Diets with corn oil and/or low protein increase acute acetaminophen hepatotoxicity compared to diets with beef tallow in a rat model

  • It has been reported that dietary polyunsaturated fats (PUFA) increase liver injury in response to ethanol feeding. We tested the hypothesis that diets rich in linoleic acid (18:2n-6) would affect acute liver injury after acetaminophen injection and that protein restriction might exacerbate the liver injury. We examined effects of feeding diets with either 15% (wt/wt) corn oil or 14% beef tallow and 1% corn oil for six weeks with either 6 or 20 g/100 g protein on acute hepatotoxicity. After the feeding period, liver injury was induced by injecting either with 600 mg/kg body weight acetaminophen suspended in gum arabic-based vehicle, or with vehicle alone during fasting status. Samples of liver and plasma were taken for analyses of hepatic glutathione (GSH) levels and liver-specific enzymes [(Glutamate-pyruvate transaminase (GPT) and glutamate-oxaloacetate transaminase (GOT)], respectively. Whereas GSH level was significantly lower in only group fed 15% corn oil with 6 g/100 g protein among acetaminophen-treated groups, activities of GPT and GOT were significantly elevated in all groups except the one fed beef tallow with 20 g/100 g protein, suggesting low protein might exacerbate drug-induced hepatotoxicity. The feeding regimens changed the ratio of 18:2n-6 to oleic acid (18:1n-9) in total liver lipids approximately five-fold, and produced modest changes in arachidonic acid (20:4n-6). We conclude that diets with high 18:2n-6 promote acetaminophen-induced liver injury compared to diets with more saturated fatty acids (SFA). In addition, protein restriction appeared to exacerbate the liver injury.

분만전후의 어미쥐의 영양부족이 새끼쥐의 뇌성장발육과 조성에 미치는 영향 (Effect of Maternal Undernutrition on the Growth and Composition of Young Rat Brain)

  • 분만전후 4주동안 Sqraque Dawley 암컷 쥐에게 양적으로 식이를 제한했다. 임의로 먹인 control group의 1일평균성취량의 1/2의 시중사로를 식이제한 group에게 주었다. Deficient I group에서는 분만후부터 이유시까지 식이제한을 했고, deficient II group에서는 임신 15일부터 이유시까지 계속 식이제한을 했다. 식이제한 group의 체중과 뇌무게는 control group의 새끼쥐보다 유의적으로 낮았지만, 체중에 대한 뇌무게의 비는 control group보다 높았다. 이유시에는 두 식이제한 group사이에서 체중과 뇌무게의 유의적인 차가 나타났다. 뇌의 DNA, RNA 및 단백질함량은 control group보다 식이제한 group에서 유의적으로 낮았지만 RNA/DNA, 뇌무게/DNA 및 단백질/DNA은 control group보다 식이제한 group에서 높았다. 이것은 새끼쥐뇌에서 세포분열이 세포의 크기성장보다 이 기간중의 어미쥐의 식이제한에 의해 더욱 심한 영향을 받았음을 시사해준다. 뇌의 DNA와 RNA는 두식이제한 group간에 유의적인 차를 나타내지만, 단백질의 경우는 유의적인 차를 나타내지 않았다.

아토피피부염 영아의 영양장애 사례보고와 채식주의에 관한 고찰 (Marasmus and Kwashiorkor by Nutritional Ignorance Related to Vegetarian Diet and Infants with Atopic Dermatitis in South Korea)

  • Infants and children with food related Atopic Dermatitis (AD) need extra dietary efforts to maintain optimal nutrition due to food restriction to prevent allergy reactions. However, nutrition ignorance and food faddism make patients even more confused and practice desirable diet more difficult. The objective of this study was to report the AD patients' malnutrition cases in Korea. We report on 2 cases of severe nutritional deficiency caused by consuming macrobiotic diets which avoid processed foods and most animal foods, i.e. one of vegetarian diet. Case 1, a 12-month-old male child, was admitted with severe marasmus. Because of a history of AD, he was started on mixed grain porridge at 3 months without any breast milk or formula feeding. His caloric intake was 66% and protein intake was 69% of the recommended dietary allowance. Patient's height and weight was under 3th percentile. On admission the patient was unable to crawl or roll over. Case 2, a 9-month-old AD female patient, was diagnosed with kwashiorkor and rickets. She was also started on mixed grain porridge at 100 days due to AD. Her caloric intake has been satisfied recommended dietary allowance until 7 months, however, she conducted sauna bath therapy and reduced both energy and protein intake at 8 months. The amount of protein intake for case 2 was higher than recommended dietary allowance, but, sauna therapy and severe AD with intakes of low guality protein may increase patient's protein requirement resulting in kwashiorkor. Case 2 patient's height and weight was on 3th percentile. Both cases showed low intake of calcium, iron, zinc, vitamin A, vitamin E and especially very low intake of vitamin B$_{12}$ and vitamin D. Allergy tests for certain foods had not done prior to admission for both cases. They followed the dietary advise operated by macrobiotic diet internet site. In conclusion, AD infants' parents and caregivers should contact a pediatrician trained as a specialist in allergy for accurate diagnosis. For infant patients, breast or formula feeding including hypoallergenic formula should be continued until their one year of age. When certain foods need to be restricted or to follow special diets such as vegetarian diet, consultation with pediatrician and dietitian is needed.d.

Effects of ᴅ-allulose on body fat accumulation in rats fed severely carbohydrate-restricted diets containing beef tallow or soybean oil

  • Purpose: The carbohydrate-restricted diet has been recognized to be effective into preventing and alleviating lifestyle-related diseases, such as obesity and type 2 diabetes. The rare sugar ᴅ-allulose is a functional monosaccharide with anti-obesity effects. In the present study, we examined the effects of dietary ᴅ-allulose on body fat accumulation in rats fed severely carbohydrate-restricted diets containing high concentrations of different fats, beef tallow, or soybean oil. Methods: Male Wistar rats (n = 35, 3-week-old) were divided into 5 groups: One chow-fed control (C) group, and four carbohydrate-restricted groups, namely, beef tallow (B), beef tallow + ᴅ-allulose (BA), soybean oil (S), and soybean oil + ᴅ-allulose (SA), with free access to the diet and water for 8 weeks. The B and BA diets contained 23% beef tallow and 2% soybean oil, whereas the S and SA diets contained 25% soybean oil. Furthermore, the BA and SA diets contained 5% ᴅ-allulose. Results: The final body weight, weight gain, and food intake were significantly higher, and food efficiency was significantly lower in the control group compared to the other carbohydrate-restricted groups. Intra-abdominal adipose tissue, carcass fat, and total body fat weights were not influenced by dietary fat type or ᴅ-allulose supplementation, except for the epididymal adipose tissue weight. In contrast, carbohydrate restriction suppressed body weight gain in rats, but remarkably increased body fat accumulation. Conclusion: Under carbohydrate-restricted conditions, no anti-obesity effects of dietary ᴅ-allulose were observed, regardless of the dietary fat type. The causes of these effects are unknown. However, they may be influenced by a very low carbohydrate and high protein diet. Further research is required to elucidate the effects of ᴅ-allulose under various nutrient compositions with different fat, carbohydrate, and protein energy ratios.

저단백식이의 투여가 만성신부전증의 진행에 미치는 영향에 관한 실험적 연구 (Effects of Dietary Protein on the Progression of Early Chronic Renal Failure in Subtotally Nephrectomizid Rats)

  • 목 적 : 초기만성신부전증에서 저단백식이의 투여가 만성신부전의 진행속도 및 혈압조절에 어떠한 영향을 미치는 지를 알기 위함이다. 방 법 : 5/6 신절제술로 만성 신부전을 유발시킨 백서를 수술 제 7일부터 무작위로 enalapril을 투여하지 않은 군과 enalapril 투여군 (식수 1L 당 50 mg)으로 나누고 각군을 정상단백식이군 ($18.5\%$ 단백식이), 저단백식이군($6\%$단백식이)으로 나누어 비교하여 보았다. 신절제술후 4주, 12주, 16주에 단백뇨의 변화, 잔여 신장무게, 신 조직의 mesangial matrix expansion score 및 morphometric analysis로 분석한 사구체용적의 변화를 비교 분석하여 다음과 같은 결과를 얻었다. 결 과 : 1) 정상단백식이군 및 저단백식이군은 신절제술후 혈압이 올라가기 시작하여 지속적인 고혈압소견을 보였다 (정상단백식이군; 4주 $147{\pm}6$mmHg, 8주 $175{\pm}11$mmHg, 12주 $180{\pm}6$mmHg, 16주 $200{\pm}6$ mmHg: 저단백식이군; 4주 $140{\pm}8$mmHg, 8주 $162{\pm}5$mmHg, 12주 $171{\pm}6$mmHg, 16주 $184{\pm}11$mmHg). Enalapril 투여군은 식이의 단백량과 관계없이 신절제술후 8주부터 혈압이 조절되기 시작하였다. 2) 16주째 저단백식이군의 24시간 뇨단백은 $74{\pm}15$mg으로 정상단백식이군 ($101{\pm}15$mg)보다 의의있게 적었다. (P<0.05). Enalapril 투여군도 저단백식이의 경우 $42{\pm}12$mg 으로. 정상단백식이($67{\pm}15$mg) 보다 의의있게 적었다. (P<0.05). 3) 신절제술후 16주째 크레아티닌 청소율은 정상단백식이군 $1.40{\pm}0.13$ ml/min, 저단백식이군은 $1.57{\pm}0.11$ml/min, enalapril 정상단백식이군은 $1.10{\pm}0.17$ml/min, enalapril 저단백식이군은 $1.37{\pm}0.14$ml/min 이었다. (P=NS). 4) 저단백식이군의 잔여 신장무게의 증가는 정상단백시이군의 신장 무게의 증가에 비해 의의있게 낮았다. (저단백식이군; 4주 $1.33{\pm}0.04g$, 12주 $1.45{\pm}0.05g$, 16주 $1.44{\pm}0.16g$: 정상단백이군; 4주 $1.58{\pm}0.19g$, 12 주$1.79{\pm}0.15g$, 16주 $1.99{\pm}0.12g$, P<0.05). 16주에 관찰한 enalapril 투여군의 신장무게를 보면 저단백식이, 정상단백식이에 따른 차이는 없었다. 5) 12주, 16주째 저단백식이군의 mesangial matrix expansion score은 정상단백식이군보다 의의있게 감소되었다. (저단백식이군; 12주 $1.91{\pm}0.02$, 16주 $1.90{\pm}0.02$; 정상단백식이군 ; 12주 $2.29{\pm}0.09$, 16주 $2.55{\pm}0.16$, P<0.05). Enalapril 투여군의 mesangial matrix expansion score는 식이의 단백량에 따른 차이는 없었다. 6) 16주째 저단백식이군의 사구체용적은 정상단백식이군보다 의의있게 감소되었다 (16주, 저단백식이군; $1.17{\pm}0.19{\times}10^6{\mu}m^3$ ; 정상단백식이군; $1.98{\pm}0.16{\times}10^6{\mu}m^3$, P<0.05). Enalapril 투여군의 사구체용적은 저단백식이의 경우 정상식이 때 보다 의의있는 감소를 보였다 (16주, 저단백식이군; $1.19{\pm}0.17{\times}10^6{\mu}m^3$: 정상단백식이군; $1.81{\pm}0.22{\times}10^6{\mi}m^3$, P<0.05). 결 론 : 항고현압제를 쓰지 않은 군에서의 저단백식이의 효과는 매우 현저하였고 항고혈압제를 쓴 군에서는 저단백식이와 정상단백식이의 차이가 없었다. 따라서 저단백식이와 항고혈압제는 각각 신조직 손상의 진행을 확실히 지연시키나 두가지를 같이 병용하면 각각 지연시키는 것에 비하여는 additional effect가 미미하였다. 그러나 혈압 조절은 저단백식이로는 만족할만하지 못하고 항고혈압제에 의한 혈압조절이 도움이 되므로_ 만성신부전 환자에서 저단백식이와 항고혈압제의 병용요법이 필요한 것으로 사료된다.

Effect of processed foods on serum levels of eosinophil cationic protein among children with atopic dermatitis

  • The prevalence of atopic dermatitis (AD) in school-age children has increased in industrialized countries. As diet is one of the main factors provoking AD, some studies have suggested that food additives in processed foods could function as pseudoallergens, which comprise the non-immunoglobulin E-mediated reaction. Eosinophil cationic protein (ECP) is an eosinophil granule protein released during allergic reactions to food allergens in patients with AD. Thus, serum ECP levels may be a useful indicator of ongoing inflammatory processes in patients with AD. The purpose of this study was to investigate the effect of consuming MSG in processed foods on serum ECP levels among children with AD. This study was performed with 13 patients with AD (age, 7-11 years) who had a normal range of total IgE levels (< 300 IU/ml). All participants ate normal diets during the first week. Then, six patients were allocated to a processed food-restricted group (PRDG) and seven patients were in a general diet group (GDG). During the second week, children in the PRDG and their parents were asked to avoid eating all processed foods. On the third week, children in the PRDG were allowed all foods, as were the children in the GDG throughout the 3-week period. The subjects were asked to complete a dietary record during the trial period. Children with AD who received the dietary restriction showed decreased consumption of MSG and decreased serum ECP levels and an improved SCORing score on the atopic dermatitis index (P < 0.05). No differences in serum ECP levels or MSG consumption were observed in the GDG. Serum total IgE levels were not changed in either group. In conclusion, a reduction in MSG intake by restricting processed food consumption may lead to a decrease in serum ECP levels in children with AD and improve AD symptoms.

Effects of chronic caloric restriction on kidney and heart redox status and antioxidant enzyme activities in Wistar rats

  • Caloric restriction (CR) has been associated with health benefits and these effects have been attributed, in part, to modulation of oxidative status by CR; however, data are still controversial. Here, we investigate the effects of seventeen weeks of chronic CR on parameters of oxidative damage/modification of proteins and on antioxidant enzyme activities in cardiac and kidney tissues. Our results demonstrate that CR induced an increase in protein carbonylation in the heart without changing the content of sulfhydryl groups or the activities of superoxide dismutase and catalase (CAT). Moreover, CR caused an increase in CAT activity in kidney, without changing other parameters. Protein carbonylation has been associated with oxidative damage and functional impairment; however, we cannot exclude the possibility that, under our conditions, this alteration indicates a different functional meaning in the heart tissue. In addition, we reinforce the idea that CR can increase CAT activity in the kidney. Moreover, CR caused an increase in CAT activity in kidney, without changing other parameters. Effects of probiotic, prebiotic, and synbiotic with and without feed restriction on performance, hematological indices and carcass characteristics of broiler chickens

  • Objective: This study was conducted to investigate the effects of probiotic, prebiotic and synbiotic with and without feed restriction on broilers performance, blood parameters, carcass characteristics, and feed cost of production from 1 to 56 days of age. Methods: Two hundred and forty unsexed one day-old chicks of Arbor Acres breed were used. Two trials, I and II, were conducted, with 120 birds in both. Each trial was divided into 4 equal groups. The birds in trial I were fed ad libitum throughout the experiment, while the chicks in trial II were fed ad libitum during the first week of age, then subjected to 5 hours/d of feed restriction from the beginning of the second week up to the end of the experiment. In both trials, the birds in group 1 were fed on a control diet while the other groups were given the same control diet supplemented either with a probiotic in group 2, prebiotic in group 3, or synbiotic in group 4. Results: It was found that chicks fed diets supplemented with probiotic, prebiotic and synbiotic (with and without feed restriction) exhibited higher body weight and feed efficiency than chicks fed the control diets. The feed additives in both trials did not affect hemoglobin, serum total protein, albumin, globulin, glucose, and total cholesterol, except the packed cell volume which was increased in the additive treatments with restriction at the end of the experiment. Moreover, the dietary treatments did not influence the carcass yield. However, the relative weights of liver, gizzard and proventriculus, small intestine and bursa of fabricius were found to be increased. The additives decreased the visible fat in the carcass, with more decreasing effect in the additive groups with restriction. The lowest feed cost per kg of weight gain was observed in the birds fed diets supplemented with synbiotic, probiotic and prebiotic. Feed restriction improved the feed conversion ratio, economic return, but decreased the feed intake, serum total cholesterol and visible fat in comparison with non-restricted groups. Conclusion: The biological feed additives could be routinely added to broiler diets, especially when a feed restriction program is followed. Finally, it can be recommended to restrict feed, and add probiotic or synbiotic to increase weight, improve feed conversion rate and reduce feed cost of production.

서울 지역 주부들의 건강과 관련된 식생활 의식구조 (Health-related Dietary Attitudes and Behaviours among Mealmanagers in Seoul Area)

  • In this survey, we investigated the way of thinking about meal management of housewives including the sincerity to meal preparation, the eating practices and cronic degenerative diseases related dietary behaviors, and studied the effects of above factors on the real food intakes. We also investigated the trends of health-foods and nutrient supplements usage. The results obtained from 506 housewives in Seoul were summarized as follows. Although our subjects prepared their meals habitually without special concern, they did not prefer the use of convenient foods. The food prefrence of housewivess who had more child and refered higher life status was similar with other family members. The dietary attitudes were good in large family, the higher income and the more child group. The highly educated group skipped breakfast more frequently. The mealmanagers with better education career and good living status considered for the restriction of salty foods, sweet foods, animal fats and pungent foods, and for the nutritionally balanced diet. The high income group showed great concerns about weight gain at meal times. The nutritional qualities expressed by the frequency of food group intakes were high in the better educated and living status groups, and their eating frequencies of animal protein foods and calcium sources were significantly high. Mealmanagers who had no job intaked vegetable oils through frying foods frequently. The use of health-foods and nutrient supplements was influenced by age, educational and economic level and self-estimated living status, but the trends in prevalence of both were not consistent. Health-foods were prefered by the groups of high educational career, affluent income and advanced living status, and low educational career, low income and low living status groups favored the nutrient supplements. The restrictive intake of animal fat and the use of health-food were positively correlated, which seemed that the subjects used health-foods as supplements in compensation for nutritional unbalance caused by the avoidance of animal protein foods.

Maternal Low-protein Diet Alters Ovarian Expression of Folliculogenic and Steroidogenic Genes and Their Regulatory MicroRNAs in Neonatal Piglets

  • Maternal malnutrition during pregnancy may give rise to female offspring with disrupted ovary functions in adult age. Neonatal ovary development predisposes adult ovary function, yet the effect of maternal nutrition on the neonatal ovary has not been described. Therefore, here we show the impact of maternal protein restriction on the expression of folliculogenic and steroidogenic genes, their regulatory microRNAs and promoter DNA methylation in the ovary of neonatal piglets. Sows were fed either standard-protein (SP, 15% crude protein) or low-protein (LP, 7.5% crude protein) diets throughout gestation. Female piglets born to LP sows showed significantly decreased ovary weight relative to body weight (p<0.05) at birth, which was accompanied with an increased serum estradiol level (p<0.05). The LP piglets demonstrated higher ratio of bcl-2 associated X protein/B cell lymphoma/leukemia-2 mRNA (p<0.01), which was associated with up-regulated mRNA expression of bone morphogenic protein 4 (BMP4) (p<0.05) and proliferating cell nuclear antigen (PCNA) (p<0.05). The steroidogenic gene, cytochrome P450 aromatase (CYP19A1) was significantly down-regulated (p<0.05) in LP piglets. The alterations in ovarian gene expression were associated with a significant down-regulation of follicle-stimulating hormone receptor mRNA expression (p<0.05) in LP piglets. Moreover, three microRNAs, including miR-423-5p targeting both CYP19A1 and PCNA, miR-378 targeting CYP19A1 and miR-210 targeting BMP4, were significantly down-regulated (p<0.05) in the ovary of LP piglets. These results suggest that microRNAs are involved in mediating the effect of maternal protein restriction on ovarian function through regulating the expression of folliculogenic and steroidogenic genes in newborn piglets.
