녹용에 관한 연구 (제3보) Cholesterol 투여가토의 혈청중 Cholesterol에 미치는 녹용의 영향 (Studies on Deer Horn (III) Effect of Deer Horn on Serum Cholesterol Level in Cholesterol-fed Rabbits)

  • Influences of deer horn on serum cholesterol level as well as growth rate of experimental cholesterol-fed rabbit were observed. The cholesterol level rose progressively in all cholesterol-fed rabbit and only after 17th day of experiment, cholesterol-level in animal treated with deer horn significantly increased at much lower rate than control group. Above results suggested that deer horn might decrease the abnormally elevated serum cholesterol.

  • This study was performed to investigate the effect of oral administration of deer horn against the asthma. Deer horn improves body metabolism and strengthens overall health, especially in elderly persons and young children. Additionally, it stimulates sexual function in females and can stimulate wound healing. Asthma was induced to Balblc mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the eosinophil number in the BALF. Concentrations of IL-4, IL-5 in BALF and splenocyte were assessed by ELISA, IgG and IgE from serum were calculated by same method. The number of eosinophil in BALF was not significantly changed in deer horn group compared with control group. Concentration of IL-4 in BALF was significantly decreased in deer horn group compared with control group. Levels of IL-5 from BALF and splenocyte were significantly decreased in deer horn group compared with control group, respectively. Concentrations of IgE and IgG in serum were significantly decreased in deer horn group compared with control group, separately. We found that the effect of deer horn extract in asthma was implicated in reductions of IL-4, IL-5 released from Th2 cell, and decreases of IgG, IgE from plasma cell. These findings suggest that deer horn extract can produce anti-asthmatic effect, which may play a role in allergen-induced asthma therapy.

녹용이 cholesterol 투여가토의 간조긱및 각장기에 미치는 영향 (The effects of deer horn on the liver and other organs of cholesterol adminstered rabbits.)

  • The oral administration of deer horn extract was proved to cause a considerable amelioration to the impairment of hepatic oxidative phosphorylation and to the decrease of ATP content in liver tissue induced by cholesterol ingestion. Deer horn extract was shown to accelerate considerably incorporation of acetate-1-C$^{14}$ into cholesterol in liver tissue and it tends to restore a decrease of GOT activity of liver tissue, caused by cholesterol administration. Histological examination showed that deer horn extract had a remarkable preventive effect against fatty infiltration of organs such as liver, heart, spleen, and adrenal gland of cholesterol given rabbits.

  • Six kinds of free amino acids, leucine, methionine, threonine, lysine, histidine and aspartic acid were found by using partition paper chromatography from water extract of deer horn which is widely used as tonic drug. The results shows that deer horn does not contain any specific free amino acid.

  • Several experiments were carried for the purpose of establishing the basis for the quality evaluation of deer horn. Deer horn originated from China, New Zealand, Soviet and Alaska were used as objectives and some constituents involved in deer horn were assayed and compared with one another. Five kinds of trace elements including Ca and Fe were determined by atomic absorbance spectroscopy. The more the region came down from the top of the deer horn, the more the ratio of Ca to Fe content (Ca/Fe) was increased. The same increasing tendancy was observed in the case of ash content. Gangliosides were isolated from deer horn by alumina column chromatography and identified by TLC using gangliosides Type II obtained from bovine brain as the standard material. The TLC chromatograms were scanned by dual wavelength TLC scanner, then those TLC profiles were compared. Deer horn from China and New Zealand were analogous but those from Soviet and Alaska were each distinguished in their TLC profiles.

  • Two kinds of gangliosides contained in deer horns were determined by integrating the peaks of TLC densitometry. Japanese deer horn originated from China showed the highest gangliosides among tested samples and the upper parts in deer horns showed higher gangliosides than the lower parts. In the case of graded samples, the best grade A showed the highest content and the worst grade E did the lowest content. Sixteen kinds of free amino acids were analyzed by auto amino acid analyzer. The lower region the deer horn was, the more the total content of free amino acids was and several kinds of amino acids were contained quite regularly.
