Crown Gall of Chrosanthemum Caused by Agrobacterium rubi and A. tumefaciens (Agrobacterium rubi와 A. tumefaciens에 의한 국화 뿌리혹병)

  • Crown gall on lower stem and root of chrysanthemum(Dendranthema grandiflorum Kitamura) was observed at Hwasung and Gumi in 2001 and 2004, respectively. Tumors were semi-round with rough surface texture of dark brown color. Nine isolates inducing gall formation on lower stem of chrysanthemum among twenty isolates from the tumor tissues were characterized. Their colonies were convex, glistening, circular with an entire edge and whitish or tannish cream in color on potato dextrose agar supplemented with 0.5% $CaCO_3$. The virulent isolates were rod-shaped with peritrichous flagellae, gram negative, aerobic and growing on D1M agar. Among nine virulent isolates, one isolate was identified as Agrobacterium rubi and eight isolates were A. tumefaciens based on biochemical and physiological characteristics, fatty acid profiles and substrate utilization patterns. A. tumefaciens had strong pathogenicity and broad host range compared with A. rubi. This is the first report on crown gall of chrysanthemum in Korea. To our knowledge, crown gall of chrysanthemum caused by A. rubi is first report in this study worldwide.

Occurrence of Blue Mold on Sweet Persimmon(Diospyros kaki) Caused by Penicillium expansum (Penicillium expansum에 의한 감 푸른곰팡이병 발생)

  • A fruit rot of sweet persimmon(Diospyros kaki cv. 'Fuyu') that infected with blue mold was found during the storage and transport in Jinju Gyeongnam Province, Korea. Fruit surfaces that infected with the fungus were formed water soaked lesion at first then gradually colonized with the fungus and formed mycelial mats. From the point of infection, fruits become sunken and mostly ruptured. The pathogenic fungus was isolated from infected fruits and cultured on potato dextrose agar. The colonies of the pathogenic fungi were white at frist then became greyish green on malt extract agar. Conidia were ellipsoidal and $2.6{\sim}3.8{\times}2.4{\sim}3.8{\mu}m$ in size. Phialides were ampulliform, verticilate of 3-7, $8.0{\sim}9.2{\times}2.0{\sim}3.0{\mu}m$ in size. Metulae were verticils of 2-4, smooth, $9.0{\sim}12.6{\times}3.0{\sim}4.6{\mu}m$ in size. Ramuli were groups 1-3, smooth, $11.0{\sim}17.6{\times}2.3{\sim}3.0{\mu}m$ in size. Rami were groups 1-2, $7.5{\sim}32.6{\times}2.6{\sim}4.2{\mu}m$ in size. Stipes were septate, smooth, thin walled, $56{\sim}302{\times}2.8{\sim}4.0{\mu}m$ in size. Penicilli were mostly quaterverticillate. Based on the cultural and mycological characteristics as well as pathogenicity test on host plants, the fungus was identified as Penicillium expansum. This is the first report on the blue mold of sweet persimmon(Diospyros kaki) caused by P. expansum in Korea.

Fusarium Wilt Caused by Fusarium oxysporum on Passionfruit in Korea (Fusarium oxysporum에 의한 패션프루트 시들음병)

  • From 2014 to 2016, Fusarium wilt disease was found on fassionfruit in Iksan and Jeju, Korea. Symptoms included wilting of foliage, drying and withering of leaves, and stunting of the plants. The infected plants eventually died during growth. Colonies on potato dextrose agar were pinkish white, and felted with cottony and aerial mycelia with 35 mm after one week. Macroconidia were falcate to almost straight, thin-walled and usually 2-3 septate. Microconidia were usually formed on monophialides of the hyphae and were hyaline, smooth, oval to ellipsoidal, aseptate or medianly 1-septate, very occasionally 2-septate, slightly constricted at the septa, $3-12{\times}2.5-6{\mu}m$. On the basis of the morphological characteristics and phylogenetic analyses of two molecular markers, internal transcribed spacer rDNA and translation elongation factor $1{\alpha}$, the fungus was identified as Fusarium oxysporum. Pathogenicity of a representative isolate was proved by artificial inoculation, fulfilling Koch's postulates. To our knowledge, this is the first report on the occurrence of F. oxysporum on fassionfruit in Korea.

Effect of Plant-growth-promoting Bacteria Inoculation on the Growth and Yield of Cucumber(Cucumis sativa L.) (식물생육촉진 세균이 오이 생육 및 수량에 미치는 영향)

  • We studied the effect of inoculation of microorganisms known to produce plant growth promoting substances, on the growth and yield of cucumber(Cucumis sativa L.), through a field experiment. The microorganisms used were isolated from the forest soil and consisted of Micrococus sp., Baccilus sustilis, Enterobacter agglomerans, Baccilus megaterium, Pseudomonas putida, Cellulomonas sp. and Staphylococus xyposus. Fotr the multiplication, microorganisms were cultured in liquid media of Pseudomonas P and Sabouraud dextrose. Inoculation of microorganisms was done by spraying the culture media after the culture of them to soil and cucumber plants, three times during the growth of cucumber at the rate of 10l/ha. The inoculation of microorganisms tended to promote the growth of cucumber plant and increase the yield of it. No sign of significant improvement of soil chemical and physical properties were observed after the harvest of crop. The population of bacteria and actinomycetes tended to be higher in the inoculated plots than in not inoculated plots, while opposite was the case in the population of fungi.

Characteristics of an Entomopathogenic Fungus Infecting Corythucha ciliata (Hemiptera: Tingidae) (버즘나무방패벌레 기생성 곰팡이의 특성 구명)

  • Overwintering adults of sycamore lace bug (Corythucha ciliata) infected by an unidentified pathogenic fungus were found on the stems of street trees of sycamore in Cheongju city. The objective of this study was to describe this entomopathogenic fungus infecting overwintering sycamore lace bug adults. This unidentified fungus colonized the insect adult body and formed white colony with subglobose clusters of conidiocarps. The size of conidiocarps was 300 to $400{\mu}m$ and each conidium was 15 to $20{\mu}m$. The conidiospore was globus and 2.5 to $3.0{\mu}m$ in diameter, and the hyphae were 1 to $5{\mu}m$ thick. This fungus was successfully isolated and cultivated on potato dextrose agar medium (PDA). The fungal colony was white and then became light yellow. When conidia from this pure culture were inoculated into the overwintering adults, the fungus formed conidiocarps with the same morphology on the insect body and the lethal rate by the fungus was $88{\pm}16%$. This fungus has over 99% homology with Cordyceps bassiana (imperfect fungal name is Beauveria bassiana) in ITS-5.8s rDNA base sequence. The fungal ecology and the infection process of the fungus into its host need to be clarified before using this fungus as a biological control agent.

Cultural characteristics of Inonotus obliquus isolated from Betula costata at Mt. Jumbong in Korea (점봉산 거제수나무에서 분리한 차가버섯의 배양특성)

  • The sterile form of Inonotus obliquus is used for preparation of the medicine befungin that is effective in the treatment of gastritis, gastric ulcers, and several tumors. The fungus is known to be produced mainly on the stems of Betula platyphylla var. japonica that grows at high altitudinal (above 1,100 m) region in Korea. But, we found the mushroom on the stem of Betula costata at Mt. Jumbong in Korea. We isolated a pure culture of the fungus from the stem of B. costata by use of potato dextrose agar (PDA) medium with streptomycin. We could isolate the fungus from plant's tissue filled with hyphae, but not from other parts. The spore collected from the sclerotia showed $6.0{\sim}10.0{\times}4.5{\sim}6.0{\mu}m$ in diameter, and the hypha was $2.5{\sim}5.0{\mu}m$ in thickness. The colony showed irregular features and scattered yellow color at the center as the culture ages. We could find brownish setae at the yellow region of colony at 20 days of culture, and the size ranged $4{\sim}6{\times}100{\sim}420{\mu}m$. The oatmeal agar (OA) provided best growth for I. obliquus among five media (CDA, CMA, MA, OA and PDA). Optimum temperature ranged $25{\sim}30^{\circ}C$, and optimum pH was relatively alkaline with the range of pH 8.0~9.5.

Effect of addition amino acids on the mycelial growth and the contents of β-glucan and γ-aminobutyric acid (GABA) in Sparassis latifolia (아미노산 첨가가 꽃송이버섯 균사체 성장 및 베타글루칸, GABA 함량 변화에 미치는 영향)

  • Sparassis latifolia (formerly S. crispa) is used in food and nutraceuticals or dietary supplements, as rich in flavor compounds and ${\beta}-glucan$. Some previous studies have reported the effects of mushroom on brain function, including its neuroprotective effect. Thus, for this mushroom to be used as an effective nutraceutical for brain function, it would be desirable for it to contain other compounds such as ${\gamma}-aminobutyric$ acid (GABA) in addition to ${\beta}-glucan$. In this study, the enhancement of growth and GABA production in the mycelium of medicinal and edible mushroom S. latifolia was investigated. Amino acids were added externally as the main source of nutrition, and the effects of amino acids were investigated using liquid medium, specifically amino acid-free potato dextrose broth (PDB). The amino acids added were L-glutamic acid (named PDBG medium) and L-ornithine (named PDBO medium). The growth of mycelia was determined to be $0.9{\pm}0.00g/L$, $2.2{\pm}0.16g/L$, and $1.93{\pm}0.34g/L$ PDBG respectively. The GABA content was $21.3{\pm}0.9mg/100g$ in PDB medium, and it in PDBG 1.4% medium, at $115.4{\pm}30.2mg/100g$. However, the PDBO medium was not effective in increasing the GABA content of mycelia. Amino acids had little effect on the ${\beta}-glucan$ content of mycelia. The ${\beta}-glucan$ content was $39.7{\pm}1.4mg/100mg$, $34.4{\pm}0.2mg/100mg$, and $35.2{\pm}9.2mg/100mg$ in PDB, PDBG 1.8% and PDBO 1.4% media, respectively. Addition of glutamic acid and ornithine positively affected the growth of S. latifolia mycelia, and glutamic acid positively affected GABA production; no degradation of GABA was observed with addition of glutamic acid.

The Effect of Chinemys reevesii Gray Pharmacopuncture for Women with Knee Osteoarthritis (구판(龜板)약침이 여성의 퇴행성 슬관절염에 미치는 영향)

  • Objectives : The aim of this study is to determine whether Chinemys reevesii Gray pharmacopuncture is a clinically effective treatment for women with knee osteoarthritis. Methods : Female patients with knee osteoarthritis were recruited from May $1^{st}$, 2014 to April $15^{th}$, 2015 at the Department of Acupuncture & Moxibustion Medicine of Dong-Eui University Korean Medical Hospital. Patients were randomly assigned to two groups. 4.0 ml of Chinemys reevesii Gray pharmacopuncture was injected at acupuncture points of the experimental group(n = 24) twice a week for three weeks. The control group(n = 24) received 5 % dextrose injections in order to identically stimulate the same acupuncture points of $ST_{35}$, $ST_{34}$, $SP_9$, $SP_{10}$, $GB_{34}$, $LR_8$, $EX-LE_4$, and $EX-LE_2$. The Short-Form McGill Pain Questionnaire(SF-MPQ), the Korean Western Ontario and McMaster Universities Osteoarthritis Index(KWOMAC), and the European Quality of Life-5 Dimensions(EQ-5D) were recorded to evaluate treatment efficacy at first visit, after two weeks and four weeks from the initial visit. Results : 40 patients completed the trial. The experimental group showed more significant improvement than the control group in the scores of SF-MPQ, KWOMAC, and EQ-5D. However, there was no significant difference between groups in a few of the assessment scales. Conclusions : Chinemys reevesii Gray pharmacopuncture can be an effective treatment in control of pain, improvement in function and health-related quality of life for women with knee osteoarthritis.

Selection of Ectomycorrhizal Isolates of Tricholoma matsutake and T. magnivelare for Inoculation on Seedlings of Pinus densiflora In Vitro (소나무 유묘에서 송이 외생균근 형성 균주의 선발)

  • We inoculated hypal suspension of Tricholoma matsutake and T. magnivelare were examined on Pinus densiflora seedlings grown in a granite soil substrate with 1/2 PDMP (12 g/l potato dextrose broth, 1.5 g/l malt extract, and 0.5 g/l peptone) medium. Four months after inoculation, the pine seedlings were examined for infection rate, matsutake aroma, and Hartig-net formation. The roots of pine seedling formed ectomycorrhizal roots in the 9 isolates from 12 isolates of T. matsutake and T. magnivelare. However, the seedlings showed different ectomycorrhizae forming rates among the 9 isolates. While matsutake aroma was confirmed from the ectomycorrhizal seedlings, the pine seedling contaminated by bacteria or fungi did not form matsutake ectomycorrhizae with sickening smell. Thus, the aroma was chosen as a good way for the verification of mycorrhizal infection. At the early stage, the mycorrhizal roots showed unramified and branched types without root hair. They also showed thin mantle layers, Hartig-nets, and turned into black color at later stage. Among the examined strains, that of Yecheon isolated in 1995 showed the best infection rate, which indicated that we need to pay attention to the selection of isolates for better result.

Decolorization of Synthetic Dyes and Ligninolytic Enzymes Production by White Rot Fungi (백색부후균에 의한 합성염료의 탈색과 리그닌분해 효소의 생산)

  • This study has been conducted to screen the decolorization of 4 aromatic synthetic dyes and production of ligninolytic enzymes by 4 white rot fungi such as Bjerkanderia adusta, Cerrena unicolor, Pleurotus pulmonarius and Abortiporus biennis. It was found that B. adusta, C. unicolor, and P. pulmonarius have the ability to efficiently decolorize congo red and moderately decolorized amaranth and orange G in solid and liquid culture media. However, the decolorization rate of 4 synthetic dyes by A. biennis was relatively low. The decolorization of congo red, amaranth, orange G were related to the growth rate of the fungal mycelia in the solid medium. But, the all fungi tested did not efficiently decolorize methylene blue in the liquid culture media. To investigate the production of ligninolytic enzymes in media containing aromatic compounds, fungi were cultured in 1% naphthalene supplemented potato dextrose broth medium. All fungi tested had the capability to produce laccase, lignin peroxidase and manganese peroxidase, and B. adusta was the best ligninolytic enzymes producing white rot fungus among other fungi tested.
