Multiple Drug Resistance and Transferable R-plasmid of Streptococci Isolated from Diseased Olive Flounder (연쇄상구균의 약제내성과 전이성 R-plasmid의 특성)

  • MICs of 8 chemotherapeutic agents against forty streptococcal isolates were determined. These strains were isolated from diseased olive flounder, Paralichthys olivaceus, and showed 2-5 multiple drug resistance against different antibacterial agents including ampicillin, doxycycline (DOXY), erythromycin, florfenicol, flumequine, norfloxacin, oxolinic acid, and oxytetracycline (OTC). In conjugation experiment, we found transferable R plasmids carrying OTC and DOXY resistance determinant in 3 drug resistance strains analyzed. Six out of 40 isolates showed positive signal in colony hybridization with the R plasmid DNA (pST9) as a probe. It suggests that other types R plasmid different from pST9 is also involving in multiple drug resistance of streptococci isolated from olive flounder.

Effect of Sub-Minimal Inhibitory Concentrations of Antibiotics on Biofilm Formation and Coaggregation of Streptococci and Actinomycetes

  • Minimal inhibitory concentration (MIC) is the lowest antibiotic concentration that inhibits the visible growth of bacteria. Sub-minimal inhibitory concentration (Sub-MIC) is defined as the concentration of an antimicrobial agent that does not have an effect on bacterial growth but can alter bacterial biochemistry, thus reducing bacterial virulence. Many studies have confirmed that sub-MICs of antibiotics can inhibit bacterial virulence factors. However, most studies were focused on Gram-negative bacteria, while few studies on the effect of sub-MICs of antibiotics on Gram-positive bacteria. In this study, we examined the influence of sub-MICs of doxycycline, tetracycline, penicillin and amoxicillin on biofilm formation and coaggregation of Streptococcus gordonii, Streptococcus mutans, Actinomyces naeslundii, and Actinomyces odontolyticus. In this study, incubation with sub-MIC of antibiotics had no effect on the biofilm formation of S. gordonii and A. naeslundii. However, S. mutans showed increased biofilm formation after incubation with sub-MIC amoxicillin and penicillin. Also, the biofilm formation of A. odontolyticus was increased after incubating with sub-MIC penicillin. Coaggregation of A. naeslundii with S. gordonii and A. odontolyticus was diminished by sub-MIC amoxicillin. These observations indicated that sub-MICs of antibiotics could affect variable virulence properties such as biofilm formation and coaggregation in Gram-positive oral bacteria.

Characterization of Antimicrobial Substance Producing Lactococcus sp. HM58 Isolated from Gastrointestinal Track of Flounder

  • A lactic acid bacterium showing antimicrobial activity against fish pathogen was isolated from gastrointestinal tract of flounder for the purpose of use as an aquaculture probiotics. From the analysis of morphological and physiological characteristics, the isolated strain was named as Lactococcus sp. HM58. Antimicrobial substance (AMS) from Lactococcus sp. HM58 showed strong growth inhibitory activity against Streptococcus sp., which is a fish pathogenic bacterium. AMS was presumed a proteinaceous compound with stability in heat and wide pH range from 2 to 10. It was started to produce in exponential growth phase and was not produced any more in stationary phase. It showed comparatively broad antimicrobial spectrum against most of gram positive bacteria used for this study. About $84\%$ of Lactococcus sp. HM58 was able to survive in the artificial gastric juice though it was low to the extent in the artificial bile juice. In the sensitivity test for various antibiotics, this strain was highly sensitive for doxycycline, erythromycin, amoxicillin clavu1anic acid and ampicillin.

Matrix Metalloproteinase Inhibitors Attenuate Neuroinflammation Following Focal Cerebral Ischemia in Mice

  • The aim of this study was to investigate whether matrix metalloproteinase (MMP) inhibitors attenuate neuroinflammation in an ischemic brain following photothrombotic cortical ischemia in mice. Male C57BL/6 mice were anesthetized, and Rose Bengal was systemically administered. Permanent focal ischemia was induced in the medial frontal and somatosensory cortices by irradiating the skull with cold white light. MMP inhibitors, such as doxycycline, minocycline, and batimastat, significantly reduced the cerebral infarct size, and the expressions of monocyte chemotactic protein-1 (MCP-1), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and indoleamine 2,3-dioxygenase (IDO). However, they had no effect on the expressions of heme oxygenase-1 and neuroglobin in the ischemic cortex. These results suggest that MMP inhibitors attenuate ischemic brain injury by decreasing the expression levels of MCP-1, TNF-${\alpha}$, and IDO, thereby providing a therapeutic benefit against cerebral ischemia.

Comparison of pretreatment methods for tetracyclines analysis by HPLC (테트라싸이클린계 잔류항생물질 분석을 위한 전처리방법 비교)

  • Solid phase extraction(SPE) and matrix solid phase dispersion(MSPD) have been studied as preparation procedures for tetracyclines analysis by high-performance liquid chromatography(HPLC) in meat. The recovery range was 74${\sim}$98% for SPE, and 72${\sim}$93% for MSPD at spiked levels of 100ng/g for oxytetracycline(OTC), tetracycline(TC), chlortetracycline(CTC), and doxycycline(DC). The detection limits were 15${\sim}$78ng/g for SPE and 25${\sim}$84ng/g for MSPD, respectively. Analytical method was HPLC with UV detector. The purpose of this study was developing a practical, accurate and precise method for rapid extraction and quantitation of tetracycline residues in meat.

Cloning and Characterization of the Tetracycline Resistant Gene, tetB, from Vibrio parahaemolyticus

  • A tetracycline resistant Vibrio parahaemolyticus, capable of growing on TCBS medium containing tetracycline, was isolated from cultivated fishes. A gene responsible for the tetracycline resistance was cloned from chromosomal DNA of the V. parahaemolyticus strain using Escherichia coli KAM3, which lacks major multi-drug efflux pumps (${\Delta}acrB$) as host cells. The nucleotide sequence and homology analysis revealed an open reading frame (ORF) for tetracycline resistance protein (TetB). In order to characterize the antibiotic resistance of TetB originated from the V. parahaemolyticus strain, the gene was sub cloned into plasmid pSTV28. The resulting plasmid was designated as pSTVTetB and transformated into E. coli KAM3. E. coli KAM3 cells harboring the recombinant plasmid pSTVTetB are able to grow on plates containing tetracycline and oxytetracycline but not doxycycline, indicating that the tetB gene confers the tetracycline- and oxytetracycline-resistance to the host cell.

Diagnosis of Anaplasmosis by PCR and Long Term Medication in a Dog (PCR을 이용한 개의 아나플라즈마병 진단과 장기간 치료)

  • A 4-year-old female Shunauzer dog was referred to the Veterinary Teaching Hospital of Chungbuk National University due to anorexia and depression. The dog had a history of regular walking on grass fields, weight loss, and hyperthermia $(40.6^{\circ}C)$. In the physical examination, lymph node enlargement was confirmed. Complete blood count result revealed leukocytosis and thrombocytopenia but there was no decreasing of red blood cells. On blood chemistry, serum ALP, GGT, CPK, and LDH were elevated. Abdominal radiograph showed splenomegaly. Anaplasma platys infection was suspected with inclusion body-like substances in platelets on blood smear. Anaplasma platys was confirmed by PCR. On the basis of laboratory examination, final diagnosis was anaplasmosis. Treatment was followed for 3 months with tetracycline and doxycycline. The patient was monitored every week during the treatment. The patient has recovered to normal condition without any clinical signs. We are going to emphasize the need of PCR technique in diagnosis and to report the possibility of long term treatment more than two months in rickettial disease.

Cell Type-Specific and Inducible PTEN Gene Silencing by a Tetracycline Transcriptional Activator-Regulated Short Hairpin RNA

  • Inducible and reversible gene silencing in desired types of cells is instrumental for deciphering gene functions using cultured cells or in vivo models. However, efficient conditional gene knockdown systems remain to be established. Here, we report the generation of an inducible expression system for short hairpin RNA (shRNA) targeted to PTEN, a well-documented dual-specificity phosphatase involved in tumor suppression and ontogenesis. Upon induction by doxycycline (DOX), the reverse tetracycline transcriptional activator (rtTA) switched on the concomitant expression of GFP and a miR-30 precursor, the subsequent processing of which released the embedded PTEN-targeted shRNA. The efficacy and reversibility of PTEN knockdown by this construct was validated in normal and neoplastic cells, in which PTEN deficiency resulted in accelerated cell proliferation, suppressed apoptosis, and increased invasiveness. Transgenic mice harboring the conditional shRNA-expression cassette were obtained; GFP expression and concurrent PTEN silencing were observed upon ectopic expression of rtTA and induction with Dox. Therefore, this study provides novel tools for the precise dissection of PTEN functions and the generation of PTEN loss of function models in specific subsets of cells during carcinogenesis and ontogenesis.

The Study on Fish Diseases with Reference to Bacterial Susceptibility to Antibiotics in the Southern Area of Kyeognam (경남 남부지역 양식어류 질병에서 항생제 감수성에 관한 연구)

  • The examination with 96 bacteria isolates for antibiotics susceptibility was resulted in exploring many antibiotics resistance isolates from the diseased fishes. Vibrio sp. isolates were demonstrated over 90% resistances for Penicillin (P), Amoxicillin (Amc), Erythromycin (I), Colistin (CL). Streptomycin (S). Novobiocin (NV) and Neomycin(N), these isolates were, however, appeared over 80% susceptibilities for Norfloxacin (Nor), CE and UB. In Edwardsiella tarda case, 90 to 100% high resistance was observed for P, Doxycycline (Do), Tetracyclin (Te), Oxytetracycline (T), E, CL, Sulfamethoxasoletrimetoprim (SXT), S, Oxolinic acid (OA), W, Neomycin(N), on the contrary, 90 to 100% high susceptibilities was found for Amc, Nor, Ciprofloxacin (Cip), Orbifloxacin (ORB), Enrofloxacin (ENR), Flumeguine (UB) and NA. CL, Pefloxacin (PEF),S, Flumeguine (UB), OA, NA, NV N was exhibited 90 to 100% resistances for Streptococcus sp., on the other hand, 100% susceptible to AMC and 80% susceptible to Do, Te, ENR and UB was recognised. Lastly, Photobacterium damsela subsp. piscicida was showed 100fe susceptible to Amc and 86% susceptible to NOR, CIP ENR and UB. As a consequence, fish bacterial pathogens isolated from Kyeongnam area, especially Tongyeong-si, Geoje-si, and Goseong-gun, were showed highly resistant to a variety of antibiotics available in the field.

Study on antimicrobial resistance of Escherichia coli isolated from domestic beef on sale (2) (유통되는 쇠고기에서 분리한 대장균의 항생제 내성 조사.연구 (2))

  • In this study, antimicrobial resistance of E. coli isolated from domestic beef on sale in Busan and Gyeongnam province was investigated from March to October 2008. A total of 400 beef samples were collected for the monitoring of antimicrobial resistance, and 39 (9.8%) strains of E. coli were isolated. Antimicrobial resistance test was carried out by agar disc diffusion method with 17 antimicrobials. In general, E. coli isolates showed the highest antimicrobial resistance to tetracycline (85.3%), followed by doxycycline (76.5%), streptomycin (61.8%) and sulfamethoxazole/trimethoprim (61.8%). Then they showed higher resistance to several antimicrobials like kanamycin and neomycin (55.9%). However, They had low antimicrobial resistance to amikacin (8.8%), amoxicillin/clavulanic acid (2.9%). Of 39 isolates, 31 (79.5%) were resistant to more than 2 antimicrobials. Among 17 antimicrobials examined, tetracyclines were the most resistant, followed by aminoglycosides, sulfonamides. The resistance was seemed to be correlated to amounts of antimicrobial use. In the result of this study, we suggest that there be need to regulate the abuse of antimicrobial on food-producing animals in Korea because the concern on antimicrobial resistant is gradually increased worldwide.
