Drosophila melanogaster (fruits fly) is a representative model system widely used in biological studies because its brain function and basic cellular processes are similar to human beings. The whole head of the fly is often used to obtain the key function in brain-related diseases like degenerative brain diseases; however the biomolecular distribution of the head may be slightly different from that of a brain. Herein, lipid profiles of the head and dissected brain samples of Drosophila were studied using electrospray ionization-mass spectrometry (ESI-MS). According to the sample types, the detection of phospholipid ions was suppressed by triacylglycerol (TAG), or the specific phospholipid signals that are absent in the mass spectrum were measured. The lipid distribution was found to be different in the wild-type and the microRNA-14 deficiency model ($miR-14{\Delta}^1$) with abnormal lipid metabolism. A few phospholipids were also profiled by comparison of the head and the brain in two fly model systems. The mass spectra showed that the phospholipid distributions in the $miR-14{\Delta}^1$ model and the wild-type were different, and principal component analysis revealed a correlation between some phospholipids (phosphatidylethanolamine (PE), phosphatidylinositol (PI), and phosphatidylserine (PS)) in $miR-14{\Delta}^1$. The overall results suggested that brain-related lipids should be profiled using fly samples after dissection for more accurate analysis.
At present, aging-related degenerative muscle diseases are considered a serious problem. However, the drug effect on the treatment and prevention of sarcopenia has not been investigated. The purpose of this study was to evaluate the value of extract of Pu'er tea as a remedy to alleviate the symptoms of sarcopenia. Pu'er tea extracts showed excellent radical scavenging ability. The expression of Myh3 was promoted and myotube formation also was increased by treatment of Pu'er tea extracts. These results suggest that Pu'er tea is a natural substance that promotes myogenesis, and is valuable as a material for pharmaceutical research on the prevention and treatment of various muscle diseases, including sarcopenia. And It is necessary to confirm specific indicator substances of puer tea and further study on this.
The knee arthritis patients are generally performed standing position radiography. However, patients with pain caused by degenerative diseases or injuries in the knee have difficulty maintaining a standing position. Therefore, this study aims to develop a knee joint standing weight bearing projection assist device to solve these problems when patients undergo knee radiography due to various diseases. The design of the knee joint weight-bearing radiography assist device is carried out with 3D design and drawing production to secure basic data, electric support and frame manufacture. A fixed device for maintaining the patient's knee standing weight-bearing projection, an electric digital height device, a digital protractor, and a safety device were designed. The arm support is made of PVC with a cushion to relieve the impact and make it easier to remove debris. The digital electric device can be moved up and down according to the patient's height, and a remote control is attached. The safety device is made to be adjustable in size by attaching metal hooks on both sides of the frame and to shield the scrotum. A digital protractor was attached to the side frame to set the desired knee angle. When a self-made assist device was used to perform a knee joint standing weight bearing projection, it helped maintain arm support and lower extremities position. In addition, the height could be adjusted using an assist device during standing projections, which helped the patient maintain the position.

Background: Pluripotent stem cells (PSCs) including embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs) offer the immense therapeutic potential in stem cell-based therapy of degenerative disorders. However, clinical trials of human ESCs cause heavy ethical concerns. With the derivation of iPSCs established by reprogramming from adult somatic cells through the transgenic expression of transcription factors, this problems would be able to overcome. In the present study, we tried to differentiate porcine iPSCs (piPSCs) into endothelial cells (ECs) for stem cell-based therapy of vascular diseases. Methods: piPSCs (OSKMNL) were induced to differentiation into ECs in four differentiation media (APEL-2, APEL-2 + 50 ng/mL of VEGF, EBM-2, EBM-2 + 50 ng/mL of VEGF) on cultured plates coated with matrigel® (1:40 dilution with DMEM/F-12 medium) for 8 days. Differentiation efficiency of these cells were exanimated using qRT-PCR, Immunocytochemistry, Western blotting and FACS. Results: As results, expressions of pluripotency-associated markers (OCT-3/4, SOX2 and NANOG) were higher observed in all porcine differentiated cells derived from piPSCs (OSKMNL) cultured in four differentiation media than piPSCs as the control, whereas endothelial-associated marker (CD-31) in the differentiated cells was not expressed. Conclusions: It can be seen that piPSCs (OSKMNL) were not suitable to differentiate into ECs in the four differentiation media unlike porcine epiblast stem cells (pEpiSCs). Therefore, it would be required to establish a suitable PSCs for differentiating into ECs for the treatment of cardiovascular diseases.

Purpose: Elderly patients with degenerative diseases undergo treatment for the hip and spine; these patients present with various symptoms. This study focused on patients with residual symptoms, predominantly pain, even after receiving treatment for their spinal lesions. Materials and Methods: Patients who underwent total hip arthroplasty (THA) between 2016 and 2022 at a single tertiary hospital were included in the study. Of the 417 patients who underwent primary THA, a retrospective review of 40 patients with previous lesions of the spine was conducted. Patients were stratified to two cohorts: Patients with symptoms related to the spine (Group A), and those with hip-related symptoms (Group B). Preand postoperative comparisons of groups A and B were performed. Results: Improvements in patients' symptoms were observed in groups A and B after THA. In Group A, the mean preoperative visual analog scale (VAS) score was 5.10±0.876, which showed a postoperative decrease to 2.70±1.767. In Group B, the mean preoperative VAS score was 5.10±1.539, which showed a postoperative decrease to 2.67±1.493. Conclusion: According to the findings, promising results were achieved with THA in treatment of debilitating diseases of the hip for both the prognosis of the disease, as well as the patients' symptoms. In addition, in some cases elderly patients with dual pathologies underwent treatment for spinal lesions without performance of any evaluation related to the hip. Thus, evaluation of a patient's hip must be performed and performance of THA in patients with symptoms even after treatment of spinal lesions is recommended.
In this study, we examined the antioxidant activity of the naturaceutical extract in vitro using total phenolic contents, total flavonoids contents, DPPH radical scavenging activity, reducing power assay, and phenolic acid contents. The total phenolic and total flavonoids contents of naturaceutical extract were 5.46 mg/g, 2.21 mg/g, respectively. The DPPH radical scavenging activity of natraceutical extract varied from 18.77 ($200{\mu}g/mL$) to 3.44 ($1,000{\mu}g/mL$). The reducing power of the natraceutical extract absorbance varied from 1.07 (0.78 mg/mL) to 3.44 (12.5 mg/mL), and reducing power of extract presented a concentration-dependent activity increase. The highest amounts of trans-ferulic acid, p-hydroxybenzoic acid, p-coumaric acid, vanillin, vanillic acid, p-hydroxybenzaldehyde, and trans-cinnamic acid were observed in the naturaceutical extract at the levels of 750.79, 619.75, 531.34, 222.04, 219.28, 107.40, and $89.56{\mu}g/g$, respectively. The results imply that this antioxidant effect of the naturaceuticals extract could be harnessed in the management and prevention of degenerative diseases associated with oxidative stress.
Insulin appears to play a role in brain physiology, and disturbances of cerebral insulin signalling and glucose homeostasis are implicated in brain pathology. The objective of the present study was to investigate the protective effects of insulin under conditions of oxidative stress induced by hydrogen peroxide ($H_2O_2$) in C6 glial cells. Insulin at concentration of $10^{-7}$ M could prevent 12 h $H_2O_2$-induced cell death. The formation of reactive oxygen species (ROS), nitric oxide (NO) and 2-thiobarbituric acid-reactive substances (TBARS) were significantly scavenged by insulin pre-treatment in C6 glial cells after $H_2O_2$-induced oxidative stress. Insulin significantly stimulated the phosphorylation of Akt in the cells and the activation of Akt was maintained in response to insulin under $H_2O_2$ incubation for 12 h. In conclusion, these results provide evidence that insulin acts as a free radical scavenger and stimulating Akt activity. These data suggest that insulin may be effective in degenerative diseases with oxidative stress.

Background: The leg length discrepancy (LLD) have been implicated with degenerative diseases since the early 1970s, which also causes musculoskeletal asymmetry problems. The LLD appears due to factors such as balance ability, response time, and muscle strength between the shorter and longer leg. In this study, the effect of manual therapy to LLD has been studied. Methods: Manual therapy has been applied on shorter leg of 30 patients with LLD. The manual therapy was conducted for 2~3 minutes 3 times a week for 4 weeks. Results: The leg length of shorter leg were measured at initial baseline and after 4 weeks. the average leg length of shorter leg increased from 94.10 cm to 94.33 cm. Conclusion: This paper reports that the manual therapy has positive effects on LLD. It was studied by conducting manual therapy on muscles and joints related to shorter leg. In the future, the muscles and joints of the longer leg should be considered and various age group should be studied to develop the understanding of the effect of manual therapy on LLD.
As the average life span has been lengthened and the rate of senile population has been raised, chronic degenerative diseases incident to aging have been increased rapidly and become a social problem. With this social background, recently, oxygen radicals(OR) have toxic effects on Central Nervous System and Peripheral Nervous System and cause neuropathy such as Parkinson's Disease, Alzheimer Disease. The purpose of this study is to examine the toxic effects caused by Xanthine Oxidase(XO) and the effects of herbal extracts such as Jokihaeatang(JHT) on the treatment of the toxic effects. For this purpose, experiments with the cultured cell from the cerebrums of new born mice were done. The results of these experiments were as follows. 1. X0, an oxygen radical, decreased the survival rate of the cultured cells on NR assay, MTT assay and amount of neurofilaments and increased the amount of lipid peroxidation. 2. JHT have efficacy of increasing the amount of neurofilaments.

An 8-year-old, castrated male Shih-tzu was referred due to relapsing pododermatitis and generalized pruritus. On physical examination, right forepaw showed swelling and serosanguinous exudates from sinus tract on dorsal paw. There were no remarkable findings on complete blood count (CBC), serum chemistry, and radiologic examination. On cytological examination of exudates from sinus tract, phagocyted bacteria and numerous degenerative neutrophils were noted. Results of deep skin scraping and plucking hair examination were unremarkable. Skin biopsy was performed and ruled out other skin diseases. Histopathology showed epidermal hyperplasia and diffuse mononuclear cell inflammation in dermal layer. Moreover, pyogranulomatous inflammation was demonstrated in subcutaneous layer. This case was clinically diagnosed as pododermatitis secondary to atopic dermatitis (AD). Clinical signs of pododermatitis were gradually improved following topical application of tacrolimus 0.1% ointment. This case report describes that tacrolimus ointment can be applicable for the treatment of refractory pododermatitis.
