SB-31$\circledR$의 일반약리작용 (General Pharmacology of SB-31$\circledR$)

  • General pharmacological effects of SB-31$^{R}$, the extracts of Pulsatilla koreana, were investigated in mice, rats and guinea-pigs. Intravenous injection of SB-31 (3 and 6 ml/kg) produced almost no effect on central nervous system no effects on the general symptom and behaviors of mice, spontaneous locomotor activity, pentobarbital- induced sleeping time , rotared performance , electroshock and pentylenetertrazole -induced seizures, acetic acid-induced writhing and normal body temperature in mice. SB-31 showed little effects on the spontaneous movement of the isolated ileum and contraction induced by agonists in isolated ileum, suggesting no influence on autonomic nervous system. Administration of SB-31 also did not show any effect on blood pressure in conscious rats. However, a slight decrease in heart rate was observed at high doses (6 and 10 ml/kg) of SB-31 in conscious rats. Similarly, a slight increase in respiratory rate was observed at 6 m1/kg of SB-31 in anesthetized rats. SB-31 did not produce any effect at the dose of 3 ml/kg, but showed a tendency to increase the urinary volume at 6 ml/kg, and produced a decrease in urinary excretions of N $a_{+}$and $K_{+}$at 6 ml/kg. However, transport capacity within the gastrointestinal tract and the secretion of the gastric juice were not influenced by 6 ml/kg of SB-31. In conclusion, these results suggest that SB-31 did not pro-duce any acute effects on the central nervous system, autonomic nervous system, respiratory and circulatory systems, digestive system and kidney function at the dose of below 3 ml/kg.ml/kg.

  • Cancer is a leading cause of death, and the number of cancer patients is increasing in Korea. The needs for the home care of cancer patients are increasing recently, but the standardized home care protocols are not developed yet. This study was designed to develop nursing assessment -intervention algorithms for the cancer chemotherapy patients at home. These algorithms suggest guidelines when we assess the patient's condition, and find appropriate nursing interventions, so that standardization and quality control of home care can be attained. The algorithms were processed by yes-no tree. Eleven common problems of cancer chemotherapy patients were identified by the literature review and oncology nurses' experience. These were digestive dysfunction, pain, fatigue, infection, respiratory difficulties, activity intolerance, hemorrhage, sensory disorder, edema, skin problem, and mucosal problem. The algorithm needs to be validated and modified by using for the cancer chemotherapy patients at home.

구아바 잎 열수추출물의 당류분해효소 저해 효과 및 OGTT에 미치는 효과 (Effect of Aqueous Extract of Guava (Psidium guajava L.) Leaves on the Oral Glucose Tolerance Test and Inhibition of Glycoside Hydrolase)

  • Guava ($Psidium$ $guajava$) contain a great deal of polyphenol compound and work on the treatment of $Diabetes$ $mellitus$ effectively. In this study, the bioactivities of aqueous extract (GLEx) of guava leaf were investigated. Total phenolic contents of GLEx was 266.9 mg tan/g. The effects of GLEx on digestive enzymes, ${\alpha}$-amylase, maltase and sucrase were investigated. $IC_{50}$ values of GLEx against ${\alpha}$-amylase, maltase and sucrase were 0.65 mg/$m{\ell}$, 2.0 mg/$m{\ell}$ and 3.5 mg/$m{\ell}$ respectively. The effect of GLEx on oral glucose tolerance test (OGTT) was performed in normal ICR mouse, control (dstilled water) and GLEx (aqueous extract of Guava leaf) were co-administered orally with glucose, showed reducing effect on the blood glucose level. The guava is likely to useful for prevention or improvement of hyperglycemia by lowering the blood glucose level and inhibiting glycoside hydrolase activity.

  • It is a recent observation that about 80 per cent of the body's immune system is localized in the gastrointestinal tract. This explains to a large extent why eating right is important for the modulation the immune response and prevention of disease. I addition it is increasingly recognized that the body has an important digestive system also in the lower gastrointestinal tract where numerous important substances are released by microbial enzymes and absorbed. Among these substances are short chain fatty acids, amino acids, various carbohydrates, polyamines, growth factors, coagulation factors, and many thousands of antioxidants, not only traditional vitamins but numerous flavonoids, carotenoids and similar plant- and vegetable produced antioxidants. Also consumption of health-promoting bacteria (probiotics) and vegetable fibres (prebiotics) from numerous sources are known to have strong health-promoting influence. It has been calculated that the intestine harbours about 300 000 genes, which is much more than the calculated about 60000 for the rest of the human body, indicating a till today totally unexpected metabolic activity in this part of the GI tract. There are seemingly several times more active enzymes in the intestine than in the rest of the body, ready to release hundred thousand or more of substances important for our health and well-being. In addition do the microbial cells produce signal molecules similar to cytokines but called bacteriokines and nitric oxide, with provide modulatory effects both on the mucosal cells, the mucosa-associated lymphoid system (MALT) and the rest of the immune system. Identification of various fermentation products, and often referred to as synbiotics, studies of their role in maintaining health and well-being should be a priority issue during the years to come.

Biochemical and Genetic Characterization of Arazyme, an Extracellular Metalloprotease Produced from Serratia proteamaculans HY-3

  • Serratia proteamaculans HY-3 isolated from the digestive tract of a spider produces an extracellular protease named arazyme, with an estimated molecular mass of 51.5 kDa. The purified enzyme was characterized as having high activities at wide pH and temperature ranges. We further characterized biochemical features of the enzymatic reactions under various reaction conditions. The protease efficiently hydrolyzed a broad range of protein substrates including albumin, keratin, and collagen. The dependence of enzymatic activities on the presence of metal ions such as calcium and zinc indicated that the enzyme is a metalloprotease, together with the previous observation that the proteolytic activity of the enzyme was not inhibited by aspartate, cysteine, or serine protease inhibitors, but strongly inhibited by 1,10-phenanthroline and EDTA. The araA gene encoding the exoprotease was isolated as a 5.6 kb BamHI fragment after PCR amplification using degenerate primers and subsequent Southern hybridization. The nucleotide sequence revealed that the deduced amino acid sequences shared extensive similarity with those of the serralysin family of metalloproteases from other enteric bacteria. A gene(inh) encoding a putative protease inhibitor was also identified immediately adjacent to the araA structural gene.

볼락, Sebastes inermis 의 섭식상태 (Feeding Ecology of Black Rockfish, Sebastes inermis)

  • 본 연구는 볼락, Sebastes inermis의 섭식생태를 파악하기 위하여 경남 사천시 신수도 연안에서 채집된 볼락의 섭식활동 주기와 위내용물을 분석하였다. 어체표본은 1984년 2월부터 1985년 5월사이에 낚시, 자망, 소형기선저인망, 낭장망 등으로서 채집하였다. 볼락의 표본은 0세군부터 4세군까지 채집되었으며, 이들의 주 섭식활동 시간대는 봄, 여름의 경우 이른 아침과 늦은 오후였고, 가을과 겨울에는 특별한 섭식활동 시간대 없이 1$\~$2시간 주기로 간헐적인 섭식활동을 하였다. 이들의 먹이생물은 어류의 연령과 계절에 따라 달랐지만, 주요 먹이생물은 요각류(copepods), 옆새우류(gammarids), 카프렐라류 (caprellids), 생 이류 (carideans), 다모류(polychaees), 단미류 (brachyurans) 등이었다 0세군은 요각류를 주먹이로 섭식하였고, 1세군 이상은 옆새우류를 주 먹이로 이용하였다. 먹이생물의 다양도와 균등도는 어체의 연령이 증가할수록 높았다. 본 종의 섭식기관, 소화기관의 형태, 먹이생물의 종류 등을 고려하였을 때, 볼락은 소형 갑각류를 주 먹이생물로 이용하는 육식성이면서 시각섭식성 식성을 지닌 어류인 것으로 판단되었다.

  • 현대사회는 1인가구가 증가함에 따라 불규칙한 식습관으로 인해 영양이 불균형하게 포진되어있다. 이러한 식습관은 위장질환, 소화기 질환 등 만성질환의 발병률을 증가시켰다. 본 논문은 위장질환 예방을 위한 다중회귀분석을 이용한 식이지식 예측을 제안한다. 제안하는 방법은 식이지식 예측을 통해 사용자의 위장질환과 식이영양을 관리하는 방법이다. 헬스 플랫폼에서 스마트 기기를 통해 수집된 사용자의 PHR을 통합한다. 통합된 데이터로부터 다중회귀분석을 이용하여 사용자의 식이와 활동량 변화를 분석한다. 사용자의 식이 성분과 소모 칼로리, 기초대사와 같은 상황정보를 입력으로 적절한 식이성분, 위장질환 수치의 변화를 예측하고 필요할 것으로 나타나는 영양성분을 사용자에게 권장한다. 이를 통해 현대인들은 균형 잡힌 식사를 통해 위장질환을 관리할 수 있다.

구강작열감증후군 환자의 임상 특징에 따른 심박변이도 분석 (Heart Rate Variability Analysis According to Clinical Characteristics in Patients with Burning Mouth Syndrome)

  • Objectives: This study aimed to identify the characteristics of the heart rate variability (HRV) index in patients with burning mouth syndrome (BMS) and to evaluate the predictive value of HRV for Korean medical treatment responsiveness to tongue pain in BMS. Methods: Among BMS patients who visited the Oral Diseases Clinic of Kyung Hee University Korean Medicine Hospital from January 1, 2018, to May 31, 2021, we reviewed the clinical records of 217 women aged 50 years and over. HRV and unstimulated saliva flow rate (USFR) measurements were carried out, and all patients were asked to indicate their degree of tongue pain using the visual analog scale (VAS). The subjects were classified into subgroups according to the presence or absence of hyposalivation and VAS improvement, and each subgroup was compared using the HRV index. Logistic regression analysis was conducted to confirm whether cause-effect relationships were present between statistically significant values. Results: BMS patients had lower LF and HF than healthy people. Subgroup analysis demonstrated that there were some statistically significant differences in the HRV index according to salivation rate and degree of pain. In addition, as the LF/HF ratio increased, the pain improvement rate decreased after treatment. Conclusion: The autonomic nerve activity of BMS patients was lower than that of healthy people; however, autonomic balance was not impaired. In addition, tongue pain in BMS patients responded more favorably to Korean medical treatment when patients fell within a normal range of the LF/HF ratio, suggesting that autonomic imbalance could be used as one of the predictable factors in clinical practice.

Anti-Wrinkling Effect of Noni (Morinda citrifolia) by Antioxidant and Anti-Inflammatory Properties

  • Noni has been used for medicinal purposes for more than 2,000 years in South Pacific Polynesia, China and India, and has been heavily ingested as an extract for its excellent antioxidant and anti-inflammatory effects. However, a recent study found that the noni extract causes digestive disorders, kidney problems, and liver diseases, which made it necessary to use it for other purposes than as an extract. In this study, we want to evaluate the potential of noni as an anti-oxidant, anti-inflammatory and anti-wrinkling agent. Methods: Noni was freeze-dried, extracted in water, and concentrated. Skin cells were treated with the noni extract for 24 hrs and then were exposed to UVB (55 mJ/cm2). After 48 hrs of incubation, pro-inflammatory cytokine, elastase, MMP-1 and type-1 procollagen levels were measured by ELISA. Results: To find out the antioxidant effect of the noni extract, the DPPH and ABTS radical scavenging activity experiments were conducted and the noni extract showed 97.0 % and 92.0 % antioxidant efficacy at 200 ㎍/mL respectively. The noni extract (50 and 100 ㎍/mL) decreased IL-6 and TNF-α in RAW 264.7 cells induced by LPS in a concentration-dependent manner. In the RT-PCR experiment involving NO production, the noni extract (50 and 100 ㎍/mL) inhibited NO production by strongly inhibiting iNOS mRNA expression, and also inhibited the elevation of MMP-1 and elastases caused by UVB irradiation by 25.0 % and 7.0 % respectively. In addition, type-1 procollagen was elevated by 20.0 % by the noni extract treatment in HaCaT cells. Conclusion: The noni extract has photoprotective ability by reducing proinflammatory mediators, elastase and MMP-1 production, and elevation of collagen synthesis. Our findings suggest that the noni extract might be a good natural substance to protect against UVB-induced premature skin aging.

Pattern of 'Concanavalin A' Synthesis during Development of Jack Bean (Canavalia ensiformia) Pods

  • Jack bean [Canavalia ensiformis (L.)], belonging to the Leguminosae family has been frequently used in edible and medicinal plants in Asian countries. Jack beans are high in protein which is approximately 30%. Concanavalin A (Con A) is a major protein of Jack bean and belongs to the family of legume lectins. It has inhibitory effect on hepatocellular carcinoma by inducing autophagy. However, Con A negatively affects nutrient utilization by other mechanisms. It binds to the glycoproteins and glycolipids of the digestive tract mucosa, inhibits the activity of the enzymes of the brush border of the enterocytes. In order to use Jack bean young seedpods, they are restricted to 'young pods (soft, pre-swelling)' according to the 'Food Code' (Ministry of Food and Drug Safety). Therefore, in this study, we investigated the quantitative change of Con A across developmental stages of Jack bean pods. Biological samples consisted of Jack bean pods and seeds in 7 stages of development. The expression pattern of Con A mRNA was monitored by quantitative reverse transcription PCR (RT-qPCR). Expression of Con A proteins was analyzed by western blotting. The expression of Con A mRNA and protein in the seeds tended to increase gradually as the seeds expanded. However, in pods, they were much less than in seeds. As the expression of Con A mRNA and protein increases as the pods thicken, it is predicted that Con A synthesis increases when the thickness growth of the pod begins after the length growth of the pod is completed. Since the expression of Con A in the pods and seeds in very low when the pods are about 2 cm, therefore 2 cm pods seem appropriate when using 'young pods'. It is also necessary to study other proteins in Jack bean, such as Urease and Canavalin. These studies will serve as the basis for processing Jack bean.
