역, 품종, 숙성 정도 등에 따라 조금씩 차이가 나는 것으로 판단된다.

Biphasic Release Characteristics of Dual Drug-loaded Alginate Beads

  • The dual drug-loaded alginate beads simultaneously containing drug in inner and outer layers were prepared by dropping plain (single-layered) alginate beads into $CaCl_2$ solution. The release characteristics were evaluated in simulated gastric fluid for 2 h followed by intestinal fluids thereafter for 12 h. The surface morphology and cross section of dual drug-loaded alginate beads was also investigated using scanning electron microscope (SEM). The poorlv water-soluble ibuprofen was chosen as a model drug. The surface of single-layered and dual drug-loaded alginate beads showed very crude and roughness, showing aggregated particles, surface cracks and rough crystals. The thickness of dual drug-loaded alginate beads surrounded by outer layer was ranged from about 57 to 329mcm. The distinct chasm between inner and outer layers was also observed. In case of single-layered alginate bead, the drug was not released in gastric fluid but was largely released in intestinal fluid. However, the release rate decreased as the reinforcing $Eudragit^{\circledR}$ polymer contents increased. When the plasticizers were added into polymer, the release rate largely decreased. The release rate of dual drug-loaded alginate beads was stable in gastric fluid for 2 h but largely increased when switched in intestinal fluid. The drug linearly released for 4 h followed by another linear release thereafter, showing a distinct biphasic release characteristics. There was a difference in the release profiles between single-layered and dual drug-loaded alginate beads due to their structural shape. However, this biphasic release profiles were modified by varying formulation compositions of inner and outer layer of alginate beads. The release rate of dual drug-loaded alginate beads slightly decreased when the outer layer was reinforced with $Eudragit^{\circledR}$ RS1OO polymers. In case of dual drug-loaded alginate beads with polymer-reinforced outer layer only, the initial amount of druc released was low but the initial release rate (slope) was higher due to more swellable inner cores when compared to polymer-reinforced inner cores. The current dual drug-loaded alginate beads may be used to deliver the drugs in a time dependent manner.

페노피브레이트 유연물질 표준품 대체시험법 개발 (Development of an Alternative Analytical Method without Related Substance Reference Standards for Fenofibrate in Korean Pharmacopoeia)

  • Analytical method for related substances can be categorized into two methods depending on the necessity of reference standard (RS). The analytical method of related substances with RS is fast and accurate, but it's very expensive and technically difficult to synthesize RS due to their complicated structure. Another method is using relative retention time (RRT) and relative response factor (RRF) which are already validated with RS. Validation of this method is not easy and time consuming, but once it has been developed, it can save cost and time. In this study, we developed the analytical method for related substances of fenofibrate using RRT and RRF. We validated the method by evaluating specificity, linearity, accuracy and precision according to the "Manual for Guideline Application for Validation of Analytical Procedures" of MFDS. Also, we calculated RRT and RRF between fenofibrate and fenofibrate related substances. The results of this study showed high specificity for fenofibrate and fenofibrate related substances. Correlation coefficient(r) of all substances were more than 0.99, and the recovery of fenofibrate, fenofibrate related substance I, II and III were 99.44%, 100.84%, 99.14% and 101.58%, respectively. Precision of fenofibrate and its related substances were ranged between RSD 0.29% and 0.93%. Quantification limits of fenofibrate, fenofibrate related substance I, II and III were determined to be $0.03{\mu}g/ml$, $0.05{\mu}g/ml$, $0.04{\mu}g/ml$ and $0.02{\mu}g/ml$, respectively by confirming signal to noise ratio of each chromatogram. The RRT for fenofibrate related substance I, II and III were determined to be 0.35, 0.41 and 1.34, respectively. Also, the RRF for fenofibrate related substance I, II and III were determined to be 1.28, 0.98 and 0.79, respectively. The developed method was applied to determine contents for fenofibrate related substances in commercial fenofibrate (active pharmaceutical ingredient). As a result, developed analytical methods of related substances will be used for revising the monograph of fenofibrate in Korean Pharmacopoeia revision and contribute quality control of drugs by improving cost and time consuming problem of RS.

뇌경색 환자에게 사용된 덱스트란 40의 부작용 분석 (Adverse Drug Reactions of Dextran 40 Treated to the Cerebral Infarction Patients)

  • 뇌경색으로 입원하여 덱스트란 40을 투여받고 약물유해반응이 발생하여 의약품 부작용 보고서에 의해 보고된 전주예수병원 환자 22명을 대상으로 하였다. 이 연구의 목적은 뇌경색으로 덱스트란 40을 투여받은 환자를 대상으로 덱스트란 40의 약물유해반응과 부작용을 조사 및 보고함으로써 약물유해반응을 줄이기 위한 방안을 제시하고자 한다. 덱스트란의 부작용은 저혈압, 오심, 호흡곤란, 전신적인 두드러기, 열이고 과량 투여 시 폐부종을 유발한다. 평균 입원 기간은 20일로 입원환자의 26.8%가 입원기간의 연장을 경험하였고 덱스트란 40의 약물유해반응 발생률은 12.4%였고, 9명(28.1%)이 itching, rash는 7명(21.9%)이었다. 주입초기에, 초회 주사에서 덱스트란 아나필락틱 반응이 나타날 수 있다고 보고되고 있으나 투여 후 4일째에 부작용이 발생한 환자가 4명으로 18.2%에 이른다. 우리나라에는 덱스트란 1이 아직 널리 알려져 있지 않지만 미국 FAD에서는 사용을 권장하고 있으며, 덱스트란 40의 약물유해반응을 줄이기 위해서는 덱스트란 1의 사용을 고려해 볼 필요가 있다.

졸피뎀의 체계적 관리 방안 연구 (A Study on the Systematic Management of Zolpidem)

  • 일반적으로 수면제로 사용되는 졸피뎀은 신경 억제 물질을 증가시키기 위해 GABA 수용체에 결합한다. 실제 졸피뎀을 4주 이상 연속으로 복용하는 경우는 많이 있다. 현재 한국에서 시행되고 있는 의약품 사용평가(DUR)은 졸피뎀의 사용 패턴을 확인할 수 있다. 수면제인 졸피뎀의 과다 사용을 예방 할 필요가 있다. 본 연구는 2019년 5월 10일부터 2019년 5월 15일까지 진행되었다. 검색된 총 125,197건의 문서 중 1차 및 2차 선별을 실시하여 진행하였다. 졸피뎀 과다 사용과 직접적인 관련이 있는 최종 254개의 문헌 사례를 선정하였다. 체계적인 문헌 연구를 통해 졸피뎀의 과다 사용을 예방 할 수 있는 방안을 모색하고자 하였다. 연구대상 선별과정에서 연구의 비뚤림 효과를 최소화 하였다. 졸피뎀의 과다 사용은 심각했으며 사용량이 계속 증가하는 추세였다. 졸피뎀을 이용한 범죄도 증가하고 있었다. 졸피뎀은 마약류 통합 관리 시스템과 의약품 사용평가를 통해 관리 될 수 있다고 연구되었다. 졸피뎀의 사용을 줄이기 위해서는 마약류 통합관리 시스템과 과다 사용자 교육을 통한 수요와 공급을 동시에 억제할 필요가 있다.

감마선 조사(照射) 생지황(生地黃)의 Catapol 성분에 대한 안전성 및 유전독성학적 연구 (Studies on the Stability of Catapol Components, and Genotoxic Safety of ${\gamma}-Irradiated$ Rehmanniae Radix crude)

  • This study is concerning to identify the hygienic problems occurring in processing, storage, and distribution of Rehmanniae radix crude, and to investigate the possibility of application of safe and hygienic ${\gamma}-irradiation$ techniques. The results are as follows. To compare the contents of catapol, index compound of Rehmanniae Radix crude, between before and after ${\gamma}-irradiated$,0,2.5,5,7.5,10,12.5,15,20, and 30 kGy of ${\gamma}-irradiation$ was applied to standard catapol and Rehmanniae radix crude. The contents of catapol in standard material and Rehmanniae radix crude were decreased with the increase of ${\gamma}-irradiation$ level. However, active components of Rehmanniae Radix crude were not changed with ${\gamma}-irradiation$. These results indicate that active components of Rehmanniae Radix crude were preserved after ${\gamma}-irradiation$ and those of Rehmanniae Radix crude did not showed significant change after irradiation of ${\gamma}-ray$ up to 20 kGy.

First Report on Isolation of Salmonella Enteritidis from Eggs at Grocery Stores in Korea

  • Salmonella Enteritidis is responsible for causing foodborne diseases upon consumption of egg products. While cases of S. Enteritidis isolation from eggs have been reported in other countries, no such cases have previously been reported in Korea. In this study, we report the first isolation and identification of S. Enteritidis from domestically distributed eggs in Korea. Eggs were collected from eight countrywide grocery stores during different seasons between 2011 and 2012. Egg contents and washing solution of egg shells were incubated in buffered peptone water, and the enriched broth was further enriched in tetrathionate broth and Rappaport-Vassiliadis. The secondary enriched broth was streaked on xylose lysine desoxycholate agar. The suspected colonies were confirmed to S. Enteritidis by a biochemical test, serotyping, and PCR test. Genetic relatedness among the isolates was analyzed using Diversilab Salmonella kit. Three strains of S. Enteritidis were isolated from egg contents and egg shells collected from grocery stores of the Eumseong-city in the fall of 2011. All three stains showed resistance to chloramphenicol, streptomycin, nalidixic acid, and ampicillin by the disk diffusion method. In addition, the isolates showed more than 99% DNA homology, indicating that they were presumably identical strains. Therefore, there is a requirement to monitor and control against S. Enteritidis from eggs in Korea.
