Estimating Saturation-paste Electrical Conductivities of Rose-cultivated Soils from their Diluted Soil Extracts (절화장미 재배토양에서 희석된 토양 침출용액으로부터 포화반죽 전기전도도 추정)

  • We examined the effect of soil:water ratio on the equivalent concentration of individual electrolyte species and the electrical conductivities (EC) of the diluted extracts of 24 soil samples (loam or silt loam) collected from rose-cultivated plastic houses to estimate the EC of saturated soil-paste extracts (ECe) from diluted soil extracts. With increasing volume ratio of water (higher dilution), the equivalent concentrations of each electrolyte species and their sum increased. The relative contribution to the EC, however, was highest for $NO_3{^-}$, irrespective of soil:water ratio. The measured ECe was 6.36 for loam and $8.09dS\;m^{-1}$ for silt loam soils and the corresponding soil:water ratio was 0.38 and 0.50, respectively. The EC_e estimated from the EC of diluted extracts at 1:1, 1:2, or 1:5 soil:water ratios using their corresponding uniform diluted factors was lower than the measured EC_e and this difference was greater with higher dilution and EC values. Therefore, the alternative diluted factors (y) for each soil: water ratio were obtained following the definition of diluted factor and were correlated significantly with volume ratios of added water (x): y=1.55x+0.5 for loam and y=1.21x+0.48 for silt loam soils. On the other hand, correlation analyses of the EC of soil extracts (y) to the volume ratio of added water (x) on log-log scale yielded linear models: logy = -0.805logx + logb, SD of slope=0.05, b=sample specific constant, n=24). With known saturation percentage of a sample representing a group and and the EC of diluted extract of a given soil, the EC_e could be predicted using the proposed logarithmic equation.

A standardized method to study immune responses using porcine whole blood

  • Background: Peripheral blood mononuclear cells (PBMCs) are commonly used to assess in vitro immune responses. However, PBMC isolation is a time-consuming procedure, introduces technical variability, and requires a relatively large volume of blood. By contrast, whole blood assay (WBA) is faster, cheaper, maintains more physiological conditions, and requires less sample volume, laboratory training, and equipment. Objectives: Herein, this study aimed to develop a porcine WBA for in vitro evaluation of immune responses. Methods: Heparinized whole blood (WB) was diluted (non-diluted, 1/2, 1/8, and 1/16) in RPMI-1640 media, followed by phorbol myristate acetate and ionomycin. After 24 h, cells were stained for interferon (IFN)-γ secreting T-cells followed by flow cytometry, and the supernatant was analyzed for tumor necrosis factor (TNF)-α. In addition, diluted WB was stimulated by lipopolysaccharide (LPS) and polyinosinic:polycytidylic acid (poly I:C), reference strain KCTC3557 (RS), field isolate (FI), of heat-killed (HK) Streptococcus suis, and porcine reproductive and respiratory syndrome virus (PRRSV). Results: The frequency of IFN-γ+CD3+ T-cells and concentration of TNF-α in the supernatant of WB increased with increasing dilution factor and were optimal at 1/8. WB TNF-α and interleukin (IL)-10 cytokine levels increased significantly following stimulation with LPS or poly I:C. Further, FI and RS induced IL-10 production in WB. Additionally, PRRSV strains increased the frequency of IFN-γ+ CD4-CD8+ cells, and IFN-γ was non-significantly induced in the supernatant of re-stimulated samples. Conclusions: We propose that the WBA is a rapid, reliable, and simple method to evaluate immune responses and WB should be diluted to trigger immune cells.

  • There are many reports on the stability of drugs in powders and various dosage forms. The author studied on the stability of ascorbic acid mixed with talcs. From the result of this experiment, it was considered that talcs markedly accelerated oxidative degredation of ascorbic acid in powders and the acceleration rate depends on the varieties of crude talcs. Iron salts seem to be the principal factor influencing the degradation of ascorbic acid Degradation rate of ascorbic acid mixed with purified talc is significantly slower than that with the crnde talcs. The more the concetration of ascorbic acid is diluted, the faster the degradation rate and degredation rate of Ascorbic acid is the function of tme.

  • Pre-foaming, one of the manufacturing way of foamed concrete, is influenced by foaming agent. When the foaming agent diluted with water, surface tension and viscosity are varied. Therefore, this study is reviewing the surface tension, viscosity and unit weight of foam by experimental factor such as foaming agent types(AES, AOS, VS FP) and foam agent dilution concentration (1, 3, 5%) and temperature of materials (5, 10, 20℃). As an expeimental result, the surface tension and viscosity slightly increased with increasing concentrations. Meanwhile, when increasing temperature, the viscosity has decreased. FP produce relatively stable foams only in case 3% or more, which produce unstable foams containing large amount of water content by decreasing only insignificant surface tension when diluted at concentration of 1%.

Rapid Determination of Caffeine in Forensic Aqueous Sample by Dilute and Shoot LC-MS/MS (시료 희석 직접 주입 LC-MS/MS를 이용한 법화학 수용액 시료 중 카페인 신속 분석)

  • A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the determination of caffeine in forensic aqueous sample. The centrifuged sample ($100{\mu}l$) was diluted 50-fold with distilled water. The diluted sample ($400{\mu}l$) was then diluted further with $200{\mu}l$ of 0.1% formic acid solution and $400{\mu}l$ of acetonitrile containing 500 ng of caffeine-(3-methyl-$^{13}C_3$) prior to LC-MS/MS analysis. The mobile phase was composed of 0.1% formic acid in distilled water (A) and acetonitrile (B). Chromatographic separation was performed by using a Zorbax SB-C18 ($100mm{\times}2.1mm$ i.d., $3.5{\mu}m$) column and caffeine was eluted within 1.1 min. Linear least-squares regression with a 1/x weighting factor was used to generate a calibration curve with the coefficients of determination ($r^2=0.9983$). The lower limit of quantification was $25ng/ml$ for the analyte. The process efficiency was 98.6~100.1%. Intra- and inter-day precisions were not more than 2.1% and 1.7%, while intra- and inter-day accuracies were ranged from -6.8 to 4.5%, respectively. The suitability of the method was examined by analyzing unknown forensic aqueous samples.

Inhallation of e-Cigarette Cartridge Solution Aggravates Allergen-induced Airway Inflammation and Hyper-responsiveness in Mice

  • Electronic cigarettes (e-cigarettes) are becoming increasingly popular worldwide and their cellular effects warrant further evaluation. In this study, we investigated the effects of an e-cigarette cartridge solution on allergen related asthmatic airway inflammation (AI) and airway hyperresponsiveness (AHR), when it is delivered by intratracheal route in mice. Asthmatic AI and AHR were induced by systemic sensitization to ovalbumin (OVA) followed by intratracheal, intraperitoneal, and aerosol allergen challenges in BALB/c mice. The cartridge solution of e-cigarette (containing 16 mg/ml nicotine) was diluted 50 times and $100{\mu}l$ of the diluted solution was intratracheally instilled to OVA-sensitized (OVA-S) mice two times a week for 10 weeks. Long-term e-cigarette inhalation elicited no remarkable changes in the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase enzymes in serum, however, increased infiltration of inflammatory cells including eosinophils, into airways from blood, aggravated the asthmatic AI and AHR, and stimulated the production of cytokines such as interleukin (IL)-4, IL-5 and IL-13, and OVA-specific IgE production. Our data suggest that the inhalation of e-cigarette solutions can function as an important factor to exacerbate the allergy-induced asthma symptoms. Further studies are needed to address the effects of e-cigarette solutions on human health.

Characteristics of MnxSi1-xTe Compound Studied by Electron Magnetic Resonance and Other Experiments

  • The magnetic and other physical characteristics of $Mn_xSi_{1-x}Te$ have been investigated by electron magnetic resonance (EMR), X-ray diffraction (XRD) and other experiments. $Mn_xSi_{1-x}Te$ is found to have corundum structure for manganese contents up to 10% and also to be ferromagnetic for temperatures below 80 K. While ferromagnetic resonance signal coexists with the usual paramagnetic resonance signal, invariance of the g-factor inferred from the electron paramagnetic resonance signals throughout all temperature ranges clearly confirms that the manganese ions are in the electronic 3d5 state. The temperature dependence of EMR line-width is the same as other diluted magnetic semiconductors. From the EMR signals relaxation times $T_2$ and $T_1$ of $Mn_xSi_{1-x}Te$ compounds are estimated to be about $4.4{\times}10^{-10}s$ and $9.3{\times}10^{-8}s$ respectively and are found to vary slightly with temperature or composition change. Exchange narrowing of the EMR line-width becomes dominant for the sample in which the substitution ratio, x = 30%. For one sample, in which x = 0.5%, spin glass-like behavior is indicated by EMR signals for temperatures lower than 60 K. This behavior may authentic for samples within a certain range of x.

Transport and Loadings of Nutrients and Dissolved Major and Trace Elements in the Yeongsan River, Korea

  • Temporal variation of nutrients and dissolved major and trace elements have been studied in the Yeongsan River, Korea. There were significant temporal fluctuations in the concentrations of these elements depending upon the flow condition. $NH_4$, $PO_4$, Na, Mg, Ca, K, Mn, Cu, Ni, Zn, Co, As and U concentrations were inversely related to the flow; that is, they are the highest at low flow and the lowest at high flow. It indicates that these elements are derived from point sources such as rock weathering and/or human activities and then diluted by increasing flow. Meanwhile, Fe and Si concentrations varied proportionally to the flow indicating that they are derived from diffuse sources including reactions within soil. The concentration-flow relationships showed that hydrology of the river is the most important factor controlling the chemical composition of the Yeongsan riverwater, which was compatible of the results of R-mode factor analysis.

An Experimental Study of Dilution Methods for Preventing Volatile Particle Generation during Measurement of Diesel Particle Number Concentration (디젤 극미세입자 개수 농도 측정시 Volatile Particle 생성을 억제할 수 있는 희석방법에 관한 실험적 연구)

  • Recently, Europe decided to start the regulation of diesel engine nanoparticles because of its well known adverse health effects. The diesel nanoparticles can be classified as solid carbon particles and volatile particles. The volatile particles generates during dilution process by condensation of gas phase volatile compounds such as hydrocarbon. The new nanoparticle regulation considers only solid particles because of difficulty of measurement of volatile particles. The aim of this study is to suggest a proper dilution method that prevent the volatile particle generation. As a result, it is found that the $1^{st}$ dilution air temperature should be above $120^{\circ}C$ in order to prevent volatile particle generation effectively. It is also found that the volatile particles can be removed effectively in the evaporation tube by the increase of evaporation tube temperature. But when exhaust gas is hot enough (>$190^{\circ}C$, in this study) and it is diluted in the first diluter with high temperature air (>$120^{\circ}C$), removal phenomenon of volatile particles by increasing of evaporation tube temperature can not be seen. It means that there are no volatile particles in the diluted exhaust gas. Additionally, dilution ratio is not an important factor for volatile particle generation compared with dilution air temperature or evaporation tube temperature.

Inhibitory Activity of Brine Mineral Water on Cancer Cell Growth, Metastasis and Angiogenesis (해양성 광천수의 암세포 성장, 전이 및 신생 혈관 생성 억제 효과)

  • Brine mineral water(BMW) has recently gained attention as a new water resource due to its biological activities. In this study, BMW from the Geumjin area(Gangneung-city, Korea) was evaluated for its growth inhibition, anti-metastasis and anti-angiogenesis activity against cancer cells. The in vitro cytotoxicity was measured by CCK assay, and the anti-metastasis activity was estimated by lung metastasis in vivo. The in vitro incubation of mouse splenic cells with BMW that had been diluted more than 4-fold showed no effect on the cell growth when compared to a control group. Additionally, BMW inhibited the growth of the EL-4, L5178Y-R and colon26-M3.1 cancer cell lines in a dose-dependent manner. In vivo evaluation of the anti-metastasis activity of BMW in BALB/c mice inoculated with the colon26-M3.1 cell line revealed dose-dependent inhibition in response to treatment with samples that were diluted by up to 9 times. Finally, treatment with BMW effectively suppressed the growth of vascular endothelial growth factor(VEGF) added human umbilical vein endothelial cells. Overall, these results suggest that BMW has anti-cancer activity.
