Effect of Sodium Caseinate Hydrolysates on Angiotensin-I Converting Enzyme Inhibition Activity (Sodium Caseinate 가수분해물의 Angiotensin-I Converting Enzyme 저해효과에 관한 연구)

  • This study was carried out to identify the ACE (Angiotensin converting enzyme) inhibitory activity of casein hydrolysates for development of anti-hypertensive hydrolysates. Sodium caseinate was treated with six kinds of commercial proteases such as Flavourzyme, Protamex, Neutrase 1.5, Alcalase, Protease M, and Protease S for 8 h individually, and was then treated with the enzyme combination for 4 h at $45^{\circ}C$. The hydrolysate which had the highest ACE inhibitory effect was then hydrolysed successively with three digestive enzymes: pepsin, trypsin, and ${\alpha}$-chymotrypsin, at $37^{\circ}C$ for 4 h under conditions mimicking those of the gastrointestinal tract. UF (ultra filtration) treatment was applied to one of the secondary hydrolysates to determine ACE inhibitory activity. When sodium caseinate was hydrolysed by commercial proteases, the degree of hydrolysis (DH) showed 2.54 to 4.25% and after secondary hydrolysis, DH showed 4.30 to 5.22%. ACE inhibitory activity and $IC_{50}$ values decreased, and inhibition rates increased during hydrolysis. Protamex treatment showed the lowest $IC_{50}$ value ($516{\mu}g/mL$) and Flavourzyme hydrolysate showed the highest $IC_{50}$value ($866{\mu}g/mL$). As the first hydrolysate was treated with Flavourzyme, the ACE inhibitory activity increased. Neutrase hydrolysate had the highest activity with an $IC_{50}$ value ($282{\mu}g/mL$). When Neutrase plus Flavourzyme treatment was hydrolyzed by digestive enzymes, the $IC_{50}$ value ($597{\mu}g/mL$) was decreased statistically (p<0.05). As Neutrase plus Flavourzyme hydrolysate is treated by UF with MW cut-off 10,000, permeate showed $273{\mu}g/mL$ of $IC_{50}$ value, showed no difference, but retentate which has over MW 10,000 showed statistically different $IC_{50}$ value, $635{\mu}g/mL$ (p<0.05).

  • Since the active center of digestive enzymetic preparations, while under storage, lose their activity and potency by the exposure to moisture, colorization, solidifying and other physical changes. It is more important than beautiful package form that protected packaging form from moisture to get a pharmaceutical safety preparations and to maintain a definite potency. Then, in order to get a desirable conditions of storage and packaging, we used shellac and $AEA^{\circledR}$ as a coating base, and blister package, foil and bottle container as a packaging material. Temperature were set on room temperature and $37^{\circ}C$, moisture was adjusted to 40% RH and 80 % RH as a accelerated conditions. Accelerated test was carried out 6 times. The results are as follows: 1) The effect of packaging conditions give great influence on the maintenace of the stable potency. 2) Best result was produced with bottle container package. 3) $AEA^{\circledR}$ is more useful than shellac as a coating base of prevention from moisture. 4) Absorption of moisture gave considerable effects on potency and it has a limited point. 5) Difference in potency between optimal and worst condition is 472 u/2T, and difference in effective period is about 44 months.

Effects of Copper-bearing Montmorillonite on Growth Performance and Digestive Function of Growing Pigs

  • A total of 96 growing barrows (Duroc${\times}$Landrace${\times}$Yorkshire) at an average BW of 20.2 kg were used to investigate the effects of montmorillonite (MMT) or copper-bearing montmorillonite (Cu-MMT) on growth performance, intestinal microflora, digestive enzyme activities of pancreas and small intestinal contents, and the apparent nutrient digestion. The pigs were allocated to three groups with 32 pigs per treatment for 42 days and the average BW at the end of the experiment was 49.7 kg. The three dietary treatments were basal diet only (control group), basal diet +1.5 g/kg MMT, and basal diet +1.5 g/kg Cu-MMT. The results showed that supplementation with Cu-MMT significantly improved growth performance as compared to control and pigs fed with Cu-MMT had higher average daily gain than those fed with MMT. As compared to control, supplementation with Cu-MMT significantly reduced the total viable counts of Escherichia coli and Clostridium in the small intestine and proximal colon. Supplementation with MMT had no significant influence on intestinal microflora, although there was a tendency for Escherichia coli and Clostridium to be lower than the control. Pigs fed with Cu-MMT had lower viable counts of Escherichia coli in colonic contents than those fed with MMT. Although supplementation with MMT improved the activities of the digestive enzymes in the small intestinal contents, the tendency was not significant. Supplementation with Cu-MMT significantly improved the activities of total protease, amylase and lipase in the small intestinal contents. Supplementation with MMT or Cu-MMT improved the apparent nutrient digestion.

Systematic Studies on Yijin-tang(Erchen-tang) for Establishment of Evidence Based Medicine (표준한방처방 EBM 구축을 위한 이진탕 문헌 연구 분석)

  • Objectives : The purpose of this report was to provide the information activity and safety of Yijin-tang by analyzing domestic/international papers and theses about Yijin-tang. Methods : Domestic/international papers and theses related to Yijin-tang were reviewed and analyzed. These papers were then classified by year, experimental method and subject. Results : 1. Administration of Yijin-tang showed therapeutic effects on diseases of the circulatory system such as thrombosis and hyperlipisemia in in vivo experiments. 2. Administration of Yijin-tang reduced hemorrhagic erosion and ulcer by inhibition of gastric juice secretion and protection effect of gastric mucosa in in vivo experiments. 3. Both clinical and basic science researches, Administration of Yijin-tang showed improvement of digestive function by up-regulated of digestive enzyme and gut-regulated protein in plasma levels. Conclusions : Administration of Yijin-tang showed therapeutic effects on diseases of the circulatory system and protection effect of gastric mucosa in in vivo experiments. Furthermore, both clinical and basic science researches, administration of Yijin-tang showed improvement of digestive function.

Effect of Salvia miltiorrhiza aerial parts on growth performance, nutrient digestibility, and digestive enzymes in rabbits

  • Objective: This study aimed to investigate the effect of Salvia miltiorrhiza (S.m.) aerial parts as an alternative ingredient on growth performance, nutrient digestibility, and digestive enzymes in growing rabbits. Methods: Treatments included five tested diets: a control (basal diet), antibiotic (basal diet+enramycin at 5 mg/kg), and S.m. aerial parts powder added at 3.0%, 6.0%, and 9.0% of feed using 300 growing rabbits. Results: The diets with S.m. aerial parts addition at 9.0% decreased (p<0.05) feed/gain compared to the control, but there were no differences in feed intake and body weight gain. In contrast with the control, the addition of antibiotic increased (p<0.05) digestibility of dry matter, crude protein, energy, fiber, and ash. The herb addition did not cause differences in the digestibility of most nutrients compared to the antibiotic, but fiber digestibility of the herb at 6.0% and 9.0% was lower (p<0.05) than that of the antibiotic. Moreover, the antibiotic and the herb also similarly increased (p<0.05) the activities of duodenal α-amylase, maltase, lipase, and trypsin, compared to the control, and the herb at 6.0% and 9.0% showed a greater (p<0.05) activity of elastase than the dose 3.0%. Conclusion: The obtained data indicate that S.m. aerial parts can be a potential forage in rabbit's diet at 9.0% with a beneficial regulation on nutrition and digestion.

Antioxidative Effect and Digestive Enzyme Inhibition of Grape Seed Extract (GSE) (포도씨 추출물의 항산화 효과 및 소화효소 저해 효과)

  • The purpose of this study is to investigate the antioxidative activity and digestive enzyme inhibition of grape seed extract (GSE). The GSE was tested for its effect on various antioxidative potentials (scavenging activities of DPPH radical, superoxide anion radical and hydroxyl radical) and inhibitory effect of various digestive enzymes (trypsin, $\alpha$-chymotrypsin, $\alpha$-amylase, $\beta$-glucosidase and lipase). DPPH radical scavenging activity ($SC_{50}$, 50% scavenging concentration) of GSE was 4.76${\pm}$0.27 ppm while those of positive controls (EGCG and vitamin C) were 2.22${\pm}$0.12 ppm and 9.50${\pm}$0.72 ppm, respectively. $SC_{50}$ value of GSE against superoxide anion radical and hydroxyl radical were 3.82${\pm}$0.07 ppm and 803.23${\pm}$27.16 ppm, respectively. In addition, $IC_{50}$ values of GSE against trypsin, $\alpha$-chymotrypsin, $\alpha$-amylase, $\alpha$-glucosidase and lipase were 2.17${\pm}$0.59 ppm, 7.46${\pm}$1.25 ppm, 18.25${\pm}$3.54 ppm, 12.30${\pm}$1.12 ppm, and 653.23${\pm}$79.34 ppm, respectively. These results suggest that GSE may be useful for the prevention or treatment of obesity.

Isolation and Characterization of Acid Protease Produced by Staphylococcus sp. CB2-3 from Digestive Organ of Harmonia axyridis (무당벌레 소화기관으로부터 산성 단백질 분해효소를 생산하는 Staphylococcus sp. CB2-3의 분리 및 특성)

  • Six protein-degrading bacteria were isolated from digestive organ of Harmonia axyridis. These isolates were categorized as Staphylococcus sciuri subsp. sciuri (3 strains), Bacillus subtilis (1 strain), and Bacillus thuringiensis (2 strains) by 16S rRNA gene sequence analysis. The Staphylococcus sp. CB2-3 was selected as a protease-producing bacterium which showed the highest protease activity of 58.5 U/ml at the pH 5.0 medium. The optimal pH and temperature of protease activity were pH 5.0 and $40^{\circ}C$, respectively. This acid protease had a relatively high stability of 80% between $30-50^{\circ}C$ at broad temperature range. The opimal medium compositions of carbon, nitrogen and mineral source for cell growth and protease activity were investigated. When sorbitol (0.5%) was used as carbon source, enzyme activity was increased about 2 times than that of the basal medium. When skim milk (0.5%) was used as nitrogen source, activity was increased about 2.5 times than that of the control. Cell growth and enzyme activity were increased by mineral source such as KCl, $K_2HPO_4$, $FeSO_4$, but was completely inhibited by divalent ions such as $Co^{2+}$, $Zn^{2+}$, $Mn^{2+}$, $Cu^{2+}$.

Influence of Essential Oil Components on Growth Performance and the Functional Activity of the Pancreas and Small Intestine in Broiler Chickens

  • To investigate the efficacy of alternatives to antibiotics, the present study was conducted to compare the effects of antibiotic, lactic acid, a blend of commercial essential oils (EOs) and EOs in combination with lactic acid on growth performance and the functional activity of the gut in broiler chickens. A total of 168 broiler chickens were given the basal diet supplemented with 10 ppm colistin (T1), 0.1% lactic acid (T2), 25 ppm EOs (T3), 25 ppm EOs+0.1% lactic acid (T4), 50 ppm EOs (T5) or 50 ppm EOs+0.1% lactic acid (T6) in the period 3 to 35 days of age. As a result, the broiler chickens assigned to T4 group throughout the experimental period had apparently (p<0.05) greater body weight and total gain than these assigned to T1, T2, T3 and T5 groups. However, there was no difference in growth performance among the birds fed the diets supplemented with antibiotic (T1), lactic acid (T2) and EOs (T3 and T5) alone. The weights of digestive organs and the number of lactobacilli and E. coli in the lower ileum were not affected by dietary treatments. Total trypsin activity was significantly (p<0.05) greater in T4 than T1, T2, T3 and T5 groups. Total and specific pancreatic $\alpha$-amylase activities were significantly (p<0.05) enhanced in the broiler chickens fed T4 diet compared with these fed T1, T2 and T3 diets. However, there were no differences in growth performance and digestive enzyme activities including pancreatic trypsin and $\alpha$-amylase between T4 and T6 groups fed the diets supplemented with either low or high EOs levels in combination of lactic acid. In conclusion, a blend of commercial EOs combined with lactic acid showed significant increases in digestive enzyme activities of the pancreas and intestinal mucosa, leading to increase in growth performance.

Oxya Chinensis Sinuosa Mishchenko Extract: Potent Glycosidase Inhibitor Alleviates Postprandial Hyperglycemia in Diabetic Mice (당뇨 모델을 이용한 벼메뚜기(O. Mistshenk) 추출물의 식후 고혈당 완화 효과)

  • This study was designed to investigate whether extracts from Oxya chinensis sinuosa Mistshenk (an edible insect considered a grasshopper) could inhibit the activity of carbohydrate digestive enzymes and alleviate postprandial hyperglycemia in diabetic mice. Oxya chinensis sinuosa Mistshenk was extracted with 80% ethanol (OEE) or water (OWE) and then concentrated. The carbohydrate digestive enzyme-inhibiting activity of the resulting extracts was evaluated by examining α-glucosidase and α-amylase. The IC50 values of OEE against α-glucosidase and α-amylase were 0.229 mg/ml and 0.106 mg/ml, respectively. This result indicated that OEE has stronger inhibitory effects than OWE and positive control. The blood glucose levels of the diabetic control mice increased after one meal. However, when OEE (300 mg/kg) was added to starch, this increase in postprandial blood glucose levels was significantly suppressed. The area under the curve also significantly decreased following the administration of OEE, which exhibited no cytotoxicity. These results indicate that OEE is more efficacious than OWE and may be used as a carbohydrate digestive enzyme inhibitor, delay carbohydrate digestion and glucose absorption, and thus alleviate postprandial hyperglycemia caused by dietary carbohydrates.

Effects of Fibrolytic Enzyme Addition on Ruminal Fermentation, Milk Yield and Milk Composition of Dairy Cows (Fibrolytic Enzyme 첨가가 반추위 발효 성상 및 착유우의 유량 및 유성분에 미치는 영향)

  • We evaluated the effects of adding fibrolytic enzyme into ruminant diets on ruminal fermentation (in vitro) and lactational performances of dairy cows (in vivo). Through the in vitro experiment that was carried out with different contents of NDF (34, 38, 43%) in diets, digestibilities of NDF in the rumen appeared not significantly different by the addition of enzyme but were different by NDF content in diets showing higher digestibility in NDF 43% diet. It could be attributed by the relatively higher amount of hemicellulose in the current experimental diets than in conventional diets that might have been digested easily by the addition of fibrolytic enzyme in the rumen. The addition of fibrolytic enzyme tended to increase NDF digestibilities to a little extent both in 0.05 and 0.1% enzyme levels. Ruminal pH, NH3-N concentrations and VFA production in the rumen were not affected by the addition of fibrolytic enzyme. Activities of CMCase and xylanase were higher in enzyme treated diets of both NDF 34 and 38%. In particular, the activities of xylanase that slowly decreased from 0 to 12 hr but rapidly after 24 hr indicates that the major action of the enzyme in the rumen occurs in early period of incubation. Through an in vivo experiment, fibrolytic enzyme addition into the diets of dairy cows indeed affected lactational performance of milk yield. The cows fed enzyme treated diets produced 8% (1.9kg/d) more amounts of milk than with no enzyme addition. Milk composition of milk fat and protein was not affected by enzyme addition. Overall, the results of this in vivo study indicates that fibrolytic enzyme can be used to improve milk production in lactating cows. In respect that animals in different treatments of this study had the same amounts of intake, the increased milk yield with enzyme addition may be attributed to the improved utilization of nutrients in the digestive tract.
