  • The identification of embryo-specific genes would provide insights into early embryonic development. However, the current methods employed to identify the genes that are expressed at a specific developmental stage are labor intensive and suffer from high rates of false positives. Here we employed a new and accurate reverse transcription-polymerase chain reaction (RT-PCR) technology that involves annealing control primers(ACPs) to identify the genes that are specifically expressed in porcine blastocysts compared to 2-cell stage embryos. Optimization of In Vitro Murine Embryo Culture Condition based on Commercial M16 Media

  • In vitro culture of murine embryos is an important step for in vitro production systems including in vitro fertilization and generations of genetically engineered mice. M16 is widely used commercialized culture media for the murine embryos. Compared to other media such as potassium simplex optimization medium, commercial M16 (Sigma) media lacks of amino acid, glutamine and antibiotics. In the present study, we optimized M16 based embryo culture system using commercialized antibiotics-glutamine or amino acids supplements. In vivo derived murine zygote were M16 media were supplemented with commercial Penicillin-Streptomycin-Glutamine solution (PSG; Gibco) or MEM Non-Essential Amino Acids solution (NEAA; Gibco) as experimental design. Addition of PSG did not improved cleavage and blastocyst rates. On the other hand, cleavage rate is not different between control and NEAA treated group, however, blastocyst formation is significantly (P<0.05) improved in NEAA treated group. Developmental competence between PSG and NEAA treated groups were also compared. Between two groups, cleavage rate was similar. However, blastocyst formation rate is significantly improved in NEAA treated group. Taken together, beneficial effect of NEAA on murine embryos development was confirmed. Effect of antibiotics and glutamine addition to M16 media is still not clear in the study.

In Vitro Production of Pig Embryos using Intracytoplasmic Injection of Flow Cytometry Sorted Boar Spermatozoa

  • The ability to preselect the sex of piglets is advantageous in the pig industry. The objective of this study was to examine the feasibility of using intracytoplasmic sperm injection (ICSI) with sorted spermatozoa to produce piglets with a preselected sex. Pig embryos were produced by ICSI of frozen X- and Y-sperm that had been separated by flow cytometry. The developmental competence of the embryos was investigated in vitro and in vivo. The populations of X- and Y-spermatozoa were 52.7% and 47.3%, respectively in our samples. The in vitro development of ICSI embryos was enhanced by longer of in vitro maturation of oocytes ($44{\sim}48\;h$ vs. $40{\sim}43\;h$). Their cleavage ($65{\sim}70%$) and blastocyst formation ($9{\sim}12%$) rates were not significantly different between male and female ICSI embryos, or between sorted and unsorted sperm-derived embryos. One pregnancy was established in a recipient that was transferred with 110 female ICSI embryos, but the pregnancy was terminated on Day 89 of gestation. Our results suggest that the separation X- and Y-spermatozoa by flow cytometric sorting can be a useful tool in combination with ICSI for the production of pig embryos and piglets of preselected sex.

Effect of 0.5 mM Dibutyryl cAMP on Meiotic Maturation during Different Incubation Time and Embryonic Development Following In Vitro Fertilization or Parthenogenetic Activation in Porcine Oocytes

  • Presently, the effect of 0.5 mM dibutyryl cAMP (dbcAMP)-supplemented maturation medium during different incubation time on meiotic arrest (germinal vesicle) and resumption (metaphase II) of porcine oocytes and embryonic development of porcine oocytes following in vitro fertilization (IVF) or parthenogenetic activation (PA) was determined. Porcine cumulus oocyte complexes (COCs) were cultured in 0.5 mM dbcAMP for 17, 22, 27, or 42 h, and an additional 22 h without 0.5 mM dbcAMP. The nuclear status was examined at each time point. Oocytes cultured from 39~49 h displayed more than 80% meiotic resumption. More than 85 % of meiotic arrest was presented at 17~22 h. Oocytes were cultured for 22 h with 0.5 mM dbcAMP and additional 22 h without dbcAMP to assess developmental potential following IVF or PA. There were no significant differences in blastocyst rates among the dbcAMPIVF, IVF, dbcAMP-PA, and PA groups, although cleavage rate of IVF group was significantly higher than those of dbcAMP-PA, and PA groups. In conclusion, 0.5 mM dbcAMP influenced meiotic maturation of porcine oocytes depending on incubation time of oocyte, although embryonic development was not improved in both IVF and PA.

Organ Culture of Ovary Isolated from Juvenile Mice (약령 마우스에서 분리한 난소의 기관배양에 관한 연구)

  • This study was to assess the developmental capacity of oocytes matured in vitro after 20, 15, 10, 5 and 0 days of organ culture when ovaries were isolated from juvenile mice at 0-, 5-, 10-,15- and 20-day old, respectively, and to develop in vitro culture system that observed a view to morphology of ovaries and nucleus maturation of oocytes. The size of ovaries decreased 35.9%, 8.7%, 1.2% and 14.4% after 20, 15, 10, 5 days of organ culture when the ovaries were isolated from 0-, 5-, 10 and 15-day old mice, respectively. After organ culture, the recovery rates, diameters of oocytes and the number of oocytes progressed from GV to MII were increased as increasing age of mice.

The Effect of P almulgunjatang gamibang on the Ovarian Functions in Female Mice (팔물군자탕(湯八物君子) 가미방(加味方)이 자성(雌性)생쥐의 생식능력에 미치는 영향)

  • Purpose: These experiments were undertaken to evaluate the effect of administration of Palmulgunjatang gamibang on ovarian functions in female mice. Methods: We administered the Palmulgunjatang gamibang to 6-week-old female CF-1 mice for 4, 8, or 12 days. After administration of Palmulgunjatang gamibang with different concentration, the female mice were injected PMSG and hCG for ovarian hyperstimulation. The mice were divided into 3 different groups for each experiment. To compare the differences, we set a control group treated with plain water at the same volume by the same way. Results: In case of 4-day, 8-day, 12-day administration of Palmulgunjatang gamibang, the mean number of total ovulated oocytes and the number of morphologically normal oocytes were increased compared with control group. We were also examined the embryonic developmental competence in vitro. In case of 4-day administration of Palmulgunjatang gamibang, the rates of blastocyst formation from 2-cell stages were higher than control group. Conclusion: From our results suggested that the medication of Palmulgunjatang gamibang has beneficial effect on reproductive functions of female mice via promotion of cell proliferation.

Developmental Ability of Enucleated Bovine Oocytes Matured In Vitro Following Fusion with a Single Blastomere of Embryos Matured and Fertilized In Vitro (소 체외수정란의 단일분할구와 제핵미수정란 융합배의 초기발생에 관한 연구)

  • This study was conducted to examine the condition of activation of the nuclear transplant bovine embryos. In vitro fertilized(IVF) and nuclear transplant embryos(NTs) were co-cultured with bovine oviduct epithelial tissue(BOET). NTs were treated with cycloheximide(CHXM) for 0 to 6 h after electrofusion to investigate the activation conditin of recipient ooplast. Then, the infljence of the CHXM treatment timing on the cleavage and development of NTs were investigated in relation to the nuclear transplant time. The cleavage rates of NTs were increased with the increasing time of the CHXM treatment from 0 to 6 h (54.7 to 91.3%, P<0.01). Similar trend was shown in the development into the morula or blastocyst stage, but very limitted. Activation of enucleated oocytes prior to fusion enhanced development of NTs compared with that post fustion. This result suggests that the frequency of activation of NTs can be greatly enhanced by treating with CHXM for 6 h. The result also suggests that if blastomeres of unknown cell cycle stage are used, activation of enucleated oocytes prior to fusion enhances development of NTs.

  • A dolescence can be defined as the period between pubescence and physical maturity, and the passage through adolescence(age of 12-19) forms a critical period. Major developmental tasks of adolescence are construction of an emancipated identity, realistic ambitions, and reasonable ideals, and the further development of sexual and social identities. In early adolescence one struggles to adjust psychologically to the spurt in growth and tensions filled with sexual and aggressive impulses. There are reshuffling of peer groups due to differing rates of maturation, early adolescence crushes, sublimation of sexuality, and the resurgence of oedipal feelings. One reaches so-called period of formal operations in cognitive development. In mid-adolescence one struggles to overcome family attachments and controls, and it is a time marked by revolt and conformity. One has to overcome sexual repression while caught in reactivation of oedipal attachment. Peer groups gains in importance, and the peer group changes into a youth group that carries the youth culture. Late adolescence is a period of achieving an ego identity and capacities for intimacy. One has to prevent premature closure : Identity foreclosure limits both opportunity and the ability to guide one's own life. Gradually adolescent begins to have a loss self-centered and narcissistic orientation to one's sexual and affectional needs, and becomes involved in love relationship.

Analysis of Environmental Effect on Reproductive Trait(Litter Size at Birth and Weaning Rate) in Swine (국내 돼지의 번식 형질(산자수 및 이유율)에 대한 환경효과 분석)

  • This study was conducted to estimate the effect of the breed, sire-breeds, farrowing year, farrowing season and parity on number of born alive (NBA), number of weaning (NW) and survival rates of weaning (SRW) in swine. The data were obtained from 46,704 litters of the Landrace, Yorkshire, Duroc and Cross breed farrowed from 1996 to 2005 at 142 GP are registered in Korean Animal Improvement Association (KAIA). There was highly significant effect of breed, sire breed, farrowing year, farrowing season and parity on NBA, NW, SRW (p<0.01). The result of this study could be available to genetic improvement of reproductive traits as a basic reference in Korean pig industry. To achieve the more effective improvement of reproductive traits, additional research such as genetic parameter evaluation should be performed.

Study of Feritilization and Developmental Rates by Hypoxia condition in Strongylocentrotue nudus (Strongylocentrotue nudus 성게에서 빈산소에 의한 배 발생률 및 수정률에 관한 연구)

  • 본 연구는 빈산소에 의한 둥근 성게 (Strongylocentrotue nudus) 수정란의 수정률 및 발생률에 관한 연구이다. 대조군 (normoxia)과 실험군 (hypoxia) 으로 나누어 수정률과 발생률의 변화를 관찰 하였으며 또한 gonad 세포의 유전자 발현의 차이를 봄으로써 스트레스 관련 유전자와 항산화 관련 유전자의 변화를 확인 할 수 있었다. 결과적으로 수정률에서는 큰 차이를 확인 할 수 없었던데 반해 발생률에 있어서 빈산소의 경우 전혀 발생이 진행되지 않는 것을 확인 할 수 있었으며 또한 빈산소에 노출된 gonad 세포의 경우도 스트레스 또는 항산화 유전자가 많이 발현 되는 것을 관찰 할 수 있었다. 앞의 실험을 토대로 빈산소 환경에서 유전자 발현량의 차이를 더욱더 수행함으로써 빈산소 상태에 따른 죽음의 바다의 증가 얼마나 위험한 것인지 더욱더 관찰 할 수 있을 것이다.
