최적이론에 의하여 설계된 최소 깊이 더블티 슬래브의 휨거동 평가 (Evaluation on Flexural Behavior of Double-tee Slabs with the Least Depth from Optimization Process)

  • Precast prestressed double-tee slab may be designed by the PCI Design Handbook. It is based on the bridge construction and is required for reorganization for the use of buildings in the domestic construction environments. Much enhanced sections are developed from the reforming process on the determined design factors in the previous experimental works on double tees. Pre-determined shape, reinforcement detail, and 5000 psi concrete strength can not be expected as the best solution for the domestic construction requirements because large amount of use on that systems are anticipated. Flexural tests are performed on four full-scale 12.5m proto-type models, "least depth double tee", which are resulted from the optimization process. Domestic superimposed live load regulation, domestic material properties which is available to product, building design requirements and economy in construction are considered as the main factors to establish. the first two sections are double tee section for 1.2 ton/$\m^2$ market live load with straight and one-point depressed strands and the second two are for 0.6 ton/$\m^2$ parking live load with those strand types. All of the specimens tested fully comply with the flexural strength requirements as specified by ACI 318-95. However, the research has shown that following improved considerations are needed for better result in practice. The locations and method of connection for the lowest bottom mild bar, connection method between precast and cast-in-place concrete, and dap-end reinforcement are need to be improved.

장·노년층의 사회적 고립감이 스마트폰 중독에 미치는 영향 : 자아통제감과 사회적지지의 매개효과 (Influence of Social Isolation on Smart Phone Addiction through Self-regulation and Social Support)

  • 본 연구는 장·노년층의 스마트폰 중독에 관련하여 이에 영향을 미치는 주요 변인들과의 인과관계를 검증하고자 하였다. 특히, 장·노년층의 스마트폰 중독에 영향을 미치는 주요 요인으로 사회적 고립감에 초점을 맞추어 장·노년층의 사회적 고립감이 자아통제감과 사회적지지에 영향을 미치는 동시에, 자아통제감과 사회적지지를 매개하여 스마트폰 중독에 영향을 미칠 것으로 보고 연구모형을 설정하였다. 이를 위하여 본 연구는 대전광역시에서 거주하는 장·노년층을 대상으로 1:1 설문조사를 실시하여 404명의 자료를 수거하였으며, 최종 404명의 자료를 분석하였다. 구조방정식모형을 통해 분석한 결과, 장·노년층의 사회적 고립감은 스마트폰 중독에 직접적인 영향을 미쳤으며, 자아통제감과 사회적지지의 이중매개 경로를 거쳐 스마트폰 중독에 영향을 미치는 것으로 나타났다. 본 연구는 이러한 연구결과에 기초하여 장·노년층의 스마트폰 의존 특성을 이해하고 관련 연구를 제고시킴은 물론, 함의 및 관련 후속연구의 필요성을 제시하였다.

Post-Translational Regulation of the RSF1 Chromatin Remodeler under DNA Damage

  • Chromatin remodeling factors are involved in many cellular processes such as transcription, replication, and DNA damage response by regulating chromatin structure. As one of chromatin remodeling factors, remodeling and spacing factor 1 (RSF1) is recruited at double strand break (DSB) sites and regulates ataxia telangiectasia mutated (ATM) -dependent checkpoint pathway upon DNA damage for the efficient repair. RSF1 is overexpressed in a variety of cancers, but regulation of RSF1 levels remains largely unknown. Here, we showed that protein levels of RSF1 chromatin remodeler are temporally upregulated in response to different DNA damage agents without changing the RSF1 mRNA level. In the absence of SNF2h, a binding partner of RSF1, the RSF1 protein level was significantly diminished. Intriguingly, the level of RSF1-3SA mutant lacking ATM-mediated phosphorylation sites significantly increased, and upregulation of RSF1 levels under DNA damage was not observed in cells overexpressing ATM kinase. Furthermore, failure in the regulation of RSF1 level caused a significant reduction in DNA repair, whereas reconstitution of RSF1, but not of RSF1-3SA mutants, restored DSB repair. Our findings reveal that temporal regulation of RSF1 levels at its post-translational modification by SNF2h and ATM is essential for efficient DNA repair.

Statistical Analysis of Gene Expression in Innate Immune Responses: Dynamic Interactions between MicroRNA and Signaling Molecules

  • MicroRNAs (miRNAs) are known to negatively control protein-coding genes by binding to messenger RNA (mRNA) in the cytoplasm. In innate immunity, the role of miRNA gene silencing is largely unknown. In this study, we performed microarray-based experiments using lipopolysaccharide (LPS)-stimulated macrophages derived from wild-type, MyD88 knockout (KO), TRIF KO, and MyD88/TRIF double KO mice. We employed a statistical approach to determine the importance of the commonality and specificity of miRNA binding sites among groups of temporally co-regulated genes. We demonstrate that both commonality and specificity are irrelevant to define a priori groups of co-down regulated genes. In addition, analyzing the various experimental conditions, we suggest that miRNA regulation may not only be a late-phase process (after transcription) but can also occur even early (1h) after stimulation in knockout conditions. This further indicates the existence of dynamic interactions between miRNA and signaling molecules/transcription factor regulation; this is another proof for the need of shifting from a 'hard-wired' paradigm of gene regulation to a dynamical one in which the gene co-regulation is established on a case-by-case basis.

Unraveling the Role of Cytochrome P450 as a Key Regulator Lantipeptide Production in Streptomyces globisporus

  • The aim of this study was to investigate the regulation of lantipeptide production in Streptomyces globisporus SP6C4, which produces the novel antifungal lantipeptides conprimycin and grisin, and to identify the role of cytochrome P450 (P450) in tis regulation. To investigate the regulation of lantipeptide production, we created gene deletion mutants, including ΔP450, ΔtsrD, ΔlanM, ΔP450ΔtsrD, and ΔP450ΔlanM. These mutants were characterized in terms of their morphology, sporulation, attachment, and antifungal activity against Fusarium oxysporum. The gene deletion mutants showed distinct characteristics compared to the wild-type strain. Among them, the ΔP450ΔlanM double mutant exhibited a recovery of antifungal activity against F. oxysporum, indicating that P450 plays a significant role in regulating lantipeptide production in S. globisporus SP6C4. Our findings highlight the significant role of P450 in the regulation of lantipeptide production and morphological processes in S. globisporus. The results suggest a potential link between P450-mediated metabolic pathways and the regulation of growth and secondary metabolism in SP6C4, thereby highlighting P450 as a putative target for the development of new antifungal agents.

의료기관의 방사선사 중 방사선 관계종사자와 방사선 작업종사자의 이원화 체계에 따른 인식도 조사 (A Survey on the Awareness of Radiation-related Workers and Radiation Workers in the Medical Institutions According to the Dual System)

  • Radiologic technologists working at the second and third medical institutions are classified as radiation-related workers and radiation workers according to their working departments, and are subject to double regulation by the Ministry of Health and Welfare and the Nuclear Safety Commission. We will try to understand the system of dualization and to understand the investigation of recognition. The dualized system of radiation-related workers and radiation workers includes the difference in name and terminology, the effective dose limit, the maintenance education and training of radiologic technologists, the period of medical examination, the radiation zone, dose of the woman whose pregnancy is confirmed in radiologic technologists, the qualification criteria of the safety officer, and the period of the regular inspection of the radiological equipment. In the questionnaire survey on the dualization system, there were various items showing significant differences between the radiation-related workers and radiation workers Overall, the radiation workers were more aware of the radiation workers' education and related terms than the radiation-related workers.

HIF-1α-Dependent Gene Expression Program During the Nucleic Acid-Triggered Antiviral Innate Immune Responses

  • Recent studies suggest a novel role of $HIF-1{\alpha}$ under nonhypoxic conditions, including antibacterial and antiviral innate immune responses. However, the identity of the pathogen-associated molecular pattern which triggers $HIF-1{\alpha}$ activation during the antiviral response remains to be identified. Here, we demonstrate that cellular administration of double-stranded nucleic acids, the molecular mimics of viral genomes, results in the induction of $HIF-1{\alpha}$ protein level as well as the increase in $HIF-1{\alpha}$ target gene expression. Whole-genome DNA microarray analysis revealed that double-stranded nucleic acid treatment triggers induction of a number of hypoxia-inducible genes, and induction of these genes are compromised upon siRNA-mediated $HIF-1{\alpha}$ knock-down. Interestingly, $HIF-1{\alpha}$ knock-down also resulted in down-regulation of a number of genes involved in antiviral innate immune responses. Our study demonstrates that $HIF-1{\alpha}$ activation upon nucleic acid-triggered antiviral innate immune responses plays an important role in regulation of genes involved in not only hypoxic response, but also immune response.

DNA Double-Strand Breaks Serve as a Major Factor for the Expression of Arabidopsis Argonaute 2

  • Argonaute 2 (AtAGO2) is a well characterized effector protein in Arabidopsis for its functionalities associated with DNA double-strand break (DSB)-induced small RNAs (diRNAs) and for its inducible expression upon ${\gamma}$-irradiation. However, its transcriptional regulation depending on the recovery time after the irradiation and on the specific response to DSBs has been poorly understood. We analyzed the 1,313 bp promoter sequence of the AtAGO2 gene ($1.3kb_{pro}$) to characterize the transcriptional regulation of AtAGO2 at various recovery times after ${\gamma}$-irradiation. A stable transformant harboring $1.3kb_{pro}$ fused with GUS gene showed that the AtAGO2 is highly expressed in response to ${\gamma}$-irradiation, after which the expression of the gene is gradually decreased until 5 days of DNA damage recovery. We also confirm that the AtAGO2 expression patterns are similar to that of ${\gamma}$-irradiation after the treatments of radiomimetic genotoxins (bleomycin and zeocin). However, methyl methanesulfonate and mitomycin C, which are associated with the inhibition of DNA replication, do not induce the expression of the AtAGO2, suggesting that the expression of the AtAGO2 is closely related with DNA DSBs rather than DNA replication.

Arabidopsis 피토크롬 돌연변이체에서 ethylene 생합성 조절 작용 (Regulation of Ethylene Biosynthesis in Phytochrome Mutants of the Arabidopsis Root)

  • 식물생장과 발달에 중요한 역할을 하는 phytochrome이 ethylene 생합성에 미치는 영향을 조사하기 위하여 여러 빛 조건에서 키운 phyA, phyB, phyAB에서 ethylene 생합성과 생합성에 관여하는 enzyme activity를 측정하였다. White light에서 키웠을 때 모든 mutant에서 ethylene 생합성이 감소되었다. 특히 double mutant에서는 wild type과 비교하여 37%가 감소하였다. Dark에서 키웠을 때에는 wild type만 감소하였고, mutant에서는 감소효과가 나타나지 않았다. Red light에서 키웠을 때 double mutant에서 급격한 감소가 일어났다. Far-red light 에서 키웠을 때는 phyB만 감소가 일어나지 않았다. Ethylene 생합성에 관여하는 enzyme인 ACO 활성 패턴과는 달리ACS 활성 패턴은 ethylene 생성 패턴과 유사하게 나타났다. 이 결과를 바탕으로 ethylene 생합성에는 phytochrome A와 B 모두 중요한 작용을 하며 특히 $P_r$ 형태의 phytochrome이 ethylene 생성량을 조절한다는 것을 제시한다. 또한 phytochrome은 ethylene 생합성 단계에서 AdoMet가 ACC로 전환되는 단계에서 조절하는 것을 제시한다.

거부민감성과 이성관계 만족의 관계에서 인지적 정서조절과 갈등해결전략의 이중매개 효과 (Double mediation effects of cognitive emotion regulation strategy and conflict resolution strategy on the association between rejection sensitivity and dating relationship satisfaction)

  • 본 연구는 거부민감성과 이성관계 만족의 관계에서 인지적 측면인 인지적 정서조절과 행동적 측면인 갈등해결전략의 매개효과를 검증하여 거부민감성 모델의 적용에 대해 검토하고자 하였다. 이를 위해 서울소재 대학 대학생 668명(남 232명, 여 436명, 평균연령, 21.59세(SD=2.08))을 대상으로 변인 간 관계에 대한 매개모형을 설정하여 관계를 검증하였다. 그 결과, 거부민감성과 이성관계 만족과의 관계에서 인지적 정서조절 전략 중 적응적 정서조절 전략과 갈등해결전략 중 통합과 타협 전략의 이중 매개효과를 확인하였다. 즉, 거부민감성이 이성관계 만족과 직접적인 부적인 상관을 보여 거부민감성이 높을 경우 이성관계 만족은 낮아질 것으로 예상할 수 있다. 다만, 상황을 긍정적으로 해석하고 초점을 확대한 적응적 인지적 정서조절전략과 서로 적극적으로 문제를 해결하고자 하는 통합과 타협의 갈등해결전략을 통해 이성관계 만족에 긍정적인 변화를 가져올 수 있음을 확인한 것이다. 이러한 결과를 바탕으로 상담실제에 대한 함의와 개입방향에 대해 논의하였다.
