  • Acyl CoA:diacylglycerol acyltransferase (DGAT) is a key enzyme involved in triacylglycerol synthesis. Too much accumulation of triacylglycerol in certain organs and tissues of the body causes high risk conditions of fatty liver. obesity and hypertriglyceridemia. leading to serious diseases of atherosclerosis.   • Protein kinase C (PK-C) represents a central element transducing signals generated by a broad range of pathways which produce the lipid second messenger sn-1,2-diacylglycerol (DAG) directly or indirectly. Many dominant oncogenes have proven to function, at least in part, through this pathway Likewise, this pathway is involved in expression of other aspects of the transformed phenotype, such as tumor invasion or multidrug resistance. As expected from its broad role in cell signaling, PK-C Is also important in a range of other physiological and pathophysiological processes, including inflammation, differentiation, and nerve function.

Synthesis of Mono-,Diacylglycerol from Rice Bran Oil using lipase and Separation by Fractionation

  • Mono-, diacylglycerol were synthesized by glycerolysis of bran oil and glycerol using IM60 (an immobilized lipase) in a stirred batch reactor for 72 hours. After glycerolysis, the composition TAG, DAG, MAG in product was 41.7%, 44.7%, and 11.1%, respectively. Glycerol was separated, then this mixture product was undergone fractionation for winterization followed. The fractionation was performed using hexane and acetone solvent to reduce palmitic acid at the low temperature for overnight individually. Temperature was set -40$^{\circ}C$, -14$^{\circ}C$, -8$^{\circ}C$ and 0$^{\circ}C$, respectively. By considering results from this experiments, fractionation with hexane at -8$^{\circ}C$ was most efficient regarding to yield without crystallization.

  • Diacylglycerol acyltransferase (DGAT) is a microsomal enzyme that plays a central role in the metabolism of cellular glycerolipid. Recently, the generation of DGA T-deficient mice has provided a better understanding of triglyceride synthesis and its relationship to obesity. Therefore DGAT is an attractive target for treatments of triglyceride metabolism disorders, such as obesity or hypertriglyceridemia. (omitted)

  • By using guinea pig lung mast cells, this study aimed to examine the effects of Aloe component(NY945) on the mediator releases caused by mast cell activation, and also aimed to assess the effects of NY945 on the mechanism of mediator releases in the mast cell activation. We partially purified mast cells from guinea pig lung tissues by using the enzyme digestion, the rough and the discontinuous density percoll gradient method. Mast cells were sensitized with $IgG_1$ (anti-OA) and challenged with ovalbumin. Histamine was assayed by fluorometric analyzer, leukotrienes by radioimmunoassay The phospholipase D activity was assessed more directly by the production of labeled phosphatidylethanol or phosphatidylbutanol which was produced by phospholipase D-mediated transphosphatidylation in the presence of ethanol or butanol. The amount of mass 1,2-diacylglycerol was measured by the [$^3H$]1,2-diacylgycerol produced when prelabeled with [$^3H$]myristic acid. In the mast cells prelabeled with L-[$^3H$]methyl methionine the phospholipid methylation was assessed by measuring the incorporation of the [$^3H$]methyl moiety into phospholipids. Pretreatment of NY945(10$\mu$g) significantly decreased histamine and leukotrienes releases during mast cell activation. The decrease of histamine release was stronger than that of leukotrienes during mast cell activation. The phospholipase D activity increased by the mast cell activation was decreased by the dose-dependent manner in the pretreatment of NY945. The amount of mass 1,2-diacylglycerol produced by activation of mast cells were decreased in the pretreatment of NY945. NY945 pretreatment strongly inhibited the incorporation of the [$^3H$]methyl moiety into phospholipids. The data suggest that NY945 purified from Aloe inhibits in part an increase of 1,2-diacylglycerol which is produced by activating mast cells with antigen-antibody complexes which is mediated via phosphatidylcholine-phospholipise D and phosphatidylinositole-phospholipise C systems, and then followed by the inhibition of histamine release. Furthermore, NY945 reduces the phosphatidylcholine production by inhibiting the methyltransfsrase I and II, which decrease the conversion of phosphatidylcholine into arachidonic acid and inhibits the production of leukotrines.

Effect of conjugated linoleic acid in diacylglycerol-rich oil on the lipid metabolism of C57BL/6J mice fed a high-fat high-cholesterol diet

  • The effect of conjugated linoleic acid (CLA) isomers esterified in diacylglycerol (DAG)-rich oil on lipid metabolism was investigated. Since dietary DAG has been known to induce the regression of atherosclerosis, CLA-DAG and olive-DAG oils containing similar levels of DAG (51.4~54.2%) were synthesized from olive oil. Hyperlipidemic C57BL/6J mice were then fed high-fat high-cholesterol diets supplemented with these oils (5% each) for 7 wk. The CLA-DAG diet containing 2.1% CLA isomers (0.78% c9,t11-CLA; 1.18% t10,c12-CLA) remarkably increased the levels of total plasma cholesterol and glutamic oxaloacetic transaminase (GOT) along with hepatic cholesterol and triacylglycerol (TAG) contents. Furthermore, the CLA-DAG diet inhibited fat uptake into adipose tissue whereas fat deposition (especially in the liver) was increased, resulting in the development of fatty livers. Hepatic fatty acid composition in the CLA-DAG mice was different from that of the olive-DAG mice, showing higher ratios of C16:1/C16:0 and C18:1/C18:0 in the liver. The activity of hepatic acyl-CoA:cholesterol acyltransferase (ACAT) was higher in CLA-DAG mice while plasma lecithin:cholesterol acyltransferase (LCAT) activity and the ferric reducing ability of plasma (FRAP) were lower in CLA-DAG mice compared to the olive-DAG animals. Results of the present study suggest that CLA incorporation into DAG oil could induce atherosclerosis in mice.

생쥐 GV난자와 1-세포기 배아의 핵막붕괴에 미치는 Protein Kinase A와 C의 작용 (Action of Protein Kinase A and C Activators on Germinal Vesicle Breakdown and One-Cell Embryos in the Mouse)

  • 난자성숙 재개와 1-세포기 배아의 세포주기에서, cAMP-의존성 protein kinase A와 diacylglycerol-의존성 protein kinase C가 핵막붕괴에 미치는 영향을 조사하였다. 난자성숙 재기는 dbcAMP, IBMX, TPA, 또는 diacyllycerol에 의해 억제되었다. 또한 protein kinase A와 protein kinase C 활성제를 같이 처리하면 난자성숙이 더욱 억제되었다. 그러나 1-세포기 배아의 전핵막붕괴에는 아무런 영향도 미치지 못하였으며, 단지 protein kinase C 활성제만이 세포질 분열을 억제하였다. 이상의 결과로부터, protein kinase A와 protein kinase C에 의한 단백질 인산화 양상이 GV난자의 핵막붕괴와 1-세포기 배아의 전핵막붕괴에 미치는 세포내 작용기작은 상이함을 알 수 있었으며, 전기영동 결과, 81 KD 단백질이 난자성숙 재개에 중요한 역할을 하리라 사료되었다.

유충 Manduca sexta 리포포린에 의한 지방체로의 디아실글리세리드 운반 (The transfer of diacylglycerol from lipophor in to fat body in larval Manduca sexta)

  • 본 논문은 리포포린에 의해 디아실글리세리드(DAG)가 Manduca sexta 유충 지방체로 운반되는 과정을 조사하였다. $[^3H]$-DAG 표지 리포포린($[^3H]$-DAG-Lp)을 시간별로 유충 지방체와 배양하여 지방체로 운반되는 DAG의 방사능을 결정하였다. $[^3H]$-DAG-Lp와 지방체를 배양하면 지방체에 DAG가 축적되며 이의 일부는 지방체에서 트리아실글리세리드(TAG)로 전환되는 것을 알 수 있었다. Suramin과 표지되지 않은 리포포린(unlabeled Lp)의 존재 하에서는 지방체로 운반되는 DAG가 억제되는 데, 이는 DAG를 운반하는 데 리포포린 수용체가 관여한다는 사실을 알 수 있었다. Suramin의 효과는 다소 복잡하지만 리포포린 수용체에 결합하여 막의 특성을 변화시켜 DAG 운반속도에 영향을 주는 것 같다. 지질 운반과정이 수용체-매개 내포작용이라는 사실을 조사하기 위하여 내포작용 억제자인 ammonium chloride와 chloroquine을 처리하였다. 그 결과 리포포린과 지방체사이에서의 지질 운반 기작이 수용체-매개 과정이라는 사실을 보여준다.

The Inhibitory Mechanism of Aloe Glycoprotein (NY945) on the Mediator Release in the Guinea Pig Lung Mast Cell Activated with Antigen-Antibody Complexes

  • It has been reported that the glycoprotein extracted from Aloe has strong anti-inflammatory response. However, there has been no research report yet about the effect of Aloe on allergic hypersensitivity reactivity. Pretreatment of NY945 (10 ${\mu}g$) significantly decreased histamine and leukotrienes releases during mast cell activation. The decrease of histamine release was stronger than that of leukotriene during mast cell activation. The phospholipase D activity increased by the mast cell activation was decreased by the dose-dependent manner in the pretreatment of NY945. The amount of DAG produced by PLC activity was decreased by NY945 pretreatment. The amount of mass 1, 2-diacylglycerol produced by activation of mast cells was decreased in the pretreatment of NY945. NY945 pretreatment strongly inhibited the incorporation of the $[^3H]methyl$ moiety into phospholipids. The data suggest that NY945 purified from Aloe inhibits in part an increase of 1, 2-diacylglycerol which is produced by activating mast cells with antigen-antibody reactions, which is mediated via phosphatidylcholine-phospholipase D and phosphatidylinositol-phospholipase C systems, and then followed by the inhibition of histamine release. Lipozyme TLIM을 효소적 반응촉매로 이용한 glyceryl monooleate로부터의 diacylglycerol의 합성 (Production of Diacylglycerol from Lipase by the Catalyzed Reaction of Soybean Oil and Glyceryl Monooleate)

  • Glycerol과 oleic ethyl ester와의 반응, 그리고 glycerol과 oleic acid와의 반응에서는 DAG가 생성되지 않았었지만 GMO와 SBO를 기질로 사용한 반응에서는 DAG가 생성됨을 확인하였다. 기질의 몰 비율에 따른 DAG 생성율을 비교하기 위하여 효소량을 기질 질량의 20%로 동일하게 설정하고 SBO와 GMO의 몰비율을 각각 2:1, 1:1과 1:2로 하여 반응한 결과, 1 hr 후 DAG의 함량이 최대로 도달 하였고 그 후에는 큰 차이를 보이지 않았다. 1hr에서 가장 큰 DAG 함량을 보인 몰비율은 1:1과 1:2(SBO:GMO)일 때로 각각 20.0, 20.4 g/100 g oil로 나타났고, 이 두 몰비율 중 TAG 함량이 낮았던 1:2 몰비율을 이용하여 DAG를 합성 하였다. 한편, 효소의 사용량에 따른 DAG 생성율을 비교하기 위하여 SBO와 GMO의 몰비율을 1:2로 동일하게 설정하고 효소량을 기질 질량의 2, 5, 10, 20%로 하여 반응한 결과, 1 hr의 DAG 함량은 각각 10.8, 14.0, 16.9, 20.4 g/100 g oil로 나타났다. 1 hr 이후의 DAG 생성량을 보면 효소량을 20% 사용한 경우, 72 hr까지 18.9-22.0 g/100 g oil로 비슷한 함량을 나타내었으나 효소량을 기질 질량의 2, 5, 10%를 사용하였을 경우, 1 hr 이후에도 DAG 함량이 증가하는 경향을 보였다.
